The present investigation dealt with the climatic and some physico-chemical conditions of the Kaptai lake with respect to their monthly variation. Air temperature was found always higher than water temperature. Vertical variation in temperature (0.8-4.7°C) was observed in all months. The water level fluctuates appreciably throughout the year. Wide seasonal fluctuations were also noted in water transparency. The lake was found to be slightly hard and alkaline pH. Dissolved oxygen (DO) (6.4-9.1 mg/l) and free carbondioxide (4.7- 6.0 mg/l) contents showed favourable condition for aquatic lives. DO at different depth has shown no wide variation (1.0-2.4 mg/1). Conductivity ranged between 91.9±7.1 and 106.4 ±5.2 mS/cm.

This communication reports the changes in physical, organoleptic and biochemical characteristics of prawn meat dip-treated with alkaline and neutral solutions of polyphosphates during frozen storage. Results are presented on changes in thawed and cooked yields, water extractable nitrogen, non-protein nitrogen, free amino-nitrogen, salt solubility, myosin and moisture in the muscle and loss of soluble nitrogenous constituents in thaw drip during frozen storage up to seven months. The salt solubility remained unchanged during storage in samples treated with neutral polyphosphate solutions and the organoleptic quality was superior to control sample. It is concluded that dip treatment with neutralized solutions of tripolyphosphate not only maintains correct drained weight and improves cooked yield during prolonged frozen storage but also protects the frozen product from denaturation as measured by the salt solubility of the proteins.

Quantitative assays of trypsin, amylase and alkaline phosphatases were made in relation to age and food during the larval development of the Indian major carp Catla catla. The responses of all the test enzymes to age and food were identical. No enzymes were detected from the fertilized eggs. Detectable amount of enzymes were first observed in the first day old hatchlings. All the test enzymes in the group fed normal feed tended to rise gradually with advancement of age till day 22 after which an asymptotic level was attained. Absence of food throughout the rearing period caused the enzymatic activity of the larva to remain at the lowest level throughout. When starvation was followed by feeding, enzymatic activity in the former group was consistently higher than that of latter, suggesting that feeding activity was primarily responsible in maintaining the enzymatic activity of carp larva. The enzymatic activity of zooplankton was significantly higher than carp larva till day 6 to 12 after which the latter exceeded the former implying that carp larva during development utilizes the exogenous enzymes of zooplankton.

During ecophysiological investigations on an intertidal gastropod, Nerita oryzarum (Recluz), of Mumbai shore, various biochemical changes could be recorded. Glycogen and lipid contents of N. oryzarum were found to decrease, whereas, water content increased with decreasing salinity. The rate of oxygen consumption declined with the decrease in salinity and also in highly acidic (pH 2) as well as highly alkaline (pH 10) sea water. The observed variations in the rate of oxygen consumption and changes in biochemical composition in the animal with changes in salinity, pH and temperature are probably the process of physiological and biochemical adjustments to the fluctuating environmental conditions in the intertidal region.

Physical characteristics of waterlogged and saline soils of five districts, namely Rohtak, Faridabad, Gurgaon, Sonipat and Jhajjar, of Haryana were studied to evaluate the aquaculture potential. The soil samples from 76 sites were procured and analysed for pH, electrical conductivity, soil particles and water retention capacity, since the knowledge of these parameters is essential for the preliminary evaluation of a site for aqua-farming. Six soil types were identified and clay-loam was observed to be the maximum at 45 (59.21%) sites, followed by silty-clay loam at 14 (18.42%), sandy-clay loam at six (7.89%), silty-loam at four (5.26%), sandy-loam at four (5.26%) and loam at three (3.94%) sites. The pH of soil indicated moderate to high alkaline conditions ranging between 8.01and 9.00 at 53 (69.73%) sites, 9.01 and 10.00 at 17 (22.36%) sites, and low between 7.01 and 8.00 at six (7.89%) sites. The electrical conductivity was found to range between <1 and 10 mmho cmˉ¹ with the value of <1.0 mmho cmˉ¹ at 36 (47.36%) sites. The water retention capacity was observed mostly to be moderate. The paper describes the interrelationship between these parameters with reference to the suitability for aqua-farming.

In the present study, an attempt was made to explore the benefits of polyphosphate for enhancement of dried prawn (Parapenaeopsis stylifera) quality commercially known as "sode" in Maharashtra coast. Dip treatment in polyphosphate solution at different concentrations (viz., 3, 5, 7 and 10%) was given to pealed and no deveined P. stylifera for different durations (viz., 1, 2, 3, 4 and 5 min). Treated prawns were dried and subjected to rehydration capacity test and organoleptic evaluation. Among the different treatments, rehydration capacity was found to increase with the increased duration and concentration of treatment. Tiny prawns treated with sodium tripolyphosphate solution at the rate of 5% concentration for 5 minutes showed an increase in pH from acidic to alkaline, and had better quality with respect to rehydration capacity and textural attributes as compared to other concentrations and durations of polyphosphate treatment.

