Background: The diagnosis of invasive candidiasis is difficult because there are no specific clinical manifestations of the disease and colonization and infection are difficult to distinguish. In the last decade, much effort has been made to develop reliable tests for rapid diagnosis of invasive candidiasis, but none of them have found widespread clinical use. Results: Antibodies against a recombinant N-terminal fragment of the Candida albicans germ tube-specific antigen hyphal wall protein 1 (Hwp1) generated in Escherichia coli were detected by both immunoblotting and ELISA tests in a group of 36 hematological or Intensive Care Unit patients with invasive candidiasis and in a group of 45 control patients at high risk for the mycosis who did not have clinical or microbiological data to document invasive candidiasis. Results were compared with an immunofluorescence test to detect antibodies to C. albicans germ tubes (CAGT). The sensitivity, specificity, positive and negative predictive values of a diagnostic test based on the detection of antibodies against the N-terminal fragment of Hwp1 by immunoblotting were 27.8 %, 95.6 %, 83.3 % and 62.3 %, respectively. Detection of antibodies to the N-terminal fragment of Hwp1 by ELISA increased the sensitivity (88.9 %) and the negative predictive value (90.2 %) but slightly decreased the specificity (82.6 %) and positive predictive values (80 %). The kinetics of antibody response to the N-terminal fragment of Hwp1 by ELISA was very similar to that observed by detecting antibodies to CAGT. Conclusion: An ELISA test to detect antibodies against a recombinant N-terminal fragment of the C. albicans germ tube cell wall antigen Hwp1 allows the diagnosis of invasive candidiasis with similar results to those obtained by detecting antibodies to CAGT but without the need of treating the sera to adsorb the antibodies against the cell wall surface of the blastospore.

Nucleic acids are most commonly associated with the genetic code, transcription and gene expression. Recently, interest has grown in engineering nucleic acids for biological applications such as controlling or detecting gene expression. The natural presence and functionality of nucleic acids within living organisms coupled with their thermodynamic properties of base-pairing make them ideal for interfacing (and possibly altering) biological systems. We use engineered small conditional RNA or DNA (scRNA, scDNA, respectively) molecules to control and detect gene expression. Three novel systems are presented: two for conditional down-regulation of gene expression via RNA interference (RNAi) and a third system for simultaneous sensitive detection of multiple RNAs using labeled scRNAs.
RNAi is a powerful tool to study genetic circuits by knocking down a gene of interest. RNAi executes the logic: If gene Y is detected, silence gene Y. The fact that detection and silencing are restricted to the same gene means that RNAi is constitutively on. This poses a significant limitation when spatiotemporal control is needed. In this work, we engineered small nucleic acid molecules that execute the logic: If mRNA X is detected, form a Dicer substrate that targets independent mRNA Y for silencing. This is a step towards implementing the logic of conditional RNAi: If gene X is detected, silence gene Y. We use scRNAs and scDNAs to engineer signal transduction cascades that produce an RNAi effector molecule in response to hybridization to a nucleic acid target X. The first mechanism is solely based on hybridization cascades and uses scRNAs to produce a double-stranded RNA (dsRNA) Dicer substrate against target gene Y. The second mechanism is based on hybridization of scDNAs to detect a nucleic acid target and produce a template for transcription of a short hairpin RNA (shRNA) Dicer substrate against target gene Y. Test-tube studies for both mechanisms demonstrate that the output Dicer substrate is produced predominantly in the presence of a correct input target and is cleaved by Dicer to produce a small interfering RNA (siRNA). Both output products can lead to gene knockdown in tissue culture. To date, signal transduction is not observed in cells; possible reasons are explored.
Signal transduction cascades are composed of multiple scRNAs (or scDNAs). The need to study multiple molecules simultaneously has motivated the development of a highly sensitive method for multiplexed northern blots. The core technology of our system is the utilization of a hybridization chain reaction (HCR) of scRNAs as the detection signal for a northern blot. To achieve multiplexing (simultaneous detection of multiple genes), we use fluorescently tagged scRNAs. Moreover, by using radioactive labeling of scRNAs, the system exhibits a five-fold increase, compared to the literature, in detection sensitivity. Sensitive multiplexed northern blot detection provides an avenue for exploring the fate of scRNAs and scDNAs in tissue culture.

