A composição química e o modo de cultivo do arroz o tornam susceptível à contaminação fúngica e, consequentemente, por micotoxinas. Considerando-se o expressivo consumo de arroz e a possibilidade de ser potencial fonte de micotoxinas, especial atenção deve ser dispensada quanto à qualidade do produto adquirido. Assim, o presente trabalho teve como objetivo avaliar espécies do gênero Aspergillus quanto à capacidade toxigênica, em diferentes subgrupos de arroz. As amostras constituíram-se de 31 marcas de arroz referentes aos subgrupos branco polido (21) e parboilizado (10), mais comumente comercializadas na cidade de Lavras - MG. Ao contrário dos outros subgrupos, a incidência de Aspergillus flavus e Aspergillus niger em amostras de arroz branco polido aumentou significativamente após a desinfecção. Pôde-se observar que, 50% dos Aspergillus flavus e 50% dos Aspergillus niger encontrados, foram considerados toxigênicos para o subgrupo branco polido. Na amostra de arroz parboilizado, 67% dos Aspergillus flavus eram potenciais produtores. O Aspergillus ochraceus não se revelou como toxigênico. Este estudo permitiu concluir que, apesar de trabalhos isolados, a presença de fungos toxigênicos em arroz é verídico, o que se torna relevante por se tratar de um cereal importante no cenário mundial.

Recently, an increased interest in antioxidant activity and health-improving capacity of cactus pear has been registered. The antioxidant capacity of the pulp of cactus-pear fruits has been previously assessed. In this work, total phenolics, flavonoids and tannins of peel and seeds of four cactus pear cultivars were examined as well as their antioxidant capacity. Tannins were the major phenolics in cactus pear seeds accounting for almost fifty percent for all cultivars. Analysis of variance revealed that ripeness, cultivar, and its interaction had highly significant effect on the total phenolics, tannin, and flavonoid contents of cactus pear peel. With regard to the seeds, only the stage of ripeness and interaction (ripeness stage x cultivar) were significant on total phenolics and tannins contents. The flavonoid content in seeds was not affected by any of the factors or their interactions. The antioxidant capacity was higher in the peel than in the seeds. Generally, fruits with light-green or yellow-brown peel have higher antiradical activity and Trolox equivalent antioxidant capacity (TEAC) values compared with those with red-purple peel. Cactus pear by-products can indeed be exploited as a good and cheap source of natural antioxidants.

The aim of this study was to assess the contamination of oysters (Crassostrea gigas), harvested in six different regions of the South Bay of Santa Catarina Island, with Coliforms at 45 ºC, Escherichia coli, Vibrio spp., positive coagulase staphylococci, and Salmonella sp. over a period of one year. One hundred eighty oyster samples were collected directly from their culture sites and analyzed. Each sample consisted of a pool of 12 oysters. All of the samples analyzed showed absence of Salmonella, 18 (10%) samples showed presence of Escherichia coli, 15 (8.3%) samples were positive for V. alginolyticus, and Vibriocholerae was detected in 4 samples (2.2%). The counts of positive-coagulase staphylococci varied from <10 to 1.9 x 102 CFU.g-1, whereas the counts of Coliforms at 45 ºC and E. coli ranged from <3 to 1.5 x 102 MPN.g-1 and <3 and 4.3 x 10 MPN.g-1, respectively. Counts of V. parahaemolyticus and V. vulnificus ranged between <3 and 7 MPN.g-1, for both microorganisms. This suggests the need for monitoring these Vibrios contamination in oysters. Based on the results of the microbiological assays, the samples analyzed showed acceptable bacteriological quality, i.e., they were within the parameters established by Brazilian Legislation.

Antimicrobial activities of two commercial disinfectants, alone or combined with heat, against three Salmonella strains and three Listeria monocytogenes strains were studied. The efficacy of disinfectants against planktonic bacteria and bacteria attached to three food contact industrial surfaces (stainless steel, polytetraflourethylene, and rubber) was investigated. The tests were conducted using the sanitizer (quaternary ammonium compounds, and alquyldiethylenediamineglycine and di-alquyldiamineethylglycine) concentrations recommended by the manufacturers, and concentrations twice and four times higher than those values. The recommended concentrations were not effective to kill bacteria, especially when they were attached to surfaces. Concentrations of disinfectants twice and four times higher than those recommended were needed to fully eliminate planktonic bacteria. These same sanitizer concentrations were not sufficient to remove attached bacteria. To remove them from the surfaces, a treatment with recommended concentrations in combination with heat was needed. Our results indicate that these two pathogenic bacteria could survive common sanitation programs used in the food industry.

