Different methods to reduce the high suction caused by conical vortices have been reported in the literature: vertical parapets, either solids or porous, placed at the roof edges being the most analysed configuration. Another method for alleviating the high suction peaks due to conical vortices is to round the roof edges. Very recently, the use of some non-standard parapet configurations, like cantilever parapets, has been suggested. In this paper, its efficiency to reduce suction loads on curved roofs is experimentally checked by testing the pressure distribution on the curved roof of a low-rise building model in a wind tunnel. El vidrio no puede ser tratado como un material estructural convencional desde el punto de vista de la resistencia mecánica. Su naturaleza, como material frágil, junto con la inevitable presencia de microfisuras en su superficie y las consecuencias de accidentes por posibles fallos, exigen métodos rigurosos que garanticen un cálculo seguro de los elementos estructurales de vidrio, cuya resistencia a rotura depende en gran medida del tamaño del elemento y del tipo de carga a la que está sometido. Por lo tanto, su cálculo debe basarse en conceptos probabilísticos y en criterios de mecánica de la fractura, en sustitución de un cálculo convencional de vidrio según tablas deducidas de programas experimentales y posterior aplicación del concepto de tensiones admisibles. Con el fin de analizar y comparar las características mecánicas de vidrios templados, termoendurecidos y recocidos, se realizó un amplio programa experimental de ensayos de flexión a cuatro puntos y de anillos concéntricos de pequeña superficie, seguido de un ajuste de los resultados mediante una función de distribución triparamétrica de Weibull. Glass cannot be handled as a conventional structural material from the point of view of the mechanical strength. Its nature as brittle material, together with the inevitable presence of micro-cracks on its surface and the consequences of eventual failures, demand rigorous methods to achieve a safe design for glass elements, whose stress resistance is very much dependent on the integrity of its surface, element size and loading pattern. Thus, its design must rely on probabilistic concepts and fracture mechanics criteria, substitutive of the conventional glass design based on charts derived from experimental programs and subsequent application of the admissible stress concept. In order to analyze and compare the strength characteristics of tempered, heat-strengthened and annealed glass, a large experimental programme based on four-point bending and coaxial double ring tests was performed and the results were fitted using a three-parameter Weibull cumulative distribution function.

The fracture behavior of rock block contacts has been studied for many years. Unfortunately, up to now, there is not a rigorous formulation or a solid theoretical foundation to support it. A mathematical development to represent the failure mechanism which occurs in the contacts between rock blocks is presented to evaluate the performance of breaking mechanism of such blocks relating it to the morphology of the contact and mechanical parameters of the material. The examined framework includes the evaluation of the surface roughness of first order in the failure mechanism of the granular particles of large size and the development of a theoretical model describing the morphology of the contact between rock blocks.

From a physical perspective, a joint experiences fracturing processes that affect the rock at both microscopic and macroscopic levels. The result is a behaviour that follows a fractal structure. In the first place, for saw-tooth roughness profiles, the use of the triadic Koch curve appears to be adequate and by means of known correlations the JRC parameter is obtained from the angle measured on the basis of the height and length of the roughnesses. Therefore, JRC remains related to the geometric pattern that defines roughness by fractal analysis. In the second place, to characterise the geometry of irregularities with softened profiles, consequently, is proposed a characterisation of the fractal dimension of the joints with a circumference arc generator that is dependent on an average contact angle with regard to the mid-plane. The correlation between the JRC and the fractal dimension of the model is established with a defined statistical ratio.

