Diuron (3-(3,4-Dichlorophenyl)-1,1-dimethylurea) is a substituted urea herbicide widely used on agricultural crops such as soy, cotton and sugar cane. In a previous long-term study this herbicide exerted carcinogenic activity on the urinary bladder and renal pelvis mucosa of Wistar rats and breast of mice. Also, it was shown to be carcinogenic to the mice skin in a initiation-promotion assay. In 1997, the northamerican EPA evaluated Diuron as a “known/likely” carcinogen for humans (USEPA, 2004). In a previous study developed at this laboratory, male Wistar rats treated with Diuron 2500 ppm during 20 weeks presented increased indices of cell proliferation and incidences of simple urothelial hyperplasia (HS) in the urinary bladder. Under scanning electron microscopy (SEM) severe urothelial necrosis and hyperplasia were observed. However, in that study the urinary bladders of animals exposed to lower doses of Diuron were not examined under SEM. Therefore, the possible dose-response influence of Diuron on the urothelium under SEM is not known. The present study aimed to analyze under SEM the urinary bladder of male Wistar rats exposed to 125 ppm, 500 ppm and 2500 ppm doses of Diuron through diet during 20 weeks and to compare to the previous histological findings in the same material. Under SEM, 125 ppm and 2500 ppm groups presented significantly (p<0,05) increased incidences of simple hyperplasia, i.e., 7/10 and 8/10 respectively, compared to control group and the 500 ppm group The sensitivity of SEM was higher since it detected a 45% incidence of hyperplasiaswhile the histological analysis found only 27%. Considering SEM as the gold-standard, histology showed a 44% sensitivity, 86.4% specificity, a positive predictive value of 72,7% and negative predictive value of 65,5% and accuracy of 67,5%. Maternal undernutrition affects the foetal development, promoting renal alterations and adult hypertension. The present study investigates, in adult male rats, the effect of food restriction in utero on arterial blood pressure changes (AP), and its possible association with the number of nephrons, renal function and angiotensin II (AT1R/AT2R), glucocorticoid (GR) and mineralocorticoid (MCR) receptors expression. The daily food supply to pregnant rats was measured and one group (n=5) received normal quantity of food (NF) while the other group received 50% of that (FR50) (n=5). The AP was measured weekly. At 16 weeks of life, fractionator’s method was used to estimate glomeruli number in histological slices. The renal function was estimate by creatinine and lithium clearances. Blood and urine samples were collected to biochemical determination of creatinine, sodium, potassium and lithium. At 90th and 23rd days of life, kidneys were also processed to AT1R, AT2R, GR and MCR immunolocalization and for western blotting analysis. FR50 offspring shows a significant reduction in BW (FR50: 5.67 ± 0.16 vs. 6.84 ± 0.13g in NF, P<0.001) and increased AP from 6th to 12nd week (6thwk FR50: 149.1 ± 3.4 vs. 125.1 ± 3.2mmHg in NF, P<0.001and, 12ndwk FR50: 164.4 ± 4.9 vs. 144.0 ± 3.3 mmHg in NF, P=0.02). Expression of AT1R and AT2R were significantly decreased in FR50 (AT1, 59080 ± 2709 vs. 77000 ± 3591 in NF, P=0.05; AT2, 27500 ± 95.50 vs. 67870 ± 1509 in NF, P=0.001) while the expression of GR increased in FR50 (36090 ± 781.5 vs. 4446 ± 364.5 in NF, P=0.0007). The expression of MCR did not change significantly. We also verified a pronounced decrease in fractional urinary sodium excretion in FR50 offspring (0.03 ± 0.02 vs. 0.06 ± 0.04 in NF, p=0.03). It is understood by Chronic Renal Failure (CRF) or Stage IV Chronic Kidney Disease (CKD) the morbid state in which there is a substantial and irreversible loss of functional capacity, metabolic and endocrine function in both kidneys, leading to a framework of dehydration, azotemia, electrolyte imbalance, anemia, uremic syndrome, secondary hyperparathyroidism, among other conditions that make the patient's life without treatment unfeasible. An adequate nutritional support to patients with CKD at any stage aims to maintain a good clinical condition and an attempt to delay the progression of the disease. The basis of a diet for patients with CKD is the restriction in this protein being of high biological value, low-phosphorus, high energy density, fiber and antioxidants combined with a good dietary practices that allow the patient a good quality of life

Chronic Renal Failure (CRF) is the most common disease among dogs of any breed. It is the cause of high levels of morbidity and death rate in dogs. It's important to have an early diagnostic of the dogs condition and immediately start a supportive treatment for CRF. In this research we are discussing the most common types of treatment focused in reducing the progression of this disease and better quality of life for the animal with IRC

