La región volcánica de Campo de Calatrava se ha interpretado como un proceso de volcanismo intraplaca desarrollado durante el Neógeno. Se han propuesto dos modelos geodinámicos contrapuestos para explicar el origen de este volcanismo: a) un proceso de rifting en un contexto extensional con un adelgazamiento localizado de corteza; b) un proceso flexural de la litosfera en un contexto compresivo débil sin adelgazamiento de corteza. El análisis de las anomalías gravimétricas de Bouguer y una modelización gravimétrica 2D a escala cortical contribuyen a discriminar entre los modelos geodinámicos propuestos para el origen del volcanismo. Los modelos gravimétricos se han constreñido en base a los estudios sísmicos profundos existentes en la zona y a la cartografía geológica regional. Los modelos gravimétricos descartan un adelgazamiento cortical, lo que cuestiona el modelo de rifting abortado y apoyan la hipótesis alternativa del proceso flexural de la litosfera en régimen compresivo débil como origen del volcanismo bético.

La investigación analiza el comportamiento de la participación electoral (PE) y su relación con las dinámicas de violencia política en Norte de Santander durante 1997-2011, con el fin de demostrar que la región del Catatumbo ha tenido un comportamiento electoral diferente al resto del departamento dado que tiene una tendencia en porcentajes de participación bajos durante las elecciones locales que se dieron en 1997, 2000 y 2003, lo cual se debe a la presencia y disputa por el control territorial entre los Grupos Armados Ilegales (GAI). Contrario a lo anterior, en las elecciones de 2007 y 2011 se presenta una variación positiva al aumentar la PE en el departamento en general. El desarrollo tecnológico y la expansión de las formas de comunicación en Colombia, no solo trajeron consigo grandes beneficios, sino también nuevos retos para el Estado Moderno. Actualmente, la oferta de espacios de difusión de propaganda electoral ha aumentado, mientras persiste un marco legal diseñado para los medios de comunicación del Siglo XX. Por tanto, este trabajo no solo realiza un diagnóstico de los actuales mecanismos de control administrativo sobre la propaganda electoral en Internet, sino también propone unos mecanismos que garanticen los principios de la actividad electoral, siendo esta la primera propuesta en Colombia. Por el poco estudio del tema, su alcance es exploratorio, se basa en un enfoque jurídico-institucional. Se utilizaron métodos cualitativos de recolección de datos (trabajo de archivo y entrevistas) y de análisis (tipologías, comparaciones, exegesis del marco legal), pero también elementos cuantitativos como análisis estadísticos.

This PhD project focuses on the study of the early stages of bone biomineralization in 2D and 3D cultures of osteoblast-like SaOS-2 osteosarcoma cells, exposed to an osteogenic cocktail. The efficacy of osteogenic treatment was assessed on 2D cell cultures after 7 days. A large calcium minerals production, an overexpression of osteogenic markers and of alkaline phosphatase activity occurred in treated samples. TEM microscopy and cryo-XANES micro-spectroscopy were performed for localizing and characterizing Ca-depositions. These techniques revealed a different localization and chemical composition of Ca-minerals over time and after treatment. Nevertheless, the Mito stress test showed in treated samples a significant increase in maximal respiration levels associated to an upregulation of mitochondrial biogenesis indicative of an ongoing differentiation process. The 3D cell cultures were realized using two different hydrogels: a commercial collagen type I and a mixture of agarose and lactose-modified chitosan (CTL). Both biomaterials showed good biocompatibility with SaOS-2 cells. The gene expression analysis of SaOS-2 cells on collagen scaffolds indicated an osteogenic commitment after treatment. and Alizarin red staining highlighted the presence of Ca-spots in the differentiated samples. In addition, the intracellular magnesium quantification, and the X-ray microscopy on mineral depositions, suggested the incorporation of Mg during the early stages of bone formation process., SaOS-2 cells treated with osteogenic cocktail produced Ca mineral deposits also on CTL/agarose scaffolds, as confirmed by alizarin red staining. Further studies are underway to evaluate the differentiation also at the genetic level. Thanks to the combination of conventional laboratory methods and synchrotron-based techniques, it has been demonstrated that SaOS-2 is a suitable model for the study of biomineralization in vitro. These results have contributed to a deeper knowledge of biomineralization process in osteosarcoma cells and could provide new evidences about a therapeutic strategy acting on the reversibility of tumorigenicity by osteogenic induction.