The superconducting properties and the microstructure of the Ag100-xPbx alloys, 1 ≤ x ≤ 5, prepared by rapid quenching from the liquid state with and without subsequent heat treatments, have been studied. The x-ray diffraction measurements show that supersaturated solid solutions of Pb in Ag can be obtained up to 3.2 at.% Pb as compared to less than 0.1 at.% Pb at equilibrium. It was found that by suitable heat treatment it is possible to vary the size and distribution of the Pb precipitates in the Ag matrix and reproducible superconducting properties in the alloy can be observed. The superconducting transition temperature of these samples can be qualitatively explained by the Silvert and Singh's theoretical calculation. The theory developed for the case of layer structure can be extended to three dimensions to explain the critical current versus temperature behavior. The critical current versus field behavior of these alloys can be explained by the modification of the Josephson effect. Combining these results together with the critical magnetic field measurements and the microstructure studies of the alloys, it can be concluded that the three-dimensional proximity effect is the main mechanism for the superconductivity in the Ag-Pb alloys. Based on the Hilsch empirical formula which was based on experimental results obtained on layer structures, the experimental data in this investigation show that the electron-phonon-electron interaction in silver is attractive. The interaction parameter NV obtained is approximately 0.06, which would lead to a value of 10-5 °K for the superconducting transition temperature of Ag. These values are in agreement with other determinations which were done on vapor-deposited metallic film sandwiches. Hence, the Hilsch empirical relation valid for layer structures is also valid in the three-dimensional case. Because the transition temperature and the critical current can be varied in a wide range by controlling the heat treatments, the Ag-Pb superconductors might have some useful applications.

A lo largo de este trabajo, realizaremos un diagnóstico de las relaciones con clientes y de las estrategias de marketing en mercados industriales. Para ello, dividimos el trabajo en dos grandes partes y lo cerramos con unas conclusiones globales. La primera parte, el marco teórico, se ha elaborado tras la revisión crítica de la bibliografía referenciada al final del trabajo y el conocimiento adquirido en las sesiones magistrales de la asignatura Marketing Industrial de 4º curso del Grado en Marketing, impartida por la profesora Mª Soledad Aguirre, a las que asistí como oyente puesto que yo no soy alumno de dicho grado. En la segunda parte, el estudio empírico, realizamos un análisis de nueve empresas y organizaciones vascas (a excepción de una de ellas) que mantienen relaciones comerciales esencialmente del tipo Business to Business, que es lo que en la bibliografía que hemos trabajado se entiende por relaciones comerciales propias de Mercados Industriales. No se habla tanto de productos industriales como de clientes industriales o empresariales (relaciones empresa-empresa). Para ello, hemos utilizado como método de análisis y contraste el Método del Caso. Por último, concluimos el trabajo con la enumeración de las principales conclusiones extraídas de nuestra investigación y con la relación de la Bibliografía y Webgrafía consultada.

We measured the differential cross section of the process γp → pƞ at the 1.5 GeV Caltech electron synchrotron, at photon energies from 0.8 to 1.45 GeV, at various angles between 45° and 100° in the center of mass. A counter-spark chamber array was used to determine the kinematics of all particles in the final state of the partial mode γp → pƞ (ƞ → 2γ). Analysis of 40,000 pictures yielded 6,000 events above a background which varied with energy from 5% to 30% of foreground. The cross section shows an energy dependence confirming earlier results up to 1000 MeV, but with improved statistics; it then remains roughly constant (at 50° C.M.), to 1.45 GeV. The data show a small angular variation, within the limited range covered, at energies between 1000 and 1100 MeV.

Este trabalho de pesquisa apresenta a festa do divino espírito santo açoriana, realizada em uma Devoção Particular (Irmandade) localizada no bairro do Encantado, zona norte da cidade do Rio de Janeiro. A Festa do Divino é uma festa popular, católica e originária da Península Ibérica que vem sendo realizada, há alguns séculos, em várias partes do mundo, entre elas o Brasil. O objetivo principal do estudo é apresentar, a partir do relato compartilhado com a própria comunidade açoriana, nossos nativos, as várias dimensões constituintes da festa, especificamente, de como ela vem sendo elaborada de forma original no subúrbio do Rio de Janeiro. Sendo a festa uma forma de reafirmação étnica em contextos transnacionais, como definido através da categoria açorianidade criada na década de 30, seus significados (da tradição) vão sendo extrapolados quando diluídos em contextos culturais transnacionais. No Rio de Janeiro tanto a cultura, como a religiosidade popular (evidenciadas através da festa), tornaram-se acessos importantes para o contato social entre açorianos e brasileiros. A irmandade açoriana do Encantado estabeleceu um diálogo ao longo de oito décadas com os moradores locais através da festa, bem marcada no calendário entre os meses de maio e junho. Constituindo-se como um verdadeiro sistema de prestações totais, um espaço privilegiado e complexo de trocas sociais, possibilitou-nos através dos dados etnográficos da pesquisa empírica uma janela para análise de temas importantes das Ciências Sociais como devoção religiosa, trabalho, cultura, tradição e espaço.

