Candida tropicalis biofilms: biomass, metabolic activity and secreted aspartyl proteinase production

According to epidemiological data, Candida tropicalis has been related to urinary tract infections and haematological malignancy. Several virulence factors seem to be responsible for C. tropicalis infections, for example: their ability to adhere and to form biofilms onto different indwelling medical devices; their capacity to adhere, invade and damage host human tissues due to enzymes production such as proteinases. The main aim of this work was to study the behaviour of C. tropicalis biofilms of different ages (24120 h) formed in artificial urine (AU) and their ability to express aspartyl proteinase (SAPT) genes. The reference strain C. tropicalis ATCC 750 and two C. tropicalis isolates from urine were used. Biofilms were evaluated in terms of culturable cells by colony-forming units enumeration; total biofilm biomass was evaluated using the crystal violet staining method; metabolic activity was evaluated by XTT assay; and SAPT gene expression was determined by real-time PCR. All strains of C. tropicalis were able to form biofilms in AU, although with differences between strains. Candida tropicalis biofilms showed a decrease in terms of the number of culturable cells from 48 to 72 h. Generally, SAPT3 was highly expressed. C. tropicalis strains assayed were able to form biofilms in the presence of AU although in a strain- and time-dependent way, and SAPT genes are expressed during C. tropicalis biofilm formation.

OBJECTIVE: To analyze associations between levels of physical activity, cardiorespiratory fitness, dietary content, and risk factors that cause a predisposition towards cardiovascular disease. METHODS: Sixty-two individuals aged between 20 and 45 years were evaluated. Levels of physical activity were established by estimates of energy demand corresponding to everyday activity; indices for cardiorespiratory fitness were obtained from estimates of maximal oxygen consumption; information about dietary content was obtained from dietary records kept on seven consecutive days. To indicate risk factors that cause a predisposition towards cardiovascular disease, use was made of body mass indexes, waist-hip circumference relationships, levels of arterial pressure and of plasma lipid-lipoprotein concentration. To establish associations between the variables studied, multiple regression analysis was used. RESULTS: Physical activity levels and cardiorespiratory fitness levels were inversely correlated with the amount and distribution of body fat and arterial pressure. Taken together, the two variables were responsible for between 16% and 19% of the variation in arterial pressure. Total and saturated fat ingestion was associated with higher serum lipid levels. Both dietary components were responsible for between 49% and 61% of the variation in LDL-cholesterol. CONCLUSION: High ingestion of food rich in total and saturated fat and decreased levels of physical activity and of cardiorespiratory fitness are associated with an increased risk of cardiovascular disease, which supports previous data.

The prevalence of obesity is increasing throughout the workforce. Manual lifting tasks are common and can produce significant muscle loading. This study compared muscular activity between obese and non-obese subjects, using surface Electromyography (EMG), during manual lifting. Six different lifting tasks (with 5, 10 and 15 kg loads in free and constrained styles) were performed by 14 participants with different obesity levels. EMG data normalization was based on the percentage of Maximum Contraction during each Task (MCT). Muscle Activation Times (AT) before each task were also evaluated. The study suggests that obesity can increase MCT and delay muscle AT. These findings reinforce the need to develop further studies focused on obesity as a risk factor for the development of musculoskeletal disorders.

This study examines physical activity patterns among women, from pre-pregnancy to the second trimester of pregnancy, and the relationship between physical activity status based on physical activity guidelines and health-related quality of life (HRQoL) and depression over pregnancy. 56 healthy pregnant women self reported physical activity, HRQoL and depression at 10-15 and 19-24 weeks of pregnancy and physical activity before pregnancy. Whereas vigorous leisure physical activity decreased after conception, moderate leisure physical activity and work related physical activity remained stable over time. The prevalence of recommended physical activity was 39.3% and 12.5% in the 1st and 2nd trimesters of pregnancy respectively, and 14.3% pre-pregnancy. From the 1st to the 2nd pregnancy trimester, most physical HRQoL dimensions scores decreased and only mental component increased, independently of physical activity status. No changes in mean depression scores were observed. These data suggest that physical activity patterns change with pregnancy and that physical and mental components are differentially affected by pregnancy course, independently of physical activity status.

