The products of corrosion reaction of electrolytic iron in 45% ammonium nitrate solution formed under various conditions of time, temperature and pH have been analysed mainly by Mössbauer spectroscopy, in combination with X-ray diffraction, infrared absorption and electron microscopy techniques. γ-Fe00H is found to be the major product of hydrolytic precipitation at pH > 5.6 while only α-FeOOH is formed at pH < 3.0. In the pH range 3.0 < pH < 5.0, α-Fe00H and ferrihydrite are both formed. However, once the nuclei of α-Fe00H are formed under low pH conditions, their growth is favoured even in the otherwise unfavourable slightly acidic medium, resulting in a hydrous α-Fe00H which has two distinct hyperfine fields at the 57Fe nucleus. Magnetite is always formed in the vicinity of the metal and its rate of formation on the surface increases with temperature. α-Fe203 is the major product of hydrolytic precipitation at temperatures >80C. The possible mechanisms for the formation of each of the corrosion products are discussed.

Objective: To examine flying foxes (Pteropus spp.) for evidence of infection with Menangle virus. Design: Clustered non-random sampling for serology, virus isolation and electron microscopy (EM). Procedure: Serum samples were collected from 306 Pteropus spp. in northern and eastern Australia and tested for antibodies against Menangle virus (MenV) using a virus neutralisation test (VNT). Virus isolation was attempted from tissues and faeces collected from 215 Pteropus spp. in New South Wales. Faecal samples from 68 individual Pteropus spp. and four pools of faeces were examined by transmission EM following routine negative staining and immunogold labelling. Results: Neutralising antibodies (VNT titres ≥ 8) against MenV were detected in 46% of black flying foxes (P. alecto), 41% of grey-headed flying foxes (P. poliocephalus), 25% of spectacled flying foxes (P. conspicillatus) and 1% of little red flying foxes (P. scapulatus) in Australia. Positive sera included samples collected from P. poliocephalus in a colony adjacent to a piggery that had experienced reproductive disease caused by MenV. Virus-like particles were observed by EM in faeces from Pteropus spp. and reactivity was detected in pooled faeces and urine by immunogold EM using sera from sows that had been exposed to MenV. Attempts to isolate the virus from the faeces and tissues from Pteropus spp. were unsuccessful. Conclusion: Serological evidence of infection with MenV was detected in Pteropus spp. in Australia. Although virus-like particles were detected in faeces, no viruses were isolated from faeces, urine or tissues of Pteropus spp.

The n=3 member of the Bi1.5Pb0.5 (Ca, Sr) n+1CunO2n+4+δ system has been prepared and characterized by X-ray diffraction and electron microscopy. High-Tc superconductivity in the n=3 member has been established by resistivity, AC susceptibility and microwave absorption measurements. It has a Tc of not, vert, similar 105K compared to a Tc of not, vert, similar 82K of the corresponding n=2 member.

Colloid of palladium nanoparticles has been prepared by the Solvated Metal Atom Dispersion (SMAD) method. Reaction of Pd(0) nanopowder obtained upon precipitation from the colloid, with ammonia borane (H3N center dot BH3, AB) in aqueous solutions at room temperature results in the generation of active hydrogen atoms. The active hydrogen atoms either combine with one another resulting in H-2 evolution or diffuse into the Pd lattice to afford PdHx. Diffusion of hydrogen atoms leads to an expansion of the Pd lattice. The diffused hydrogen atoms are distributed uniformly over the entire particle. These features were established using powder XRD and electron microscopy studies. The H-1 NMR spectral studies of PdHx before and after desorption of H-2 revealed that the hydrogen atoms trapped inside Pd lattice are hydridic in nature. Desorption of hydrogen from PdHx did not result in complete reversibility suggesting that some hydrogen atoms are strongly trapped inside the Pd lattice. (C) 2010 Professor T. Nejat Veziroglu. Glass composites of strontium tetraborate, SrB4O7 (SBO) with bismuth vanadate, Bi2VO5.5 (BiV) of the composition (1-x) SBO-x BN (0 less than or equal to x less than or equal to 0.75), have been synthesized. X-ray powder diffraction and electron microscopy indicate as-quenched composites to be amorphous and the annealed samples showed the presence of nanometer sized particles of BN dispersed in the glassy matrix of SBO. The dielectric constant of these composites increases with increase in the volume fraction of BiV, at 300 K. The measured dielectric constant of the composite very nearly obeys the Maxwell's relationship. Freestanding crystalline PbZrO3 nanoparticles with an average size of 15 nm were synthesized by the modified sot gel method and characterized by X-ray diffraction and electron microscopy. Dielectric studies indicated that the paraelectric to antiferroelectric phase transition in the PbZrO3 nanoparticles was observed around at 205 degrees C which was at 233 degrees C for PbZrO3 bulk material. A single leaky ferroelectric loop was observed instead of an antiferroelectric double hysteresis loop which may be because of the defects such as grain boundaries and the pores in the sample because the sample was not sintered at higher temperatures to retain the nanoscale dimension of the PbZrO3 particles.

