Three methods of extraction of lipopolysaccharide (LPS) were compared-the conventional hot phenol-water method with 40% phenol, a modified form of this method using 10% phenol, and the hot saline method. Good recovery of LPS was achieved by each of the three methods, with the LPS found in the aqueous phase with the two phenol-based procedures. The application of SDS-PAGE to the LPS extracts, followed by silver staining, showed similar banding with all three methods of extraction. When the hot saline extraction LPS fraction from eight strains of Bradyrhizobium spp. and eight strains of Bradyrhizobium japonicum was compared with SDS-PAGE, characteristic profiles were achieved. Serological analysis of eight strains of Bradyrhizobium spp., using antisera prepared against whole cells in agglutination reactions, showed extensive sharing of antigens. When antisera was prepared using outer membrane LPS, extracted by the hot saline method, the amount of cross-reaction was reduced greatly. The results indicated that LPS provide an efficient means of obtaining monospecific antisera to be used for serological identification of strains of Bradyrhizobium spp. and that the hot saline extraction method is recommended for a fast, simple and efficient way to obtain LPS and characterize this bacterium.

Diagnostic insect damage to cocoa tree leaves and fruits were compared in situations with and without the presence of the little fire ants, Wasmannia auropunctata and the closely related and sympatric W. cf rochai. No significant differences in thrips, lepidopteran, or chrysomelid beetle damage to fruits, or to young and old leaves, were associated with these ants. However, significant increases of pseudococcids Planococcus citrii, associated with areas dominated by W. auropunctata, and to a lesser degree with W. cf rochai, were present. W. auropunctata has been reported to be a canopy mosaic dominant in cocoa farms, but the lack of reduced herbivore incidences and its lack of spatial permanence do not support favoring its populations for the management of phytophagous insect control.

A cave bat community from a semiarid region in eastern Brazil was studied during the dry season. A high diversity was observed, with thirteen species recorded. Except for Micronycteris minuta and Lionycteris spurrelli, these bats were also found in other Brazilian karstic areas. The commonest species were the hematophague Desmodus rotundus, the omnivore Carollia perspicillata, and the insectivore Natalus stramineus. The bats formed itinerant colonies, moving frequently inside the cave. L. spurrelli was previously known from Amazonia, usually associated with evergreen forests. The present record extends its range 1600 km to the south. A disjunct distribution explains some morphological differentiation between Amazonia and Olhos d'Agua cave populations. The latter may be a relict of former continuous forest vegetation, which has been fragmented after the climatic optimum. Dispersion through the Cerrado forest gallery is also possible.

We examined the prevalence and the in vitro susceptibility to antifungal drugs of Candida spp isolated from clinical specimens at our university hospital in São Paulo, Brazil. Among 6,417 samples studied, positive cultures, were obtained from 222 (3.5%) most of them (68%) from the pediatric unit and nursery. Candida albicans and Candida parapsilosis were the most frequent species and the susceptibility patterns of a panel of 130 isolates to amphotericin B, ketoconazole and fluconazole. showed that the order of antifungal efficacy was amphotericin B > ketoconazole > fluconazole.

Canine brains infected with rabies virus were submitted to decomposition by being left at room temperature of 25 to 29 degrees C for up to 168 h. At 24 h intervals, brain fragments were analyzed by immunofluorescence (IF) and by the mouse intracerebral inoculation (MI) test to confirm the diagnosis of rabies and to measure the putrefaction effect on the accuracy of the diagnosis. Forty eight h after the beginning of the experiment, the MI test showed signs of impairment with four negative results, while after 72 h, 100% of the results were negative to the MI test and only one result was negative to the IF test, indicating that the threshold period for accurate diagnosis is 24 to 48 h before putrefaction. The authors recommend the shipment of suspected cases of rabies to the laboratory for confirmation, but the use of putrid materials for diagnosis is meaningless because of false-negative results.

