En Chile, el gastrópodo Concholepas concholepas conocido popularmente como ‘loco’, es un importante recurso bentónico de las pesquerías artesanales, con el que el Estado introdujo en la segunda mitad de los 90s un régimen de co-manejo denominado Áreas de Manejo y Explotación de Recursos Bentónicos (AMERB). En él se asignan a pescadores artesanales formalmente organizados, derechos exclusivos de acceso a stocks de recursos bentónicos en áreas marino costeras geográficamente delimitadas, para su explotación y manejo. Actualmente, 758 AMERB decretadas se extienden en 38º latitudinales de la línea costera chilena, con un total de más de 124.000Ha asignadas, y más de 31.000 pescadores artesanales participantes; siendo consideradas por la comunidad académica internacional como uno de los más grandes experimentos de comanejo a escala global. A pesar de los importantes avances logrados con las AMERB, el régimen enfrenta todavía retos y amenazas, entre los que la captura Ilegal y/o No Declarada (captura IND), a pesar de ser considerada especialmente grave por los pescadores usuarios del sistema, es omitida por la administración central, que establece Capturas Totales Permisibles (CTPs) suponiendo niveles de captura IND nulos, lo que conlleva a arriesgar la sustentabilidad tanto económica como biológica del sistema, además de distorsionar y devaluar la información proveniente del sector formal. En la presente Tesis, se evalúa el impacto de la captura ilegal (robos) del recurso ‘loco’ sobre la sustentabilidad biológica y económica de un sistema AMERB en la Isla Mocha. Con tal propósito se realizaron, consultas formales a las principales entidades públicas fiscalizadoras, una evaluación socio-económica extractiva con los usuarios de AMERB de la Isla Mocha, además de la recopilación de data secundaria. En vista de la escasa respuesta obtenida con las consultas formales, basamos el estudio principalmente en la evaluación socio-económica extractiva. Ésta consistió en la aplicación de entrevistas semi-estructuradas, dirigidas a caracterizar al sector extractivo ilegal, a reconstruir una serie histórica de capturas ilegales intra-AMERB de ‘loco’ en Isla Mocha, y a sistematizar las percepciones de los pescadores sobre los factores causales y las propuestas de solución en torno al problema. Adicionalmente, se desarrolló un modelo bioeconómico simple para analizar el impacto de diferentes niveles de captura ilegal (robo) de ‘loco’, sobre la sustentabilidad económica y ecológica de una AMERB particular. El modelo supone un único stock cerrado, cuya abundancia mensual, es afectada positivamente por un reclutamiento. denso-dependiente y negativamente por la captura formal y la captura ilegal; y además incorpora aspectos económicos resultantes de calcular los costos asociados al despliegue del esfuerzo pesquero formal, los ingresos brutos asociados a la venta de las capturas formales, y los consiguientes beneficios netos obtenidos. Se evaluaron los impactos en 48 diferentes escenarios de simulación (combinación de 4 niveles de Robo, 3 Reglas de Cosecha, y 4 Precios en playa de ‘loco’), mediante el análisis de 4 variables anuales además de 4 indicadores descriptivos de las dinámicas bio-pesquera y económica del sistema en un lapso de simulación de 12 años, generándose 100 salidas por año, cuyos estadígrafos se compararon mediante ANOVA y Pruebas de Tukey. Los resultados indican que los factores más explicativos de la varianza de las salidas de simulación para los 4 indicadores propuestos, son el ‘Robo’, la ‘Regla-Cosecha’, seguidos de la interacción ‘Robo-Regla-Cosecha’; mientras que las proyecciones de abundancia a enero, reclutamiento, y capturas anuales formal e ilegal, muestran que el reclutamiento y dinámica poblacional del stock simulado de ‘loco’, serían resilientes en todos los escenarios, excepto en aquellos resultantes de combinar el nivel de ‘Robo Descontrolado’ con cualquier nivel de ‘Regla-Cosecha’, donde la pesquería formal colapsa. En un escenario “idóneo” para los usuarios del AMERB (ausencia de robos, una elevada tasa de cosecha formal, y buen precio del recurso), el indicador económico generaría promedios de hasta $8,8 millones/año por embarcación formal participante, sin afectar la sostenibilidad del stock simulado. En cuanto a las percepciones, los usuarios de AMERB de Isla Mocha consideran más grave y más compleja la problemática asociada a la captura ilegal de ‘loco’ intra-AMERB imputada a foráneos con respecto a aquella imputada a isleños; no obstante, identifican causas comunes a ambas, agrupadas en los componentes ‘distorsiones socioeconómicas’, ‘ineficacia del régimen de manejo pesquero’, ‘debilidades internas a la organización’, y ‘productividad pesquera artesanal reducida’. Dentro de las ‘distorsiones socioeconómicas’, el ítem “necesidad económica” es considerado como el principal incentivo para la captura ilegal de ‘loco’. En el componente ‘ineficacia del régimen manejo pesquero’, para el robo de infractores foráneos se considera como principal ítem causal a una “fiscalización ineficaz de la Armada”; mientras que para el robo de infractores isleños se consideran como principal ítem causal a una “fiscalización interna ineficaz”. Los ítems de solución propuestos para afrontar a los infractores foráneos, expresan que las acciones deben provenir más del lado institucional normativo público, identificando principalmente a la Armada como infractor protagónico de acción. Asimismo, las propuestas para afrontar a los infractores isleños, expresan que la mayor expectativa de solución de los usuarios de AMERB está puesta en mejoras de la sanción y vigilancia de sus propias organizaciones, algunas de ellas apoyadas por la Armada.

