A repeated DNA element in Xenopus laevis is described that is present in about 7500 copies dispersed throughout the genome. It was first identified in the 5' flanking region of one vitellogenin gene and was therefore named the Vi element. Seven copies are present within the vitellogenin gene region, three of them within introns of the genes A1, A2 and B2, and the other four copies in the gene flanking regions. Four of these copies have been sequenced. The Vi element is bounded by a well-conserved 13 base-pair inverted repeat; in addition, it is flanked by a three base-pair direct repeat that appears to be site-specific. The length of these four copies varies from 112 to 469 base-pairs; however, sequence homology between the different copies is very high. Their structural characteristics suggest that length heterogeneity may have arisen by either unequal recombinations, deletions or tandem duplications. Altogether, the characteristics and properties of the Vi element indicate that it might represent a mobile genetic element. One of the four copies sequenced is inserted close (position -535) to the transcription initiation site of the vitellogenin gene B2 in a region otherwise showing considerable homology with the closely related gene B1. Nevertheless, the presence of the Vi element does not seem to influence significantly the estrogen-controlled expression of gene B2. In addition, three alleles of this gene created by length polymorphism in intron 3 and in the Vi element inserted near the transcription initiation site are described.

Vitellogenin is synthesized under estrogen control in the liver, extensively modified, transported to the ovary, and there processed to the yolk proteins lipovitellin and phosvitin. In the frog Xenopus laevis there are at least four distinct but related vitellogenin genes. The two genes A1 and A2 have a 95 percent sequence homology in their messenger RNA coding regions, and contain 33 introns that interrupt the coding region (exons) at homologous positions. Sequences and lengths of analogous introns differ, and many introns contain repetitive DNA elements. The introns in these two genes that have apparently arisen by duplication have diverged extensively by events that include deletions, insertions, and probably duplications. Rapid evolutionary change involving rearrangements and the presence of repeated DNA suggests that the bulk of the sequences within introns may not have any specific function.

Estrogen receptors regulate transcription of genes essential for sexual development and reproductive function. Since the retinoid X receptor (RXR) is able to modulate estrogen responsive genes and both 9-cis RA and fatty acids influenced development of estrogen responsive tumors, we hypothesized that estrogen responsive genes might be modulated by RXR and the fatty acid receptor (peroxisome proliferator-activated receptor, PPAR). To test this hypothesis, transfection assays in CV-1 cells were performed with an estrogen response element (ERE) coupled to a luciferase reporter construct. Addition of expression vectors for RXR and PPAR resulted in an 11-fold increase in luciferase activity in the presence of 9-cis RA. Furthermore, mobility shift assays demonstrated binding of RXR and PPAR to the vitellogenin A2-ERE and an ERE in the oxytocin promoter. Methylation interference assays demonstrated that specific guanine residues required for RXR/PPAR binding to the ERE were similar to residues required for ER binding. Moreover, RXR domain-deleted constructs in transfection assays showed that activation required RXR since an RXR delta AF-2 mutant completely abrogated reporter activity. Oligoprecipitation binding studies with biotinylated ERE and (35)S-labeled in vitro translated RXR constructs confirmed binding of delta AF-2 RXR mutant to the ERE in the presence of baculovirus-expressed PPAR. Finally, in situ hybridization confirmed RXR and PPAR mRNA expression in estrogen responsive tissues. Collectively, these data suggest that RXR and PPAR are present in reproductive tissues, are capable of activating estrogen responsive genes and suggest that the mechanism of activation may involve direct binding of the receptors to estrogen response elements.