To examine effects of mother's anxiety and depression and associated risk factors during early pregnancy on fetal growth and activity. Repeated measures of mother's anxiety (State-Anxiety Inventory (STAI-S)) and depression (Edinburgh Postnatal Depression Scale (EPDS)) and related socio demographics and substance consumption were obtained at the 1st and 2nd pregnancy trimesters, and fetus' (N = 147) biometric data and behavior was recorded during ultrasound examination at 20-22 weeks of gestation. Higher anxiety symptoms were associated to both lower fetal growth and higher fetal activity. While lower education, primiparity, adolescent motherhood, and tobacco consumption predicted lower fetal growth, coffee intake predicted lower fetal activity. Vulnerability of fetal development to mother's psychological symptoms as well as to other sociodemographic and substance consumption risk factors during early and mid pregnancy is suggested.

OBJECTIVE: To study the effect of propafenone on the contractile function of latissimus dorsi muscle isolated from rats in an organ chamber. METHODS: We studied 20 latissimus dorsi muscles of Wistar rats and divided them into 2 groups: group I (n=10), or control group - we studied the feasibility of muscle contractility; group II (n=10), in which the contralateral muscles were grouped - we analyzed the effect of propafenone on muscle contractility. After building a muscle ring, 8 periods of sequential 2-minute baths were performed, with intervals of preprogrammed electrical stimulation using a pacemaker of 50 stimuli/min. In group II, propafenone, at the concentration of 9.8 µg/mL, was added to the bath in period 2 and withdrawn in period 4. RESULTS: In group I, no significant depression in muscle contraction occurred up to period 5 (p>0.05). In group II, a significant depression occurred in all periods, except between the last 2 periods (p<0.05). Comparing groups I and II only in period 1, which was a standard period for both groups, we found no significant difference (p>0.05). CONCLUSION: Propafenone had a depressing effect on the contractile function of latissimus dorsi muscle isolated from rats and studied in an organ chamber.

OBJECTIVE: Physical exercise helps to prevent cardiovascular disorders. Campaigns promoting exercise have taken many people to the parks of our city. The most appropriate exercise for preventing cardiovascular disorders is the aerobic modality; inappropriate exercise acutely increases cardiovascular risk, especially in individuals at higher risk. Therefore, assessing the cardiovascular risk of these individuals and their physical activities is of practical value. METHODS: In the Parque Fernando Costa, we carried out the project "Exercício e Coração" (Exercise and Heart) involving 226 individuals. Assessment of the cardiovascular risk and of the physical activity practiced by the individuals exercising at that park was performed with a questionnaire and measurement of the following parameters: blood pressure, weight, height, and waist/hip ratio. The individuals were lectured on the benefits provided by exercise and how to correctly exercise. Each participant received a customized exercise prescription. RESULTS: In regard to risk, 43% of the individuals had health problems and 7% of the healthy individuals had symptoms that could be attributed to heart disorders. High blood pressure was observed in a large amount of the population. In regard to the adequacy of the physical activity, the individuals exercised properly. The project was well accepted, because the participants not only appreciated the initiative, but also reported altering their exercise habits after taking part in the project. CONCLUSION: Data obtained in the current study point to the need to be more careful in assessing the health of individuals who exercise at parks, suggesting that city parks should have a sector designated for assessing and guiding physical activity.

