The regulation of carotenoid biosynthesis in a high-carotenoid-accumulating Fe’i group Musa cultivar, “Asupina”, has been examined and compared to that of a low-carotenoid-accumulating cultivar, “Cavendish”, to understand the molecular basis underlying carotenogenesis during banana fruit development. Comparisons in the accumulation of carotenoid species, expression of isoprenoid genes, and product sequestration are reported. Key differences between the cultivars include greater carotenoid cleavage dioxygenase 4 (CCD4) expression in “Cavendish” and the conversion of amyloplasts to chromoplasts during fruit ripening in “Asupina”. Chromoplast development coincided with a reduction in dry matter content and fruit firmness. Chromoplasts were not observed in “Cavendish” fruits. Such information should provide important insights for future developments in the biofortification and breeding of banana.

BACKGROUND: Earlier we reported that an oral administration of two mannose-specific dietary lectins, banana lectin (BL) and garlic lectin (GL), led to an enhancement of hematopoietic stem and progenitor cell (HSPC) pool in mice. STUDY DESIGN AND METHODS: Cord blood–derived CD34+ HSPCs were incubated with BL, GL, Dolichos lectin (DL), or artocarpin lectin (AL) for various time periods in a serum- and growth factor–free medium and were subjected to various functional assays. Reactive oxygen species (ROS) levels were detected by using DCHFDA method. Cell fractionation was carried out using lectin-coupled paramagnetic beads. RESULTS: CD34+ cells incubated with the lectins for 10 days gave rise to a significantly higher number of colonies compared to the controls, indicating that all four lectins possessed the capacity to protect HSPCs in vitro. Comparative analyses showed that the protective ability of BL and GL was better than AL and DL and, therefore, further experiments were carried out with them. The output of long-term culture-initiating cell (LTC-IC) and extended LTC-IC assays indicated that both BL and GL protected primitive stem cells up to 30 days. The cells incubated with BL or GL showed a substantial reduction in the ROS levels, indicating that these lectins protect the HSPCs via antioxidant mechanisms. The mononuclear cell fraction isolated by lectin-coupled beads got enriched for primitive HSPCs, as reflected in the output of phenotypic and functional assays. CONCLUSION: The data show that both BL and GL protect the primitive HSPCs in vitro and may also serve as cost-effective HSPC enrichment tools.

Tämän pro gradu-tutkielman tarkoitus on tutkia, miten reilun kaupan banaania tuottavan El Guabon pienten banaaninviljelijöiden järjestön Asoguabon reilun turismin projekti on rakennettu. Projektin muotoutumista tutkitaan tässä työssä kahdesta eri näkökulmasta: siitä, miksi ja miten Asoguabo on monipuolistanut viljelystä turismiin, sekä siitä, millainen kuva projektista on rakennettu markkinoinnin kautta ja miten reilun kaupan mainonnasta tuttuja ilmiöitä on hyödynnetty reilun turismin rakentamisessa. Tutkielman teoreettisena taustana on maaseudun muutoksia tarkasteleva uuden ruraliteetin käsite. Turismi on viime vuosikymmenten aikana muuttunut, ja matkailijat etsivät yhä enemmän aitoja ja autenttisia matkailukokemuksia. Samanaikaisesti turismia on tuotteistettu korostamalla sen tiettyjä, kestävän kehityksen mukaisia piirteitä, ja erityisesti kehittyvissä maissa turismia markkinoidaan usein vaihtoehtoisena, yhteisöpohjaisena tai ekoturismina. Reilu turismi on uutena käsitteenä tullut mukaan tähän laajaan kirjoon, ja tämä tutkielma käsitteleekin Asoguabon projektia nimenomaan reilun turismin näkökulmasta. Tämä tutkielma on tapaustutkimus Asoguabon turismiprojektista, ja pohjautuu kuukauden pituiseen kenttätyöhön Ecuadorissa tammikussa 2010 sekä kirjoittajan aiempiin kokemuksiin Asoguabosta. Aineisto koostuu 21 puoli-strukturoidusta laadullisesta haastattelusta kuudentoista informantin kanssa, joista suurin osa oli hyvin läheisesti tekemisissä projektin kanssa. Tämän lisäksi aineistonkeruussa on käytetty havainnointia sekä projektin markkinointimateriaalien sisällönanalyysia. Tutkielma osoittaa reilun turismin projektin sisältävän monia haasteita ennen kuin se voi saavuttaa tavoitteensa tuottaa lisätuloja Asoguabon toimintaan. Tutkimushetkellä projekti hyödytti suoraan pääasiassa muutamia järjestön viljelijöitä, lähinnä niitä, jotka toimivat projektissa oppaina. Nämä oppaat hyötyvät projektista saamalla pieniä lisätuloja, kasvattamalla sosiaalista pääomaansa, sekä saamalla mahdollisuuden oppia muun muassa osallistumalla kursseille. Tutkimuksen keskeiset tulokset osoittavat myös, kuinka vuorovaikutus-ongelmat projektin eri toimijoiden välillä vaikeuttavat tiedonkulkua ja täten reilun turismin toimintaa. Nämä ongelmat myös lisäävät epätietoisuutta Asoguabon muiden viljelijöiden parissa. Lisäksi tutkimus osoittaa, kuinka reilun turismin projektin mainonnassa käytetään osittain samoja keinoja kuin reilun kaupan tuotteiden mainonnassa, joskin tuottajia on kuvattu mainosmateriaaleissa yllättävän vähän ja pääosassa ovat usein eurooppalaiset turistit.