The envelope protein (E1-E2) of Hepatitis C virus (HCV) is a major component of the viral structure. The glycosylated envelope protein is considered to be important for initiation of infection by binding to cellular receptor(s) and also known as one of the major antigenic targets to host immune response. The present study was aimed at identifying mouse monoclonal antibodies which inhibit binding of virus like particles of HCV to target cells. The first step in this direction was to generate recombinant HCV-like particles (HCV-LPs) specific for genotypes 3a of HCV (prevalent in India) using the genes encoding core, E1 and E2 envelop proteins in a baculovirus expression system. The purified HCV-LPs were characterized by ELISA and electron microscopy and were used to generate monoclonal antibodies (mAbs) in mice. Two monoclonal antibodies (E8G9 and H1H10) specific for the E2 region of envelope protein of HCV genotype 3a, were found to reduce the virus binding to Huh7 cells. However, the mAbs generated against HCV genotype 1b (D2H3, G2C7, E1B11) were not so effective. More importantly, mAb E8G9 showed significant inhibition of the virus entry in HCV JFH1 cell culture system. Finally, the epitopic regions on E2 protein which bind to the mAbs have also been identified. Results suggest a new therapeutic strategy and provide the proof of concept that mAb against HCV-LP could be effective in preventing virus entry into liver cells to block HCV replication.

We have developed a unique single-step chemical vapor deposition (CVD) route for the synthesis of composite thin films containing carbon nanotubes (CNTs). CVD was carried out in an inert ambient using only iron(III) acetylacetonate as the precursor. Depositions were conducted at 700 degrees C on stainless steel substrates in argon ambient in the absence of any reactive gases (such as oxygen, hydrogen). By changing the deposition parameters, especially the pressure in the CVD reactor, the form of carbon deposited could be changed from amorphous to carbon nanotubes, the latter resulting in Fe-Fe3O4-CNT films. X-ray diffraction, Raman spectroscopy, X-ray photoelectron spectroscopy, and electron microscopy together confirm the formation of the three-component composite and illustrate the nanoscale mixing of the components. Elemental iron formed in this process was protected from oxidation by the co-deposited carbon surrounding it. Irrespective of the substrate used, a composite coating with CNTs was formed under optimum conditions, as verified by analyses of the film formed on polycrystalline alumina and silicon substrates.

Melanosomes are a class of lysosome-related organelles produced by melanocytes. Biogenesis of melanosomes requires the transport of melanin-synthesizing enzymes from tubular recycling endosomes to maturing melanosomes. The SNARE proteins involved in these transport or fusion steps have been poorly studied. We found that depletion of syntaxin 13 (STX13, also known as STX12), a recycling endosomal Qa-SNARE, inhibits pigment granule maturation in melanocytes by rerouting the melanosomal proteins such as TYR and TYRP1 to lysosomes. Furthermore, live-cell imaging and electron microscopy studies showed that STX13 co-distributed with melanosomal cargo in the tubular-vesicular endosomes that are closely associated with the maturing melanosomes. STX family proteins contain an N-terminal regulatory domain, and deletion of this domain in STX13 increases both the SNARE activity in vivo and melanosome cargo transport and pigmentation, suggesting that STX13 acts as a fusion SNARE in melanosomal trafficking pathways. In addition, STX13-dependent cargo transport requires the melanosomal R-SNARE VAMP7, and its silencing blocks the melanosome maturation, reflecting a defect in endosome-melanosome fusion. Moreover, we show mutual dependency between STX13 and VAMP7 in regulating their localization for efficient cargo delivery to melanosomes.