As Maehly and Chance point out in their excellent review] the advantages of these methods is "... that they can be used for any kind of biological material, and purification of the enzyme is not required. The assay is independent of small amounts of peroxidase activity. It is fairly simple to perform, it is rapid and it can be adapted to continuous reading of the reaction". A variety of drawbacks are also listed by the same authors, viz, the inactivation of the enzyme under the experimental conditions and the time lag before a constant rate of oxygen evolution is reached. OBJECTIVE: To evaluate the level of satisfaction with body weight and the self-perception of the weight/height ratio and to verify the influence of the frequency of present and past physical activity on these variables. METHODS: Using questionnaires or interviews, we obtained height data, reported and desired weight, self-perception of the weight/height ratio, and the frequency of current physical activity in 844 adults (489 women). Of these, evaluated the frequency of physical activity during high school of 193 individuals,and we measured their height and weight. RESULTS: Less than 2/3 of the individuals had body mass index between 20 and 24.9 kg/m2. A tendency existed to overestimate height by less than 1 cm and to underestimate weight by less than 1kg. Desired weight was less than that reported (p<0.001), and only 20% were satisfied with their current weight. Only 42% of men and 25% of women exercised regularly. No association was found between the frequency of physical activity and the variables height, weight, and body mass index, and the level of satisfaction with current weight. CONCLUSION: Height and weight reported seem to be valid for epidemological studies, and great dissatisfaction with body weight and a distorted self-perception of height/weight ratio exists, especially in women, regardless of the frequency of physical activity.

TEMA I. MECANISMOS NO CLÁSICOS EN EL CONTROL DE LA BIOSÍNTESIS DE HORMONAS TIROIDEAS. El objetivo general de este aspecto del proyecto es el estudio de los mecanismos bioquímicos y moleculares que regulan la hormonogénesis tiroidea en diferentes condiciones funcionales. En proyectos anteriores hemos demostrado que la endotoxina bacteriana lipopolisacárido (LPS) y el oxido nítrico (NO) inducen modificaciones en la biosíntesis de hormonas tiroideas. En base a estos resultados se propone investigar el mecanismo responsable de la estimulación de la captación de ioduro y la expresión de NIS ejercida por LPS y los factores que regulan la producción de NO en la célula tiroidea. Por otra parte se investigarán posibles factores hormonales reguladores de la absorción de ioduro a nivel intestinal y su relación con el eje hipófiso-tiroideo, así como los mecanismos involucrados en la expresión de NIS en enterocitos. Como extensión con aplicación clínica y en base a la experiencia del grupo en el estudio de proteínas que participan en la biosíntesis hormonal tiroidea, se realizará una pesquisa de posibles mutaciones en el transportador de ioduro en pacientes con Hipotiroidismo congénito.TEMA II. ESTUDIO DEL EFECTO DE LAS HORMONAS TIROIDEAS EN LA INICIACIÓN DE LA RESPUESTA INMUNE EN RATÓN. INTERACCIÓN CON GLUCOCORTICOIDES. En nuestro grupo demostramos recientemente un efecto novel de las hormonas tiroideas sobre la maduración/función de células presentadoras de antígenos especializadas: células dendríticas (DC), derivadas de médula ósea de ratón. En este proyecto se propone: 1) profundizar el mecanismo molecular involucrado en el efecto de triiodotironina (T3) sobre DC: rol del receptor de T3-señalamiento intracelular; capacidad de DC tratadas con T3 de estimular la citotoxicidad antígeno-específica; estrategia de vacunación en el tratamiento antitumoral. Por su parte, también previamente demostramos la característica de los glucocorticoides (GC) de interaccionar con el mecanismo de acción de las hormonas tiroideas en la expresión final de los efectos T3-específicos. Considerando el uso terapéutico de los GC en diversos estados clínico-patológicos inmunes, se propone: 2) Estudio del efecto de dexametasona (GC de síntesis) sobre el mecanismo de acción de T3 a nivel de DC.