The latitudinal diversity gradient (LDG) is one of the most striking and consistent biodiversity patterns across taxonomic groups. We investigate the species richness gradient in the buckwheat family, Polygonaceae, which exhibits a reverse LDG and is, thus, decoupled from dominant gradients of energy and environmental stability that increase toward the tropics and confound mechanistic interpretations. We test competing age and evolutionary diversification hypotheses, which may explain the diversification of this plant family over the past 70 million years. Our analyses show that the age hypothesis, which posits that clade richness is positively correlated with the ecological and evolutionary time since clade origin, fails to explain the richness gradient observed in Polygonaceae. However, an evolutionary diversification hypothesis is highly supported, with diversification rates being 3.5 times higher in temperate clades compared to tropical clades. We demonstrate that differences in rates of speciation, migration, and molecular evolution insufficiently explain the observed patterns of differential diversification rates. We suggest that reduced extinction rates in temperate clades may be associated with adaptive responses to selection, through which seed morphology and climatic tolerances potentially act to minimize risk in temporally variable environments. Further study is needed to understand causal pathways among these traits and factors correlated with latitude.

Hepatitis C virus (HCV) infections are the major cause of chronic liver disease, cirrhosis and hepatocellular carcinoma worldwide. Both spontaneous and treatment-induced clearance of HCV depend on genetic variation within the interferon-lambda locus, but until now no clear causal relationship has been established. Here we demonstrate that an amino-acid substitution in the IFNλ4 protein changing a proline at position 70 to a serine (P70S) substantially alters its antiviral activity. Patients harbouring the impaired IFNλ4-S70 variant display lower interferon-stimulated gene (ISG) expression levels, better treatment response rates and better spontaneous clearance rates, compared with patients coding for the fully active IFNλ4-P70 variant. Altogether, these data provide evidence supporting a role for the active IFNλ4 protein as the driver of high hepatic ISG expression as well as the cause of poor HCV clearance.

