O objetivo deste trabalho é o desenvolvimento de frameworks de testes automáticos de software. Este tipo de testes normalmente está associado ao modelo evolucionário e às metodologias ágeis de desenvolvimento de software, enquanto que os testes manuais estão relacionados com o modelo em cascata e as metodologias tradicionais. Como tal foi efetuado um estudo comparativo sobre os tipos de metodologias e de testes existentes, para decidir quais os que melhor se adequavam ao projeto e dar resposta à questão "Será que realmente compensa realizar testes (automáticos)?". Finalizado o estudo foram desenvolvidas duas frameworks, a primeira para a implementação de testes funcionais e unitários sem dependências a ser utilizada pelos estagiários curriculares da LabOrders, e a segunda para a implementação de testes unitários com dependências externas de base de dados e serviços, a ser utilizada pelos funcionários da empresa. Nas últimas duas décadas as metodologias ágeis de desenvolvimento de software não pararam de evoluir, no entanto as ferramentas de automação não conseguiram acompanhar este progresso. Muitas áreas não são abrangidas pelos testes e por isso alguns têm de ser feitos manualmente. Posto isto foram criadas várias funcionalidades inovadoras para aumentar a cobertura dos testes e tornar as frameworks o mais intuitivas possível, nomeadamente: 1. Download automático de ficheiros através do Internet Explorer 9 (e versões mais recentes). 2. Análise do conteúdo de ficheiros .pdf (através dos testes). 3. Obtenção de elementos web e respetivos atributos através de código jQuery utilizando a API WebDriver com PHP bindings. 4. Exibição de mensagens de erro personalizadas quando não é possível encontrar um determinado elemento. As frameworks implementadas estão também preparadas para a criação de outros testes (de carga, integração, regressão) que possam vir a ser necessários no futuro. Foram testadas em contexto de trabalho pelos colaboradores e clientes da empresa onde foi realizado o projeto de mestrado e os resultados permitiram concluir que a adoção de uma metodologia de desenvolvimento de software com testes automáticos pode aumentar a produtividade, reduzir as falhas e potenciar o cumprimento de orçamentos e prazos dos projetos das organizações.

OBJECTIVE: Local heating increases skin blood flow SkBF (thermal hyperemia). In a previous study, we reported that a first local thermal stimulus could attenuate the hyperemic response to a second one applied later on the same skin spot, a phenomenon that we termed desensitization. However, other studies found no evidence for desensitization in similar conditions. The aim of the present work was to test whether it was related to differences in instrumentation. METHODS: Twenty-eight healthy young males were studied. Two pairs of heating chambers, one custom-made (our study) and one commercial (other groups), were affixed to forearm skin. SkBF was measured with single-point laser-Doppler flowmetry (LDF) (780nm) in one pair, and laser-Doppler imaging (LDI) (633nm) in the other. A temperature step from 34 to 41°C, was applied for 30minutes and repeated after two hours. RESULTS: During the second thermal challenge, the plateau SkBF was lower than during the first thermal and was observed with each of the four combinations of SkBF measurement techniques and heating equipment (p<0.05 for all conditions, range -9% to -16% of the initial value). CONCLUSION: Desensitization of thermal hyperemia is not specific to peculiar operating conditions.

BACKGROUND & AIMS: Hepatitis C virus (HCV) induces chronic infection in 50% to 80% of infected persons; approximately 50% of these do not respond to therapy. We performed a genome-wide association study to screen for host genetic determinants of HCV persistence and response to therapy. METHODS: The analysis included 1362 individuals: 1015 with chronic hepatitis C and 347 who spontaneously cleared the virus (448 were coinfected with human immunodeficiency virus [HIV]). Responses to pegylated interferon alfa and ribavirin were assessed in 465 individuals. Associations between more than 500,000 single nucleotide polymorphisms (SNPs) and outcomes were assessed by multivariate logistic regression. RESULTS: Chronic hepatitis C was associated with SNPs in the IL28B locus, which encodes the antiviral cytokine interferon lambda. The rs8099917 minor allele was associated with progression to chronic HCV infection (odds ratio [OR], 2.31; 95% confidence interval [CI], 1.74-3.06; P = 6.07 x 10(-9)). The association was observed in HCV mono-infected (OR, 2.49; 95% CI, 1.64-3.79; P = 1.96 x 10(-5)) and HCV/HIV coinfected individuals (OR, 2.16; 95% CI, 1.47-3.18; P = 8.24 x 10(-5)). rs8099917 was also associated with failure to respond to therapy (OR, 5.19; 95% CI, 2.90-9.30; P = 3.11 x 10(-8)), with the strongest effects in patients with HCV genotype 1 or 4. This risk allele was identified in 24% of individuals with spontaneous HCV clearance, 32% of chronically infected patients who responded to therapy, and 58% who did not respond (P = 3.2 x 10(-10)). Resequencing of IL28B identified distinct haplotypes that were associated with the clinical phenotype. CONCLUSIONS: The association of the IL28B locus with natural and treatment-associated control of HCV indicates the importance of innate immunity and interferon lambda in the pathogenesis of HCV infection.