The aim of this study was to verify the efficiency of the BAX® system for the detection of Salmonella spp. in raw chicken meat. The conventional culture method (IN 62, MAP) was used as a reference method. A total of 8,813 chicken carcass samples were analyzed. In the first part of the study, 1,200 samples were analyzed using the BAX® System and the conventional culture method. In the second part, 7,613 samples were analyzed by the BAX® system, and the conventional method was used only for samples that tested positive for Salmonella spp. by the BAX® system. The sensitivity, specificity, relative accuracy, positive predictive value, and negative predictive value obtained in the first part of this study were 100%, 92.3%, 96.4%, 53.3% and 100%, respectively. The BAX® system showed no false-negative results and reduced the time to obtain presumptive positive results. It is a suitable method for use in laboratories that perform a large number of food samples analyses daily. However, the conventional method is still required to confirm the presence of Salmonella spp. in samples that test positive using the BAX® system.

Staphylococcus spp. can survive in biofilms for long periods of time, and they can be transferred from one point to another and cause environmental contamination in food processing. The aim of this study was to detect Staphylococcus strains isolated from a poultry processing plant by the presence of adhesion genes and the phenotypic production of exopolysaccharide. In the present study, the production of exopolysaccharide and the presence of adhesion genes in 65 strains of Staphylococcus spp. were evaluated. All strains of Staphylococcus spp. produced exopolysaccharide, as confirmed by formation of black and opaque colonies in Congo Red Agar. The variation of sucrose content was critical for the production of exopolysaccharide in Congo Red Agar since at low sucrose concentrations all strains presented a characteristic result, i.e., there was no exopolysaccharide production. The atl gene was found in all strains, and the icaA and icaD genes were found in 97% of them. The data obtained suggest that Staphylococcus spp. isolated from the poultry processing plant evaluated has a potential for biofilm formation. An efficient control of this microorganism in food processing environment is necessary as they may represent a potential risk to consumers.

The objectives of this study were to physicochemically characterize and determine the antioxidant activities and anthocyanin contents of organic Rabbiteye blueberries grown in Southern Brazil and its derived products, in order to investigate the utility of food processing wastes as raw materials for developing products with beneficial health properties. The antioxidant capacity of the blueberries was superior to that of other fruits and juices. The pomace exhibited high activity, albeit lower than that of the fruit, while the flour and the dried blueberries lost 66% and 46% of the original antioxidant activity, respectively. The average anthocyanin contents of the fruits were moderate compared to other sources and species of blueberries. The pomace contains a large amount of anthocyanins while the flour and dried blueberries exhibited a 32% and 42% loss in anthocyanin content, respectively. The use of agro-industrial residues, in addition to adding value and minimizing the impact caused by the accumulation in the environment, can be directed toward the development of new products with bioactive properties.

Antioxidant activities and total phenolic content (TPC) were analyzed in ethanol extracts of 11 marigold cultivars grown in Thailand. Antioxidant activity assays performed in this study were the 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical cation scavenging activity, ferric ion reducing antioxidant power (FRAP), 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, oxygen radical absorbance activity (ORAC), and superoxide anion radical scavenging activity (SRSA) assays. ‘Optiva Orange’ showed the best activity in the ORAC assay and in % SRSA, as well as the highest content of lutein (20.59 mg), gallic acid (25.77 mg), and quercetin (12.61 mg) per gram dry marigold petal among the 11 cultivars. Furthermore, ‘Optiva Orange’ showed the highest lutein yield per plant, as compared to other cultivars. In contrast, ‘Rodeo Gold’ showed the highest activity by ABTS testing (0.92 mmol of trolox/g dry sample), as well as an 89.90% inhibition of DPPH. Lutein content showed a positive correlation with TPC and all antioxidant activity assays. In conclusion, ‘Optiva Orange’ and ‘Rodeo Gold’ could be utilized as a good lutein source for functional food products and cosmetics.

Plant breeding is generally done through sexual reproduction even when the species is propagated asexually for commercial exploitation, as for example, in sugarcane. Therefore, the development of procedures to evaluate sugarcane seed viability is important for plant breeding programs. The objective of this research was to develop a methodology for analyzing the viability of sugarcane seeds (Saccharum spp.). Three crosses were used, two biparental crosses and one polycross. For the germination test study, two substrates (paper and sand) and three constant incubation temperatures (25 ºC, 30 ºC and 35 ºC), in the presence of constant light and also an alternating temperatures (20-30 ºC), with 8 hours light (30 ºC) and 16 hours darkness (20 ºC), were studied. Seedlings were evaluated every five days. The results demonstrated that temperature affected sugarcane seed germination with the most favorable conditions being the alternating temperature (20-30 ºC) and the constant temperature of 30 ºC on a paper substrate.

