Background Entry into mitosis is regulated by cyclin dependent kinases that in turn are phosphoregulated. In most eukaryotes, phosphoregulation is through WEE1 kinase and CDC25 phosphatase. In higher plants a homologous CDC25 gene is unconfirmed and hence the mitotic inducer Schizosaccharomyces pombe (Sp) cdc25 has been used as a tool in transgenic plants to probe cell cycle function. Expression of Spcdc25 in tobacco BY-2 cells accelerates entry into mitosis and depletes cytokinins; in whole plants it stimulates lateral root production. Here we show, for the first time, that alterations to cytokinin and ethylene signaling explain the rooting phenotype elicited by Spcdc25 expression in Arabidopsis. Results Expressing Spcdc25 in Arabidopsis results in increased formation of lateral and adventitious roots, a reduction of primary root width and more isodiametric cells in the root apical meristem (RAM) compared with wild type. Furthermore it stimulates root morphogenesis from hypocotyls when cultured on two way grids of increasing auxin and cytokinin concentrations. Microarray analysis of seedling roots expressing Spcdc25 reveals that expression of 167 genes is changed by > 2-fold. As well as genes related to stress responses and defence, these include 19 genes related to transcriptional regulation and signaling. Amongst these was the up-regulation of genes associated with ethylene synthesis and signaling. Seedlings expressing Spcdc25 produced 2-fold more ethylene than WT and exhibited a significant reduction in hypocotyl length both in darkness or when exposed to 10 ppm ethylene. Furthermore in Spcdc25 expressing plants, the cytokinin receptor AHK3 was down-regulated, and endogenous levels of iPA were reduced whereas endogeous IAA concentrations in the roots increased. Conclusions We suggest that the reduction in root width and change to a more isodiametric cell phenotype in the RAM in Spcdc25 expressing plants is a response to ethylene over-production. The increased rooting phenotype in Spcdc25 expressing plants is due to an increase in the ratio of endogenous auxin to cytokinin that is known to stimulate an increased rate of lateral root production. Overall, our data reveal important cross talk between cell division and plant growth regulators leading to developmental changes.

This study determined annual and monthly fluctuations in concentration of 20 fungal genera. The selection of taxa was made based upon their high frequency in the air as well as their well-known allergenic properties. Air samples were collected using a spore trap of Hirst design at an urban site where the trap continuously worked throughout a 5-year survey. Weather data were acquired from a meteorological station co-located with the air sampler. Influence of several meteorological parameters was then examined to reveal speciesenvironment interactions and the potential location of fungal spore sources within the urban area. The maximum monthly sum of mean daily spore concentration varied between genera, and the earliest peaks were recorded for Pleospora sp. in April and Ustilago sp. in June. However, the majority of investigated spore types occurred in the greatest concentrations between August and September. Out of the 20 studied taxa, the most dominant genus was Cladosporium sp., which exceeded an allergenic threshold of 3000 s m-3 40 times during very rainy years and twice as much during dry years. A Spearman’s rank test showed that statistically significant (p B 0.05) relationships between spore concentration and weather parameters were mainly rs B 0.50. Potential sources of spores at Worcester were likely to be localised outside the city area.

G.H. Mead (1863-1931) oriented much of his intellectual efforts around three unavoidable questions for anyone living in a modern society: how are selfhood, knowledge, and politics understood and organized in such a society? Modern individuals continually seek answers to questions although nobody has ever come up with a definitive answer to them. Modernity, in other words, confronts us with inevitable problematics that fundamentally shape the way in which we think about certain topics. For the purposes of my discussion of Mead, I focus upon three of these modern problematics: science, selfhood, and democratic politics. But before I discuss Mead’s treatment of these problem areas, allow me to briefly situate Mead as a pragmatist in relation to Dewey and James within pragmatism.

In 1968, Herbert Marcuse believed that a Great Refusal was possible, one that would deny the exploitative power of corporate capitalism. Marcuse's vision was never realised. This essay argues that society today is in an advanced state of that which the Frankfurt School termed repressive desublimation and questions whether a liberationary praxis is still possible. It claims that Bret Easton Ellis's fiction choreographs an internalising of the forms of critique that marked 1968 and about which Marcuse writes. It is Ellis's act of double voicing that allows him to develop a duplicitous recalcitrant voice within the state of assimilation and it is double voicing which emerges as the key technique in Ellis's work that effects an ongoing critique in commodity society. Looking at Slavoj iek's recent revisionism of the notion of repressive desublimation, which connects Marxism and psychoanalysis, the essay considers how Ellis's novels, American Psycho, Glamorama and Lunar Park, function to address and reconfigure the relationship between the status of the Marxist fetishised object and the psychoanalytic phobic object in the present-day era of late capitalism. This essay seeks to illuminate how Ellis's fiction, through an involution of Marcuse's political theories, enacts a contemporary refusal from within the state of reification.

