The Australian beef industry places the greatest value in bulls, in comparison to cows, for prime beef production. Male carcasses can be sold for a larger profit due to their increased muscle mass. This project aims to demonstrate the feasibility of producing male animals that can sire male only offspring, through a transgenic approach in mice that could later be translated into livestock production systems. The mouse Sry (Sex determining region on the Y) gene has been shown to provide the initiating molecular signal leading to male sex determination in mammals. Sry has also been shown to cause sex reversal in XX mice transgenic for the gene. In this project Sry will be targeted to a locus not subject to X-inactivation on the X chromosome of XY mice. These mice will be bred to determine how the transgene is passed on, to determine expression of the transgene, and to assess its activity in causing XX sex reversal. The male mice transgenic for the Sry gene on their X chromosome will be produced using tetraploid aggregation, which in a single step produces 100% ES cell derived embryos. The same target locus can later be used to introduce the bovine SRY gene onto the X chromosome of bovidae species and using germ cell transplantation produce sex reversed animals. This would bypass the need for expensive chimera crosses and provide farmers with a stud bull capable of producing only sons.

A presente pesquisa tem como propósito maior identificar as consequências sócioreligiosas que se dão no encontro entre a prédica protestante de missão e os principais marcos de referência da identidade brasileira. Para tanto, num primeiro momento, analisa os movimentos religiosos que fomentaram a matriz teológica do protestantismo de missão responsável pela caracterização de sua prédica por meio de revisão bibliográfica e estudo dos sermões de alguns dos primeiros missionários americanos e pregadores nacionais. Num segundo momento, por meio de pesquisa de campo e análise de sermões contemporâneos, a pesquisa identifica as variações da matriz teológica do protestantismo de missão e os reflexos consequentes em sua prédica atual. Ao definir, num terceiro momento, os principais aspectos da identidade nacional, confronta-os com as principais características da prédica protestante de missão brasileira, identificando algumas das consequências sócio-religiosas provenientes dos encontros e desencontros que se dão entre os dois fenômenos. Por fim, baseado nos resultados da pesquisa e no que se tem escrito sobre o tema, aponta-se alguns caminhos para uma ciência homilética que considera a identidade brasileira como elemento basilar de sua reflexão e práxis.

A Constituição de 1988 e leis subsequentes determinam que o Estado preste informações aos cidadãos e favoreça a sua participação nas questões públicas trata-se do princípio legal da Transparência Administrativa, que compreende os seguintes subprincípios: (1) Informação; (2) Motivação e, o mais importante, (3) Participação e interatividade cidadãs. O alto investimento na Comunicação Estatal e os avanços tecnológicos, por si sós, não garantem a prática da transparência pública ou da democratização da informação. Sob uma perspectiva multidisciplinar, esta pesquisa discutiu o princípio legal de Transparência Administrativa, comparativamente à Teoria da Comunicação, com o objetivo de propor um conceito de Comunicação Estatal que, de fato, corresponda aos ideais e à ética necessários à Comunicação Pública. Para o desenvolvimento deste estudo foi investigada a relação da comunicação com o grau de transparência alcançado no portal do Senado Federal. O estudo analisou a tramitação da reforma do Poder Judiciário no período de 2000 a 2004, tendo em vista os três subprincípios legais da Transparência Pública. A análise contemplou, no portal do Senado, o trabalho jornalístico e a disponibilização on-line de textos digitais referentes a documentos originais, tais como atas públicas e notas taquigráficas. A metodologia, de enfoques quantitativo e qualitativo, teve como instrumento principal a Nova Retórica, para análise de matérias jornalísticas e textos documentais. Para averiguação da interatividade conceito que fundamenta o ideal de justiça , foram estabelecidos critérios analíticos a partir da intersecção entre os conceitos de transparência e E-parliaments. Constatou-se que o portal do Senado, no referente à reforma da Justiça, alcançou graus de transparência, atendendo mais aos subprincípios da informação e da motivação em detrimento aos da participação e interatividade cidadãs.(AU)