O conhecimento dos processos germinativos de sementes com tegumentos resistentes, como ocorre em algumas espécies pertencentes à família Fabaceae, pode fornecer subsídios para a produção de mudas e recomposição de áreas degradadas. As sementes de Parkia apresentam germinação natural lenta e irregular, o que pode ser atribuído à impermeabilidade do tegumento. O trabalho foi desenvolvido com o objetivo de avaliar diferentes métodos de superação de dormência física das sementes de Parkia multijuga, P. velutina e P. panurensis. Foram utilizadas sementes coletadas na Base de Operações Geólogo Pedro de Moura (BOGPM), em Coari/AM, submetidas aos seguintes tratamentos: testemunha (T0), desponte (T1), desponte seguido da imersão em água por 8 horas (T2), escarificação mecânica do tegumento em esmeril elétrico (T3), escarificação mecânica do tegumento em esmeril elétrico seguido pela imersão em água por 8 horas (T4), punção com agulha quente (T5), punção com agulha quente seguido pela imersão em água por 8 horas (T6), imersão em ácido sulfúrico (H2SO4) por 30 minutos (T7), imersão em ácido sulfúrico (H2SO4) por 20 minutos (T8), imersão em ácido sulfúrico (H2SO4) por 15 minutos (T9). As sementes de cada tratamento foram submetidas ao teste de germinação, avaliando-se a porcentagem de germinação, tempo médio de emergência de plântulas, índice de velocidade de emergência de plântulas e índice de sincronização. Em geral, todos os tratamentos pré-germinativos aplicados às sementes das três espécies, foram eficientes para aumentar a porcentagem de germinação em relação à testemunha. Os tratamentos de escarificação mecânica com esmeril elétrico na lateral da semente foram mais eficientes na superação da dormência de sementes de P. panurensis e P. multijuga, enquanto a escarificação química com ácido sulfúrico em maior tempo de imersão foi mais eficiente para as sementes de P. panurensis e P. velutina tanto para emergência de plântulas quanto para a formação de plântulas normais.

Tutkimuksen päätarkoituksena oli selvittää puutiaisten (Ixodes spp.) levinneisyyttä Suomessa vuonna 2014. Tarkoituksena oli samalla selvittää puutiaisten mahdollista runsastumista ja esiintyvyyttä ennakoivia tekijöitä sekä puutiaislevitteisten tautien (borrelioosin ja puutiaisaivokuumeen) esiintyvyyttä. Menetelmänä käytettiin valtakunnallista kyselytutkimusta, jossa ihmiset kertoivat internetissä olleelle kyselytutkimuslomakkeelle puutiaishavainnoistaan vuonna 2014. Kyselylomakkeeseen oli mahdollista vastata touko–marraskuun välisenä aikana. Vastauksista analysoitua puutiaistilannetta verrattiin viimeiseen kattavaan puutiaisten levinneisyyttä koskevaan tutkimukseen Suomessa vuosilta 1956–1958. Vertailun avulla saatiin tietoa puutiaisten levinneisyysalueen muutoksista. Tulokset osoittivat, että puutiaiset ovat levinneet yhä pohjoisemmaksi Suomessa viimeisen viidenkymmenen vuoden aikana. Havaintojen ja ihmisten kertomien kokemusten perusteella voitiin olettaa, että puutiaiskanta on myös paikoin runsastunut Suomessa. Puolet puutiaishavainnoista oli tehty eläimestä, joista suurin osa oli koirasta. Eniten havaintoja kertyi niityiltä ja heinikoilta, ja suurin osa näistä oli eläimestä tehtyjä havaintoja. Ihmisistä tehtyjä havaintoja tehtiin eniten puistoilta tai pihoilta. Eniten puutiaishavaintoja tehtiin touko–kesäkuussa, jonka jälkeen havaintomäärät vähenivät. Puutiaishavaintojen määrät eri säätiloissa ja kellonaikoina olivat todennäköisesti yhteydessä ihmisten aktiivisuuteen, sillä havaintoja kertyi eniten päiväsaikaan aurinkoisella ja puolipilvisellä säällä. Borrelioosin diagnoosin oli ilmoittanut saaneensa 110 havainnoitsijaa ja puutiaisaivokuumeen diagnoosin 1 havainnoitsija. Eniten diagnoosiin johtaneita havaintoja tehtiin niinä kuukausina ja niissä ympäristöissä, joissa puutiaishavaintojakin tehtiin eniten. Syyksi puutiaisen yleistymiseen on epäilty ilmastonmuutosta ja sen vaikutuksia suoraan puutiaisiin tai välillisesti isäntäeläimiin. Ilmaston yhä lämmetessä ja kasvukauden pidentyessä, puutiaiset todennäköisesti jatkavat levittäytymistä pohjoisemmaksi ja puutiaisten vuosittainen aktiivinen aika pidentyy. Tämän myötä voidaan myös olettaa, että puutiaisten levittämät taudit yleistyvät ja leviävät yhä laajemmalle Suomessa.