This study shows that the relaxivity and optical properties of functionalised lanthanide-DTPA-bis-amide complexes (lanthanide=Gd3+ and Eu3+, DTPA=diethylene triamine pentaacetic acid) can be successfully modulated by addition of specific anions, without direct Ln3+/anion coordination. Zinc(II)-dipicolylamine moieties, which are known to bind strongly to phosphates, were introduced in the amide “arms of these ligands, and the interaction of the resulting Gd–Zn2 complexes with a range of anions was screened by using indicator displacement assays (IDAs). Considerable selectivity for polyphosphorylated species (such as pyrophosphate and adenosine-5′-triphosphate (ATP)) over a range of other anions (including monophosphorylated anions) was apparent. In addition, we show that pyrophosphate modulates the relaxivity of the gadolinium(III) complex, this modulation being sufficiently large to be observed in imaging experiments. To establish the binding mode of the pyrophosphate and gain insight into the origin of the relaxometric modulation, a series of studies including UV/Vis and emission spectroscopy, luminescence lifetime measurements in H2O and D2O, 17O and 31P NMR spectroscopy and nuclear magnetic resonance dispersion (NMRD) studies were carried out.

The RHPP policy provided subsidies for private householders, Registered social landlords and communities to install renewable heat measures in residential properties. Eligible measures included air and ground-source heat pumps, biomass boilers and solar thermal. Around 18,000 heat pumps were installed via this scheme. DECC funded a detailed monitoring campaign, which covered 700 heat pumps (around 4% of the total). The aim of this monitoring campaign was to assess the efficiencies of the heat pumps and to estimate the carbon and bill savings and amount of renewable heat generated. Data was collected from 31/10/2013 to 31/03/2015. This report represents the analysis of this data and represents the most complete and reliable data in-situ residential heat pump performance in the UK to date.

A new procedure for determining eleven organochlorine pesticides in soils using microwave-assisted extraction (MAE) and headspace solid phase microextraction (HS-SPME) is described. The studied pesticides consisted of mirex, α- and γ-chlordane, p,p’-DDT, heptachlor, heptachlor epoxide isomer A, γ-hexachlorocyclohexane, dieldrin, endrin, aldrine and hexachlorobenzene. The HS-SPME was optimized for the most important parameters such as extraction time, sample volume and temperature. The present analytical procedure requires a reduced volume of organic solvents and avoids the need for extract clean-up steps. For optimized conditions the limits of detection for the method ranged from 0.02 to 3.6 ng/g, intermediate precision ranged from 14 to 36% (as CV%), and the recovery from 8 up to 51%. The proposed methodology can be used in the rapid screening of soil for the presence of the selected pesticides, and was applied to landfill soil samples.

A procedure for the determination of seven indicator PCBs in soils and sediments using microwave-assisted extraction (MAE) and headspace solid-phase microextraction (HS-SPME) prior to GC-MS/MS is described. Optimization of the HS-SPME was carried out for the most important parameters such as extraction time, sample volume and temperature. The adopted methodology has reduced consumption of organic solvents and analysis runtime. Under the optimized conditions, the method detection limit ranged from 0.6 to 1 ng/g when 5 g of sample was extracted, the precision on real samples ranged from 4 to 21% and the recovery from 69 to 104%. The proposed method, which included the analysis of a certified reference material in its validation procedure, can be extended to several other PCBs and used in the monitoring of soil or sediments for the presence of PCBs.