El presente trabajo ahonda en el conocimiento del viento urbano. La investigación pasa revista a la historia de la relación del viento y la ciudad y revisa tres pares de disciplinas implicadas en comprender mejor dicha relación: la arquitectura y el urbanismo, la meteorología y la climatología y, por último, la ingeniería aeroespacial y la aerodinámica civil. Se estudian el comportamiento y la fluidez del viento al desplazarse por cuerpos romos no fuselados (los edificios y la trama urbana), así como sus efectos dentro de la ciudad. Asimismo, se examinan las metodologías existentes para comprenderlo, medirlo y analizarlo, desde los estudios de proporción y modelamiento en túneles de viento hasta las simulaciones virtuales y las dinámicas de fluidos CFD. Posteriormente se reconoce un caso de estudio que permite analizar el viento como un factor aislado, pero desde los parámetros morfológicos de una ciudad en la que se generan patrones aerodinámicos muy característicos: Punta Arenas, la ciudad más austral del mundo, donde los vientos corren casi siempre desde la misma dirección, el “oeste”, a más de 33,3 m/s, lo que equivale a 120 Km/h. La hipótesis de la investigación es que la morfología del casco histórico de Punta Arenas genera patrones aerodinámicos que condicionan el bienestar en los espacios públicos. El objetivo general de la investigación es estudiar los efectos aerodinámicos presentes en la morfología urbana para mejorar la permanencia en los espacios públicos, proponiendo estrategias para el desarrollo morfológico y volumétrico de los cuerpos edificados. En el desarrollo del caso de estudio se reconocen, al interior del cañón urbano, las temperaturas, los índices de asoleamiento y sus conos de sombra, la dirección del viento y la visualización del vórtice al interior del cañón urbano, para determinar cómo estos factores impactan en el espacio público. Las conclusiones indican que los patrones aerodinámicos presentes en la morfología urbana conducen el viento hacia los espacios públicos que se encuentran o desprotegidos del viento o con excesiva turbulencias, por tanto, los patrones aerodinámicos inciden en el uso estancial de los espacios públicos, generando problemas mecánicos al peatón e incidiendo en la sensación térmica en dichos espacios. Ello permite confirmar que es posible modificar y mejorar el uso de los espacios públicos si somos capaces de modelar la morfología urbana con el fin de reorientar los patrones aerodinámicos que afectan significativamente a dichos espacios. ABSTRACT This work deepens into the knowledge of urban wind. The study reviews the history of the relationship between the wind and the city and reviews three pairs of disciplines involved in understanding better these relationship: Architecture and Urbanism, Meteorology and Climatology and, finally, Aerospace and Civil Aerodynamics. The behavior and flow of wind through blunt bodies not fairings (the buildings and the urban fabric) and its effects within the city are studied. Also, existing methodologies to understand, measure and analyze the wind are examined, from the studios of proportion and modeling in wind tunnels to virtual simulations and fluid dynamics CFD. Subsequently, a case study to analyze the wind as an isolated factor is recognized, but from the morphological parameters of a city where very characteristic aerodynamic patterns are generated: Punta Arenas, the southernmost city in the world, where the winds run almost always from the same direction, the "West", at more than 33.3 m/s, which is equivalent to 120 km/h. The research hypothesis is that the morphology of the historic center of Punta Arenas generates aerodynamic patterns that determine the well-being in public spaces. The overall objective of the research is to study the aerodynamic effects present in the urban morphology to improve retention in public spaces, proposing strategies for morphological and volumetric development of the built bodies. In developing the case study are recognized, within the urban canyon, temperatures, rates of sunlight and shadow cones, wind direction and visualization of the vortex into the urban canyon, to determine how these factors impact in public space. The findings indicate that the aerodynamic patterns in urban morphology lead wind to public spaces that are unprotected or find themselves in a condition of excessive wind or turbulence; therefore, aerodynamic patterns affect the use of public spaces, generating mechanical problems for pedestrians and affecting the thermal sensation in such spaces. This confirms that it is possible to modify and improve the use of public spaces if we are able to model the urban morphology in order to reorient the aerodynamic patterns that significantly affect those spaces.