考虑到空间滤波器系统在激光装置中的稳定性要求,分析了影响稳定性的各种因素。对现行装置中的SF4空间滤波器系统进行振动模态测试,初步确定系统的工作状况,并结合振动理论明确系统稳定性的设计参数。利用有限元软件ANSYS对空间滤波器系统及相关的设计进行仿真验证计算。结果表明:SF4系统的一阶固有频率达到83Hz,与实验结果相符合,满足系统稳定性的要求;在系统中引入支撑不但能有效增加系统的刚度,同时能转移系统的振型;系统基础支撑平台的选择要求平台独立,并且选择较低的高度以便于控制上层光学元件的变形;在系统中的引入

No presente trabalho, buscou-se identificar os conflitos de interesses existentes nos três níveis do poder executivo que dificultam a gestão integrada de políticas públicas voltadas para a recuperação ambiental dos rios urbanos, considerando-se, como área de estudo, o Município de São Gonçalo (RJ). A pesquisa exploratória consistiu em revisão bibliográfica e trabalho empírico. No referencial teórico de análise, abordou-se a formação do Estado brasileiro, as políticas públicas e as relações de poder entre os agentes na tomada de decisão e na administração da coisa pública. Através de revisão na literatura, levantou--se o contexto histórico da ocupação e da degradação dos rios de São Gonçalo e da Baixada Fluminense, os planos e projetos para o setor de saneamento da região, bem como se empreendeu a compilação das normas legais pertinentes à gestão do território, aos recursos hídricos e ao saneamento. O trabalho de campo consistiu em levantamento de dados sobre a degradação das bacias hidrográficas no Município de São Gonçalo e das inter-relações existentes nas políticas públicas referentes à Região Hidrográfica da Bacia da Baía de Guanabara, onde o município em comento se insere. O estudo do caso evidenciou que a degradação dos rios urbanos é resultante, entre outros fatores, da falta de articulação das três esferas de governo, da descontinuidade das ações públicas, de interesses políticos e financeiros desarticulados das demandas socioambientais, da falta de infraestrutura técnica e financeira dos municípios, da pouca participação social no planejamento e tomada de decisões face às relações desiguais de poder, além da inconsistência e desarticulação dos planos e projetos governamentais, a exemplo dos planos diretores de uso ocupação do solo. Conclui-se ser fundamental o fortalecimento dos comitês de bacia e das instituições que os integram, possibilitando a articulação entre as políticas públicas municipais e as do governo estadual e federal, em relação às condicionantes ambientais, bem como ao uso do solo e ao saneamento. A solução dos problemas relativos aos rios urbanos só será possível através da gestão integrada e participativa, envolvendo efetivamente os diferentes setores usuários da bacia hidrográfica. O controle social das ações é imprescindível para manter a coerência, a efetividade, a eficácia e a continuidade dos planos, projetos e políticas do Estado. Assim, poderá ser contido o processo contínuo de degradação ambiental, em particular, dos recursos hídricos em bacias urbanas como acontece no Município de São Gonçalo e em diversos outros municípios brasileiros. Espera-se que esse estudo contribua para o aprimoramento do conhecimento e de soluções no que concerne à morte iminente dos rios urbanos do país.