Congenital hypogonadotropic hypogonadism (CHH) and its anosmia-associated form (Kallmann syndrome [KS]) are genetically heterogeneous. Among the >15 genes implicated in these conditions, mutations in FGF8 and FGFR1 account for ∼12% of cases; notably, KAL1 and HS6ST1 are also involved in FGFR1 signaling and can be mutated in CHH. We therefore hypothesized that mutations in genes encoding a broader range of modulators of the FGFR1 pathway might contribute to the genetics of CHH as causal or modifier mutations. Thus, we aimed to (1) investigate whether CHH individuals harbor mutations in members of the so-called "FGF8 synexpression" group and (2) validate the ability of a bioinformatics algorithm on the basis of protein-protein interactome data (interactome-based affiliation scoring [IBAS]) to identify high-quality candidate genes. On the basis of sequence homology, expression, and structural and functional data, seven genes were selected and sequenced in 386 unrelated CHH individuals and 155 controls. Except for FGF18 and SPRY2, all other genes were found to be mutated in CHH individuals: FGF17 (n = 3 individuals), IL17RD (n = 8), DUSP6 (n = 5), SPRY4 (n = 14), and FLRT3 (n = 3). Independently, IBAS predicted FGF17 and IL17RD as the two top candidates in the entire proteome on the basis of a statistical test of their protein-protein interaction patterns to proteins known to be altered in CHH. Most of the FGF17 and IL17RD mutations altered protein function in vitro. IL17RD mutations were found only in KS individuals and were strongly linked to hearing loss (6/8 individuals). Mutations in genes encoding components of the FGF pathway are associated with complex modes of CHH inheritance and act primarily as contributors to an oligogenic genetic architecture underlying CHH.

The identification of associations between interleukin-28B (IL-28B) variants and the spontaneous clearance of hepatitis C virus (HCV) raises the issues of causality and the net contribution of host genetics to the trait. To estimate more precisely the net effect of IL-28B genetic variation on HCV clearance, we optimized genotyping and compared the host contributions in multiple- and single-source cohorts to control for viral and demographic effects. The analysis included individuals with chronic or spontaneously cleared HCV infections from a multiple-source cohort (n = 389) and a single-source cohort (n = 71). We performed detailed genotyping in the coding region of IL-28B and searched for copy number variations to identify the genetic variant or haplotype carrying the strongest association with viral clearance. This analysis was used to compare the effects of IL-28B variation in the two cohorts. Haplotypes characterized by carriage of the major alleles at IL-28B single-nucleotide polymorphisms (SNPs) were highly overrepresented in individuals with spontaneous clearance versus those with chronic HCV infections (66.1% versus 38.6%, P = 6 × 10(-9) ). The odds ratios for clearance were 2.1 [95% confidence interval (CI) = 1.6-3.0] and 3.9 (95% CI = 1.5-10.2) in the multiple- and single-source cohorts, respectively. Protective haplotypes were in perfect linkage (r(2) = 1.0) with a nonsynonymous coding variant (rs8103142). Copy number variants were not detected. We identified IL-28B haplotypes highly predictive of spontaneous HCV clearance. The high linkage disequilibrium between IL-28B SNPs indicates that association studies need to be complemented by functional experiments to identify single causal variants. The point estimate for the genetic effect was higher in the single-source cohort, which was used to effectively control for viral diversity, sex, and coinfections and, therefore, offered a precise estimate of the net host genetic contribution.

According to most political scientists and commentators, direct democracy seems to weaken political parties. Our empirical analysis in the 26 Swiss cantons shows that this thesis in its general form cannot be maintained. Political parties in cantons with extensive use of referendums and initiatives are not in all respects weaker than parties in cantons with little use of direct democratic means of participation. On the contrary, direct democracy goes together with more professional and formalized party organizations. Use of direct democracy is associated with more fragmented and volatile party systems, and with greater support for small parties, but causal interpretations of these relationships are difficult.

