Objectives: The incretin-based therapy might be effective in patients possessing certain levels of preserved pancreatic beta-cells. However, doubts still exist regarding the efficacy of this atment in the recovery of tissues damaged by type 1 diabetes. Thus, the objective of this study was to evaluate the treatment with MK0431 in salivary glands of spontaneously diabetic mice, focusing mainly on the possible therapeutic and hypoglycaemic effects of this dipeptidyl peptidase IV inhibitor in the recovery of these salivary tissues. Methods and results: Twenty mice were divided into two groups of 10 animals each: group I (NOD diabetic/untreated) and group II (NOD diabetic MK0431/treated). The group II was treated during 4 weeks with MK0431 mixed in the food. The group I was maintained in the same way without receiving, however, any treatment. Glucose levels were monitored during treatment and salivary glands samples were collected at the end of treatment for the histological examination under both transmitted and polarized light microscopy. High glucose levels were observed in untreated animals, while in animals with treatment, reduction of these levels was observed. Tissue restructuring was also observed in animals submitted to therapy with MK0431, mainly in relation to the attempt to extracellular matrix reorganization. Conclusions: According to results, the treatment with this dipeptidyl peptidase IV inhibitor contributed to the general homeostasis of the organism and to the reestablishment of both epithelial and stromal compartments which were damaged by the hyperglycaemic condition, demonstrating that the incretin-based therapy may be an important complementary treatment for the type 1 diabetic condition. © 2012 Elsevier Ltd.

The cysteine proteinase inhibitor cystatin C inhibited RANKL-stimulated osteoclast formation in mouse bone marrow macrophage cultures, an effect associated with decreased mRNA expression of Acp5, Calcr, Ctsk, Mmp9, Itgb3, and Atp6i, without effect on proliferation or apoptosis. The effects were concentration dependent with half-maximal inhibition at 0.3 μM. Cystatin C also inhibited osteoclast formation when RANKL-stimulated osteoclasts were cultured on bone, leading to decreased formation of resorption pits. RANKL-stimulated cells retained characteristics of phagocytotic macrophages when cotreated with cystatin C. Three other cysteine proteinase inhibitors, cystatin D, Z-RLVG-CHN2 (IC50 0.1 μM), and E-64 (IC 50 3 μM), also inhibited osteoclast formation in RANKL-stimulated macrophages. In addition, cystatin C, Z-RLVG-CHN2, and E-64 inhibited osteoclastic differentiation of RANKL-stimulated CD14+ human monocytes. The effect by cystatin C on differentiation of bone marrow macrophages was exerted at an early stage after RANKL stimulation and was associated with early (4 h) inhibition of c-Fos expression and decreased protein and nuclear translocation of c-Fos. Subsequently, p52, p65, IκBα, and Nfatc1 mRNA were decreased. Cystatin C was internalized in osteoclast progenitors, a process requiring RANKL stimulation. These data show that cystatin C inhibits osteoclast differentiation and formation by interfering intracellularly with signaling pathways downstream RANK. © FASEB.

Despite the improvements in neoadjuvant chemotherapy, the outcome of patients with advanced bladder cancer has changed very little over the past 30 years. In the present study we tested and compared the in vitro antitumor activities of four different inhibitors of Polo-like kinase 1 (PLK1) (BI 2536, BI 6727, GW843682X, and GSK461364), against 3 bladder carcinoma cell lines RT4, 5637 and T24. The impact on radiosensitivity and drug interactions in simultaneous treatments with cisplatin, methotrexate, and doxorubicin were also investigated. Our results showed that PLK1 inhibition prevented cell proliferation and clonogenicity, causing significant inhibition of invasion of tumor cells, though modest differences were observed between drugs. Moreover, all PLK1 inhibitors induced G2/M arrest, with the subsequent induction of death in all 3 cell lines. Drug interactions studies showed auspicious results for all PLK1 inhibitors when combined with the commonly used cisplatin and methotrexate, though combinations with doxorubicin showed mostly antagonistic effects. Comparably, the four PLK1 inhibitors efficiently sensitized cells to ionizing radiation. Our findings demonstrate that irrespective of the inhibitor used, the pharmacological inhibition of PLK1 constrains bladder cancer growth and dissemination, providing new opportunities for future therapeutic intervention. However, further laboratorial and preclinical tests are still needed to corroborate the usefulness of using them in combination with other commonly used chemotherapeutic drugs. © 2013 Landes Bioscience.

A resistência às drogas antirretrovirais é o inevitável resultado da incompleta supressão da replicação do HIV-1. No presente estudo foi caracterizado o perfil de resistência genética aos antirretrovirais em amostras sorológicas provenientes dos estados do Amazonas e Pará, Região Norte do Brasil, no período de 2002 a 2006. Um total de 127 amostras de plasmas obtidas de pacientes HIV positivos e/ou Aids foram submetidas ao teste de resistência pelo ViroSeqTM Genotyping System (Celera Diagnostic-Abbott, USA). Considerando a informação genética derivada das regiões da protease e/ou transcriptase reversa do HIV-1, o subtipo B foi observado em 85% dos casos; seguido por ambos subtipo F1 e forma recombinante BF1 (4,6%) e CF1 (0,8%). A mutação M184V (81,1%) foi a mais comumente observada associada aos NRTI, em indivíduos positivos com TARV no estado do Pará, e a mutação T215F/Y (56,3%) em indivíduos do estado do Amazonas. A mutação K103N foi a mais prevalente (em torno de 33,5%) para os NNRTI em ambos os estados. O perfil de mutação de resistência associado ao gene da protease mostrou a mutação minor L63P como a mais frequente em ambos os estados. O estudo revelou a importância da identificação de mutações associadas com resistência às drogas antirretrovirais para o uso em futuros esquemas terapêuticos. Os resultados deste estudo foram similares aos outros realizados em várias regiões do Brasil.