Vascular endothelium is an important transducer and integrator of both humoral and biomechanical stimuli within the cardiovascular system. Utilizing a differential display approach, we have identified two genes, Smad6 and Smad7, encoding members of the MAD-related family of molecules, selectively induced in cultured human vascular endothelial cells by steady laminar shear stress, a physiologic fluid mechanical stimulus. MAD-related proteins are a recently identified family of intracellular proteins that are thought to be essential components in the signaling pathways of the serine/threonine kinase receptors of the transforming growth factor β superfamily. Smad6 and Smad7 possess unique structural features (compared with previously described MADs), and they can physically interact with each other, and, in the case of Smad6, with other known human MAD species, in endothelial cells. Transient expression of Smad6 or Smad7 in vascular endothelial cells inhibits the activation of a transfected reporter gene in response to both TGF-β and fluid mechanical stimulation. Both Smad6 and Smad7 exhibit a selective pattern of expression in human vascular endothelium in vivo as detected by immunohistochemistry and in situ hybridization. Thus, Smad6 and Smad7 constitute a novel class of MAD-related proteins, termed vascular MADs, that are induced by fluid mechanical forces and can modulate gene expression in response to both humoral and biomechanical stimulation in vascular endothelium.

Phosphorylation of the α-subunit of Na+,K+-ATPase plays an important role in the regulation of this pump. Recent studies suggest that insulin, known to increase solute and fluid reabsorption in mammalian proximal convoluted tubule (PCT), is stimulating Na+,K+-ATPase activity through the tyrosine phosphorylation process. This study was therefore undertaken to evaluate the role of tyrosine phosphorylation of the Na+,K+-ATPase α-subunit in the action of insulin. In rat PCT, insulin and orthovanadate (a tyrosine phosphatase inhibitor) increased tyrosine phosphorylation level of the α-subunit more than twofold. Their effects were not additive, suggesting a common mechanism of action. Insulin-induced tyrosine phosphorylation was prevented by genistein, a tyrosine kinase inhibitor. The site of tyrosine phosphorylation was identified on Tyr-10 by controlled trypsinolysis in rat PCTs and by site-directed mutagenesis in opossum kidney cells transfected with rat α-subunit. The functional relevance of Tyr-10 phosphorylation was assessed by 1) the abolition of insulin-induced stimulation of the ouabain-sensitive 86Rb uptake in opossum kidney cells expressing mutant rat α1-subunits wherein tyrosine was replaced by alanine or glutamine; and 2) the similarity of the time course and dose dependency of the insulin-induced increase in ouabain-sensitive 86Rb uptake and tyrosine phosphorylation. These findings indicate that phosphorylation of the Na+,K+-ATPase α-subunit at Tyr-10 likely participates in the physiological control of sodium reabsorption in PCT.

The kidneys of patients with autosomal dominant polycystic kidney disease become massively enlarged due to the progressive expansion of myriad fluid-filled cysts. The epithelial cells that line the cyst walls are responsible for secreting the cyst fluid, but the mechanism through which this secretion occurs is not well established. Recent studies suggest that renal cyst epithelial cells actively secrete Cl across their apical membranes, which in turn drives the transepithelial movement of Na and water. The characteristics of this secretory flux suggest that it is dependent upon the participation of an apical cystic fibrosis transmembrane conductance regulator (CFTR)-like Cl channel and basolateral Na,K-ATPase. To test this hypothesis, we have immunolocalized the CFTR and Na,K-ATPase proteins in intact cysts and in cyst epithelial cells cultured in vitro on permeable filter supports. In both settings, cyst epithelial cells were found to possess Na,K-ATPase exclusively at their basolateral surfaces; apical labeling was not detected. The CFTR protein was present at the apical surfaces of cyst epithelial cells that had been stimulated to secrete through incubation in forskolin. CFTR was detected in intracellular structures in cultured cyst epithelial cells that had not received the forskolin treatment. These results demonstrate that the renal epithelial cells that line cysts in autosomal dominant polycystic kidney disease express transport systems with the appropriate polarity to mediate active Cl and fluid secretion.