The way colleagues and supervisors acknowledge specific contribution and efforts of individuals is crucial for occupational mental health and well being. It contributes to improve the self image of employees and it gives a sense to the activities performed. We carried out a study about occupational health in police officers with a special emphasis on acknowledgment and reward. A questionnaire was sent to 1000 police officers working for a cantonal administration in Switzerland. In total, 695 participants answered the questionnaire. We used the French version of the Langner's questionnaire on psychiatric symptoms to identify cases characterized by potential mental health problems. Multiple choice items (6 modalities ranging from "not at all" to "tremendously") to measure acknowledgment were used. Answers were later dichotomized (low annoyance- high annoyance). Questions we used are: "Do you feel annoyed due to a lack of support and attention from your supervisors?" "Do you feel annoyed because the authorities (politics, judges, etc.) have a low consideration of your occupation?" "Do you feel annoyed due to a low appreciation by the public?" and "Do you feel annoyed due to a lack of acknowledgment by the hierarchy?". The score for psychiatric symptoms was high for 86 police officers for whom health might be at risk. Acknowledgment aspects associated with a high score for psychiatric symptoms are : high annoyance due to a lack of support and attention from supervisors (odds ratio [OR] 3.2, 95% confidence interval [CI] 2.0 to 5.1), high annoyance because authorities seems to have a low consideration for police officers (OR 2.7, 95% CI 1.7 to 4.3), high annoyance due to a low appreciation by the public (OR 1.8, 95% CI 1.2 to 2.9), and high annoyance due to a lack of acknowledgment by the hierarchy (OR 3.0, 95% CI 1.9 to 4.8). Preserving mental health in occupations characterized by high emotional demand is challenging. The results from our study suggest that appropriate acknowledgment might contribute to the prevention of mental health problems. Further research should address a potential causal relation of acknowledgment on mental health.

Staphylococcal enterotoxin B (SEB) is a bacterial superantigen that binds to major histocompatibility complex class II molecules and selectively interacts with T cells that bear certain T cell receptor (TCR) V beta domains. Administration of SEB in adult mice results in initial proliferation of V beta 8+ T cells followed by a state of unresponsiveness resulting from a combination of clonal deletion and clonal anergy in the SEB-reactive population. At this time, it is unclear what relationship exists between the T cells that have proliferated and those that have been deleted or have become anergic. Here we show that only a fraction of the potentially reactive V beta 8+ T cells proliferate in response to SEB in vivo, and that all the cells that have proliferated eventually undergo apoptosis. Virtually no apoptosis can be detected in the nonproliferating V beta 8+ T cells. These data demonstrate a causal relationship between proliferation and apoptosis in response to SEB in vivo, and they further indicate that T cells bearing the same TCR V beta segment can respond differently to the same superantigen. The implications of this differential responsiveness in terms of activation and tolerance are discussed.

Rapport de synthèse : Introduction: la prévalence de l'insuffisance rénale chronique (IRC) augmente et malgré les traitements de remplacement rénal telle que la transplantation ou la dialyse, la mortalité chez des patients atteints d'une IRC reste très élevée. Les maladies cardiovasculaires sont la cause principale de mortalité chez ces patients, et le risque de décès dú à une complication cardiovasculaire est chez eux accru de 10 à 20 fois par rapport à la population générale. Méme si les facteurs de risque cardiovasculaires «traditionnels », principalement l'hypertension artérielle et le diabète sont très prévalents chez les patients avec IRC, ils sont insuffisants pour expliquer l'excès de mortalité cardiovasculaire. D'autres facteurs de risques « nontraditionnels » comme l'accumulation du diméthylarginine asymétrique (ADMA), un inhibiteur endogène de la synthase d'oxyde d'azote (NO), semblent aussi être importants. Chez les patients avec IRC, des taux élevés d'ADMA sont un puissant facteur prédictif indépendant de la mortalité cardiovasculaire. Il a également été démontré chez des souris que l'ADMA peut étre une cause directe de dysfonction endothéliale. Cette dernière joue un rôle primordial dans le développement de l'athérosclérose, cause principale des complications cardiovasculaires. Le but du présent travail est de tester l'hypothèse qu'une réduction du taux d'ADMA après une séance unique d'hémodialyse améliore la dysfonction endothéliale. Méthodes: la dysfonction endothéliale peut être évaluée dans les microvaisseaux de la péan de façon non invasive par fluxmétrie laser Doppler. La vasodilatation cutanée induite par un échauffement local de 34° à 41 °C (hyperémie thermique) est connue pour être dépendante de la production endothéliale de NO et a été utilisée dans plusieurs études cliniques pour évaluer la dysfonction endothéliale. Nous avons recruté 24 patients traités par hémodialyse chronique et également 24 sujets contrôles du même âge et sexe. Chez les patients dialysés, l'hyperémie thermique est mesuré une fois directement avant une séance d'hémodialyse, et une fois directement après une autre séance, toutes deux distantes de 2 à 7 jours. En même temps, les taux plasmatiques d'ADMA sont mesurés par la méthode de spectrométrie de masse en tandem. Chez les sujets contrôle, l'hyperémie thermique est également mesurée à deux reprises, à un intervalle de 2 à 7 jours comme chez les patients dialysés et les taux d'ADMA sont déterminés qu'une seule fois. Résultats: chez les patients dialysés, les réactions d'hyperémie thermique étaient superposables avant et après dialyse, mais moindre que chez les sujets contrôles. Par contre, les taux d'ADMA étaient plus élevés avant qu'après dialysé. Les taux d'ADMA après dialyse étaient similaires aux taux chez les sujets contrôles. Conclusion: cette étude montre que la vasodilatation dépendante de la production endothéliale de NO dans la microcirculation cutanée n'est pas influencée par les taux plasmatiques d'ADMA chez les patients dialysés. Ces résultats suggèrent que d'autres mécanismes sont responsables de la dysfonction endothéliale chez ces patients. Ceci met en question le concept que l'accumulation d'ADMA est un facteur causal du risque cardiovasculaire élevé et suggère que l'ADMA est juste un marqueur du milieu très athérogénique causé par l'IRC.