Enterobacter spp. are considered important causing agents of infection, specially in hospitalized individuals. The natural resistance of these microorganisms and the great facility to develop resistance to new antimicrobial agents make this genus an important object of study. In this work, 176 strains isolated from various clinical samples were used from hospitalized patients (University Hospital Domingos Leonardo Cerávolo) and from clinic patients (Clinical Laboratory from Unoeste), both situated in Presidente Prudente - SP. E. cloacae (78.9%), E. aerogenes (7.9%) and E. (pantoea) agglomerans (3.9%) were the ones more frequently isolated. Eleven antimicrobial agents were tested by the disk diffusion method and around 90% of the strains presented resistance to the cephalotin, ampicillin and cefaclor. Strains of E. (pantoea) agglomerans presented wide profile of sensibility However one strain of E. cloacae presented resistance to all the antimicrobial agents. The antimicrobial agents with greater inhibitory activity were imipenem and cefepime, for this reason these antimicrobial agents could be the treatment of choice in emergencial therapeutic. This emergencial therapy can be applied with relative security, whereas the data obtained in this study show homogeneity in the profile of sensibility to these antimicrobial agents, independent of the infection site and from the isolated species. The ESBL enzyme could not be detected in no one of the strains by the double diffusion test.

Using molecular markers, this work compares the genetic diversity in Colletotrichum gloeosporioides infecting species of the tropical forage legume Stylosanthes at the center of origin in Brazil and Colombia with that of Australia, China, and India, where Srylosanthes spp. have been introduced for commercial use. There was extensive diversity in the pathogen population from Brazil, Colombia, China, and India. The Australian pathogen population was least diverse probably due to its geographical isolation and effective quarantine. The extensive diversity in China and India means that threats from exotic pathogen races to Stylosanthes pastures can potentially come from countries outside the South American center of origin. In Brazil and India, both with native Stylosanthes populations, a high level of genetic differentiation in the pathogen population was associated with sites where native or naturalized host population was widely distributed. There was limited genetic diversity at germplasm evaluation sites, with a large proportion of isolates having identical haplotypes. This contrasts recent pathogenicity results for 78 of the Brazilian isolates that show hot spots of complex races are more common around research stations where host germplasm are tested, but few are found at sites containing wild host populations. For a pathogen in which the same races arise convergently from different genetic backgrounds, this study highlights the importance of using both virulence and selectively neutral markers to understand pathogen population structure.

The pariparoba (Piper cernuum Vell.), a native plant from the Atlantic Tropical Forest (ATF), has been commonly used in folk medicine. Works seeking the exploitation of P. cernuum natural populations need to be accomplished, seeking compatibility in obtaining income rates and preservation of ATF. The present work aimed to study the reproductive phenology and seed dispersal in P. cernuum natural population in the counties of Sete Barras/SP (Parque Estadual Intervales - PEI). The population of P. cernuum studied in PEI presented continuously bud-inflorescences, green-infrutescences and bud-inflorescence emission, with variation among individuals. Fruit-set average was of 9.1%, varying among individuals. Shading and foliar damage presented negative effect on the fruit-set. Three species of bats were identified as potential dispersors of the P. cernuum seeds: Carollia perspicillata, Anoura geoffroyi and Artibeus sp. The foragement strategy observed was the withdrawal of some seeds per infrutescence per flight. This strategy increases the potential in establishing new plants as well as in increase the potential of gene flow in comparison with the entire infrutescence plant eaten at the night perches. In consequence, the results of reproductive phenology and seed dispersal ecology presented in this work can help to establish management and conservation strategies of P. cernuum natural populations in the ATF.

Territorial behavior in hummingbirds minimizes competition through aggressive interactions, resulting in a dominance hierarchy among species and individuals. Interactions among seven hummingbird species visiting flowering eucalyptus in the Floresta Estadual near Rio Claro, São Paulo, in southeastern Brazil, were studied. Dominance was determined by weight and size with the largest species being the most dominant. Time spent in defense and the number of aggressive interactions were greater than recorded in literature, perhaps due to the relatively greater density of hummingbirds in the study area. Daily activity patterns differed among dominant and subordinate species, but were not correlated with either the quantity of available nectar or with nectar sugar concentration.

