Soil community genomics or metagenomics is employed in this study to analyze the evolutionary related - ness of mangrove microbial community. The metagenomic DNA was isolated from mangrove sediment and 16SrDNA was amplified using universal primers. The amplicons were ligated into pTZ57R/T cloning vector and transformed onto E. coli JM109 host cells. The recombinant plasmids were isolated from positive clones and the insert was confirmed by its reamplification. The amplicons were subjected to Amplified Ribosomal DNA Restriction Analysis (ARDRA) using three different tetra cutter restriction enzymes namely Sau3A1, Hha1 and HpaII. The 16SrDNA insert were sequenced and their identity was determined. The sequences were submitted to NCBI database and accession numbers obtained. The phylo - genetic tree was constructed based on Neighbor-Joining technique. Clones belonged to two major phyla of the bacterial domain, namely Firmicutes and Proteobacteria, with members of Firmicutes predominating. The microbial diversity of the mangrove sediment was explored in this manner.
Continuation and discontinuation of local institution in community based natural resource management

Currently the push toward frontier areas, which until twenty years ago were still largely untouched by commercial agriculture, is taking place on a massive scale. This push is being driven not the least by global economic developments, such as the price increase of agriculture commodities like coffee and cocoa. In most cases the indigenous communities become trapped between the state monopoly in natural resource management and the competition for resources by external actors. In this processes the indigenous communities start to lose their access to resources. Another victim in this process is the environment where the natural resources are imbedded. International and national organizations working to conserve environment have became conscious of the important role that indigenous people could fulfill as partners in this endeavour. This partnership in struggle has produced a new discourse on the relationship between indigenous people and their environment. As a further consequence, programs were set up to develop what became known as Community Based Natural Resource Management (CBNRM) with its numerous variations. Based on a case study in a village on the eastern border of the Lore Lindu National Park in Central Sulawesi, this study questioned the basic assumption behind the concept of Community Based Natural Resource Management (CBNRM). Namely the assumption that communities living at the margin of forest are socially and culturally homogenous, still more or less egalitarian, and basically living in harmony with their natural environment. This study was inspired by the persistent critique – although still a minority – on the basic assumption the CBNRM from academicians and practitioners working through the Entitlement perspective. Another inspiration was the mounting critique toward the participatory approach. In its effort the study explore further the usefulness of certain approaches. One of the approach much relied on in this study was the local history of the community studied, through exerting oral and local written documents on local history, legends and local stories. These sources proofed quite capable in bringing the local history into the light. Another was the actor oriented approach, which later came to be supported by the concept of Social Pool Resources. The latter concept proofed to be useful as analytical instrument to integrate social institutions and the common pool resources, as a field of action for the different actors as human agencies.

This is an empirical study with theoretical interpretation and elaboration simultaneously on the migration process and the related spatial development in contemporary China. In so doing, there is always a combination of series of studies of the modernization of the migrants themselves with accumulation of forms of capital and changes of lebenswelt (life world) as well as the regions of their origins by the effective use of the gained resources from outgoing migration and remigration. With great efforts made to put the issues together for analysis, the author has taken three approaches to the study based on the political and economic institutional arrangements, the field work data and the elaboration of respective findings. First, as the analytical parts of the institutional changes, which have gone through the whole research, many of the policies from state level to townships involved in the migration, remigration and spatial development have been interpreted with Chinese political and cultural insight. The making of these, as the means of understanding the contexts of macro level and micro level cases is served as key linkages between scholarly imagination and social reality. Indeed most of the discussions made to explain the phenomena such as the sudden upsurge of migration flows, the emergence of three generations, the strong and weak trends of remigration as well as the related spatial development planning, etc are mainly due to the domination, at least the impact of governments decision-making in spite of growing market functioning in often operative manners. Secondly, case studies of the effects of migration and remigration are carried out between the years of 1995 and 2005 in the costal urban regions as designations and the interior rural regions as origins. Conducted mainly by the author, the cases drawn in the research focus on the process of migration with an accumulation of forms of capital away from home and the effective use of the resources flowing back to home areas. As a result, ways of accumulation and utilization of the economic, social and cultural capital are described and interpreted in terms of the development and modernization of both the migrants themselves and the regions where they come out from or move to in the future. Thirdly, in accordance with the findings generated from the cases, the author proposes in the final chapter an important argumentation as conclusion that the duel social-economic structure will inevitably be broken up and reformulated with flows of migrants and forms of capital they possess as types of future spatial development that will be put into practice. With scenarios and all the other conclusions worked out in the end, the research concludes that the pluralistic spatial development in the condition of constant space flows between regions can be a decisive line of thinking in the process of urbanization, industrialization and modernization in the long run in the future. Since this is an exploratory study of the past and present, the author has left some space open for academic debates and put forward suggestions on the inclusion of future research before implementing policies necessary for migration associated spatial practice and development.