BACKGROUND: There is an emerging knowledge base on the effectiveness of strategies to close the knowledge-practice gap. However, less is known about how attributes of an innovation and other contextual and situational factors facilitate and impede an innovation's adoption. The Healthy Heart Kit (HHK) is a risk management and patient education resource for the prevention of cardiovascular disease (CVD) and promotion of cardiovascular health. Although previous studies have demonstrated the HHK's content validity and practical utility, no published study has examined physicians' uptake of the HHK and factors that shape its adoption. OBJECTIVES: Conceptually informed by Rogers' Diffusion of Innovation theory, and Theory of Planned Behaviour, this study had two objectives: (1) to determine if specific attributes of the HHK as well as contextual and situational factors are associated with physicians' intention and actual usage of the HHK kit; and (2), to determine if any contextual and situational factors are associated with individual or environmental barriers that prevent the uptake of the HHK among those physicians who do not plan to use the kit. METHODS: A sample of 153 physicians who responded to an invitation letter sent to all family physicians in the province of Alberta, Canada were recruited for the study. Participating physicians were sent a HHK, and two months later a study questionnaire assessed primary factors on the physicians' clinical practice, attributes of the HHK (relative advantage, compatibility, complexity, trialability, observability), confidence and control using the HHK, barriers to use, and individual attributes. All measures were used in path analysis, employing a causal model based on Rogers' Diffusion of Innovations Theory and Theory of Planned Behaviour. RESULTS: 115 physicians (follow up rate of 75%) completed the questionnaire. Use of the HHK was associated with intention to use the HHK, relative advantage, and years of experience. Relative advantage and the observability of the HHK benefits were also significantly associated with physicians' intention to use the HHK. Physicians working in solo medical practices reported experiencing more individual and environmental barriers to using the HHK. CONCLUSION: The results of this study suggest that future information innovations must demonstrate an advantage over current resources and the research evidence supporting the innovation must be clearly visible. Findings also suggest that the innovation adoption process has a social element, and collegial interactions and discussions may facilitate that process. These results could be valuable for knowledge translation researchers and health promotion developers in future innovation adoption planning.

This paper uses microdata to evaluate the impact on the steady-state unemployment rate of an increase in maximum benefit duration. We evaluate a policy change in Austria that extended maximum benefit duration and use this policy change to estimate the causal impact of benefit duration on labor market flows. We find that the policy change leads to a significant increase in the steady-state unemployment rate and, surprisingly, most of this increase is due to an increase in the inflow into rather than the outflow from unemployment.