The presented study aimed to evaluate the productive and physiological behavior of a 2D multileader apple training systems in the Italian environment both investigating the possibility to increase yield and precision crop load management resolution. Another objective was to find valuable thinning thresholds guaranteeing high yields and matching fruit market requirements. The thesis consists in three studies carried out in a Pink Lady®- Rosy Glow apple orchard trained as a planar multileader training system (double guyot). Fruiting leaders (uprights) dimension, crop load, fruit quality, flower and physiological (leaf gas exchanges and fruit growth rate) data were collected and analysed. The obtained results found that uprights present dependence among each other and as well as a mutual support during fruit development. However, individual upright fruit load and upright’s fruit load distribution on the tree (~ plant crop load) seems to define both upright independence from the other, and single upright crop load effects on the final fruit quality production. Correlations between fruit load and harvest fruit size were found and thanks to that valuable thinning thresholds, based on different vegetative parameters, were obtained. Moreover, it comes out that an upright’s fruit load random distribution presents a widening of those thinning thresholds, keeping un-altered fruit quality. For this reason, uprights resulted a partially physiologically-dependent plant unit. Therefore, if considered and managed as independent, then no major problems on final fruit quality and production occurred. This partly confirmed the possibility to shift crop load management to single upright. The finding of the presented studies together with the benefits coming from multileader planar training systems suggest a high potentiality of the 2D multileader training systems to increase apple production sustainability and profitability for Italian apple orchard, while easing the advent of automation in fruit production.

In political debates, the media[tisation] can determine the use of language with the aim to increase their spectacularisation and polarisation, possibly by means of criticism and humour, respectively. These linguistic strategies are often used in order to shape what was defined by Goffman as one’s face. Politicians, in particular, can recur to facework in a double sense: shaping their own face positively and/or that of their opponents negatively. Starting from the sociologic theory of face by Goffman and Levinson, with the help of corpus analysis tools, this research investigated the ways in which various forms of criticism and forms of humour were conducted in 3 electoral debates on a national scale (Germany, Ireland, and New Zealand) and 1 debate for the municipal election in Rome. The transcripts were revised after automatic transcriptions were extracted or found online, of which the audio-visual content is available on the Internet. The CADS research aimed to investigate the role that criticism and humour played within each participant’s discourse, and to identify differences and similarities among the strategies used by political leaders and moderators in different countries, and in different cultural, political, and media contexts.

The study of ultra-cold atomic gases is one of the most active field in contemporary physics. The main motivation for the interest in this field consists in the possibility to use ultracold gases to simulate in a controlled way quantum many-body systems of relevance to other fields of physics, or to create novel quantum systems with unusual physical properties. An example of the latter are Bose-Fermi mixtures with a tunable pairing interaction between bosons and fermions. In this work, we study with many-body diagrammatic methods the properties of this kind of mixture in two spatial dimensions, extending previous work for three dimensional Bose-Fermi mixtures. At zero temperature, we focus specifically on the competition between boson condensation and the pairing of bosons and fermions into molecules. By a numerical solution of the main equations resulting by our many-body diagrammatic formalism, we calculate and present results for several thermodynamic quantities of interest. Differences and similarities between the two-dimensional and three-dimensional cases are pointed out. Finally, our new results are applied to discuss a recent proposal for creating a p-wave superfluid in Bose-Fermi mixtures with the fermionic molecules which form for sufficiently strong Bose-Fermi attraction.