La autooxidación es la forma de deterioro de los productos grasos más importante después de las alteraciones producidas por microorganismos, lo que representa un tema de gran interés económico para las industrias alimenticia y cosmética, ya que da lugar a la aparición de sabores y olores desagradables lo que hace que estos productos sean inaceptables para el consumidor o que reduzcan su vida útil. Dicho proceso se inicia a partir de la reacción de ácidos grasos con oxígeno y puede ser desencadenado por la exposición del producto graso a la luz medioambiental. En estos casos ocurre un proceso de fotooxidación sensibilizada, con la participación de especies reactivas de oxígeno (ROS). Por esta razón, la preservación de producto graso al efecto de las ROS es un punto de capital importancia. Las industrias intentan evitar la oxidación de los productos grasos mediante diferentes técnicas, como el envasado al vacío o en recipientes opacos, pero también utilizando antioxidantes agregados ex-profeso. En particular, los fenoles son secuestradores no enzimáticos de ROS y radicales libres. Actúan como antioxidantes secundarios o interruptores de la cadena oxidativa de lípidos, desactivando las especies reactivas en sus etapas iniciales y evitando que el proceso oxidativo continúe. Por tal motivo, para el presente Proyecto hemos escogido, como potenciales antioxidantes, dos fenoles estructuralmente relacionados con el hidroxitirosol-un antioxidante natural del aceite de oliva-. A través de reacciones fotosensibilizadas, mediante un estudio cinético, mecanístico, de relaciones estructura-reactividad y de dilucidación de fotoproductos se intentará obtener la información que satisface los objetivos específicos de este Proyecto, a saber: a) la resistencia de dHT frente a la oxidación fotopromovida, y en particular a los procesos fotosensibilizados; b) la propensión de dHT para generar especies oxidantes ya sea por irradiación directa o por interacciones específicas con estados excitados de otras moléculas; c) la influencia del medio sobre la capacidad antioxidante de dHT; d) el establecimiento de relaciones estructura-reactividad en lo referida a la actividad antioxidante de dHT. Se trabajará con distintos tipos de sensibilizadores como generadores de diferentes ROS. Para establecer y dilucidar los aspectos cinéticos y mecanísticos mencionados es necesario obtenerinformación acerca de las constantes cinéticas de los diferentes procesos involucrados. La estrategia de trabajo consistirá en abordar condiciones experimentales tales que inhiban determinadas reacciones competitivas y permitan el desarrollo de otras. Se espera que el conocimiento que se genere a partir de los resultados del presente Proyecto, constituya un importante aporte para el diseño y desarrollo de nuevos antioxidantes liposolubles que posean exacerbadas sus propiedades como fotoprotectores frente a eventuales oxidaciones a las que pueda estar expuesto un producto graso.