The renin-angiotensin system plays a critical role in sodium and fluid homeostasis. Genetic or acquired alterations in the expression of components of this system are strongly implicated in the pathogenesis of hypertension. To specifically examine the physiological and genetic functions of the type 1A receptor for angiotensin II, we have disrupted the mouse gene encoding this receptor in embryonic stem cells by gene targeting. Agtr1A(-/-) mice were born in expected numbers, and the histomorphology of their kidneys, heart, and vasculature was normal. AT1 receptor-specific angiotensin II binding was not detected in the kidneys of homozygous Agtr1A(-/-) mutant animals, and Agtr1A(+/-) heterozygotes exhibited a reduction in renal AT1 receptor-specific binding to approximately 50% of wild-type [Agtr1A(+/+)] levels. Pressor responses to infused angiotensin II were virtually absent in Agtr1A(-/-) mice and were qualitatively altered in Agtr1A(+/-) heterozygotes. Compared with wild-type controls, systolic blood pressure measured by tail cuff sphygmomanometer was reduced by 12 mmHg (1 mmHg = 133 Pa) in Agtr1A(+/-) mice and by 24 mmHg in Agtr1A(-/-) mice. Similar differences in blood pressure between the groups were seen when intraarterial pressures were measured by carotid cannulation. These studies demonstrate that type 1A angiotensin II receptor function is required for vascular and hemodynamic responses to angiotensin II and that altered expression of the Agtr1A gene has marked effects on blood pressures.

Given the historical rates of combat post-traumatic stress disorder (PTSD), one can expect 30% of soldiers returning from current military conflicts to suffer from PTSD. For these individuals, various cognitive behavioral therapies (CBT) are the most commonly employed treatments. Unfortunately, however, symptom relapse can be expected with the various CBT approaches, as traumatic memories remain. Soldiers are imbued with a militarized identity, and the identity loss experienced by those soldiers who suffer from PTSD is particularly painful for this population, as the militarized identity effectively disavows personal suffering. For this reason, many combat veterans diagnosed with post-traumatic stress disorder experience undue, prolonged suffering as they struggle to make sense of the different person they fear they have become. This paper contrasts certain versions of Western philosophy, which view the self as a fixed and reified entity with certain versions of Eastern philosophy, which view the self as more contextual and fluid, in order to illuminate the value of employing third wave behavioral treatments, specifically Acceptance and Commitment Therapy (ACT), to treat the identity loss experienced by military veterans with PTSD. ACT echoes the Buddhist principle that attachment to verbally-constructed conceptual notions of self contribute to undue suffering, and that more vital living can be achieved by assuming a more contextual and experiential perspective on identity. Research and anecdotal accounts are cited to illustrate why treatment for identity loss associated with combat PTSD should be less focused on reconstructing a historically substance-oriented self and more focused on an epistemological reorientation to a deconstructed, contextual self.

Esta tese apresenta o desenvolvimento e aplicação de modelos de turbulência, transição laminar-turbulenta e de interações fluido-estrutura ao escoamento externo em cilindro rígido estacionário e em vibrações induzidas por vórtices. Tais desenvolvimentos foram realizados no código ReFRESCO, baseado em técnicas de dinâmica de fluidos computacional (CFD). Realizou-se um estudo quanto ao desempenho do modelo k- SST em extensa faixa de números de Reynolds, segundo o qual se identificaram as deficiências de modelagem para este escoamento. A modelagem adaptativa das escalas (SAS) e o modelo de transição por correlações locais (LCTM), ambos combinados ao SST, melhoraram a aderência aos resultados experimentais para este escoamento, em uma contribuição original deste trabalho. A aplicação de técnicas de verificação e validação possibilitou a estimação de incertezas e erros para os modelos e números de Reynolds e também de identificada como outra contribuição deste trabalho. A combinação da modelagem em SST, SAS e LCTM com movimentos impostos de realizada para números de Reynolds moderados, diferentes frequências e amplitudes de vibração, algo que poucas publicações abordam em detalhes. Com relação aos movimentos livres, este trabalho traz contribuições com a aplicação dos modelos SST e SAS ao estudo de vibrações induzidas por vórtices em dois graus de liberdade, baixa razão de massa e números de Reynolds moderados, mais altos do que normalmente observados na literatura. Por fim, a investigação da importância relativa de efeitos da turbulência aos casos de movimentos livres e impostos, com relação ao caso de cilindro estacionário, comprovou a conjetura formulada na parte inicial deste trabalho, no que tange à escolha do modelo de turbulência em determinadas aplicações. Tal escolha mostrou-se menos decisiva no caso do cilindro em movimento imposto e ainda menos nos movimentos livres, em comparação ao caso estacionário, uma vez que a resposta em movimentos do corpo filtra grande parte dos efeitos turbulentos de ordem superior. Esta observação mostra-se relevante, uma vez que pode permitir simplificações na modelagem e aplicação de ferramentas de CFD em uma classe importante de projetos de engenharia.