A conceptual information system consists of a database together with conceptual hierarchies. The management system TOSCANA visualizes arbitrary combinations of conceptual hierarchies by nested line diagrams and allows an on-line interaction with a database to analyze data conceptually. The paper describes the conception of conceptual information systems and discusses the use of their visualization techniques for on-line analytical processing (OLAP).

Die Verordnung des Europäischen Rates (EC) 834/2007 erkennt das Recht des Konsumenten auf eine Entscheidung basierend auf vollständiger Information bezüglich der enthaltenen Zutaten im Produkt und deren Herkunft (Qualität der Verarbeitung). Die primäre Kennzeichnungsverordnung betont „organische“ Produktionsstandards ebenso wie die Notwendigkeit zur Kontrolle und Aufsicht. Jedoch ist zurzeit keine validierte Methode zur analytischen Diskriminierung zwischen „organischer“ und „konventioneller“ Herkunft von angebotenen Lebensmitteln verfügbar. Das Ziel der Dissertationsarbeit war die Überprüfung der Möglichkeit mit ausgewählten analytischen und holistischen Methoden zwischen organisch und konventionell angebautem Weizen objektiv zu unterscheiden. Dies beinhaltete die Bestimmung des Gesamtstickstoff (Protein) nach Dumas, zweidimensionale Fluoreszenzdifferenz Gelelektrophorese (2D DIGE) und die Kupferchloridkristallisation. Zusätzlich wurde die Anzahl der Körner pro Ähre (Kornzahl) bestimmt. Alle Bestimmungen wurden an rückverfolgbaren in den Jahren 2005 – 2007 in Belgien gesammelten Proben des Winterweizen (Triticum aestivum L. cv. Cubus) durchgeführt. Statistisch signifikante (p < 0.05) Unterschiede wurden innerhalb der untersuchten Probengruppen sowohl in der Kornzahl, dem Gesamtsticksoff (Eiweißgehalt), als auch in der Gesamtausbeute gefunden, wobei in den meisten Fällen die konventionellen Proben höhere Kornzahlen und Gesamtsticksoff (Eiweißgehalte) aufwiesen. Eine mit der 2D DIGE kompatible Probenvorbereitungsmethode für Winterweizen wurde entwickelt und auf einen internen Winterweizenstandard sowie die entsprechenden Proben angewendet. Die organischen Proben waren im Vergleich mit den konventionellen Gegenstücken in allen Fällen durch eine kleinere Anzahl von signifikant (p < 0.05) stärker exprimierten Proteinspots gekennzeichnet. Gewisse Tendenzen in Richtung der Bevorzugung bestimmter Regionen von stärker ausgeprägten Proteinspots auf aufeinanderfolgenden 2D Abbildungen in Abhängigkeit von der landwirtschaftlichen Methode konnten zwar beobachtet werden, jedoch konnte kein universelles Markerprotein zur Unterscheidung von konventionell und biologisch angebautem Winterweizen identifiziert werden. Die rechnergestützte Verarbeitung der digitalisierten Kristallisierungsbilder mittels multivariater statistischer Analyse und der Regression partieller kleinster Quadrate ermöglichte eine 100%ig korrekte Vorhersage der landwirtschaftlichen Methode unbekannter Proben sowie der Beschreibung der Kristallisierungsbilder. Diese Vorhersage bezieht sich nur auf den hier verwendeten Datensatz (Proben einer Sorte von drei Standorten über zwei Jahre) und kann nicht ohne weiteres übertragen (generalisiert) werden. Die Ergebnisse deuten an, dass die Quantifizierung der beschriebenen Parameter ein hohes Potential zur Lösung der gestellten Aufgabe besitzt.
