Lime Creek is a sub-watershed of the Cedar River above; approximately 25 miles from Cedar Rapids. The lower half of the stream is on the Iowa 2004 Section 303(d) impaired waters list. Monitoring by the Cedar River Watershed Monitoring Coalition documents that Lime Creek delivers above average amounts of nitrate+ nitrite-N, ammonia-Nand total phosphorus (above the 901 percentile) compared to other Cedar River sub-watersheds. The Cedar Rapids water utility is concerned about increasing delivery of nitrate+nitrate to the Cedar River, which provides drinking water for about 125,000 people in the area. A group of local citizens has formed the Lime Creek watershed council with the goal of reducing pollutant delivery to the creek and promoting sustainable, watershed-wide action by producers, urban and rural residents for improved environmental management. The council has established a performance-based program that rewards cooperators for improvement in research-based test and index scores which directly measure environmental impact of BMPs. The Iowa Com Growers Association is funding the performance rewards. The Watershed Coalition is contributing in-kind monitoring. Council and performance cooperators participate primarily with commitment of their own resources. WIRB funds will be used to increase program cooperators and for staff support. In addition to improvement of water quality in Lime Creek, the project will establish baseline values for arket-based a pro ch to valuing pollutant reduction by intensive livestock operations in eastern Iowa.

In cases of transjugular liver biopsies, the venous angle formed between the chosen hepatic vein and the vena cava main axis in a frontal plane can be large, leading to technical difficulties. In a prospective study including 139 consecutive patients who underwent transjugular liver biopsy using the Quick-Core biopsy set, the mean venous angle was equal to 49.6 degrees. For 21.1% of the patients, two attempts at hepatic venous catheterization failed because the venous angle was too large, with a mean of 69.7 degrees. In all of these patients, manual reshaping of the distal curvature of the stiffening metallic cannula, by forming a new mean angle equal to 48 degrees , allowed successful completion of the procedure in less than 10 min.

Wiss, Janney, Elstner Associates, Inc. (WJE) evaluated potential nondestructive evaluation (NDE) methodologies that may be effective in 1) identifying internal defects within slip formed concrete barriers and 2) assessing the corrosion condition of barrier dowel bars. The evaluation was requested by the Bridge Maintenance and Inspection Unit of the Iowa Department of Transportation (IaDOT) and the Bureau of Bridges and Structures of the Illinois Department of Transportation (IDOT). The need arose due to instances in each Department’s existing inventory of bridge barriers where internal voids and other defects associated with slip forming construction methods were attributed to poor barrier performance after completion of construction and where, in other barrier walls, unintentional exposure of the dowel bars revealed extensive corrosion-related section loss at previously uninspectable locations, reducing the capacity of the barriers to resist traffic impact loads. WJE trial tested potential NDE techniques on laboratory mock-up samples built with known defects, trial sections of cast-in-place barriers at in-service bridges in Iowa, and slip formed and cast-in-place barrier walls at in-service bridges in Illinois. The work included review of available studies performed by others, field trial testing to assess candidate test methods, verification of the test methods in identifying internal anomalies and dowel bar corrosion, and preparation of this report and nondestructive evaluation guidelines.

PURPOSE: To study VP22 light controlled delivery of antisense oligonucleotide (ODN) to ocular cells in vitro and in vivo. METHODS: The C-terminal half of VP22 was expressed in Escherichia coli, purified and mixed with 20 mer phosphorothioate oligonucleotides (ODNs) to form light sensitive complex particles (vectosomes). Uptake of vectosomes and light induced redistribution of ODNs in human choroid melanoma cells (OCM-1) and in human retinal pigment epithelial cells (ARPE-19) were studied by confocal and electron microscopy. The effect of vectosomes formed with an antisense ODN corresponding to the 3'-untranslated region of the human c-raf kinase gene on the viability and the proliferation of OCM-1 cells was assessed before and after illumination. Cells incubated with vectosomes formed with a mismatched ODN, a free antisense ODN or a free mismatched ODN served as controls. White light transscleral illumination was carried out 24 h after the intravitreal injection of vectosomes in rat eyes. The distribution of fluorescent vectosomes and free fluorescent ODN was evaluated on cryosections by fluorescence microscopy before, and 1 h after illumination. RESULTS: Overnight incubation of human OCM-1 and ARPE-19 cells with vectosomes lead to intracellular internalization of the vectosomes. When not illuminated, internalized vectosomes remained stable within the cell cytoplasm. Disruption of vectosomes and release of the complexed ODN was induced by illumination of the cultures with a cold white light or a laser beam. In vitro, up to 60% inhibition of OCM-1 cell proliferation was observed in illuminated cultures incubated with vectosomes formed with antisense c-raf ODN. No inhibitory effect on the OCM-1 cell proliferation was observed in the absence of illumination or when the cells are incubated with a free antisense c-raf ODN and illuminated. In vivo, 24 h after intravitreal injection, vectosomes were observed within the various retinal layers accumulating in the cytoplasm of RPE cells. Transscleral illumination of the injected eyes with a cold white light induced disruption of the vectosomes and a preferential localization of the "released" ODNs within the cell nuclei of the ganglion cell layer, the inner nuclear layer and the RPE cells. CONCLUSIONS: In vitro, VP22 light controlled delivery of ODNs to ocular cells nuclei was feasible using white light or laser illumination. In vivo, a single intravitreal injection of vectosomes, followed by transscleral illumination allowed for the delivery of free ODNs to retinal and RPE cells.

Iowa's secondary roads contain nearly 15,000 bridges which are less than 40 feet (12 m) in length. Many of these bridges were built several decades ago and need to be replaced. Box culvert construction has proven to be an adequate bridge replacement technique. Recently a new bridge replacement alternative, called the Air-0-Form method, has emerged which has several potential advantages over box culvert construction. This new technique uses inflated balloons as the interior form in the construction of an arch culvert. The objective of research project HR-314 was to construct an air formed arch culvert to determine the applicability of the Air-O-Form technique as a county bridge replacement alternative.

There are still many vintage portland cement concrete (PCC) pavements, 18 ft wide (5.4 m), dating back to pre-World War II era in use today. Successive overlays have been placed to cover joints and to improve rideability. The average thickness of the existing asphalt cement concrete (ACC) along route E66 in Tama County, Iowa, was 6.13 in. (15.6 cm). The rehabilitation strategy called for widening the base using the top 3 in. (7.6 cm) of the existing ACC by a recycling process involving cold milling and mixing with additional emulsion/rejuvenator. The material was then placed into a widening trench and compacted to match the level of the milled surface. This project was undertaken to develop a rehabilitation methodology to widen these older pavements economically and to have a finished surface capable of carrying traffic with little or no additional work.

The object of this research project is to construct and evaluate several bituminous concrete base overlays with various high penetration bituminous products which have the potential to reduce future maintenance and construction costs. In September of 1981 nine different bituminous test sections were constructed on Osceola County roads A-34 and A-46 east of Ashton. Asphalt binders used were AC-5 asphalt cement, SC-800 liquid asphalt, MC-3000 cutback asphalt, CSS-1 emulsion and HFMS-2 emulsion. Aggregate for four of the test sections consisted of salvaged bituminous pavement and new aggregate combined. Each section was road mixed using a different binder arid placed 2-1/4" thick with a motor patrol. The other five test sections consisted of an AC-5 Type B asphalt concrete control section and four plant-mixed, cold-laid bituminous concrete overlay sections using new aggregate. The material was mixed in a continuous drum mix plant and laid with a full width paver.