Context. Turbulent fluxes of angular momentum and heat due to rotationally affected convection play a key role in determining differential rotation of stars. Aims. We compute turbulent angular momentum and heat transport as functions of the rotation rate from stratified convection. We compare results from spherical and Cartesian models in the same parameter regime in order to study whether restricted geometry introduces artefacts into the results. Methods. We employ direct numerical simulations of turbulent convection in spherical and Cartesian geometries. In order to alleviate the computational cost in the spherical runs and to reach as high spatial resolution as possible, we model only parts of the latitude and longitude. The rotational influence, measured by the Coriolis number or inverse Rossby number, is varied from zero to roughly seven, which is the regime that is likely to be realised in the solar convection zone. Cartesian simulations are performed in overlapping parameter regimes. Results. For slow rotation we find that the radial and latitudinal turbulent angular momentum fluxes are directed inward and equatorward, respectively. In the rapid rotation regime the radial flux changes sign in accordance with earlier numerical results, but in contradiction with theory. The latitudinal flux remains mostly equatorward and develops a maximum close to the equator. In Cartesian simulations this peak can be explained by the strong 'banana cells'. Their effect in the spherical case does not appear to be as large. The latitudinal heat flux is mostly equatorward for slow rotation but changes sign for rapid rotation. Longitudinal heat flux is always in the retrograde direction. The rotation profiles vary from anti-solar (slow equator) for slow and intermediate rotation to solar-like (fast equator) for rapid rotation. The solar-like profiles are dominated by the Taylor-Proudman balance.

A 4 A electron-density map of Pf1 filamentous bacterial virus has been calculated from x-ray fiber diffraction data by using the maximum-entropy method. This method produces a map that is free of features due to noise in the data and enables incomplete isomorphous-derivative phase information to be supplemented by information about the nature of the solution. The map shows gently curved (banana-shaped) rods of density about 70 A long, oriented roughly parallel to the virion axis but slewing by about 1/6th turn while running from a radius of 28 A to one of 13 A. Within these rods, there is a helical periodicity with a pitch of 5 to 6 A. We interpret these rods to be the helical subunits of the virion. The position of strongly diffracted intensity on the x-ray fiber pattern shows that the basic helix of the virion is right handed and that neighboring nearly parallel protein helices cross one another in an unusual negative sense.

Dimeric banana lectin and calsepa, tetrameric artocarpin and octameric heltuba are mannose-specific beta-prism I fold lectins of nearly the same tertiary structure. MD simulations on individual subunits and the oligomers provide insights into the changes in the structure brought about in the protomers on oligomerization, including swapping of the N-terminal stretch in one instance. The regions that undergo changes also tend to exhibit dynamic flexibility during MD simulations. The internal symmetries of individual oligomers are substantially retained during the calculations. Energy minimization and simulations were also carried out on models using all possible oligomers by employing the four different protomers. The unique dimerization pattern observed in calsepa could be traced to unique substitutions in a peptide stretch involved in dimerization. The impossibility of a specific mode of oligomerization involving a particular protomer is often expressed in terms of unacceptable steric contacts or dissociation of the oligomer during simulations. The calculations also led to a rationale for the observation of a heltuba tetramer in solution although the lectin exists as an octamer in the crystal, in addition to providing insights into relations among evolution, oligomerization and ligand binding.

