[EN] In the frame of the restoration of natural populations of Cymodocea nodosa of the Canary Islands, seeds are being collected at natural populations where germination is rather scarce and seasonal after dormancy. We have developed techniques of propagation in vitro of collected seeds, consisting in forced seed germination and seedlings propagation to obtain mature 20-30 cm plantlet, which eventually are being used for restoration. In order to improve the developed methodology, several experiments were conducted to adjust conditions for seed storage under different regimes of temperature without loosing germinative potential, fertilize during propagation with controlled released NPK fertilizers, and increase growth by dipping seedlings in solutions of the most common plant hormones.

[EN] The seagrass Cymodocea nodosa (Ucria) Ascherson is the most abundant seagrass species in the Canary Islands (Spain), where it forms dense submerged, ecologically relevant communities as stable and protected habitats. As with other seagrasses, concern has arisen due to a decline in the number and extension of the communities as the result of adverse activities in coastal areas. Seed germination and planting are assumed as cost-effective method for restoration. In the frame of the restoration of natural populations of Cymodocea nodosa, pilot experiences not tested so far in the Canary Islands have been carried out to developed in vitro techniques to produce viable seedlings and its transference to the natural environment.

[EN] Plant Tissue Culture, also called “micropropagation”, is the propagation of plants from different tissues (or explants) in a shorter time than conventional propagation, making use of the ability that many plant cells have to regenerate a whole plant (totipotency).There are two alternative mechanisms by which an explant can regenerate an entire plant, namely organogenesis and somatic embryogenesis. Since the last decades, the number of higher terrestrial plants species from which these techniques have been successfully applied has continually increased. However, few attempts have been carried out in marine plants. Previous seagrasses authors have focused their studies on i) vegetative propagation of rhizome fragments as explants in Ruppia maritima, Halophila engelmannii, Cymodocea nodosa and Posidonia oceanica; ii) culture of meristems in Heterozostera tasmanica, C. nodosa or P. oceanica; and iii) culture of germinated seeds on aseptic conditions, in Thalassia testudinum, H. ovalis, P. coriacea, P. oceanica, and H. decipiens. All these studies determine the most adequate culture medium for each species (seawater, nutrients, vitamins, carbon sources, etc...), often supplemented with different plant growth regulators and the necessary conditions for the culture maintenance, such as light and temperature. On the other hand, several studies have previously established protocols for cell or protoplast isolation in the species Zostera marina, Z. muelleri, P. oceanica, and C. nodosa, using shoots collected from natural meadows as original vegetal source, but further cell growth was never accomplished. Due to the absence of somatic embryogenesis or organogenetic studies in seagrasses we wonder: IS THE SUCCESSFUL APPLICATION OF TISSUE CULTURE TECHNIQUES POSSIBLE IN SEAGRASSES?

[ES] Cymodocea nodosa (Ucria) Ascherson es una fanerógama marina que crece en fondos arenosos bien iluminados, dando lugar a las praderas submarinas (sebadales), hábitat que tiene una importante función ecológica y repercusiones económicas, por su beneficio para las poblaciones de peces. Las actividades humanas en el litoral amenazan a las comunidades de fanerógamas marinas, lo que ha llevado a la regresión de estos ecosistemas y al detrimento de la calidad de sus aguas. De forma natural, la especie sólo responde mediante la propagación vegetativa a estas alteraciones, ya que, estudios sobre la germinación de las semillas en Cymodocea nodosa muestran que, en relación con el alto número de frutos generalmente encontrados en el sedimento, sólo son detectadas un bajo número de plántulas con un rizoma desarrollado, y la viabilidad natural de estas nuevas plántulas es, además, baja. Ante tal situación natural, el empleo de técnicas de micropropagación puede contribuir a las labores que se realizan para la restauración o reimplantación de sebadales. Habiéndose comprobado que existen limitaciones para la propagación in vitro a partir de fragmentos de la planta (ej. rizoma, se ha pensado en incrementar la capacidad germinativa, venciendo i) la dormancia que presenta la semilla, aclimatando en acuarios y transplantando al mar nuevas plántulas, como primera medida, y ii) la inducción de embriogénesis somática, como segunda medida estratégica que asegure la provisión permanente de nuevas plántulas.