A transmissão vertical é a principal fonte de infecção pelo HIV em crianças, e pode ocorrer antes, durante e depois do nascimento, dessa forma sendo um grande problema de saúde pública mundial. O presente trabalho teve como objetivo descrever a epidemiologia molecular e o perfil de susceptibilidade/resistência das cepas de HIV-1 identificadas em mulheres nos estados do Acre e do Tocantins. Coletou-se amostra de sangue de 36 mulheres, sendo 9 grávidas e 16 mães de Palmas (Tocantins) e 1 grávida e 10 mães do Rio Branco (Acre) entre abril de 2007 a outubro de 2008. Realizou-se a técnica molecular Reação em Cadeia mediada pela Polimerase (PCR) Nested, para a amplificação de duas regiões genômicas (pro e tr) do DNA proviral, seguido de sequenciamento nucleotídico e análise filogenética. No Acre, tanto em relação ao segmento da protease quanto da transcriptase reversa, todas as amostras foram do subtipo B. Em Tocantins, quanto ao segmento da protease, todas as amostras pertenceram ao subtipo B, já em relação ao segmento da transcriptase reversa 87,5% foram do subtipo B e 12,5% pertencente ao subtipo F. No estado do Acre, todas as cepas analisadas foram suscetíveis aos inibidores de protease (IP) e apenas uma grávida de Tocantins (4,7%) apresentou cepa com resistência aos IP utilizados atualmente. Além disso, verificou-se uma baixa prevalência de cepas com mutações de resistência aos inibidores de transcriptase reversa nucleosídicos (ITRN) e não nucleosídicos (ITRNN), sendo que a resistência as três classes desses ARV foi observada em apenas uma amostra proveniente do estado do Tocantins. Dessa forma, a maioria das cepas de HIV isoladas mostrou susceptibilidade aos ARV utilizados, indicando que há baixa circulação de cepas do HIV resistentes a estes medicamentos nesses estados.

As cisteína-proteases estão entre os alvos mais promissores para o desenvolvimento de novos agentes terapêuticos, visto que participam de eventos fundamentais do ciclo de vida de muitos microorganismos, inclusive Giardia. Como a atividade das proteases pode ser controlada por inibidores específicos, essas substâncias têm sido avaliadas quanto ao potencial antiparasitário. Diante disso, o presente estudo teve por objetivo avaliar o efeito in vitro do inibidor de cisteína-proteases E-64 sobre o crescimento, a aderência e a viabilidade de trofozoítos de cepa de Giardia isolada em Botucatu. Nos ensaios de crescimento e aderência, o número de trofozoítos foi estimado microscopicamente em hemocitômetro, enquanto que a viabilidade celular foi avaliada pelo método do MTT. No presente estudo, embora o metronidazol tenha se apresentado bastante efetivo, o E-64 mostrou ser capaz de inibir o crescimento, a aderência e a viabilidade em taxas superiores a 50%, especialmente nos cultivos expostos à concentração de 100 µM. A despeito de preliminares, esses resultados demonstram que o inibidor E-64 pode interferir em processos primordiais para a sobrevivência do parasita, além do que, abrem novas perspectivas para investigações futuras a fim de se avaliar o real potencial giardicida dos inibidores de proteases.

Chikungunya virus (CHIKV) is a mosquito-borne arthrogenic alphavirus that causes acute febrile illness in humans accompanied by joint pains and in many cases, persistent arthralgia lasting weeks to years. The re-emergence of CHIKV has resulted in numerous outbreaks in the eastern hemisphere, and threatens to expand in the foreseeable future. Unfortunately, no effective treatment is currently available. The present study reports the use of resazurin in a cell-based high-throughput assay, and an image-based high-content assay to identify and characterize inhibitors of CHIKV-infection in vitro. CHIKV is a highly cytopathic virus that rapidly kills infected cells. Thus, cell viability of HuH-7 cells infected with CHIKV in the presence of compounds was determined by measuring metabolic reduction of resazurin to identify inhibitors of CHIKV-associated cell death. A kinase inhibitor library of 4,000 compounds was screened against CHIKV infection of HuH-7 cells using the resazurin reduction assay, and the cell toxicity was also measured in non-infected cells. Seventy-two compounds showing >= 50% inhibition property against CHIKV at 10 mu M were selected as primary hits. Four compounds having a benzofuran core scaffold (CND0335, CND0364, CND0366 and CND0415), one pyrrolopyridine (CND0545) and one thiazol-carboxamide (CND3514) inhibited CHIKV-associated cell death in a dose-dependent manner, with EC50 values between 2.2 mu M and 7.1 mu M. Based on image analysis, these 6 hit compounds did not inhibit CHIKV replication in the host cell. However, CHIKV-infected cells manifested less prominent apoptotic blebs typical of CHIKV cytopathic effect compared with the control infection. Moreover, treatment with these compounds reduced viral titers in the medium of CHIKV-infected cells by up to 100-fold. In conclusion, this cell-based high-throughput screening assay using resazurin, combined with the image-based high content assay approach identified compounds against CHIKV having a novel antiviral activity -inhibition of virus-induced CPE - likely by targeting kinases involved in apoptosis.
