The unicellular amoeba Dictyostelium discoideum embarks on a developmental program upon starvation. During development, extracellular oscillatory cAMP signaling orchestrates the chemotaxis-mediated aggregation of ∼105 amoebae and is required for optimal induction of so-called pulse-induced genes. This requirement for pulsatile CAMP reflects adaptation of the cAMP-receptor-mediated pathways that regulate these genes. Through examination of a collection of pulse-induced genes, we defined two distinct gene classes based on their induction kinetics and the impact of mutations that impair PKA signaling. The first class (represented by D2 and prtA) is highly dependent on PKA signaling, whereas the second class (represented by carA, gpaB, and acaA) is not. Analysis of expression kinetics revealed that these classes are sequentially expressed with the PKA-independent genes peaking in expression before the PKA-dependent class. Experiments with cycloheximide, an inhibitor of translation, demonstrated that the pulse induction of both classes depends on new protein synthesis early in development. carA and gpaB also exhibit pulse-independent, starvation-induced expression which, unlike their pulse induction, was found to be insensitive to cycloheximide added at the outset of starvation. This result indicates that the mechanism of starvation induction pre-exists in growing cells and is distinct from the pulse induction mechanism for these genes. In order to identify cis-acting elements that are critical for induction of carA, we constructed a GFP reporter controlled by a 914-base-pair portion of its promoter and verified that its expression was PKA-independent, pulse-inducible, and developmentally regulated like the endogenous carA gene. By a combination of truncation, internal deletion, and site-directed mutation, we defined several distinct functional elements within the carA promoter, including a 39-bp region required for pulse induction between base pairs -321 and -282 (relative to the transcription start site), a 131-bp region proximal to the start site that is sufficient for starvation induction, and two separate enhancer domains. Identification of factors that interact with these promoter elements and genetic approaches exploiting the GFP reporter described here should help complete our understanding of the mechanisms regulating these genes, including adaptation mechanisms that likely also govern chemotaxis of Dictyostelium and mammalian cells. ^

Chronic inflammation is an established risk factor in the pathogenesis of many cancers. Pancreatic ductal adenocarcinoma, a malignancy with a particularly dismal prognosis, is no exception. Cyclooxygenase-2, a key enzyme induced by tissue injury, has a critical role in the generation of bioactive lipids known as prostaglandins. COX-2 overexpression is a frequent finding in pancreatic cancer, chronic pancreatitis and pancreatic intraepithelial neoplasias. To explore mechanisms through which chronic inflammation establishes and maintains a protumorigenic environment, we designed a mouse model overexpressing COX-2 in pancreatic parenchyma (BK5.COX-2 mice). We discovered that constitutive expression of COX-2 has a number of important sequelae, including upregulation of additional eicosanoid-generating enzymes and proinflammatory cytokines. Many of these molecular alterations precede the onset of significant histopathological changes. Increased levels of prostaglandins E2, D2, and F2α, 5-, 12-, and 15-hydroxyeiosatetraenoic acid (HETEs) were documented in tumors and pancreata of younger transgenic mice. Using a TaqMan™ Mouse Immune Panel, we detected elevated mRNAs for a number of proinflammatory cytokines (e.g., TNFα, IL-1β, IL-6). ^ Histological examination revealed early changes in the pancreas with similarities to human chronic pancreatitis, including loss of acinar cells, appearance of metaplastic ducts, and increased deposition of stroma. As the lesions progress, features typical of dysplastic and neoplastic cells emerged within the metaplastic ductal complexes, including cellular and nuclear atypia, crowding of cells, and loss of normal tissue architecture. The amount of fibroinflammatory stroma increased considerably; numerous small vessels were evident. A number of immunocytes from both the myeloid and lymphoid lineages were identified in transgenic pancreata. Neutrophils were the earliest to infiltrate, followed shortly by macrophages and mast cells. B and T cells generally began to appear by 8–12 weeks, and organized aggregates of lymphoid cells were often found in advanced lesions. ^ We tested the efficacy of several chemopreventive agents in this model, including celecoxib, a COX-2 selective inhibitor, pentoxifylline, a cytokine inhibitor, curcumin, a polyphenol with antioxidant and anti-inflammatory properties, and GW2974, a dual EGFR/ErbB2 inhibitor. Effects on lesion development were modest in the GW2974 and pentoxifylline treated groups, but significant prevention effects were observed with curcumin and celecoxib. ^