[ES]La charla trata de dar a conocer aspectos de la vegetación marina, desde las diminutas algas microscópicas que forman el fitoplancton hasta las que construyen bosques marinos con talos de metros de longitud. Las algas son importantes e insustituibles en la alimentación humana, pero es que sostinenen el funcionamiento del ecosistema marino, es decir, la existencia de peces y del mismo ser humano. Las sebas son plantas con flor, Cymodocea nodosa, que forman sebadales se analizan y sus beneficios ambientales hacen que sea necesario conocerlas y emplear técnicas de biotecnología para recuperarlas.

[EN] Several weeks of intense endurance training enhances mitochondrial biogenesis in humans. Whether a single bout of exercise alters skeletal muscle mitochondrial DNA (mtDNA) content remains unexplored. Double-stranded mtDNA, estimated by slot-blot hybridization and real time PCR and expressed as mtDNA-to-nuclear DNA ratio (mtDNA/nDNA) was obtained from the vastus lateralis muscle of healthy human subjects to investigate whether skeletal muscle mtDNA changes during fatiguing and nonfatiguing prolonged moderate intensity [2.0-2.5 h; approximately 60% maximal oxygen consumption (Vo(2 max))] and short repeated high-intensity exercise (5-8 min; approximately 110% Vo(2 max)). In control resting and light exercise (2 h; approximately 25% Vo(2 max)) studies, mtDNA/nDNA did not change. Conversely, mtDNA/nDNA declined after prolonged fatiguing exercise (0.863 +/- 0.061 vs. 1.101 +/- 0.067 at baseline; n = 14; P = 0.005), remained lower after 24 h of recovery, and was restored after 1 wk. After nonfatiguing prolonged exercise, mtDNA/nDNA tended to decline (n = 10; P = 0.083) but was reduced after three repeated high-intensity exercise bouts (0.900 +/- 0.049 vs. 1.067 +/- 0.071 at baseline; n = 7; P = 0.013). Our findings indicate that prolonged and short repeated intense exercise can lead to significant reductions in human skeletal muscle mtDNA content, which might function as a signal stimulating mitochondrial biogenesis with exercise training.

[EN] OBJECTIVES: To investigate to what extent bone mass accrual is determined by physical activity and changes in lean, fat, and total body mass during growth. METHODS: Twenty six physically active and 16 age matched control boys were followed up for three years. All subjects were prepubertal at the start of the survey (mean (SEM) age 9.4 (0.3) years). The weekly physical activity of the active boys included compulsory physical education sessions (80-90 minutes a week), three hours a week of extracurricular sports participation, and occasional sports competitions at weekends. The physical activity of the control group was limited to the compulsory physical education curriculum. Bone mineral content (BMC) and areal density (BMD), lean mass, and fat mass were measured by dual energy x ray absorptiometry. RESULTS: The effect of sports participation on femoral bone mass accrual was remarkable. Femoral BMC and BMD increased twice as much in the active group as in the controls over the three year period (p < 0.05). The greatest correlation was found between the increment in femoral bone mass and the increment in lean mass (BMC r = 0.67 and BMD r = 0.69, both p < 0.001). Multiple regression analysis revealed enhancement in lean mass as the best predictor of the increment in femoral bone BMC (R = 0.65) and BMD (R = 0.69). CONCLUSIONS: Long term sports participation during early adolescence results in greater accrual of bone mass. Enhancement of lean mass seems to be the best predictor of this bone mass accumulation. However, for a given muscle mass, a greater level of physical activity is associated with greater bone mass and density in peripubertal boys.

[EN] The aim of this study was to find out whether the efficiency of concentric muscle contraction is impaired by eccentric squatting exercise. The study involved 25 male physical education students in two experiments. In the first experiment 14 subjects undertook cycling exercise at 65% VO(2)max until exhaustion on two occasions. During the experimental condition their cycling was interrupted every 10 min so they could perform eccentric squatting exercise, whereas in the control condition they rested seated on the bike during the interruptions. Eccentric squatting consisted of 10 series of 25 reps with a load equivalent to 150% of the subject's body mass on the shoulders. During the first experiment gross efficiency decreased (mean +/- SE) from 17.1 +/- 0.3 to 16.0 +/- 0.4%, and from 17.2 +/- 0.3 to 16.5 +/- 0.4%, between the 2nd and 9th cycling bouts of the experimental and control conditions, respectively (both p < 0.05). The reduction in cycling efficiency was similar in both conditions (p = 0.10). Blood lactate concentration [La] was higher during the experimental than in the control condition (p < 0.05), but substrate oxidation was similar. MVC was decreased similarly (25-28%) in both conditions. The 11 subjects participating in the second experiment undertook 25 reps of eccentric squatting exercise only, each with a load equivalent to 95% of his maximal voluntary contraction (MVC), repeated every 3 min until exhaustion. One hour after the end of the eccentric squatting exercise series cycling, VO(2) and gross cycling efficiency were comparable to the values observed before the eccentric exercise. Both experimental protocols with eccentric exercise elicited similar muscle soreness 2 days later; however, at this time cycling efficiency was similar to that observed prior to eccentric exercise. The interposition of cycling exercise between the eccentric exercise bouts accelerated the recovery of MVC. We conclude that eccentric exercise does not alter or has only a marginal effect on gross cycling efficiency even in presence of marked muscle soreness.