Background: Aligning similar molecular structures is an important step in the process of bio-molecular structure and function analysis. Molecular surfaces are simple representations of molecular structure that are easily constructed from various forms of molecular data such as 3D atomic coordinates (PDB) and Electron Microscopy (EM) data. Methods: We present a Multi-Scale Morse-Smale Molecular-Surface Alignment tool, MS3ALIGN, which aligns molecular surfaces based on significant protrusions on the molecular surface. The input is a pair of molecular surfaces represented as triangle meshes. A key advantage of MS3ALIGN is computational efficiency that is achieved because it processes only a few carefully chosen protrusions on the molecular surface. Furthermore, the alignments are partial in nature and therefore allows for inexact surfaces to be aligned. Results: The method is evaluated in four settings. First, we establish performance using known alignments with varying overlap and noise values. Second, we compare the method with SurfComp, an existing surface alignment method. We show that we are able to determine alignments reported by SurfComp, as well as report relevant alignments not found by SurfComp. Third, we validate the ability of MS3ALIGN to determine alignments in the case of structurally dissimilar binding sites. Fourth, we demonstrate the ability of MS3ALIGN to align iso-surfaces derived from cryo-electron microscopy scans. Conclusions: We have presented an algorithm that aligns Molecular Surfaces based on the topology of surface curvature. A study of the two-dimensional flow pattern of particles in consolidation process under explosive-implosive shock waves has been performed to further understand the mechanism of shock-wave consolidation of metal powder, in which bunched low-carbon steel wires were used instead of powder. Pressure in the compact ranges from 6 to 30 GPa. Some wires were electroplated with brass, some pickled. By this means, the flow pattern at particle surfaces was observed. The interparticle bonding and microstructure have been investigated systematically for the consolidated specimens by means of optical and electron microscopy, as well as by microhardness. The experimental results presented here are qualitatively consistent with Williamson's numerical simulation result when particle arrangement is close packed, but yield more extensive information. The effect of surface condition of particle on consolidation quality was also studied in order to explore ways of increasing the strength of the compacts. Based on these experiments, a physical model for metal powder shock consolidation has been established.

Ao longo do século XX, poucos estudos de dendrocronologia foram desenvolvidos com espécies de ambientes tropicais, em função da crença de que as condições climáticas nessas regiões não apresentavam variações suficientemente marcantes e regulares para induzir um ritmo anual de crescimento radial. A realização de trabalhos sobre esse tema nas últimas décadas revelou que a formação de anéis de crescimento anuais nos trópicos pode estar associada a fatores diversos, como: existência de estação seca bem definida, ocorrência de inundações sazonais, respostas ao comportamento fenológico, respostas ao fotoperíodo e a ritmos endógenos. O presente estudo tem por objetivo compreender a dinâmica de crescimento radial de uma espécie da Mata Atlântica se desenvolvendo em ambiente natural. Para tanto, propôs-se: i) investigar a periodicidade da atividade cambial e dos fatores que a influenciam; ii) estimar a idade e taxa de crescimento diamétrico e iii) correlacionar os fatores ambientais com os anéis de crescimento, em indivíduos de Cedrela odorata L. Para o estudo da atividade cambial, foram obtidas amostras de caule a 1,30 m do solo, contendo periderme, faixa cambial e xilema e floema secundários, por métodos não destrutivos. A fenologia vegetativa e a frutificação dos indivíduos amostrados foram acompanhadas durante todo o período do experimento. O material coletado foi processado segundo técnicas usuais em Anatomia Vegetal e analisado sob microscopia óptica e de fluorescência. Os dados de fotoperíodo, precipitação, temperatura e fenologia vegetativa foram correlacionados à atividade cambial. Para o estudo dos anéis de crescimento, as coletas também foram realizadas a 1,30 m do solo, por meio de sonda de Pressler. As amostras obtidas foram polidas e analisadas sob microscópio estereoscópio, para demarcação e aferição do número de anéis de crescimento, e a largura dos anéis foi mensurada para a determinação das taxas de crescimento radial. A série histórica de temperatura e precipitação foi correlacionada à cronologia dos anéis de crescimento. Os resultados indicaram que a atividade cambial segue um ritmo anual de crescimento, correlacionado à sazonalidade do fotoperíodo, da precipitação e da fenologia vegetativa. A análise dos anéis de crescimento permitiu estimar a idade dos indivíduos e determinar a taxa média de incremento e as taxas de incremento diamétrico acumulado e incremento médio anual para a espécie no sítio de estudo. Os dados de incremento radial evidenciaram a ausência de relação entre a idade e o diâmetro das árvores. A análise da variação na largura dos anéis não apresentou correlações significativas com os fatores climáticos analisados.