Uma topossequência apresenta solos com características estruturais e morfológicas distintas que podem alterar a susceptibilidade à deformação estrutural em curtas distâncias espaciais. Os objetivos deste estudo foram avaliar a capacidade de suporte de carga e a suscetibilidade à compactação e mensurar o efeito que cargas acima e abaixo da pressão de preconsolidação causam na permeabilidade ao ar de solos numa topossequência da Depressão Central do Rio Grande do Sul, Brasil. Amostras indeformadas foram coletadas nos horizontes de três perfis de Argissolos (Ap, A1, AB, BA, Bt1 e Bt2; Ap, A1, A2, AB, BA, Bt1 e Bt2; e Ap, A1, BA, Bt1 e Bt2) e dois de Gleissolos (A1, A2, Bg; A, Bg e Cg), no longo de uma topossequência. Essas amostras foram submetidas ao teste de compressão uniaxial, sendo avaliada a permeabilidade ao ar antes e após compressão. Na condição de capacidade de campo, os Gleissolos apresentaram menor suscetibilidade à compactação e maior capacidade de suporte de carga que os Argissolos. O teor de argila esteve diretamente correlacionado com a capacidade de suporte de carga. Portanto, foi inversamente proporcional à suscetibilidade à compactação, exceto em condições de elevada umidade do solo quando a água lubrificou as superfícies aumentando a susceptibilidade à compactação de quaisquer solos. Até 25 kPa de carga ao solo não resultou degradação física do solo, mas quando foram aplicadas cargas de 200 kPa, mesmo na condição de capacidade de campo, houve redução significativa do volume de macroporos e porosidade total em níveis inferiores ao limite crítico (0,10 m3 m-3).

Distortions of polyacene polymers are studied within a many-body valence-bond framework using a powerful transfer-matrix technique for the valence-bond (or Heisenberg) model of the system. The computations suggest that the ground-state geometry is either totally symmetric or possibly exhibits a slight (A2 or B2 symmetry) bond-alternation distortion. The lowest-energy (nonsymmetric, in-plane) distortions are the A2 and B2 modes, which, within our approximations, are degenerate.

RESUMO O uso de dejetos líquidos de suínos como fertilizante do solo é uma prática comum na Região Sul do Brasil. Apesar de ter benefícios na reutilização dos dejetos, essa prática apresenta sérios riscos ambientais. Os indicadores microbiológicos de qualidade do solo são bastante sensíveis e permitem o monitoramento das condições do ambiente edáfico. Objetivou-se avaliar a qualidade microbiológica do solo de pastagens com diferentes históricos de uso sucessivo de dejetos líquidos de suínos. Determinaram-se o teor de C da biomassa microbiana, a respiração microbiana do solo, o quociente metabólico (qCO2) e a atividade das enzimas β-glicosidase, urease e hidrólise do diacetato de fluoresceína (FDA), em áreas de pastagem com uso de dejetos de suínos há dois anos (A2) e 14 anos (A14) e em área com mata nativa (MN). O uso sucessivo de dejetos de suínos em pastagem não influenciou o C da biomassa e a respiração microbiana do solo, que variaram conforme a época de coleta. O qCO2 não foi influenciado pelo uso de dejetos de suínos no solo; a atividade enzimática do solo foi influenciada pelo uso de dejetos de suínos, sendo que a urease e a FDA foram sensíveis na detecção de diferenças na atividade dos solos com uso de dejetos de suínos, enquanto a β-glicosidase não permitiu a diferenciação entre as áreas estudadas.

RESUMO A matéria orgânica do solo é uma importante fonte primária de nutrientes às plantas e influencia a infiltração, retenção de água, suscetibilidade à erosão e agregação do solo. No entanto, esse processo está condicionado à qualidade e quantidade da matéria orgânica aportada. O objetivo deste trabalho foi avaliar a influência do desastre ambiental causado pela chuva em áreas de produção de oleráceas na região Serrana do Rio de Janeiro. Foram selecionadas as seguintes áreas: A1: sem impacto (testemunha); A2: soterramento total e cultivo de aveia + ervilhaca; A3: transbordamento do rio e cultivo de milheto + girassol; A4: transbordamento do rio e grande deposição de areia com cultivo de aveia-preta; A5: transbordamento do rio com grande deposição de areia e sem cultivo; e A6: transbordamento do rio em pequena escala e sem cultivo. As propriedades edáficas analisadas foram: textura; densidade do solo (Ds); densidade de partículas (Dp); diâmetro médio ponderado (DMP) e diâmetro médio geométrico (DMG) de agregados estáveis em água; pH(H2O), Al3+, Ca2++Mg2+, Na+, H+Al, K, P e carbono orgânico total (COT); e frações granulométricas e químicas da matéria orgânica. Os maiores valores de DMP e DMG foram observados nas áreas A3 e A6, que seguem a mesma tendência dos maiores teores de argila. Os teores de COT e suas frações químicas e físicas apresentaram diferenças em relação à testemunha com os menores valores verificados nas áreas que foram submetidas ao impacto pelo transbordamento do rio e grande deposição de areia. As áreas que foram impactadas pelo soterramento com predomínio da fração argila apresentaram valores semelhantes ou superiores aos da testemunha. As variáveis foram selecionadas e avaliadas por meio da análise de componentes principais, que evidenciou distinção entre as áreas estudadas, com separação entre elas associada à textura do solo e densidade de partículas.