A necessidade de construir que se manifesta em todos os sectores, desde o da habitação, em que o homem recupera da fadiga física e intelectual, consequência da sua actividade diária, até àquele em que exerce a sua actividade de produção ou onde realiza as suas ocupações sociais dos tempos livres, obriga a procurar soluções, para a limitação de espaços - cada vez mais económicos em tempo, dinheiro, materiais e mão de obra. Este objectivo levou ao desenvolvimento de técnicas de pré-fabricação,e bem assim a industrializar a construção, por elementos cada vez maiores - de acordo com os meios mecânicos de que se pode dispor na zona industrial, nos transportes e no estaleiro. Assim, conforme os elementos de construção a colocar em obra so manuseáveis ou, pelas suas dimenses e peso, careçam de meios mecânicos para poderem ser utilizados, a pré fabricação está diferenciada em: a) Pré fabricação ligeira b) Pré fabricação pesada Por outro lado, os diversos materiais que se utilizam na construção permitem a pré fabricação industrial de elementos de construção: metálicos, de madeira, de betão, de pedra, de argila cozida, de vidro, etc.

The integration of wind power in eletricity generation brings new challenges to unit commitment due to the random nature of wind speed. For this particular optimisation problem, wind uncertainty has been handled in practice by means of conservative stochastic scenario-based optimisation models, or through additional operating reserve settings. However, generation companies may have different attitudes towards operating costs, load curtailment, or waste of wind energy, when considering the risk caused by wind power variability. Therefore, alternative and possibly more adequate approaches should be explored. This work is divided in two main parts. Firstly we survey the main formulations presented in the literature for the integration of wind power in the unit commitment problem (UCP) and present an alternative model for the wind-thermal unit commitment. We make use of the utility theory concepts to develop a multi-criteria stochastic model. The objectives considered are the minimisation of costs, load curtailment and waste of wind energy. Those are represented by individual utility functions and aggregated in a single additive utility function. This last function is adequately linearised leading to a mixed-integer linear program (MILP) model that can be tackled by general-purpose solvers in order to find the most preferred solution. In the second part we discuss the integration of pumped-storage hydro (PSH) units in the UCP with large wind penetration. Those units can provide extra flexibility by using wind energy to pump and store water in the form of potential energy that can be generated after during peak load periods. PSH units are added to the first model, yielding a MILP model with wind-hydro-thermal coordination. Results showed that the proposed methodology is able to reflect the risk profiles of decision makers for both models. By including PSH units, the results are significantly improved.

O presente texto assinala os 75 anos da formação da Polaroid e celebra a memória da popularização da fotografia instantânea e do inventor Edwin Herbert Land, fundador da Polaroid Corporation, sendo um exemplo do espírito empreendedor norte-americano que conseguia tornar as invenções em produtos de acesso fácil, chegando aos utilizadores através da sua produção massificada. A constituição da empresa é também contemporânea do nascimento do design industrial nos EUA. Os primeiros exemplares foram concebidos pelos mais notáveis pioneiros do design industrial norte-americano, nomeadamente Walter Dorwin Teague, Henry Dreyfuss e Albrecht Goertz, bem como os últimos modelos so também representativos do design de produto contemporâneo deste país, com projectos liderados pelo inovador gabinete IDEO.

À medida que o mundo Ocidental se apresenta cada vez mais subjugado ao primado da tecnologia (Forman, 2007:1), a possibilidade de ignorar o crescente impacte da Ludologia vai-se gradualmente inviabilizando. Através da adopção de uma perspectiva anglo-portuguesa, a presente dissertação focar-se-á no amplamente popular jogo de vídeo Sid Meier’s Civilization V: Brave New World (2013). Ao (re)interpretar as relações luso-britânicas no espaço de tempo compreendido entre 1890 (Ultimatum britânico) e 1910 (implantação da Primeira República Portuguesa), a presente dissertação tem em vista não s estudar a imprensa periódica portuguesa e inglesa, mas também demonstrar o modo como o jogo de vídeo em questão permite ao jogador desconstruir o “prepotente” domínio britânico sobre o “little Portugal”. Neste contexto, atribuir-se-á um lugar de destaque ao evolucionismo de Herbert Spencer. Profundamente respeitado por toda a Europa, durante a segunda metade do sculo XIX e princípios do sculo XX, o pensamento spenceriano revelou-se fundamental para compreender, por um lado, e em toda a sua extenso, o discurso jornalístico de finais do sculo XIX e inícios do XX, e, por outro, a narrativa e a estética do jogo Sid Meier’s Civilization V: Brave New World. De um modo geral, a presente análise visa, acima de tudo, questionar o cariz pós-colonial do jogo de vídeo sob observação e, simultaneamente, a sua capacidade de desafiar, de forma subversiva, o que pode ser tido como uma arrogante soberania britânica sobre um Portugal militar, económica e politicamente indefeso.