Uremic syndrome, arising from kidney malfunction, consists of a set of systemic changes caused by the accumulation of toxic substances to the body. Since, with the advance of medicine, the animals lived more reaching advanced age and entering thus on track of senility, chronic renal disease, became a common complaint in the routine of the ambulatory. This high rate of morbidity generates an increased need for clarification of pathophysiology involved in this disease. The components of the uremic syndrome include water and sodium imbalance, anemia, intolerance to carbohydrate, neurological disorders, disorders of the gastrointestinal tract, osteoarthritis, immunological incompetence and metabolic acidosis. The clinical manifestations occur in isolation or in combination. In most cases canines patients are subject to an assessment when the kidney disease has evolved to the final stage with uremic syndrome and installed already, under these conditions, the prognosis is reserved

Diuron (3-(3,4-dichlorophenyl)-1,1-dimethylurea) is a substituted urea herbicide widely used in crops of sugar cane, cotton and soybeans. In 1997, this agent has been classified by the United States Environmental Protection Agency as known/likely human carcinogen because it induced tumors in the urinary bladder and renal pelvis of rats, and breast and skin of mice exposed to 2500 ppm for feed for two years. A previous study from our group demonstrated dose-response relationship in the gene expression profile associated with severe necrosis on bladder urothelium and increased incidence of simple hyperplasia in male Wistar rats treated with different concentrations of diuron for 20 weeks. To check how early the molecular changes occurs, rats were fed for 7 days with diets containing diuron at 0, 125, 500 or 2500 ppm. The main observations recorded were urothelium ultrastructural alterations and disruptions of molecular pathways associated with cell-cell interaction and the tissue organization maintenance. Particularly, the gene Glypican 3 (Gpc3), a surface proteoglycan related to cellular adhesion and apoptosis induction, was down regulated on urothelium exposed to 2500ppm diuron for 7 days and 20 weeks. The aim of this study was validate by quantitative RT-PCR real time, the reduced Gpc3 gene expression in epithelial cells of the urinary bladder of male Wistar rats treated with different concentrations of diuron for 7 days and 20 weeks. The endogenous control of the quantitative PCR real time technique was the β-actin gene and the target was the gene Gpc3. The relative quantification (RQ) was obtained by the method of relative quantification 2-ΔΔCt . Animals exposed to diuron for 7 days or for 20 weeks presented reduction of Gpc3 gene expression compared to the control group. This reduction was statistically significant only for the 7 days study. A Doença Renal Crônica (DRC) é considerada um problema mundial de saúde pública. O paciente com DRC apresenta baixa tolerância ao exercício físico e como conseqüência apresenta limitações na capacidade funcional e força muscular respiratória. O paciente com DRC em programa de hemodiálise pode sofrer impactos negativos na sua capacidade funcional. Objetivo: Avaliar a capacidade funcional e a força muscular respiratória associada ao exercício físico em pacientes com doença renal crônica que realizam hemodiálise. Metodologia: 28 pacientes de ambos os sexos com idade entre 40 e 60 anos participaram do estudo, em programa de hemodiálise no Instituto do Rim da Santa Casa de Misericórdia de Presidente Prudente-SP. Os pacientes foram avaliados pela força muscular inspiratória (PImax) e força muscular expiratória (PEmax) pela manovacuometria e Teste de Caminhada de Seis Minutos (TC6’) para avaliar a capacidade funcional. Após as avaliações, os pacientes iniciaram o programa de atividade física, com duração de oito semanas, realizado 3 vezes por semana durante 40 minutos na hemodiálise. Ao final do programa de exercícios físicos, os pacientes foram reavaliados. Resultados: Na avaliação da força muscular respiratória o valor da PImax pós foi significativamente maior que o valor obtido na avaliação pré programa (p<0,05), para a variável PEmax não foi encontrada diferença significativa entre os momentos pré e pós intervenção (p<0,05). As avaliações da capacidade funcional ou Teste de Caminhada de Seis Minutos (TC6’) inicial e após o programa de exercícios, não apresentaram diferenças significativas (p<0,05). Dados da Escala de Borg indicaram redução significativa do cansaço e dispnéia (p<0,05), após o programa de exercícios. The pets, mostly dogs and cats are susceptible to a variety of situations that can lead to acute kidney failure, such as infection and intoxication, besides other common causes such as dehydration or other pre-existing diseases. The abrupt decrease in renal function in these animals characterizes a severe clinical condition and requires specialized veterinary medical care since it increases the death. Early diagnosis is an important factor for the survival of these animals and so is the appropriate treatment. New technologies on diagnosis and treatment of serious diseases in veterinary medicine have made acute renal failure successfully reversed by hemodialysis in cases where conventional therapy may not be effective. In cases like that, hemodialysis treatment may be the only chance of survival for the animals, which makes this therapy more and more important and necessary, especially in referral centers of veterinary assistance