In this article we review the current status in the modelling of both thermotropic and lyotropic Liquid crystal. We discuss various coarse-graining schemes as well as simulation techniques such as Monte Carlo (MC) and Molecular dynamics (MD) simulations.In the area of MC simulations we discuss in detail the algorithm for simulating hard objects such as spherocylinders of various aspect ratios where excluded volume interaction enters in the simulation through overlap test. We use this technique to study the phase diagram, of a special class of thermotropic liquid crystals namely banana liquid crystals. Next we discuss a coarse-grain model of surfactant molecules and study the self-assembly of the surfactant oligomers using MD simulations. Finally we discuss an atomistically informed coarse-grained description of the lipid molecules used to study the gel to liquid crystalline phase transition in the lipid bilayer system.

beta-Prism I fold lectins constitute one of the five widely occurring structural classes of plant lectins. Each single domain subunit is made up of three Greek key motifs arranged in a threefold symmetric fashion. The threefold symmetry is not reflected in the sequence except in the case of the lectin from banana, a monocot, which carries two sugar-binding sites instead of the one in other lectins of known three-dimensional structure, all from dicots. This is believed to be a consequence of the different evolutionary paths followed by the lectin in monocots and dicots. The galactose-specific lectins among them have two chains produced by posttranslational proteolysis and contain three aromatic residues at the binding site. The extended binding sites of galactose- and mannose-specific lectins have been thoroughly characterized. Ligand binding at the sites involves both conformational selection and induced fit. Molecular plasticity of some of the lectins in the family has been characterized. The plasticity appears to be such as to promote variability in quaternary association which could be dimeric, tetrameric, or octameric. Structural and evolutionary reasons for the variability have been explored, and the relation of oligomerization to ligand binding and conformational selection investigated.

We evaluated the use of strip-transect survey methods for manatees through a series of replicate aerial surveys in the Banana River, Brevard County, Florida, during summer 1993 and summer 1994. Transect methods sample a representative portion of the total study area, thus allowing for statistical extrapolation to the total area. Other advantages of transect methods are less flight time and less cost than total coverage, ease of navigation, and reduced likelihood of double-counting. Our objectives were: (1) to identify visibility biases associated with the transect survey method and to adjust the counts accordingly; (2) to derive a population estimate with known variance for the Banana River during summer; and (3) to evaluate the potential value of this survey method for monitoring trends in manatee population size over time. In Nigeria, the culture of fish is gaining importance, but local fish farmers face a set back because of the stoppage on importation of fish feed. Locally available raw materials such as yam, plantain, banana, cowpeas, macuna, maize, cassava, millet, sorghum, groundnut, sunnhemp seed and brewery wastes are considered as potential materials for fish feed. These have been examined on their minimum protein contributions since this is the most expensive part of the fish feed. Alternative sources to animal proteins are also examined. Plant protein from groundnut, melon, mucuna and others compare favourably with bloodmeal mixture and thus can be used to replace the more expensive animal proteins. Pellet feed can be produced on a small scale or commercial basis from the locally available raw materials and the fish farmer is advised to seek assistance from qualified fisheries personnel

The authors present quantitative information on the shrimp resources of Sierra Leone waters. Four of the nine species present have been studied, of which Paenaeus duorarum notialis is dominant in the fishery. Synoptic surveys were undertaken in June 1977, and March 1978, to determine the abundance of the shrimp stock on the inshore shelf. The temperature-salinity-depth curves for the fishing ground show the existence of three water masses. The majority of fish caught were sciaenids, with some sparids also being taken. Detailed discussion of distribution and abundance of individual species of shrimp is given. The surveys have shown that the Banana Islands are the most productive shrimp grounds in the country, and the authors believe that they can support a viable shrimp industry for several years to come at present rates of exploitation

Atualmente observa-se uma expressiva perda de biodiversidade global resultante de atividades antrópicas, sendo a introdução de espécies exóticas uma das mais impactantes. A jaqueira Artocarpus heterophyllus é uma espécie exótica introduzida no Brasil durante o período colonial, sendo considerada invasora em diversas localidades. Na Mata Atlântica invade áreas de mata aberta e de borda, habitualmente associadas a ambientes antrópicos. Na Ilha Grande é encontrada em grande abundância em decorrência do histórico de ocupação humana. Para compreender como a mastofauna responde a presença da jaqueira, o Laboratório de Ecologia de Mamíferos da Universidade do Estado do Rio de Janeiro (UERJ) vem desenvolvendo um estudo ao longo de seis anos nos arredores da Vila Dois Rios, localizada na face oceânica da Ilha Grande. A partir dos resultados prévios iniciou-se uma segunda etapa do estudo no mesmo local que buscou avaliar diferentes métodos de controle das jaqueiras. O presente estudo é uma continuação direta desses dois trabalhos anteriores e teve como objetivo acompanhar as respostas da comunidade de pequenos mamíferos no período imediatamente posterior ao controle. Durante 18 meses foram amostradas bimestralmente 18 grades, 10 aonde foi efetuado o controle das jaqueiras e 8 aonde não foi constatada a presença desta árvore. Em cada grade foram colocadas 11 armadilhas de captura viva sendo banana a isca utilizada. Os mamíferos capturados foram medidos e suas fezes coletadas. A quantidade de jacas em cada área também foi anotada bimensalmente. As fezes foram analisadas em laboratório e as sementes encontradas identificadas. Os resultados obtidos indicam que a influência de A. heterophyllus sobre a estrutura da comunidade de pequenos mamíferos foi menor após o tratamento de controle. A única espécie que parece ainda responder a abundância de jaqueiras é o roedor Trinomys dimidiatus, que apresentou densidades mais elevadas nas áreas em tratamento, porém mais próximas a resultados obtidos para espécies congêneres em áreas pouco antropizadas. Utilizando uma abordagem de redes complexas observamos que, embora T. dimidiatus seja a espécie mais abundante em termos de número de indivíduos, o gambá Didelphis aurita parece ser a espécie de mamífero mais importante para dispersão de sementes nativas, aparecendo como espécie com maior número de conexões com espécies de sementes nas redes contruídas para as áreas sem jaqueiras e com jaqueiras antes e após o tratamento. Finalmente, a partir dos dados obtidos criamos um modelo matemático para a população de T. dimidiatus dos arredores da Vila Dois Rios, baseado em um crescimento logístico. Os resultados do modelo proposto se mostraram correlacionados com os dados de abundância reais, de modo que ele parece ser um simulador adequado da população local.