This thesis deals with the problematic of the business systems systemic purpose definition. The definition of the systemic purpose, which is regarded as the utmost expression of the system's purposefulness, is to be achieved by ensuring the participation of all the stakeholders, if possible, who affect or they are affected by the business system's operations. The nature of participation, defined as a process of the stakeholders' perceptual exchanges, is deemed to be problematic in itself due to the influence exerted upon it by organisational power, coercion and false consciousness. The main focus of the thesis then is to make aware and provide the stakeholders with an explicit philosophical pedestal and a set of principles upon which a meta- epistemological framework for the enquiry of the business system's purposeful behaviour is developed. In addition, the thesis focuses on the development of a methodology that can be used by the stakeholders to achieve self-knowledge through the critical and systemic examination of their normative presuppositions, about the business system, at both sociological as well as the psychological levels concurrently and the subsequent development of an organisational intrinsically motivated information system. According to the critical systems philosophy and principles, developed in this thesis, normative presuppositions define the stakeholders' perceptions about the purposeful behaviour of the business system they perceived as having a material, an informational and/or an emacipatory stake (human interest) in. The methodology will provide Information Systems that demonstrably improve coordination of organisational activities by enabling the development and maintenance of a single/multifaceted view of purpose throughout organisations.

This article considers the role of accounting in organisational decision making. It challenges the rational nature of decisions made in organisations through the use of accounting models and the problems of predicting the future through the use of such models. The use of accounting in this manner is evaluated from an epochal postmodern stance. Issues raised by chaos theory and the uncertainty principle are used to demonstrate problems with the predictive ability of accounting models. The authors argue that any consideration of the predictive value of accounting needs to change to incorporate a recognition of the turbulent external environment, if it is to be of use for organisational decision making. Thus it is argued that the role of accounting as a mechanism for knowledge creation regarding the future is fundamentally flawed. We take this as a starting-point to argue for the real purpose of the use of the predictive techniques of accounting, using its ritualistic role in the context of myth creation to argue for the cultural benefits of the use of such flawed techniques.

In 2004, the Royal Pharmaceutical Society of Great Britain (RPSGB) funded research on teaching, learning and assessment within the UK undergraduate pharmacy degree (MPharm), including the compulsory final year project. Documentary analysis showed that all schools met the project requirement, although there were wide variations in the relative contribution of the project to the final year mark and the degree classification. Interviews with staff revealed that organisation of research projects was complex and time consuming and exacerbated by increasing student numbers and the impact of research ethics. 61% of students, surveyed via a self-completion questionnaire (response rate 50.6%) perceived the research project to be very or fairly important. Whilst 47% considered that they had enough choice of topic and 37% said that their training in research methods provided a good foundation for their project, this suggests scope for improvement. In the UK, there are legislative changes impending which may provide an opportunity to review the future purpose and feasibility of a "significant" final year project within the MPharm.

The success of mainstream computing is largely due to the widespread availability of general-purpose architectures and of generic approaches that can be used to solve real-world problems cost-effectively and across a broad range of application domains. In this chapter, we propose that a similar generic framework is used to make the development of autonomic solutions cost effective, and to establish autonomic computing as a major approach to managing the complexity of today’s large-scale systems and systems of systems. To demonstrate the feasibility of general-purpose autonomic computing, we introduce a generic autonomic computing framework comprising a policy-based autonomic architecture and a novel four-step method for the effective development of self-managing systems. A prototype implementation of the reconfigurable policy engine at the core of our architecture is then used to develop autonomic solutions for case studies from several application domains. Looking into the future, we describe a methodology for the engineering of self-managing systems that extends and generalises our autonomic computing framework further.

The initial aim of this research was to investigate the application of expert Systems, or Knowledge Base Systems technology to the automated synthesis of Hazard and Operability Studies. Due to the generic nature of Fault Analysis problems and the way in which Knowledge Base Systems work, this goal has evolved into a consideration of automated support for Fault Analysis in general, covering HAZOP, Fault Tree Analysis, FMEA and Fault Diagnosis in the Process Industries. This thesis described a proposed architecture for such an Expert System. The purpose of the System is to produce a descriptive model of faults and fault propagation from a description of the physical structure of the plant. From these descriptive models, the desired Fault Analysis may be produced. The way in which this is done reflects the complexity of the problem which, in principle, encompasses the whole of the discipline of Process Engineering. An attempt is made to incorporate the perceived method that an expert uses to solve the problem; keywords, heuristics and guidelines from techniques such as HAZOP and Fault Tree Synthesis are used. In a truly Expert System, the performance of the system is strongly dependent on the high quality of the knowledge that is incorporated. This expert knowledge takes the form of heuristics or rules of thumb which are used in problem solving. This research has shown that, for the application of fault analysis heuristics, it is necessary to have a representation of the details of fault propagation within a process. This helps to ensure the robustness of the system - a gradual rather than abrupt degradation at the boundaries of the domain knowledge.