The Austrian-Ceylonese hydrobiological mission studied 38 biotopes; 28 of which contain Odonata. From the Zygoptera the Calopterydoidea seem to be the dominant form (22 habitats), while the Coenagrionoidea are scarcer (11 habitats). The most frequent species was Euphaea splendens (Epallagidae - 16 habitats) followed by Vestolis apicalis nigrescens (Calopterygidae, 8 habitats) and Neurobasis chinensis (Calopterygidae, 6 habitats). From the Anisoptera Zygonyx ceylanica (Libellulidae: Zygonictinae) was the dominant form (8 habitats), but some Libellulinae remain undescribed. The number of species varied greatly between different biotopes. The biotopes containing Odonata are small brooks, in which the pH was mostly on the limit between acid and alkaline reaction. They are fast running waters, situated in most cases on lower or middle elevations, only three species being found in higher elevations (1800-2000 m). Adaptations to fast currents and other factors are described.

Neurotrypsin is one of the extra-cellular serine proteases that are predominantly expressed in the brain and involved in neuronal development and function. Mutations in humans are associated with autosomal recessive non-syndromic mental retardation (MR). We studied the molecular evolution of neurotrypsin by sequencing the coding region of neurotrypsin in 11 representative non-human primate species covering great apes, lesser apes, Old World monkeys and New World monkeys. Our results demonstrated a strong functional constraint of neurotrypsin that was caused by strong purifying selection during primate evolution, an implication of an essential functional role of neurotrypsin in primate cognition. Further analysis indicated that the purifying selection was in fact acting on the SRCR domains of neurotrypsin, which mediate the binding activity of neurotrypsin to cell surface or extracellular proteins. In addition, by comparing primates with three other mammalian orders, we demonstrated that the absence of the first copy of the SRCR domain (exon 2 and 3) in mouse and rat was due to the deletion of this segment in the murine lineage. The density of benthic macro-biota in number over weight of biomass at three sampling stations was 29 units m²/0.614 g m² to 171 units m²/11.346 m² at Station I; 22 units m²/0.410 g m² to 155 units m²/8.717 m² at Station II; 15 units m² 10.321 g m² to 122 units m²/6.793 g m² at Station III. The Caddis fly larvae were the most dominant component and contributed 52.41% in the macro-biota. These animals were abundant in Gaula River when benthic algae were abundant; water is well oxygenated, alkaline and contains sufficient nutrients but observed to be less abundant during high velocity of water, high river depth and higher turbid waters.

The bed Shakla, comprising an average area of 75.0 ha is located in the northeastern region (Brahmanbaria district) of Bangladesh. The study was carried out to assess the ecological aspects of bed ecosystem. Surface run-off and increase inflow of rain water from the upper stretch during monsoon cause inundation and resumption of connection between beel and parent rivers. The range of dissolved oxygen (DO) content ( 4.5-8.9 mg/L) was found congenial for aquatic life. pH was in the alkaline range (7.3-8.5) and free C02 was reletavely high. Lower values of total hardness and total alkalinity indicated less nutrients in the beel water. A wide variation (1.4-27.2 x 103 ceHs/L) in the standing crop of total plankton was recorded during study period of which phytoplankton alone contributed about 90%. Phytoplankton diversity in the beel represented by three groups viz. Chlorophyceae, Myxophyceae and Bacillariophyceae in order of abundance. A total of 52 fish species belonging to 36 genera, 20 families and 1 species of prawn were identified so far from the beel. About l3 types of fishing method were found in operation. Seine nets (moshari berja, ghono berjal) and gill net (current jal) were identified as detrimental gear killing juveniles of different species during post spawning period. Kua fishing was also found harmful due to dewatering nature. A total of 11 species belonging to 11 genera and 10 families of aquatic weeds were identified from the beet The eggs of Macrobrachium Jamarrei were identified into the Najas najas vegetation during April-September.

A specific blood coagulation factor X activator was purified from the venom of Ophiophagus hannah by gel filtration and two steps of FPLC Mono-Q column ion-exchange chromatography. It showed a single protein band both in sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and alkaline polyacrylamide gel electrophoresis. The mol. wt was estimated to be 62,000 in non-reducing conditions and 64,500 in reducing conditions by SDS-PAGE. The isoelectric point was found to be pH 5.6. The enzyme had weak amidolytic activities toward CBS 65-25, but it showed no activities on S-2266, S-2302, thrombin substrate S-2238, plasmin substrate S-2251 or factor Xa substrate S-2222. It had no arginine esterase activity toward substrate benzoylarginine ethylester (BAEE). The enzyme activated factor X in vitro and the effect was absolutely Ca2+ dependent, with a Hill coefficient of 6.83. It could not activate prothrombin nor had any effect on fibrinogen and thus appeared to act specifically on factor X. The procoagulant activity of the enzyme was almost completely inhibited by serine protease inhibitors like PMSF, TPCK and soybean trypsin inhibitor; partially inhibited by L-cysteine. Metal chelator EDTA did not inhibit its procoagulant activity. These results suggest that the factor X activator from O. hannah venom is a serine protease.

