The objective of this work were apply and provide a preliminary evaluation of the Weather Research and Forecasting model coupled with Chemistry (WRF-Chem) performance, for Londrina region. We performed comparison with measurements obtained in meteorological stations. The model was configured to run with three domains with 27,9 and 3 km of grid resolution, using the ndown program and also was realized a simulation with the model configured to run with a single domain using a land use file based in a classified image for region of MODIS sensor. The emission files to supply the chemistry run were generated based in the work of Martins et al., 2012. RADM2 chemical mechanism and MADE/SORGAM modal aerosol models were used in the simulations. The results demonstrated that model was able to represent coherently the formation and dispersion of the pollution in Metropolitan Region of Londrina and also the importance of using the appropriate land use file for the region.

A análise de elementos traço, tais como terras raras, TH, U, Ta, Hf, Ba, Rb e Ba, é uma ferramenta muito importante para estudos petrogenéticos. No intuito de estudar tais processos em diques do Enxame Serra do Mar (litoral de São Paulo e Rio de Janeiro), pertencente à Provincia Magmática do Paraná (PMP), uma das províncias de basaltos continentais mais expressivas do mundo, foram realizadas análises por ativação com nêutrons nestes diques. A técnica, empregada no Centro de Reator de pesquisa do Instituto de Pesquisas Energéticas e Nucleares, forneceu concentrações de elementos traço com os níveis de precisão de 10% e exatidão 9% os quais são adequadas para estudos petrogenéticos. Devido as baixas concentrações dos elementos analisados, a rotina experimental de preparação das amostras abrangeu processos bastantes cuidadosos para evitar contaminação. O uso de radionuclídeos em hidrologia, como a concentração de urânio e a razão de atividades (234U/238U), tornou-se ultimamente uma ferramenta de grande importância nas pesquisas relacionadas à água subterrânea. Estes parâmetros têm sido usados para identificar os principais mecanismos de mobilização, precipitação e transporte dos isótopos de um aquífero, ou seja, esses isótopos têm sido empregados como traçadores para água. A área de estudo deste trabalho compreende os corpos graníticos fraturados da Suíte Intrusiva de Itu, que afloram a leste do Estado de São Paulo junto à Bacia Sedimentar do Paraná, fazendo parte da Bacia Hidrográfica do Médio Tietê. Os poços de onde foram coletadas as águas subterrâneas investigadas foram perfurados nos granitos desta Suíte, localizados nos municípios de Salto (poço YVC), Itu (HND), Itupeva (poço ITUP) e Indaiatuba (poços BDN 1 e BDN 2), sendo estes utilizados no abastecimento residencial ou para irrigação. As coletas foram mensais e o tempo de amostragem foi de um ano e meio com o objetivo de avaliar a sazonalidade destas águas. A técnica utilizada, para discriminar os isótopos de urânio dissolvidos, bem como para determinar sua razão de atividade (234U/238U), nas amostras das águas subterrâneas, foi a de espectrometria alfa. Os resultados mostram que os parâmetros físico-químicos e composição química da água exercem um papel muito importante no comportamento dos isótopos analisados. As importantes variações observadas tanto entre as águas de um mesmo poço, como também entre poços diferentes, mostram que os processos de recarga e/ou de interação rocha – água ocorrem de maneira distinta.