We assessed the effect of abandonment of sylvo-pastoral practices in chestnut orchards (Castanea sativa) on bats in southern Switzerland to determine practical recommendations for bat conservation. We compared bat species richness and foraging activities between traditionally managed and unmanaged chestnut orchards, testing the hypothesis that managed orchards provide better foraging opportunities and harbour more bat species. Echolocation calls of foraging bats were sampled simultaneously at paired sites of managed and unmanaged orchards using custom made recorders. Vegetation structure and aerial insect availability were sampled at the recording sites and used as explanatory variables in the model. In a paired sampling design, we found twice the number of bat species (12) and five times higher total foraging activity in the managed chestnut orchards compared to the unmanaged ones. Bat species with low flight manoeuvrability were 14 times more common in managed orchards, whereas bats with medium to high manoeuvrability were only 5 times more common than in abandoned orchards. The vegetation structure was less dense in managed orchards. However, management did not affect relative insect abundance. Bats primarily visited the most open orchards, free of undergrowth. As a result of restricted access into the overgrown forests, the abandonment of chestnut orchards leads to a decline in bat species richness and foraging activities. Continued management of chestnut orchards to maintain an open structure is important for the conservation of endangered bat species in the southern Swiss Alps.

Disease characteristics. Perry syndrome is characterized by parkinsonism, hypoventilation, depression, and weight loss. The mean age at onset is 48 years; the mean disease duration is five years. Parkinsonism and psychiatric changes (depression, apathy, character changes, and withdrawal) tend to occur early; severe weight loss and hypoventilation manifest later. Diagnosis/testing. The diagnosis is based on clinical findings and molecular genetic testing of DCTN1, the only gene known to be associated with Perry syndrome. Management. Treatment of manifestations: Dopaminergic therapy (particularly levodopa/carbidopa) should be considered in all individuals with significant parkinsonism. Although response to levodopa is often poor, some individuals may have long-term benefit. Noninvasive or invasive ventilation support may improve quality of life and prolong life expectancy. Those patients with psychiatric manifestations may benefit from antidepressants and psychiatric care. Weight loss is managed with appropriate dietary changes. Surveillance: routine evaluation of weight and calorie intake, respiratory function (particularly at night or during sleep), strength; and mood. Agents/circumstances to avoid: Central respiratory depressants (e.g., benzodiazepines, alcohol). Genetic counseling. Perry syndrome is inherited in an autosomal dominant manner. The proportion of cases attributed to de novo mutations is unknown. Each child of an individual with Perry syndrome has a 50% chance of inheriting the mutation. No laboratories offering molecular genetic testing for prenatal diagnosis are listed in the GeneTests Laboratory Directory; however, prenatal testing may be available through laboratories offering custom prenatal testing for families in which the disease-causing mutation has been identified.

The antigen-presenting cell-expressed CD40 is implied in the regulation of counteractive immune responses such as induction of pro-inflammatory and anti-inflammatory cytokines interleukin (IL)-12 and IL-10, respectively. The mechanism of this duality in CD40 function remains unknown. Here, we investigated whether such duality depends on ligand binding. Based on CD40 binding, we identifed two dodecameric peptides, peptide-7 and peptide-19, from the phage peptide library. Peptide-7 induces IL-10 and increases Leishmania donovani infection in macrophages, whereas peptide-19 induces IL-12 and reduces L. donovani infection. CD40-peptide interaction analyses by surface plasmon resonance and atomic force microscopy suggest that the functional differences are not associated with the studied interaction parameters. The molecular dynamic simulation of the CD40-peptides interaction suggests that these two peptides bind to two different places on CD40. Thus, we suggest for the first time that differential binding of the ligands imparts functional duality to CD40.