This study considers the function and complexity of tasks during foraging of three Acromyrmex species. Foraging was classified as a team task composed of 2 or 3 processes: recruitment, selection, and collection. Each process was subdivided into different subtasks. Points were attributed to subtasks considering their hierarchical level to compare the complexity of foraging among species. Total scores obtained were 19 for A. balzani and 14 for A. crassispinus and A. rugosus, indicating different degrees of social complexity for grass-cutting and leaf-cutting ant species. Acromyrmex balzani, a grass-cutting ant species, shows a behavioral repertoire composed of more variable subtasks during foraging.

Superinfection by Candida can be refractory to conventional periodontal treatments in specific situations, such as in immunocompromised patients. In these cases, the systemic therapy with antifungal drugs could be indicated. The aim of this study was to analyse antifungal susceptibility of Candida spp. strains isolated from chronic periodontitis patients and from control individuals. A total of 39 C. albicans isolates, 9 C. tropicalis, 2 C. glabrata and 5 Candida spp. from control individuals and 30 C. albicans, 3 C. tropicalis and 2 C. glabrata from periodontitis patients were tested. In the control group, 1 isolate of C. glabrata was resistant to ketoconazole and 1 Candida spp. was resistant to amphotericin B, ketoconazole and miconazole. Among the isolates of periodontitis group, 1 (3.33%) C. albicans isolate was resistant to flucytosine and ketoconazole. According to the obtained results, it could be concluded that fluconazole was the most effective drug against the several Candida species studied. There were not expressive differences in the susceptibility of isolates from periodontitis patients or from control individuals.

In Brazil, garden and cut rose bushes are propagated through cuttings or grafting. The rootstocks used in most of the traditional areas of cultivation are not identified, being known just by the vulgar name. There is not any information in the literature on the morphologic characteristics, nor about the productivity of different cultivars on the rootstocks available in Brazil. The objective of this work was to characterize and determine productivity of nine rootstocks (Rosa multiflora 'Paulista'; R. multiflora 'Japones'; R. multiflora 'Iowa'; R. multiflora 'Kopmans'; R. indica 'Mayor'; R. indica x multiflora; R. sp. 'Natual Brier'; R. manetti and R. canina 'Inermis') grafted with 'Versilia' and 'Tineke' scions. Morphologic studies were undertaken to characterize branches of adult plants to determine shape and dimensions of the leaf; number of prickles/15 cm of stem; vigor of the stems; incidence of Diplocarpon rosae (blackspot) and coloration of the leaf. The productivity (number of commercial flowers/plant/month) was evaluated for 6 months (October, 2000 to March, 2001), in the city of Andradas, Minas Gerais State, located at 1251 meters above sea level. Since there was significant difference in just two months for both rootstocks, 'Tineke' and 'Versilia', no rootstocks could be considered inadequate. Nonetheless, in general, the rootstocks that showed a better performance with 'Tineke' were, R. multiflora 'Kopmans' and R. manetti and with 'Versilia', R. multiflora 'Kopmans' and Rosa sp. 'Natual Brier'.

A hundred and ninety-five (195) strains of Enterobacter spp., isolated from diverse clinical specimens - urine, feces, cateter, blood, wound, tracheal aspirate, vaginal fluid - were submitted to the conventional identification by biochemical tests, and were also submitted to the identification by panels NegCombo 20 of the system automated MicroScan - AutoScan- 4 (Dade Behring Inc., West Sacramento, CA, USA). The samples were from patients of the Clinical Laboratory from the School of Pharmacy and Biochemistry of UNOESTE, Presidente Prudente, SP, and from patients hospitalized at the University Hospital Domingos Leonardo Cerávolo, UNOESTE. Of the total of strains tested, 191 (97.9%) presented agreement between the two approaches utilized and 4 strains (2.1%) presented identification disagreement, that is, the genus identified was different in each approach. By this study, the conclusion is that both the approaches utilized for the identification presented advantages and disadvantages related to the cost, facility of execution, quickness, reliability and some other characteristics. Even so, our results showed that conventional methods represent a reliable tool for Enterobacter identification.