Although a substantial amount of research has been done on all aspects ofHeliconius biology and their ecological interactions with Passiflora, there has not hitherto been a phylogenetic examination of this association for coevolution. To test the HeliconiuslPassilfora association for coevolutionary congruence, phylogenies for each group were established and compared. The phylogeny for 14 species ofHeliconiinae from Costa Rica was based on combined sequence data from rRNA ITS 2 and partial EF-1a gene regions. For the Passifloraceae, 17 host plant species were utilized to establish a phylogeny based on tRNALeucine and ITS 1/5.8S1 ITS 2 sequence data. The phylogenies for both groups were largely in agreement with current classification (for Passifloraceae) and previously established phylogenies. Associations with the large subgenera Passiflora and Decaloba correspond with the two major Advanced Radiation groups in Heliconius. Although strict congruence above subgenus level was not observed, broad scale congruence was evident. One main host shift as well as other possible explanations for lack of strict congruence are suggested.

Many species of Anopheles mosquitoes (Diptera: Culicidae) are now recognized as species complexes whose members are often indistinguishable morphologically but identifiable based on ecological, genetic, or behavioural data. Because the members of species complexes often differ in their vector potential, accurate identification of vector species is essential for successful mosquito control. To investigate the cryptic species status of Anopheles mosquitoes in Canada, specimens were collected from across the country and examined using morphological, molecular, and ecological data. Six of the seven traditionally recognised species from Canada were collected from locations in British Columbia, Quebec, Newfoundland and Labrador, and throughout Ontario, including Anopheles barberi, An. earlei, An. freeborni, An. punctipennis, An. quadrimaculatus s.l., and An. walkeri. Variation in polymorphic traits within An. earlei, An. punctipennis, and An. quadrimaculatus s.l. were quantified and egg morphology examined using scanning electron microscopy. Morphological identification of adult and larval specimens suggested that two described cryptic species, An. perplexens and An. smaragdinus, were present in Canada. DNA sequence data were analysed for evidence of cryptic species using three molecular markers: COl, ITS2, and ITS!. Intraspecific COl variation was very low in most species «1 %), except for An. punctipennis with 2% sequence divergence between those from British Columbia (BC) and Ontario (ON), and An. walkeri with 7% sequence divergence between populations from Manitoulin Island (NO) and Long Point Provincial Park (LP). Similar patterns were also seen using ITS2 and ITS 1. Therefore, molecular data revealed the presence of two putative cryptic species within two species examined (i.e., An. walkeri and An. punctipennis), corresponding to collection location (i.e., NO vs. LP and BC vs. ON, respectively). Surprisingly, there was no molecular support for the presence of either An. perplexens or An. smaragdinus in Canada despite the morphological assessments. Ecological data from all collection sites were recorded and are available in an online database designed to manage all collection and identification data. Current bionomic information, including regional abundance, larval habitat, and species associations, was determined for each species. This multidisciplinary study of Anopheles mosquitoes is the first detailed investigation of these potential disease vectors in Canada and demonstrates the importance of an integrated approach to anopheline systematics that includes molecular data.