BACKGROUND:: Mechanical stretch has been shown to induce vascular remodeling and increase vessel density, but the pathophysiologic mechanisms and the morphologic changes induced by tensile forces to dermal vessels are poorly understood. METHODS:: A custom computer-controlled stretch device was designed and applied to the backs of C57BL/6 mice (n = 38). Dermal and vascular remodeling was studied over a 7-day period. Corrosion casting and three-dimensional scanning electron microscopy and CD31 staining were performed to analyze microvessel morphology. Hypoxia was assessed by immunohistochemistry. Western blot analysis of vascular endothelial growth factor (VEGF) and mRNA expression of VEGF receptors was performed. RESULTS:: Skin stretching was associated with increased angiogenesis as demonstrated by CD31 staining and vessel corrosion casting where intervascular distance and vessel diameter were decreased (p < 0.01). Immediately after stretching, VEGF dimers were increased. Messenger RNA expression of VEGF receptor 1, VEGF receptor 2, neuropilin 1, and neuropilin 2 was increased starting as early as 2 hours after stretching. Highly proliferating epidermal cells induced epidermal hypoxia starting at day 3 (p < 0.01). CONCLUSIONS:: Identification of significant hypoxic cells occurred after identification of neovessels, suggesting an alternative mechanism. Increased expression of angiogenic receptors and stabilization of VEGF dimers may be involved in a mechanotransductive, prehypoxic induction of neovascularization.

Langattomien lähiverkkojen yleistyessä nopeasti suurten verkkojen teknologiana käyttösääntöjen valvonta tulee tarpeelliseksi. Tässä työssä kuvataan, kuinka käyttäjät voidaan pakottaa noudattamaan käyttösääntöjä julkisissa WLAN-verkoissa. Työssä käsiteltävät ongelmat koskevat menetelmiä epäluotettavien DHCP-palvelinten paljastamiseksi sekä omia IP-osoitteita käyttävien käyttäjien paljastamiseksi tilanteissa, jolloin IP-osoite ei ole virallisen DHCP-palvelimen myöntämä. Jokaisen menetelmän kohdalla pohditaan, kuinka tällaisia käyttäjiä voidaan estää rikkomasta käyttösääntöjä. Lisäksi pohditaan keskitetyn tietojen keruun hyödyntämistä kuvattujen tehtävien suorittamiseksi. Esitetyt ratkaisut on erityisesti suunniteltu testiverkkoa varten, mutta yleiset ideat ovat toimivia missä tahansa langattomassa verkossa.

Utredningen "Servicenivå för Västra Nylands kollektivtrafik" är en fortsättning till Nylands ELY-centrals förstudie om bestämning av kollektivtrafikens servicenivå på Nylands ELY-centrals område, nr 29/2010. Enligt förstudiet blev definieringsarbetet för kollektivtrafikens servicenivå på Nylands ELY-centrals område delat upp för att förverkligas i sju helheter som baserar sig på regionbiljettområden. Det blev sju rapporter om definieringsarbetet av vilka denna som rör Västra Nyland är en. Utredningen är en del av processen där ELY-centralen i samarbete med kommuner, behöriga myndigheter, landskapsförbund och trafikidkare förberedde Nylands ELY-centrals bestämmelse över bestyrkning av kollektivtrafikens servicenivå enligt lagen om kollektivtrafik. ELY-centralen bestyrkte med sitt beslut i december 2011 den åsyftade servicenivån enligt utredningen för åren 2012 - 2016. I utredningen har man beskrivit både den nutida och den åsyftade servicenivån som sträcker sig till år 2016. I huvudsak har man gjort beskrivningen som förbindelsestreckanalys. De valda förbindelsestreckorna har klassificerats enligt trafikens kvantitativa och kvalitativa faktorer i sex klasser: konkurrensnivå, lockande nivå, medelnivå, grundservicenivå, miniminivå och lagstadgad nivå. Som viktigaste klassificeringsfaktorer användes trafikeringstid och turantal. I klassificeringen har Trafikverkets anvisning nr 15/2011 tillämpats.