El presente proyecto apunta a buscar una posible solución a dos problemáticas de salud que afectan a un importante grupo de la población mundial y son los asociados a la enfermedad de Alzheimer (EA) y a la disfunción eréctil. La EA es una enfermedad neurodegenerativa progresiva caracterizada por la pérdida de memoria a corto plazo, entre otras manifestaciones. En la actualidad la terapia más utilizada son los inhibidores de la acetilcolinesterasa (AChE), ya que acetilcolina es el principal neurotransmisor afectado. No obstante, los agentes terapéuticos utilizados presentan efectos no deseados a nivel gastrointestinal, lo que motiva la búsqueda de nuevos agentes. A partir del estudio de la especie autóctona Huperzia saururus aislamos hasta el presente 10 alcaloides, algunos de los cuales resultaron activos sobre la retención de memoria, investigación realizada mediante ensayos tanto in vitro como in vivo. A partir de los promisorios resultados obtenidos se continuarán profundizando estos estudios y también se buscará el farmacóforo, la parte de la estructura responsable de la actividad. Para ello, se realizarán modificaciones químicas de los grupos funcionales y los productos obtenidos serán igualmente evaluados por su actividad sobre memoria. Los que resulten activos serán investigados en relación a su toxicidad, en la búsqueda de potenciales agentes terapéuticos. Paralelamente se estudiarán las diferentes familias de compuestos presentes en la especie, dado que el uso popular proclama su efecto afrodisíaco. La búsqueda de un afrodisíaco perfecto que incremente el deseo sexual, el placer y el desempeño, ha sido una constante desde los tiempos remotos. En todos los casos, se ha buscado mejorar el rendimiento sexual o combatir la disfunción eréctil (DE). Este término se describe como “la incapacidad constante para alcanzar o mantener una erección que es satisfactoria para el rendimiento sexual”. Para investigar la actividad biológica de H. saururus, los extractos y compuestos aislados serán evaluados por la generación de NO que produzcan, la que será estimada mediante la acumulación de nitrito, metabolito estable del NO, en sobrenadantes de cultivo de células endoteliales (HUVEC). Los que resulten efectivos serán evaluados mediante ensayos in vivo tales como, el de conducta sexual en machos y de comportamiento de monta. Del mismo modo que para los alcaloides, se estudiará la toxicidad de los compuestos que resulten activos a los fines de proseguir la metodología en la investigación preclínica de potenciales medicamentos. The present Project aims to look for a possible solution for two health problems that affect an important group of world population and they are those associated to Alzheimer Disease (AD) and Erectile Dysfunction. AD is a neurodegenerative progressive disease characterized by short term memory loss, among other signs. Nowadays the most used therapy are the acethylcholinesterase inhibitors (AChE), in view that the acethylcholine is the main neurotransmitter affected. Nevertheless, the therapeutic agents used present collateral effects at gastrointestinal level, so it motivates the search for new agents. From the autochthonous species Huperzia saururus we have isolated 10 alkaloids until the present, some of which were active on memory retention, investigation developed in vitro and in vivo. After these promising results, these studies will be continued, as well as the search for the pharmacophore. For this reason, chemical modifications on functional groups will be done and the obtained products will be evaluated on their activity on memory as well. In line with this, the different families of compounds present in H. saururus will be studied in view that the popular use claims its aphrodisiac effect. The search for a perfect aphrodisiac that increase the sexual desire, the pleasure and performance was a constant since the old times. In all the cases it was looked a better sexual performance or fight against the erectile dysfunction (DE). For investigate the biological activity of H. saururus, the extracts and the isolated compounds will be evaluated on the NO generation, which will be estimated by means of nitrite accumulation, stable metabolite of NO, in supernatants of endothelial cell cultures (HUVEC). The compounds that appear effectives will be evaluated by in vivo assays such as sexual male behavior, and mount behavior. As for alkaloids, toxicity of active compounds will be studied to follow the pre-clinic methodology research of potential medicines.