As formulações baseadas na mecânica do contínuo, embora precisas até certo ponto, por vezes não podem ser utilizadas, ou não são conceitualmente corretas para o entendimento de fenômenos em escalas reduzidas. Estas limitações podem aparecer no estudo dos fenômenos tribológicos em escala nanométrica, que passam a necessitar de novos métodos experimentais, teóricos e computacionais que permitam explorar estes fenômenos com a resolução necessária. Simulações atomísticas são capazes de descrever fenômenos em pequena escala, porém, o número necessário de átomos modelados e, portanto, o custo computacional - geralmente torna-se bastante elevado. Por outro lado, os métodos de simulação associados à mecânica do contínuo são mais interessantes em relação ao custo computacional, mas não são precisos na escala atômica. A combinação entre essas duas abordagens pode, então, permitir uma compreensão mais realista dos fenômenos da tribologia. Neste trabalho, discutem-se os conceitos básicos e modelos de atrito em escala atômica e apresentam-se estudos, por meio de simulação numérica, para a análise e compreensão dos mecanismos de atrito e desgaste no contato entre materiais. O problema é abordado em diferentes escalas, e propõe-se uma abordagem conjunta entre a Mecânica do Contínuo e a Dinâmica Molecular. Para tanto, foram executadas simulações numéricas, com complexidade crescente, do contato entre superfícies, partindo-se de um primeiro modelo que simula o efeito de defeitos cristalinos no fenômeno de escorregamento puro, considerando a Dinâmica Molecular. Posteriormente, inseriu-se, nos modelos da mecânica do contínuo, considerações sobre o fenômeno de adesão. A validação dos resultados é feita pela comparação entre as duas abordagens e com a literatura.

The Amon mud volcano (MV), located at 1250 m water depth on the Nile Deep Sea Fan, is known for its active emission of methane and non-methane hydrocarbons into the hydrosphere. Previous investigations showed a low efficiency of hydrocarbon-degrading anaerobic microbial communities inhabiting the Amon MV center in the presence of sulphate and hydrocarbons in the seeping subsurface fluids. By comparing spatial and temporal patterns of in situ biogeochemical fluxes, temperature gradients, pore water composition and microbial activities over three years, we investigated why the activity of anaerobic hydrocarbon degraders can be low despite high energy supplies. We found that the central dome of the Amon MV, as well as a lateral mud flow at its base, showed signs of recent exposure of hot subsurface muds lacking active hydrocarbon degrading communities. In these highly disturbed areas, anaerobic degradation of methane was less than 2% of the methane flux. Rather high oxygen consumption rates compared to low sulphide production suggest a faster development of more rapidly growing aerobic hydrocarbon degraders in highly disturbed areas. In contrast, the more stabilized muds surrounding the central gas and fluid conduits hosted active anaerobic hydrocarbon-degrading microbial communities. Furthermore, within three years, cell numbers and hydrocarbon degrading activity increased at the gas-seeping sites. The low microbial activity in the hydrocarbon-vented areas of Amon mud volcano is thus a consequence of kinetic limitations by heat and mud expulsion, whereas most of the outer mud volcano area is limited by hydrocarbon transport.