[EN] The tight relation between arterial oxygen content and maximum oxygen uptake (Vv(o2max)within a given person at sea level is diminished with altitude acclimatization. An explanation often suggested for this mismatch is impairment of the muscle O(2) extraction capacity with chronic hypoxia, and is the focus of the present study. We have studied six lowlanders during maximal exercise at sea level (SL) and with acute (AH) exposure to 4,100 m altitude, and again after 2 (W2) and 8 weeks (W8) of altitude sojourn, where also eight high altitude native (Nat) Aymaras were studied. Fractional arterial muscle O(2) extraction at maximal exercise was 90.0+/-1.0% in the Danish lowlanders at sea level, and remained close to this value in all situations. In contrast to this, fractional arterial O(2) extraction was 83.2+/-2.8% in the high altitude natives, and did not change with the induction of normoxia. The capillary oxygen conductance of the lower extremity, a measure of oxygen diffusing capacity, was decreased in the Danish lowlanders after 8 weeks of acclimatization, but was still higher than the value obtained from the high altitude natives. The values were (in ml min(-1) mmHg(-1)) 55.2+/-3.7 (SL), 48.0+/-1.7 (W2), 37.8+/-0.4 (W8) and 27.7+/-1.5 (Nat). However, when correcting oxygen conductance for the observed reduction in maximal leg blood flow with acclimatization the effect diminished. When calculating a hypothetical leg V(o2max)at altitude using either the leg blood flow or the O(2) conductance values obtained at sea level, the former values were almost completely restored to sea level values. This would suggest that the major determinant V(o2max)for not to increase with acclimatization is the observed reduction in maximal leg blood flow and O(2) conductance.

[EN] During maximal whole body exercise VO2 peak is limited by O2 delivery. In turn, it is though that blood flow at near-maximal exercise must be restrained by the sympathetic nervous system to maintain mean arterial pressure. To determine whether enhancing vasodilation across the leg results in higher O2 delivery and leg VO2 during near-maximal and maximal exercise in humans, seven men performed two maximal incremental exercise tests on the cycle ergometer. In random order, one test was performed with and one without (control exercise) infusion of ATP (8 mg in 1 ml of isotonic saline solution) into the right femoral artery at a rate of 80 microg.kg body mass-1.min-1. During near-maximal exercise (92% of VO2 peak), the infusion of ATP increased leg vascular conductance (+43%, P<0.05), leg blood flow (+20%, 1.7 l/min, P<0.05), and leg O2 delivery (+20%, 0.3 l/min, P<0.05). No effects were observed on leg or systemic VO2. Leg O2 fractional extraction was decreased from 85+/-3 (control) to 78+/-4% (ATP) in the infused leg (P<0.05), while it remained unchanged in the left leg (84+/-2 and 83+/-2%; control and ATP; n=3). ATP infusion at maximal exercise increased leg vascular conductance by 17% (P<0.05), while leg blood flow tended to be elevated by 0.8 l/min (P=0.08). However, neither systemic nor leg peak VO2 values where enhanced due to a reduction of O2 extraction from 84+/-4 to 76+/-4%, in the control and ATP conditions, respectively (P<0.05). In summary, the VO2 of the skeletal muscles of the lower extremities is not enhanced by limb vasodilation at near-maximal or maximal exercise in humans. The fact that ATP infusion resulted in a reduction of O2 extraction across the exercising leg suggests a vasodilating effect of ATP on less-active muscle fibers and other noncontracting tissues and that under normal conditions these regions are under high vasoconstrictor influence to ensure the most efficient flow distribution of the available cardiac output to the most active muscle fibers of the exercising limb.