El 17 de mayo de 1984, dos personas de la Conadep entraron a la cárcel de Mendoza a tomar un testimonio clave. Habían recibido una llamada pidiendo su presencia, pero de la Conadep no sabían que encontrarían. Allí conocen a Carmello Sivera Paredes, un ex-suboficial de la Policía Federal preso por los delitos cometidos antes y durante la dictadura. En su declaración, Paredes detalla su actividad dentro de la institución, las torturas, los fusilamientos y el funcionamiento del D2. Este testimonio, se convierte en el primer caso en que un ex represor habla de lo sucedido.

The surface distributions of dissolved silicic acid, chlorophyll and diatom abundance were measured in the plume of the Mississippi River and adjacent waters during spring (late April and early May 1993) and summer (July 1992). In spring, the time of maximum river flow, there was an intense diatom bloom with a mean diatom abundance of 1.5 x 10**7 cells/l, more than an order of magnitude higher than in summer. Mixing curves of silicic acid concentration ([Si(OH)4]) versus salinity indicate that biological uptake within the river plume removed >99% of the Si(OH)4 supplied by the river in spring and 80 to 95% in summer. In spring [Si(OH)4] was occasionally depleted to <0.2 µM-among the lowest values ever reported from the ocean-with extensive depletion to >=0.5 µM over the shelf. In summer [Si(OH)4] was less severely depleted; the lowest measured was 0.93 µM and all others were >=2.4 µM. 30Si kinetic experiments were performed during both spring and summer to measure the degree to which the rate of Si uptake by the natural diatom assemblages was limited in situ by substrate availability. In spring the dependence of the specific uptake rate (V) on extracellular [Si(OH)4] conformed much more closely to the Michaelis-Menten saturation function than has been observed in past studies. Strong dependence of V on [Si(OH)4] was observed throughout the most Si(OH)4-depleted (<0.5 µM) region, where V was limited to 12 to 45% of the diatom assemblages' maximum uptake rate (Vmax). Half-saturation concentrations for Si uptake (Ks) averaged 0.85 uM (range = 0.48 to 1.71; n = 7) in spring, with the lowest values equal to the lowest previously reported for natural diatom assemblages. There was only 1 station in summer where V was limited by [Si(OH)4], and at that station Ks was 5.3 µM-quite high in comparison with previous studies. At stations where V was limited by [Si(OH)4], in both spring and summer, Chaetoceros spp. were numerically dominant; where there was no Si limitation other diatoms, usually Skeletonema costatum, dominated. The data thus indicate strong Si limitation in spring, with diatom assemblages well adapted to low [Si(OH)4], but little or no Si limitation in summer. Historical data suggest that coastal Si(OH)4 depletion and Si limitation may be recent phenomena in the northern Gulf of Mexico, resulting from increasing [NO3-] and decreasing [Si(OH)4] in the Mississippi River during the past 30 to 50 yr.

Microscopic and electron probe examination of some manganese nodules show that they consist of segregations of manganese-iron oxides in an interstitial material almost free of manganese but rich in iron and silicates. The segregations are widely spaced in the volcanic cores of the nodules but become more abundant towards their outer crusts where they form the centres of linked polygons of interstitial materials. Most of the minor elements are concentrated in the segregations compared to the interstitial materials. It is suggested that the structures observed result partly from solution and reprecipitation of elements in the original volcanic cores of the nodules and partly from the replacement and coating of these cores by manganese-iron oxides precipitated from sea water.