Dopamine is a neurotransmitter which has a role in several psychiatric and neurological disorders. In-vivo detection of its concentration at the microscopic scale would benefit the study of these conditions and help in the development of therapies. The ideal sensor would be biocompatible, able to probe concentrations in microscopic volumes and sensitive to the small physiological concentrations of this molecule (10 nM - 1 μM). The ease of oxidation of dopamine makes it possible to detect it by electrochemical methods. An additional requirement in this kind of experiments when run in water, though, is to have a large potential window inside which no redox reactions with water take place. A promising class of materials which are being explored is the one of pyrolyzed photoresists. Photoresists can be lithographically patterned with micrometric resolution and after pyrolysis leave a glassy carbon material which is conductive, biocompatible and has a large electrochemical water window. In this work I developed a fabrication procedure for microelectrode arrays with three dimensional electrodes, making the whole device using just a negative photoresist called SU8. Making 3D electrodes could be a way to enhance the sensitivity of the electrodes without occupying a bigger footprint on the device. I characterized the electrical, morphological, and electrochemical properties of these electrodes, in particular their sensitivity to dopamine. I also fabricated and tested a two dimensional device for comparison. The three dimensional devices fabricated showed inferior properties to their two dimensional counter parts. I found a possible explanation and suggested some ways in which the fabrication could be improved.

Low-density nanostructured foams are often limited in applications due to their low mechanical and thermal stabilities. Here we report an approach of building the structural units of three-dimensional (3D) foams using hybrid two-dimensional (2D) atomic layers made of stacked graphene oxide layers reinforced with conformal hexagonal boron nitride (h-BN) platelets. The ultra-low density (1/400 times density of graphite) 3D porous structures are scalably synthesized using solution processing method. A layered 3D foam structure forms due to presence of h-BN and significant improvements in the mechanical properties are observed for the hybrid foam structures, over a range of temperatures, compared with pristine graphene oxide or reduced graphene oxide foams. It is found that domains of h-BN layers on the graphene oxide framework help to reinforce the 2D structural units, providing the observed improvement in mechanical integrity of the 3D foam structure.

Chemical cross-linking has emerged as a powerful approach for the structural characterization of proteins and protein complexes. However, the correct identification of covalently linked (cross-linked or XL) peptides analyzed by tandem mass spectrometry is still an open challenge. Here we present SIM-XL, a software tool that can analyze data generated through commonly used cross-linkers (e.g., BS3/DSS). Our software introduces a new paradigm for search-space reduction, which ultimately accounts for its increase in speed and sensitivity. Moreover, our search engine is the first to capitalize on reporter ions for selecting tandem mass spectra derived from cross-linked peptides. It also makes available a 2D interaction map and a spectrum-annotation tool unmatched by any of its kind. We show SIM-XL to be more sensitive and faster than a competing tool when analyzing a data set obtained from the human HSP90. Chronic pain has been often associated with myofascial pain syndrome (MPS), which is determined by myofascial trigger points (MTrP). New features have been tested for MTrP diagnosis. The aim of this study was to evaluate two-dimensional ultrasonography (2D US) and ultrasound elastography (UE) images and elastograms of upper trapezius MTrP during electroacupuncture (EA) and acupuncture (AC) treatment. 24 women participated, aged between 20 and 40 years (M ± SD = 27.33 ± 5.05) with a body mass index ranging from 18.03 to 27.59 kg/m2 (22.59 ± 3.11), a regular menstrual cycle, at least one active MTrP at both right (RTPz) and left trapezius (LTPz) and local or referred pain for up to six months. Subjects were randomized into EA and AC treatment groups and the control sham AC (SHAM) group. Intensity of pain was assessed by visual analogue scale; MTrP mean area and strain ratio (SR) by 2D US and UE. A significant decrease of intensity in general, RTPz, and LTPz pain was observed in the EA group (p = 0.027; p < 0.001; p = 0.005, respectively) and in general pain in the AC group (p < 0.001). Decreased MTrP area in RTPz and LTPz were observed in AC (p < 0.001) and EA groups (RTPz, p = 0.003; LTPz, p = 0.005). Post-treatment SR in RTPz and LTPz was lower than pre-treatment in both treatment groups. 2D US and UE effectively characterized MTrP and surrounding tissue, pointing to the possibility of objective confirmation of subjective EA and AC treatment effects.
