Recent studies have shown that ingestion by the army worm Spodoptera frugiperda of Cry1Ac toxin from Bt cotton promotes histochemical and ultrastructural changes in the digestive cells of the predatory pentatomid bug Podisus nigrispinus. Therefore, mindful of the changes in the midgut of the predator, which represents the first line of defence in this insect, our aim was to test the hypothesis that the consumption of Bt cotton-fed S. frugiperda by P. nigrispinus might lead to alterations in components of the immune system of P. nigrispinus. The Cry1Ac toxin level in the leaves of Bt cotton, nitric oxide, phenoloxidase activity, and total proteins were quantified by ELISA. Total and differential hemocyte counts were evaluated, and hemocyte ultrastructure analysis was undertaken. We found that ingestion of the prey fed daily with approximately 23 ± 0.70 ng g-1 Cry1Ac by wet weight of leaves, and expressed by the Bt cotton, induces small ultrastructural changes in the predator's granulocytes and plasmatocytes. However, these changes did not affect the total number and differential and humoral variables analyzed for the bug's hemocytes. © 2013 Copyright Taylor and Francis Group, LLC.

Objectives: To present some immunological aspects of fresh-frozen allogeneic bone grafting for lateral bone augmentation, based on the quantitative evaluation of IL-10, IL-1β, IFN- γ and TNF- α in patients sera. Material and methods: Thirty-three partially or totally edentulous patients received fresh-frozen allogeneic bone (AL - 20 patients) or autologous bone onlay block grafts (AT - 13 patients) prior to oral implant placement. Blood samples were collected from each patient at various time-points during a 6 month-period (baseline, 14, 30, 90 and 180 days postoperatively). Quantitative evaluation of IL-10, IL-1β, IFN- γ and TNF- α was performed by enzyme linked immunosorbent assay (ELISA). Results: For all evaluated markers and at all evaluated periods, inter-group comparisons showed no statistically significant differences between the groups, while the observed values were within normal levels. For AL-treated patients, intra-group evaluation showed statistically significant increase of TNF-α from baseline to 90 (P < 0.001) and 180 (P < 0.01) days, and from 14 to 90 (P < 0.01) and 180 (P < 0.05) days. IFN- γ showed intercalated results, with a decrease from baseline to 14 days (P < 0.05), and increase from 14 to 90 days (P < 0.001) and 180 (P < 0.05) days. No differences between the periods of evaluation were found for the AT group. Conclusions: AL grafting for lateral bone augmentation, similar to AT grafting, does not seem to challenge the immune system significantly. © 2012 John Wiley & Sons A/S.

Development of vaccines against canine visceral leishmaniasis (CVL) may provide a prophylactic barrier, but antibody response detected by standard diagnostic techniques may not separate vaccinated from naturally infected dogs. Moreover, anti-Leishmania antibody levels in vaccinated dogs may be detectable for months. Accordingly, the aim of the present study was to comparatively evaluate an in-house ELISA with three serological tests officially adopted by the Brazilian Ministry of Health for the diagnosis of CVL in dogs vaccinated with Leishmune®. A total of 18 mongrel dogs were submitted to a complete protocol of the vaccine, monitored and evaluated in 5 times (T0-T4) up to 180 days after T0. Twenty-one days after the first dose (T1), 50% of the dogs were seropositive by the in-house ELISA and 5.5% by IFAT, while by the official ELISA and DPP® CVL rapid test all dogs tested negative. At time T2, 42 days after of the first dose, 100%, 83.3%, 11.1%, and 5.5% of the dogs were seropositive by the in-house ELISA, IFAT, official ELISA kit and the DPP® CVL rapid test, respectively. Ninety days after the first dose (T3), 100%, 83.3%, 72.2% and 33.3% of the dogs were seropositive by the in-house ELISA, official ELISA kit, IFAT, and the DPP® CVL rapid test, respectively. Finally, at time T4, 88.8%, 33.3%, 11.1% and 5.5% of the dogs were seropositive by the in-house ELISA, official ELISA kit, DPP® CVL rapid test and IFAT, respectively. In conclusion, dogs vaccinated with Leishmune® cross-react by an in-house ELISA and by the three official Brazilian serological tests for the diagnosis of canine visceral leishmaniasis up to six months after the first vaccine dose, and may be mistakenly diagnosed and removed. © 2013 Elsevier B.V.

Parametric VaR (Value-at-Risk) is widely used due to its simplicity and easy calculation. However, the normality assumption, often used in the estimation of the parametric VaR, does not provide satisfactory estimates for risk exposure. Therefore, this study suggests a method for computing the parametric VaR based on goodness-of-fit tests using the empirical distribution function (EDF) for extreme returns, and compares the feasibility of this method for the banking sector in an emerging market and in a developed one. The paper also discusses possible theoretical contributions in related fields like enterprise risk management (ERM). © 2013 Elsevier Ltd.

A infecção pelo Vírus da imunodeficiência humana 1 (HIV -1) em associação com a do Vírus da hepatite C (HCV) representa, atualmente, uma comorbidade que pode interferir principalmente na história natural da hepatite C. Este trabalho tem como objetivo descrever aspectos demográfico, clínico e laboratorial, incluindo exame histopatológico de pacientes coinfectados HIV/RCV. No período de agosto de 2004 a dezembro de 2006, 36 pacientes coinfectados, foram selecionados para o estudo. Noventa e dois por cento desses pacientes eram procedentes de Belém, com média de idade de 42 anos. Entre as principais informações demográficas da população estudada, foram identificados 72,52% solteiros, 83,5% do sexo masculino e 61,1% relataram ser heterossexuais. Entre os fatores de risco para o HCV o uso de drogas ilícitas injetáveis foi identificado em 41,7% dos casos, o uso de cocaína intranasal foi relatado por 38,9% dos pacientes, e o compartilhamento de seringas e material pessoal, em 38,9% dos casos. A história de etilismo em 77,8% e o uso de TARV foram os possíveis fatores agravantes mais frequentes para a doença hepática. Apenas um paciente apresentou sinais clínicos de insuficiência hepática crônica. Entre os testes bioquímicos hepáticos, a mediana de ALT e AST foi de 68UI/L e 61UI/L, respectivamente. Os níveis de linfócitos T CD4+ apresentaram mediana de 327 células/mm³, a carga viral do HIV com mediana de 2,53 logl0 cópias/mL (ep=0,34), carga viral do HCV com mediana de 5,9 log10UI/mL. O genótipo 1 do HCV foi o mais frequente (58,82%). Cinqüenta e sete por cento dos pacientes submetidos à biópsia hepática apresentavam fibrose de moderada a severa, e 11% não apresentaram fibrose pela classificação MET AVIR. Houve associação entre níveis de linfócitos T CD4+ e níveis de ALT e de AST (p=0,0009 e p=0,0002, respectivamente), assim como associação entre genótipo 1 do HCV e HCV-RNA maior ou igual a 6 log10 UI/mL (p=0.0039). Foi observada também associação entre HCV-RNA e HIV-RNA (p=0,039). Os pacientes apresentam estado geral bom, imunologicamente estáveis, sem sinais de descompensação hepática, mas com alterações estruturais hepáticas importantes, sendo portanto bons candidatos à terapia antiviral para o HCV. Futuros estudos, talvez de caso controle, com casuística maior são necessários para melhor entendimento da co-infecção HIV/HCV.