Exome sequencing of an individual with congenital cataracts, hypertrophic cardiomyopathy, skeletal myopathy, and lactic acidosis, all typical symptoms of Sengers syndrome, discovered two nonsense mutations in the gene encoding mitochondrial acylglycerol kinase (AGK). Mutation screening of AGK in further individuals with congenital cataracts and cardiomyopathy identified numerous loss-of-function mutations in an additional eight families, confirming the causal nature of AGK deficiency in Sengers syndrome. The loss of AGK led to a decrease of the adenine nucleotide translocator in the inner mitochondrial membrane in muscle, consistent with a role of AGK in driving the assembly of the translocator as a result of its effects on phospholipid metabolism in mitochondria.

The process of eliciting client language toward change (change talk [CT]) is implicated as a causal mechanism in motivational interviewing (MI) and brief motivational interventions (BMI). We investigated the articulation of counselor behaviors and CT during BMI with young men. We coded 149 sessions using the Motivational Interviewing Skill Code and summarized these codes into three counselor categories (MI-consistent [MICO], MI-inconsistent [MIIN], other) and three client categories (CT, counter CT [CCT], follow/neutral [F/N]). We then computed immediate transition frequencies and odds ratios using sequential analysis software. CT was significantly more likely following MICO behaviors, whereas MIIN behaviors only led to CCT and F/N. This strongly supports the use of MI skills to elicit CT during BMI with young men, whose speech also predicted counselor behaviors (particularly CT to MICO and CCT to MIIN). Additional analyses showed that among MICO behaviors, reflective listening may be a particularly powerful technique to elicit CT.

This article designs what it calls a Credit-Risk Balance Sheet (the risk being that of default by customers), a tool which, in principle, can contribute to revealing, controlling and managing the bad debt risk arising from a company¿s commercial credit, whose amount can represent a significant proportion of both its current and total assets.To construct it, we start from the duality observed in any credit transaction of this nature, whose basic identity can be summed up as Credit = Risk. ¿Credit¿ is granted by a company to its customer, and can be ranked by quality (we suggest the credit scoring system) and ¿risk¿ can either be assumed (interiorised) by the company itself or transferred to third parties (exteriorised).What provides the approach that leads to us being able to talk with confidence of a real Credit-Risk Balance Sheet with its methodological robustness is that the dual vision of the credit transaction is not, as we demonstrate, merely a classificatory duality (a double risk-credit classification of reality) but rather a true causal relationship, that is, a risk-credit causal duality.Once said Credit-Risk Balance Sheet (which bears a certain structural similarity with the classic net asset balance sheet) has been built, and its methodological coherence demonstrated, its properties ¿static and dynamic¿ are studied.Analysis of the temporal evolution of the Credit-Risk Balance Sheet and of its applications will be the object of subsequent works.

This article has an immediate predecessor, upon which it is based and with which readers must necessarily be familiar: Towards a Theory of the Credit-Risk Balance Sheet (Vallverdú, Somoza and Moya, 2006). The Balance Sheet is conceptualised on the basis of the duality of a credit-based transaction; it deals with its theoretical foundations, providing evidence of a causal credit-risk duality, that is, a true causal relationship; its characteristics, properties and its static and dynamic characteristics are analyzed. This article, which provides a logical continuation to the previous one, studies the evolution of the structure of the Credit-Risk Balance Sheet as a consequence of a business¿s dynamics in the credit area. Given the Credit-Risk Balance Sheet of a company at any given time, it attempts to estimate, by means of sequential analysis, its structural evolution, showing its usefulness in the management and control of credit and risk. To do this, it bases itself, with the necessary adaptations, on the by-now classic works of Palomba and Cutolo. The establishment of the corresponding transformation matrices allows one to move from an initial balance sheet structure to a final, future one, to understand its credit-risk situation trends, as well as to make possible its monitoring and control, basic elements in providing support for risk management.