The paper is devoted to a marine geophysical-geological research in the West Antarctic. This researche contributed to establishing the base geodesic network of the West Antarctic and supplemented geokinematic monitoring based on this network with geophysical and geologic information on structure and features of geomorphological and tectonic development of the South Ocean floor. Collected materials allow to conclude about the inhomogeneity of the Scotia Sea floor and about combination of fragments of a continental massif with young rift structures in conditions of the upwelling mantle. The ancient continental bridge, faunal connections between the South America and the West Antarctic has been destroyed by processes of destruction, taphrogeny and sea floor spreading. Structures of the Scotia and Caribbean Seas, North Fiji and Arctic Basins are similar.

In 1995-1997 three oceanographic cruises to the White Sea were undertaken in the framework of the INTAS project 94-391, and a multi-disciplinary geochemical study of the major North Dvina estuary has been carried out. Distribution of temperature, salinity and concentration of suspended matter in water columm, as well as contents of Al, Fe, Mn, Co, Cu, Ni, Cr, Pb, Zn, and organic carbon contents in suspended matter and sediments of the North Dvina estuary were determined. Most of the metals and organic matter studied appear to be of terrestrial origin, since the main source of investigated elements in the estuary is river run-off. It was found that metals incorporated in minerals are absolutely prevailing forms in estuarine sediments, they comprise up to 60-99% of total metal contents. Two zones of metal accumulation in the sediments were found in the North Dvina estuary. These zones are considered as local geochemical barriers within a major river-sea barrier. Distribution of most elements studied in the sediments of the North Dvina estuary is controlled by grain size variability in the sediments. Analysis of data on heavy metal contents in the sediments and bivalves of the North Dvina estuary did not reveal any anthropogenic heavy metal pollution in the region.

Chinese scientists will start to drill a deep ice core at Kunlun station near Dome A in the near future. Recent work has predicted that Dome A is a location where ice older than 1 million years can be found. We model flow, temperature and the age of the ice by applying a three-dimensional, thermomechanically coupled full-Stokes model to a 70 × 70 km**2 domain around Kunlun station, using isotropic non-linear rheology and different prescribed anisotropic ice fabrics that vary the evolution from isotropic to single maximum at 1/3 or 2/3 depths. The variation in fabric is about as important as the uncertainties in geothermal heat flux in determining the vertical advection which in consequence controls both the basal temperature and the age profile. We find strongly variable basal ages across the domain since the ice varies greatly in thickness, and any basal melting effectively removes very old ice in the deepest parts of the subglacial valleys. Comparison with dated radar isochrones in the upper one third of the ice sheet cannot sufficiently constrain the age of the deeper ice, with uncertainties as large as 500 000 years in the basal age. We also assess basal age and thermal state sensitivities to geothermal heat flux and surface conditions. Despite expectations of modest changes in surface height over a glacial cycle at Dome A, even small variations in the evolution of surface conditions cause large variation in basal conditions, which is consistent with basal accretion features seen in radar surveys.

ZooScan with ZooProcess and Plankton Identifier (PkID) software is an integrated analysis system for acquisition and classification of digital zooplankton images from preserved zooplankton samples. Zooplankton samples are digitized by the ZooScan and processed by ZooProcess and PkID in order to detect, enumerate, measure and classify the digitized objects. Here we present a semi-automatic approach that entails automated classification of images followed by manual validation, which allows rapid and accurate classification of zooplankton and abiotic objects. We demonstrate this approach with a biweekly zooplankton time series from the Bay of Villefranche-sur-mer, France. The classification approach proposed here provides a practical compromise between a fully automatic method with varying degrees of bias and a manual but accurate classification of zooplankton. We also evaluate the appropriate number of images to include in digital learning sets and compare the accuracy of six classification algorithms. We evaluate the accuracy of the ZooScan for automated measurements of body size and present relationships between machine measures of size and C and N content of selected zooplankton taxa. We demonstrate that the ZooScan system can produce useful measures of zooplankton abundance, biomass and size spectra, for a variety of ecological studies.