Brazil is a country with the largest world´s vegetal genetic diversity and the Environmental Protection Area (APA) of the “Serra da Mantiqueira” is a very heterogeneous region, representing one of the richest sources of pharmacologically actives materials. The population uses medicinal plants and according to the OMS, 80% of the population uses them in primary treatment of several diseases. Nevertheless, the loss of traditional knowledge associated with medicinal plants is occurring quickly. The ethnopharmacological strategy uses traditional knowledge to the search for medicinal plants that can have bioactive substances against diseases that afflicting the population and thus protect traditional knowledge. The “cipó-prata” (Trigonia nívea Cambess.) is a native plant normally found in the “Bacia do Paraná” region and present in the flora in the neighborhood of “Marins”, Piquete-SP and usually, said for the treatment of renal and urinary diseases. So, the objective of this study was test if the “cipóprata” (Trigonia nívea Cambess.) has effects on the renal excretion of water and salt, in anesthetized Wister rats. The tests were made in males Wistar rats and randomly distributed into 4 experimental groups: Group I – aqueous control, Group II – treated with aqueous extract (EA) of “cipó-prata”, Group III – water control + “tween 80”, Group IV – treated with ethanol extract (EE) and “cipó-prata”. All groups were subjected to experimental protocol, composed of three periods: Balance (40 minutes), Basal (30 minutes) and Experimental (90 minutes), occurring the urine collection every 30 minutes, from the basal period and measuring blood pressure every 10 minutes. The kidneys, for his anatomical and functional characteristics, are sensitive to affections that take the liberation of renal toxins or the blood supply, causing irreversible injuries to his renoparenchymal tissue that is substituted by fibrous tissue. Even after the resolution of the basic cause, there will be the loss of a significant number of his functional unity, renal adaptations will take place in the attempt of maintaining the renal function. These adaptations produce additional injuries, perpetuating to loss of renoparenchymal tissue and the reduction of the renal function. The renal insufficiency (IR) takes place after the loss of 3/4 of the number of his functional unities. Before the progression to the phase of IR, the animal shows up practically without symptoms, but for the gradual increase of the urinary and of the ingestion of water. The reduction of the degree of renal function leads to alterations system compensatory for the accumulation of substances that would suffer renal excretion. The progression of IR leads to the phase of the syndrome urêmica. In this phase the animal presents innumerable clinical signs that can take it to the death. The treatment is symptomatic and dietetic, but depending on the phase not much efficient. Because of being progressive and insidious, the IRC demands the preparation of campaigns and programs of explanation for the veterinary doctors who aim for the consciousness and/or sensibility of the owners to carry out periodic examinations of selection that precociously detect the renal dysfunction. The diagnosis in the beginning IR enables the efficiency of the treatment in stop or slowing his progression, extending the time and quality of life of the patient

It is known that exposure to substances in the environment can contribute to various reproductive disorders, especially if such exposure occurs during critical periods of development such as the intra-uterine and postnatal. The female reproductive system may be the target of androgens, both as a result of exposure to environmental chemicals, or by pathological conditions (polycystic ovary syndrome or congenital adrenal hyperplasia).Usually, little attention is given off in relation to the study of androgenic effects in the female reproductive axis. This study aims to evaluate the effects of exposure to androgens on the development, structure and reproductive function in rats whose mothers were exposed to testosterone propionate from gestational day 12 (DG12) after weaning - postnatal day 21 (DPN21) . For this purpose, pregnancy rats were divided into four groups: a control group that received corn oil (vehicle) and three groups receiving testosterone propionate in doses of 0.05 mg / kg / day, 0.1 mg / kg / day and 0.2 mg / kg / day, all under the same experimental conditions. The possible effects of exposure were assessed using reproductive parameters, such as a measure of anogenital distance, count areolas / nipples, age at vaginal opening and first estrus (puberty indicative installation), weight and histological evaluation of the reproductive organs ( uterus and ovaries), weight of the kidneys, liver and pituitary hormone levels, regularity of the estrous cycle, sexual behavior and fertility. Such analysis is important in understanding the effects of androgen exposure on the female genital system, especially on the reproductive potential, and processes that may involve morphofunctional changes. In these experimental conditions, it is concluded that treatment with PT caused reduction in body weight and initial masculinization in females without cubs, however, commit further sexual development