The resurgence of malaria in highland regions of Africa, Oceania and recently in South America underlines the importance of the study of the ecology of highland mosquito vectors of malaria. Since the incidence of malaria is limited by the distribution of its vectors, the purpose of this PhD thesis was to examine aspects of the ecology of Anopheles mosquitoes in the Andes of Ecuador, South America. A historical literature and archival data review (Chapter 2) indicated that Anopheles pseudopunctipennis transmitted malaria in highland valleys of Ecuador prior to 1950, although it was eliminated through habitat removal and the use of chemical insecticides. Other anopheline species were previously limited to low-altitude regions, except in a few unconfirmed cases. A thorough larval collection effort (n=438 attempted collection sites) in all road-accessible parts of Ecuador except for the lowland Amazon basin was undertaken between 2008 - 2010 (Chapter 3). Larvae were identified morphologically and using molecular techniques (mitochondrial COl gene), and distribution maps indicated that all five species collected (Anopheles albimanus, An. pseudopunctipennis, Anopheles punctimacula, Anopheles oswaldoi s.l. and Anopheles eiseni) were more widespread throughout highland regions than previously recorded during the 1940s, with higher maximum altitudes for all except An. pseudopunctipennis (1541 m, 1930 m, 1906 m, 1233 m and 1873 m, respectively). During larval collections, to characterize species-specific larval habitat, a variety of abiotic and biotic habitat parameters were measured and compared between species-present and species-absent sites using chi-square tests and stepwise binary logistic regression analyses (Chapter 4). An. albimanus was significantly associated with permanent pools with sand substrates and An. pseudopunctipennis with gravel and boulder substrates. Both species were significantly associated with floating cyanobacterial mats and warmer temperatures, which may limit their presence in cooler highland regions. Anopheles punctimacula was collected more often than expected from algae-free, shaded pools with higher-than-average calculated dissolved oxygen. Anopheles oswaldoi s.l., the species occurring on the Amazonian side of the Andes, was associated with permanent, anthropogenic habitats such as roadside ditches and ponds. To address the hypothesis that human land use change is responsible for the emergence of multiple highland Anopheles species by creating larval habitat, common land uses in the western Andes were surveyed for standing water and potential larval habitat suitability (Chapter 5). Rivers and road edges provided large amounts of potentially suitable anopheline habitat in the western Andes, while cattle pasture also created potentially suitable habitat in irrigation canals and watering ponds. Other common land uses surveyed (banana farms, sugarcane plantations, mixed tree plantations, and empty lots) were usually established on steep slopes and had very little standing water present. Using distribution and larval habitat data, a GIS-based larval habitat distribution model for the common western species was constructed in ArcGIS v.l 0 (ESRI 2010) using derived data layers from field measurements and other sources (Chapter 6). The additive model predicted 76.4 - 97.9% of the field-observed collection localities of An. albimanus, An. pseudopunctipennis and An. punctimacula, although it could not accurately distinguish between species-absent and speciespresent sites due to its coarse scale. The model predicted distributional expansion and/or shift of one or more anopheline species into the following highland valleys with climate warming: Mira/Chota, Imbabura province, Tumbaco, Pichincha province, Pallatanga and Sibambe, Chimborazo province, and Yungilla, Azuay province. These valleys may serve as targeted sites of future monitoring to prevent highland epidemics of malaria. The human perceptions of malaria and mosquitoes in relation to land management practices were assessed through an interview-based survey (n=262) in both highlands and lowlands, of male and female land owners and managers of five property types (Chapter 7). Although respondents had a strong understanding of where the disease occurs in their own country and of the basic relationship among standing water, mosquitoes and malaria, about half of respondents in potential risk areas denied the current possibility of malaria infection on their own property. As well, about half of respondents with potential anopheline larval habitat did not report its presence, likely due to a highly specific definition of suitable mosquito habitat. Most respondents who are considered at risk of malaria currently use at least one type of mosquito bite prevention, most commonly bed nets. In conclusion, this interdisciplinary thesis examines the occurrence of Anopheles species in the lowland transition area and highlands in Ecuador, from a historic, geographic, ecological and sociological perspective.

Larval habitat for three highland Anopheles species: Anopheles albimanus Wiedemann, Anopheles pseudopunctipennis Theobald, and Anopheles punctimacula Dyar and Knab was related to human land uses, rivers, roads, and remotely sensed land cover classifications in the western Ecuadorian Andes. Of the five commonly observed human land uses, cattle pasture (n = 30) provided potentially suitable habitat for A. punctimacula and A. albimanus in less than 14% of sites, and was related in a principal components analysis (PCA) to the presence of macrophyte vegetation, greater surface area, clarity, and algae cover. Empty lots (n = 30) were related in the PCA to incident sunlight and provided potential habitat for A. pseudopunctipennis and A. albimanus in less than 14% of sites. The other land uses surveyed (banana, sugarcane, and mixed tree plantations; n = 28, 21, 25, respectively) provided very little standing water that could potentially be used for larval habitat. River edges and eddies (n = 41) were associated with greater clarity, depth, temperature, and algae cover, which provide potentially suitable habitat for A. albimanus in 58% of sites and A. pseudopunctipennis in 29% of sites. Road-associated water bodies (n = 38) provided potential habitat for A. punctimacula in 44% of sites and A. albimanus in 26% of sites surveyed. Species collection localities were compared to land cover classifications using Geographic Information Systems software. All three mosquito species were associated more often with the category “closed/open broadleaved evergreen and/or semi-deciduous forests” than expected (P ≤ 0.01 in all cases), given such a habitat’s abundance. This study provides evidence that specific human land uses create habitat for potential malaria vectors in highland regions of the Andes.