El láser de baja y media energía y la magnetoterapia son utilizados en desórdenes osteomioarticulares por sus efectos analgésico, antiinflamatorio y trófico, entre los más destacados. Sin embargo, son insuficientes las investigaciones sobre su mecanismo de acción y antecedentes científicos que avalen sus efectos. Es por ello, que la determinación de acontecimientos celulares y moleculares que ocurren durante la interacción de estos tipos de energía con el sistema muscular, sería relevante para el conocimiento y optimización de tales terapias en las ciencias biomédicas. En las miopatías inflamatorias idiopáticas, se encuentra afectada la estructura, morfología y bioquímica del tejido muscular. La energía que éste requiere para el normal funcionamiento es generada en la mitocondria. Esta organela también es la responsable de la generación de especies oxidantes provocando estrés oxidativo y el inicio de los procesos de apoptosis. Por lo antes dicho, consideramos que la determinación de los biomarcadores inflamatorios asociados a estrés oxidativo, realizando el análisis histomorfométrico ultraestructural y valorando la actividad de los complejos enzimáticos mitocondriales, permitiría una evaluación de la acción terapéutica del láser y la magnetoterapia en un modelo experimental de miopatía. Para ello se propone evaluar el efecto de la magnetoterapia y del láser de baja energía (He-Ne y As.Ga) en miopatía experimental determinando indicadores inflamatorios asociados a estrés oxidativo, análisis histomorfométrico y valoración de la actividad enzimática mitocondrial. Específicamente: -Determinar indicadores inflamatorios y de estrés oxidativo: Oxido Nítrico, Grupos carbonilos, L-citrulina, Fibrinógeno, Superóxido dismutasa, Glutation peroxidasa y Catalasa por espectrofotometría. -Identificar los cambios anatomopatológicos del músculo esquelético por microscopía óptica (MO): cuantificación del infiltrado inflamatorio; MO de alta resolución (MOAR) y por microscopía electrónica: histomorfometría de la ultraestructura miofibrilar y mitocondrial. -Valorar las actividades enzimáticas de la citrato sintasa y de los complejos: I (NADH-ubiquinona reductasa), II (succinato-ubiquinona-reductasa) III (ubiquinona-citocromo c-reductasa) y IV (citocromo c-oxidasa); en mitocondrias de tejido muscular por espectrofotometría. -Evaluar la actividad apoptótica en las fibras musculares de los diferentes grupos por ténica de T.U.N.E.L. Las mediciones mitocondriales (por ME) y de infiltrado inflamatorio (por MO) se realizarán en un total de 5 fotos de aumentos similares en forma aleatoria por grupo estudiado (n=10). Los cambios estructurales observados se analizarán en el programa Axiovision 4.8, para cuantificar el área total ocupada, número total y grado de alteración de las mitocondrias y el porcentaje de infiltrado inflamatorio determinando el grado de inflamación. Los resultados de los datos cuantitativos se analizarán aplicando ANAVA (test de Fisher para comparaciones múltiples); y para los datos categóricos se utilizará Chi cuadrado (test de Pearson), estableciéndose un nivel de significación de p < 0.05 para todos los casos. Importancia del Proyecto: La salud y el bienestar del hombre son los logros perseguidos por las ciencias de la salud. La obtención de terapias curativas o paliativas con un mínimo de efectos colaterales para el enfermo se incluye en estos logros. Por esto y todo lo anteriormente expuesto es que consideramos de gran importancia poder esclarecer desde las ciencias básicas los efectos celulares y moleculares en modelos experimentales la acción de la terapia con láser y magnetoterapia para una aplicación clínica con base científica en todas las áreas de las Ciencias Médicas. In the idiopathic inflammatory myopathies, is affected the structure, morphology and biochemistry of muscle tissue. The mitochondria is responsible for the generation of oxidizing species leading to oxidative stress and the beginning of the process of apoptosis. As said before, we consider the determination of inflammatory biomarkers related to oxidative stress, by ultrastructural morphometric analysis and assessing the activity of mitochondrial enzyme complexes, permit an evaluation of the therapeutic action of laser and magnetic therapy in an experimental model myopathy. We propose to evaluate the effect of the treatment identifying indicators in experimental inflammatory myopathy associated with oxidative stress, histomorphometric analysis and assessment of mitochondrial enzyme activity. Specifically -determining: Nitric oxide, carbonyl groups, L-citrulline, fibrinogen, superoxide dismutase, glutathione peroxidase and catalase by spectrophotometry. -Identify the pathological changes in skeletal muscle by optical microscopy (OM): quantification of the inflammatory infiltrate, OM high resolution (MOAR) and electron microscopy, histomorphometry of myofibrillar and mitochondrial ultrastructure. -Evaluate the enzymatic activity of citrate synthase and complexes: I, II, III and IV in mitochondria muscle tissue by spectrophotometry. -Evaluate apoptotic activity in muscle fibers by TUNEL technique of Mitochondrial measurements and inflammatory infiltration (by OM) was performed in a total of 5 photos of similar increases in random by the study group (n = 10). The structural changes observed are discussed in the program Axiovision 4.8, to quantify number, degree of alteration of mitochondria and the percentage of inflammatory infiltrate determining the degree of inflammation. The results of the quantitative data were analyzed using ANOVA (Fisher test), and categorical data with Chi-square (Pearson test), establishing a significance level of p <0.05.