A novel plasminogen activator from Trimeresurus stejnegeri venom (TSV-PA) has been identified and purified to homogeneity. It is a single chain glycoprotein with an apparent molecular weight of 33,000 and an isoelectric point of pH 5.2. It specifically activates plasminogen through an enzymatic reaction. The activation of human native GIu-plasminogen by TSV-PA is due to a single cleavage of the molecule at the peptide bond Arg(561)-Val-(562). Purified TSV-PA, which catalyzes the hydrolysis of several tripeptide p-nitroanilide substrates, does not activate nor degrade prothrombin, factor X, or protein C and does not clot fibrinogen nor show fibrino(geno)lytic activity in the absence of plasminogen. The activity of TSV-PA was readily inhibited by phenylmethanesulfonyl fluoride and by p-nitrophenyl-p-guanidinobenzoate. Oligonucleotide primers designed on the basis of the N-terminal and the internal peptide sequences of TSV-PA were used for the amplification of cDNA fragments by polymerase chain reaction. This allowed the cloning of a full-length cDNA encoding TSV-PA from a cDNA library prepared from the venom glands. The deduced complete amino acid sequence of TSV-PA indicates that the mature TSV-PA protein is composed of 234 amino acids and contains a single potential N-gIycosylation site at Asn(1G1). The sequence of TSV-PA exhibits a high degree of sequence identity with other snake venom proteases: 66% with the protein C activator from Aghistrodon contortrix contortrix venom, 63% with batroxobin, and 60% with the factor V activator from Russell's viper venom. On the other hand, TSV-PA shows only 21-23% sequence similarity with the catalytic domains of u-PA and t-PA. Furthermore, TSV-PA lacks the sequence site that has been demonstrated to be responsible for the interaction of t-PA (KHRR) and u-PA (RRHR) with plasminogen activator inhibitor type 1.

Characterization of three fibrinogenolytic enzymes from Chinese green tree viper (Trimeresurus stejneger ) venom. Toxicon 36, 457-467, 1998.-From the venom of Chinese green tree viper (Trimeresurus stejnegeri), three distinct fibrinogenolytic enzymes: stejnefibrase-l, stejnefibrase-2 and stejnefibrase-3, were purified by gel filtration, ion-exchange chromatography and reverse-phase high-performance chromatograghy (HPLC). SDS-PAGE analysis of those three enzymes showed that they consisted of a single polypeptide chain with mel. wt of -50 000, 31 000 and 32 000, respectively. Like TSV-PA (a specific plasminogen activator) and stejnobin (a fibrinogen-clotting enzyme) purified from the same venom, stejnfibrase-1, -2 and -3 were able to hydrolyze several chromogenic substrate. On the other hand, different from TSV-PA. and stejnobin, stejnefibrase-l, -2 and -3 did not activate plasminogen and did not possess fibrinogen-clotting activity. The three purified enzymes directly degraded fibrinogen to small fragments and rendered it unclottable by thrombin. Stejnefibrase-2 degraded preferentially BE-chain while stejnefibrase-l and -3 cleaved concomitantly Ax and B beta-chains of fibrinogen. None of these proteases degraded the gamma-chain of fibrinogen. When correlated with the loss of clottability of fibrinogen, the most active enzyme was stejnefibrase-l. The activities of the three enzymes were inhibited by phenylmethylsulfonyl fluoride (PMSF) and p-nitrophenyl-p-guanidinobenzoate (NPGB), indicating that like TSV-PA and stejnobin, they are venom serine proteases. A novel peptide inhibitor (OGTI) of serine protease with a molecular weight of 1949.8, was purified from the skin secretion of the frog, Odorrana grahami. Of the tested serine proteases, OGTI only inhibited the hydrolysis activity of trypsin on synthetic chromogenic substrate. This precursor deduced from the cDNA sequence is composed of 70 amino acid residues. The mature OGTI contains 17 amino acid residues including a six-residue loop disulfided by two half-cysteines (AVNIPFKVHFRCKAAFC). In addition to its unique six-residue loop, the overall structure and precursor of OGTI are different from those of other serine protease inhibitors. Snake venom Kunitz/BPTI members are good tools for understanding of structure-functional relationship between serine proteases and their inhibitors. A novel dual Kunitz/BPTI serine proteinase inhibitor named OH-TCI (trypsin- and chymotrypsin-dual inhibito