La tesi analizza, nel quadro del secondo dopoguerra, quattro casi studio scelti tra le opere di ricostruzione dell’architetto Josef Wiedemann (1910-2001) nel centro di Monaco di Baviera: Odeon (1951-1952), Alte Akademie (1951-1955), Siegestor (1956-1958) e Glyptothek (1961-1972). L’architetto si occupa di opere simbolo della città di Monaco, affrontando la loro ricostruzione come un tema fondante per la storia e l’identità del popolo bavarese, ma soprattutto come un’occasione per definire un metodo d’intervento sulle rovine della guerra. Il suo lavoro è caratterizzato infatti per la ricerca costante di una sintesi tra interesse per la conservazione dell’antico e apertura al nuovo; ispirandosi all’insegnamento del maestro Hans Döllgast, Wiedemann traccia una nuova originale strada per l’intervento sull’antico, segnata da una profonda capacità tecnico-progettuale e dall'attenzione alle nuove esigenze a cui deve rispondere un’architettura contemporanea. Partendo dai suoi scritti e dalle sue opere, si può rilevare un percorso coerente che, partendo dalla conoscenza della storia dell'edificio, ripercorrendone l’evoluzione dallo stato che potremmo definire “originario” allo stato di rovina, giunge a produrre nel progetto realizzato una sintesi tra il passato e il futuro. L'architetto, nella visione di Wiedemann, è chiamato a un compito di grande responsabilità: conoscere per progettare (o ri-progettare) un edificio che porta impressi su di sé i segni della propria storia. Nel metodo che viene messo progressivamente a punto operando nel corpo vivo dei monumenti feriti dalla guerra, è percepibile fino a distinguerlo chiaramente l’interesse e l’influenza del dibattito italiano sul restauro. La conservazione “viva” dell'esistente, così come viene definita da Wiedemann stesso, si declina in modo diverso per ogni caso particolare, approdando a risultati differenti tra loro, ma che hanno in comune alcuni principi fondamentali: conoscere, ricordare, conservare e innovare.

La tesi affronta la tematica del sessismo linguistico e analizza le proposte elaborate dalle autorità dei paesi francofoni (Belgio, Francia, Québec, Svizzera) per far fronte a questo problema e garantire una migliore rappresentazione delle donne nella lingua. Dopo una breve spiegazione sul sessismo linguistico, la tesi si sofferma sui decreti, le guide e le circolari ministeriali proposti nel tempo dai singoli paesi, mettendo in luce una sempre maggiore attenzione da parte delle autorità a questa tematica delicata. L'elaborato si conclude con un confronto fra le proposte dei vari paesi, da cui emerge come il Québec sia il paese più all'avanguardia in materia di femminizzazione dei nomi di mestiere, titolo, grado e funzione e la Svizzera lo stato più attivo in quanto a redazione non discriminatoria dei testi.

Genome-wide association studies and candidate gene studies in ulcerative colitis have identified 18 susceptibility loci. We conducted a meta-analysis of six ulcerative colitis genome-wide association study datasets, comprising 6,687 cases and 19,718 controls, and followed up the top association signals in 9,628 cases and 12,917 controls. We identified 29 additional risk loci (P < 5 × 10(-8)), increasing the number of ulcerative colitis-associated loci to 47. After annotating associated regions using GRAIL, expression quantitative trait loci data and correlations with non-synonymous SNPs, we identified many candidate genes that provide potentially important insights into disease pathogenesis, including IL1R2, IL8RA-IL8RB, IL7R, IL12B, DAP, PRDM1, JAK2, IRF5, GNA12 and LSP1. The total number of confirmed inflammatory bowel disease risk loci is now 99, including a minimum of 28 shared association signals between Crohn's disease and ulcerative colitis.

PURPOSE: Neoadjuvant treatment is an accepted standard approach for treating locally advanced esophago-gastric adenocarcinomas. Despite a response of the primary tumor, a significant percentage dies from tumor recurrence. The aim of this retrospective exploratory study from two academic centers was to identify predictors of survival and recurrence in histopathologically responding patients. METHODS: Two hundred thirty one patients with adenocarcinomas (esophagus: n = 185, stomach: n = 46, cT3/4, cN0/+, cM0) treated with preoperative chemotherapy (n = 212) or chemoradiotherapy (n = 19) followed by resection achieved a histopathological response (regression 1a: no residual tumor (n = 58), and regression 1b < 10 % residual tumor (n = 173)). RESULTS: The estimated median overall survival was 92.4 months (5-year survival, 56.6 %) for all patients. For patients with regression 1a, median survival is not reached (5-year survival, 71.6 %) compared to patients with regression 1b with 75.3 months median (5-year survival, 52.2 %) (p = 0.031). Patients with a regression 1a had lymph node metastases in 19.0 versus 33.7 % in regression 1b. The ypT-category (p < 0.001), the M-category (p = 0.005), and the type of treatment (p = 0.04) were found to be independent prognostic factors in R0-resected patients. The recurrence rate was 31.7 % (n = 66) (local, 39.4 %; peritoneal carcinomatosis, 25.7 %; distant metastases, 50 %). Recurrence was predicted by female gender (p = 0.013), ypT-category (p = 0.007), and M-category (p = 0.003) in multivariate analysis. CONCLUSION: Response of the primary tumor does not guarantee recurrence-free long-term survival, but histopathological complete responders have better prognosis compared to partial responders. Established prognostic factors strongly influence the outcome, which could, in the future, be used for stratification of adjuvant treatment approaches. Increasing the rate of histopathological complete responders is a valid endpoint for future clinical trials investigating new drugs.