Tradicionalmente, na análise e dimensionamento de estruturas de aço, assume-se que as ligações viga-coluna são rígidas ou flexíveis (rotuladas). Por outro lado, é de conhecimento geral que a grande maioria das ligações viga-coluna apresenta um comportamento intermediário, ou seja, semi-rígido. Inúmeros trabalhos de pesquisa têm sido desenvolvidos nos últimos vinte e cinco anos, de forma a estudar o comportamento desse tipo de ligação. Um dos principais objetivos desta investigação é o de propor uma metodologia de análise que represente de forma apropriada a influência do comportamento semi-rígido de placas de base e de ligações viga-coluna, sobre a resposta dinâmica (linear e não-linear) de estruturas de aço. Outra contribuição desta dissertação diz respeito à investigação do comportamento dinâmico (linear e não-linear) de pórticos de aço, a partir da consideração de ligações viga-coluna simétricas e não-simétricas e especialmente das placas de base. A análise estrutural é desenvolvida com base no emprego do programa de elementos finitos ANSYS [27]. Nos modelos em elementos finitos foram considerados os efeitos de não-linearidade geométrica (efeitos de segunda ordem), o comportamento não-linear das placas de base e das ligações viga-coluna e, bem como, o efeito de histerese que ocorre quando a estrutura é submetida a cargas cíclicas. Os resultados alcançados indicaram que o fenômeno físico da ressonância não ocorre no que se refere à resposta dinâmica dos modelos semi-rígidos não-lineares. A ressonância não ocorre na resposta dos modelos devido ao fato de que, na análise dinâmica não-linear, o efeito de histerese presente nas ligações (placas de base e viga-coluna), essencialmente com comportamento não-linear, provoca um amortecimento na resposta dinâmica da estrutura.

The von Bertalanffy growth function is used for length based analysis of growth and mortality patterns for management of fisheries. However, certain fish have growth patterns that the VBGF may not be able to describe adequately.e.g. the Acanthurus lineatus in Samoa. In such cases a two phase VBGF may be a useful approach.

Emergent properties of global political culture were examined using data from the World History Survey (WHS) involving 6,902 university students in 37 countries evaluating 40 figures from world history. Multidimensional scaling and factor analysis techniques found only limited forms of universality in evaluations across Western, Catholic/Orthodox, Muslim, and Asian country clusters. The highest consensus across cultures involved scientific innovators, with Einstein having the most positive evaluation overall. Peaceful humanitarians like Mother Theresa and Gandhi followed. There was much less cross-cultural consistency in the evaluation of negative figures, led by Hitler, Osama bin Laden, and Saddam Hussein. After more traditional empirical methods (e.g., factor analysis) failed to identify meaningful cross-cultural patterns, Latent Profile Analysis (LPA) was used to identify four global representational profiles: Secular and Religious Idealists were overwhelmingly prevalent in Christian countries, and Political Realists were common in Muslim and Asian countries. We discuss possible consequences and interpretations of these different representational profiles.

William Francis Thompson (1888–1965) was a preeminent fishery scientist of the early to mid twentieth century. Educated at Stanford University in California (B.A. 1911, Ph.D. 1930), Thompson conducted pioneering research on the Pacific halibut, Hippoglossus stenolepis, from 1914 to 1917 for the British Columbia Provincial Fisheries Department. He then directed marine fisheries research for the State of California from 1917 to 1924, was Director of Investigations for the International Fisheries Commission from 1924 to 1939, and Director of the International Pacific Salmon Fisheries Commission from 1937 to 1942. He was also Director of the School of Fisheries, University of Washing-ton, Seattle, from 1930 to 1947. Thompson was the founding director in 1947 of the Fisheries Research Institute at the University of Washington and served in that capacity until his retirement in 1958. He was a dominant figure in fisheries research of the Pacific Northwest and influenced a succession of fishery scientists with his yield-based analysis of fishery stocks, as opposed to studying the fishes’environment. Will Thompson was also a major figure in education, and many of his former students attained leadership positions in fisheries research and administration.

William Francis Thompson (1888–1965), an early fishery biologist, joined the California Fish and Game Commission in 1917 with a mandate to investigate the marine fisheries of the state. He initiated studies on the albacore tuna, Thunnus alalunga, and the Pacific sardine, Sardinops sagax, as well as studies on other economically important marine organisms. Thompson built up a staff of fishery scientists, many of whom later attained considerable renown in their field, and he helped develop, and then direct, the commission’s first marine fisheries laboratory. During his tenure in California, he developed a personal philosophy of research that he outlined in several publications. Thompson based his approach on the yield-based analysis of the fisheries as opposed to large-scale environmental studies. He left the state agency in 1925 to direct the newly formed International Fisheries Commission (now the International Pacific Halibut Commission). William Thompson became a major figure in fisheries research in the United States, and particularly in the Pacific Northwest and Alaska, during the first half of the 20th cent

A closed-loop control technique based on monitoring phase current risetime for switched reluctance (SR) motors without direct rotor-position sensors has been studied and implemented successfully. In this technique the variation in incremental phase inductance in a SR motor is used to detect rotor position. A control circuit for current-waveform-based rotor position detection has been implemented using hard-wire digital circuits. Torque-speed and system-efficiency characteristics resulting from the application of the method to a 4-kW, four-phase SR motor with an IGBT drive are presented.