Os anfíbios anuros que habitam o bioma Mata Atlântica, especialmente em sua floresta ombrófila densa, são pouco conhecidos sob diversos aspectos de sua ecologia, existindo poucas informações disponíveis na literatura científica. Estes dados estão limitados a poucas localidades, geralmente estudos realizados em fragmentos remanescentes do sudeste brasileiro. A mata bem conservada que recobre os 3300 ha da Serra do Mendanha, apesar de localizada na região metropolitana do Estado do Rio de Janeiro, ainda não tinha sua anurofauna conhecida. No presente estudo, recolhemos informações ecológicas sobre as espécies que habitam a região, especialmente da assembléia encontrada no folhiço que recobre o solo, com o uso de diferentes metodologias: armadilhas de queda; parcelas cercadas; procura visual e auditiva em transecções e encontros ocasionais; assim como dados abióticos do ambiente regional (profundidade do folhiço; níveis de pH; taxa de oxigênio dissolvido; temperaturas do ar e da água; umidade do ar). Exemplares-testemunho foram fixados e depositados na coleção de anfíbios do Museu Nacional, Rio de Janeiro. A anurofauna da Serra do Mendanha é composta por pelo menos 44 espécies que estão distribuídas em 12 famílias, sendo a família Hylidae com o maior número de espécies (50%, n = 22), enquanto que Physalaemus signifer foi a espécie mais abundante (18%, n = 272), sendo habitante do folhiço. A espécie Rhinella ornata contribuiu com a maior biomassa (m = 548 g). Identificamos uma nova espécie de anuro (Brachycephalidae), que também habita o folhiço das cotas altimétricas acima de 700 m. Biogeograficamente a comunidade de anuros da área estudada indicou ser mais similar (68%) com a comunidade da região da Serra da Tiririca e arredores. Comparando-se os três tipo de fisionomias, ou mesohábitats, existentes na Serra do Mendanha, em termos de riqueza e diversidade, a floresta secundária indicou ter os mais elevados índices, seguido pela floresta pouco perturbada e pela monocultura de bananeiras. A assembléia de anuros que habita o folhiço da Serra do Mendanha é composta por nove espécies, que pouco diferiu ao longo de um gradiente altitudinal (0 a 900 m), com o registro das espécies Euparkerella brasiliensis, H. binotatus, Leptodactylus marmoratus e Zachaenus parvulus na maioria das cotas altimétricas. A altitude, a declividade e profundidade do folhiço foram as variáveis abióticas que mais influenciaram na distribuição de abundância e de riqueza de espécies do folhiço. Na estação úmida (setembro a março) a densidade de anuros no folhiço foi de 10,4 ind./100m2, enquanto que na estação seca (abril a agosto) houve uma redução de 34,6% (6,8 ind./100m2). As assembléias de anuros utilizam os recursos hídricos disponíveis na Serra do Mendanha (água da chuva, córregos, fitotelmas, rios e umidade do ar), de diferentes formas, associadas diretamente ao modo reprodutivo de cada espécie. A altitude, a temperatura da água e o pH afetaram a distribuição de espécies de girinos nos diferentes sítios reprodutivos. O modo reprodutivo 1 foi o mais freqüente (n = 18) entre os 14 modos identificados. O estudo de 12 poças (lênticas e lóticas) indicou que estas diferiram consistentemente entre si, seja nas suas dimensões, na composição de suas assembléias de girinos e nos seus componentes abióticos. Diversos predadores de girinos foram encontrados em algumas poças. A assembléia de girinos de cada poça indicou ser moldada pela interação de diferentes fatores ambientais, ecológicos e filogenéticos de cada espécie