Evidence from animal and human studies imply the amygdala as the most critical structure involved in processing of fear-relevant stimuli. In phobias, the amygdala seems to play a crucial role in the pathogenesis and maintenance of the disorder. However, the neuropathology of specific phobias remains poorly understood. In the present study, we investigated whether patients with spider phobia show altered amygdala volumes as compared to healthy control subjects.

Deep brain stimulation (DBS) for Parkinson's disease often alleviates the motor symptoms, but causes cognitive and emotional side effects in a substantial number of cases. Identification of the motor part of the subthalamic nucleus (STN) as part of the presurgical workup could minimize these adverse effects. In this study, we assessed the STN's connectivity to motor, associative, and limbic brain areas, based on structural and functional connectivity analysis of volunteer data. For the structural connectivity, we used streamline counts derived from HARDI fiber tracking. The resulting tracks supported the existence of the so-called "hyperdirect" pathway in humans. Furthermore, we determined the connectivity of each STN voxel with the motor cortical areas. Functional connectivity was calculated based on functional MRI, as the correlation of the signal within a given brain voxel with the signal in the STN. Also, the signal per STN voxel was explained in terms of the correlation with motor or limbic brain seed ROI areas. Both right and left STN ROIs appeared to be structurally and functionally connected to brain areas that are part of the motor, associative, and limbic circuit. Furthermore, this study enabled us to assess the level of segregation of the STN motor part, which is relevant for the planning of STN DBS procedures.

Crohn's disease and ulcerative colitis, the two common forms of inflammatory bowel disease (IBD), affect over 2.5 million people of European ancestry, with rising prevalence in other populations. Genome-wide association studies and subsequent meta-analyses of these two diseases as separate phenotypes have implicated previously unsuspected mechanisms, such as autophagy, in their pathogenesis and showed that some IBD loci are shared with other inflammatory diseases. Here we expand on the knowledge of relevant pathways by undertaking a meta-analysis of Crohn's disease and ulcerative colitis genome-wide association scans, followed by extensive validation of significant findings, with a combined total of more than 75,000 cases and controls. We identify 71 new associations, for a total of 163 IBD loci, that meet genome-wide significance thresholds. Most loci contribute to both phenotypes, and both directional (consistently favouring one allele over the course of human history) and balancing (favouring the retention of both alleles within populations) selection effects are evident. Many IBD loci are also implicated in other immune-mediated disorders, most notably with ankylosing spondylitis and psoriasis. We also observe considerable overlap between susceptibility loci for IBD and mycobacterial infection. Gene co-expression network analysis emphasizes this relationship, with pathways shared between host responses to mycobacteria and those predisposing to IBD.

We characterized changes in the visual behavior of mice in which a loss of the retinal pigment epithelium (RPE) was experimentally induced with intravenous (i.v.) administration of sodium iodate (NaIO3). We compared and correlated these changes with alterations in neural retinal structure and function. RPE loss was induced in 4-6 week old male C57BL/6 mice with an i.v. injection of 1% NaIO3 at three concentrations: 35, 50, or 70 mg/kg. At 1, 3, 7, 14, 21, and 28 days (d) as well as 6 months post injection (PI) a behavioral test was performed in previously trained mice to evaluate visual function. Eye morphology was then assessed for changes in both the RPE and neural retina. NaIO3-induced RPE degeneration was both dose and PI time dependent. Our low dose showed no effects, while our high dose caused the most damage, as did longer PI times at our intermediate dose. Using the intermediate dose, no changes were detectable in either visual behavior or retinal morphology at 1 d PI. However, at 3 d PI visual behavior became abnormal and patchy RPE cell loss was observed. From 7 d PI onward, changes in retinal morphology and visual behavior became more severe. At 6 months PI, no recovery was seen in any of these measures in mice administered the intermediate dose. These results show that NaIO3 dosage and/or time PI can be varied to produce different, yet permanent deficits in retinal morphology and visual function. Thus, this approach should provide a unique system in which the onset and severity of RPE damage, and its consequences can be manipulated. As such, it should be useful in the assessment of rescue or mitigating effects of retinal or stem cell transplantation on visual function.