Neste trabalho, foram utilizadas três resinas reticuladas comerciais de troca iônica e caráter ácido à base de estireno e divinilbenzeno: AmberliteGT73 da Rohm and Haas Co. com grupo tiol, Lewatit VPOC1800 da Bayer Co. com grupo sulfônico e Amberlyst 15WET da Rohm and Haas Co. também com grupo sulfônico. As citadas resinas comerciais foram escolhidas por apresentarem grande capacidade de troca iônica, estabilidade e grupos funcionais de interesse para a introdução de íons Ag+. As resinas foram tratadas com ácido clorídrico para garantir as formas ácidas de seus grupos funcionais e em seguida a redução dos íons Ag+, provenientes de solução de nitrato de prata, foi realizada in situ pela hidroxilamina em presença de solução protetora de colóide composta por 2-hidróxi-etil-celulose e gelatina 1:1. Alguns parâmetros foram modificados durante a redução dos íons Ag+ a Ag0, como por exemplo, o tempo de adição da solução redutora de hidroxilamina, a solução utilizada para controle do pH, e condições do repouso após o controle do pH. Após a incorporação das nanopartículas de prata, tanto as resinas comerciais quanto o produto final foram caracterizados por titulometria, fluorescência de raios-x, análise termogravimétrica, análise elementar, grau de inchamento, difração de raios-x, microscopias ótica e eletrônica. A avaliação da atividade biocida foi realizada através do método da contagem em placas utilizando-se uma cepa de Escherichia Coli ATCC25922TM em concentrações de 103 a 107 células/mL. Todos os compósitos obtidos mostraram atividade bactericida significante, sendo que foi possível perceber que a ação bactericida dos compósitos está relacionada com a presença de prata na forma metálica e a características como tamanho, formato e dispersão das partículas na matriz polimérica. Para efeito de comparação, foram realizados ensaios bactericidas com os copolímeros de partida e assim foi comprovado que a ação bactericida pôde ser atribuída somente às nanopartículas de prata

A intervenção farmacológica pode minimizar ou até mesmo reverter o remodelamento adverso em órgãos num modelo de síndrome metabólica. Este trabalho teve como objetivo avaliar os efeitos das monoterapias e associações medicamentosas sobre a morfologia do tecido adiposo, remodelamento hepático e pancreático em camundongos C57bl/6 alimentados com dieta very high-fat. Camundongos C57bl/6 machos foram alimentados com dieta very high-fat (HF, 60% de lipídios) ou dieta padrão (SC, 10% de lipídios) por 10 semanas, quando foram iniciados os tratamentos: HF-T (HF + Telmisartana, 5.2mg/Kg/dia), HF-S (HF + Sitagliptina, 1.08g/Kg/dia), HF-M (HF + Metformina, 310.0mg/Kg/dia) e as associações medicamentosas HF-TM, HF-TS e HF-SM. Os grupos tratados também tiveram livre acesso à dieta high fat e os tratamentos duraram 6 semanas. Técnicas morfométricas, estereológicas, imunohistoquímicas, ELISA, western blotting e microscopia eletrônica foram utilizadas. A dieta high-fat causou sobrepeso, intolerância oral à glucose, hiperinsulinemia, hipertrofia de ilhotas e adipócitos, grau 2 de esteatose hepatica (<33%) redução da expressão de PPAR-alfa e de GLUT-2, concomitante com aumento da expressão de SREBP-1 no grupo HF (P<0.0001). Por outro lado, todos os tratamentos resultaram em perda de peso significativa, reversão da resistência à insulina, hipertrofia de ilhotas e adipócitos e alívio da esteatose hepática. Somente os grupos HF-T e HF-TS apresentaram massa corporal similar ao grupo SC ao final do experimento, sendo que o ultimo também apresentou reversão da esteatose hepática. O aumento da expressão do PPAR-alfa paralelamente ao decréscimo da expressão do SREBP-1 explica os achados favoráveis para o fígado. A normalização do tamanho do adipócito foi consistente com os níveis maiores de adiponectina e com a redução dos níveis de TNF-alfa (P<0.0001) nos grupos tratados.Todos os tratamentos foram eficazes para controlar a síndrome metabólica. Os melhores resultados foram alcançados com a telmisartana e sitagliptina como monoterapias ou como associação entre essas, combinando a ativação parcial do PPAR-alfa no fígado com a extensão do tempo de ação das incretinas