Phobias are characterized by excessive fear, cued by the presence or anticipation of a fearful situation. Whereas it is well established that glucocorticoids are released in fearful situations, it is not known whether these hormones, in turn, modulate perceived fear. As extensive evidence indicates that elevated glucocorticoid levels impair the retrieval of emotionally arousing information, they might also inhibit retrieval of fear memory associated with phobia and, thereby, reduce phobic fear. Here, we investigated whether acutely administrated glucocorticoids reduced phobic fear in two double-blind, placebo-controlled studies in 40 subjects with social phobia and 20 subjects with spider phobia. In the social phobia study, cortisone (25 mg) administered orally 1 h before a socio-evaluative stressor significantly reduced self-reported fear during the anticipation, exposure, and recovery phase of the stressor. Moreover, the stress-induced release of cortisol in placebo-treated subjects correlated negatively with fear ratings, suggesting that endogenously released cortisol in the context of a phobic situation buffers fear symptoms. In the spider phobia study, repeated oral administration of cortisol (10 mg), but not placebo, 1 h before exposure to a spider photograph induced a progressive reduction of stimulus-induced fear. This effect was maintained when subjects were exposed to the stimulus again 2 days after the last cortisol administration, suggesting that cortisol may also have facilitated the extinction of phobic fear. Cortisol treatment did not reduce general, phobia-unrelated anxiety. In conclusion, the present findings in two distinct types of phobias indicate that glucocorticoid administration reduces phobic fear.

The "Schema-focussed Emotive Behavioral Therapy" (SET) was developed by our research group as a new group therapy approach for patients with personality disorders from all clusters (A to C; DSM-IV). It was evaluated in a randomised controlled study (n = 93). Data were collected before and after treatment as well as one year after study entry. A completer analysis was conducted with matched subgroups (n = 60). After therapy, SET patients improved in the outcome domains interactional behavior, strain, and symptomatic complaints (IIP-D, GAF, VEV-VW, BSI-P). Furthermore, they showed a significant lower dropout rate. At the follow-up assessment, Cluster C patients of the experimental group deteriorated with regard to symptomatic complaints (BSI-P). In contrast, cluster B patients improved more over time compared to control subjects. SET seems to be an adequate and effective group therapy with effects that seem to be stable over time, especially for patients with Cluster B diagnosis.

In schizophrenia there is a consistent epidemiological finding of a birth excess in winter and spring. Season of birth is thought to act as a proxy indicator for harmful environmental factors during foetal maturation. There is evidence that prenatal exposure to harmful environmental factors may trigger pathologic processes in the neurodevelopment, which subsequently increase the risk of schizophrenia. Since brain white matter alterations have repeatedly been found in schizophrenia, the objective of this study was to investigate whether white matter integrity was related to the season of birth in patients with schizophrenia. Thirty-four patients with schizophrenia and 33 healthy controls underwent diffusion tensor imaging. Differences in the fractional anisotropy maps of schizophrenia patients and healthy controls born in different seasons were analysed with tract-based spatial statistics. A significant main effect of season of birth and an interaction of group and season of birth showed that patients born in summer had significantly lower fractional anisotropy in widespread white matter regions than those born in the remainder of the year. Additionally, later age of schizophrenia onset was found in patients born in winter months. The current findings indicate a relationship of season of birth and white matter alterations in schizophrenia and consequently support the neurodevelopmental hypothesis of early pathological mechanisms in schizophrenia.