Many new types of vaccines against infectious or malignant diseases are currently being proposed. Careful characterization of the induced immune response is required in assessing their efficiency. While in most studies human tumor antigen-specific T cells are analyzed after in vitro re-stimulation, we investigated these T cells directly ex vivo using fluorescent tetramers. In peripheral blood lymphocytes from untreated melanoma patients with advanced disease, a fraction of tumor antigen (Melan-A/MART-1)-specific T cells were non-naive, thus revealing tumor-driven immune activation. After immunotherapy with synthetic peptides plus adjuvant, we detected tumor antigen-specific T cells that proliferated and differentiated to memory cells in vivo in some melanoma patients. However, these cells did not present the features of effector cells as found in cytomegalovirus specific T cells analyzed in parallel. Thus, peptide plus adjuvant vaccines can lead to activation and expansion of antigen specific CD8(+) T cells in PBL. Differentiation to protective CD8(+) effector cells may, however, require additional vaccine components that stimulate T cells more efficiently, a major challenge for the development of future immunotherapy.

In oviparous vertebrates vitellogenin, the precursor of the major yolk proteins, is synthesized in the liver of mature females under the control of estrogen. We have established the organization and primary structure of the 5' end region of the Xenopus laevis vitellogenin A2 gene and of the major chicken vitellogenin gene. The first three homologous exons have exactly the same length in both species, namely 53, 21 and 152 nucleotides, and present an overall sequence homology of 60%. In both species, the 5'-non-coding region of the vitellogenin mRNA measures only 13 nucleotides, nine of which are conserved. In contrast, the corresponding introns of the Xenopus and the chicken vitellogenin gene show no significant sequence homology. Within the 500 nucleotides preceding the 5' end of the genes, at least six blocks with sequence homologies of greater than 70% were detected. It remains to be demonstrated which of these conserved sequences, if any, are involved in the hormone-regulated expression of the vitellogenin genes.

Cholesterol regulates plasma membrane (PM) association and functioning of syntaxin-4 and soluble N-ethylmaleimide-sensitive fusion protein 23 (SNAP23) in the secretory pathway. However, the molecular mechanism and cellular cholesterol pools that determine the localization and assembly of these target membrane SNAP receptors (t-SNAREs) are largely unknown. We recently demonstrated that high levels of annexin A6 (AnxA6) induce accumulation of cholesterol in late endosomes, thereby reducing cholesterol in the Golgi and PM. This leads to an impaired supply of cholesterol needed for cytosolic phospholipase A2 (cPLA2) to drive Golgi vesiculation and caveolin transport to the cell surface. Using AnxA6-overexpressing cells as a model for cellular cholesterol imbalance, we identify impaired cholesterol egress from late endosomes and diminution of Golgi cholesterol as correlating with the sequestration of SNAP23/syntaxin-4 in Golgi membranes. Pharmacological accumulation of late endosomal cholesterol and cPLA2 inhibition induces a similar phenotype in control cells with low AnxA6 levels. Ectopic expression of Niemann-Pick C1 (NPC1) or exogenous cholesterol restores the location of SNAP23 and syntaxin-4 within the PM. Importantly, AnxA6-mediated mislocalization of these t-SNAREs correlates with reduced secretion of cargo via the SNAP23/syntaxin-4¿dependent constitutive exocytic pathway. We thus conclude that inhibition of late endosomal export and Golgi cholesterol depletion modulate t-SNARE localization and functioning along the exocytic pathway.

The Formica rufa group (red wood ants) currently includes six species. Nevertheless, during previous work based on molecular markers, we showed the existence of one population morphologically identified as F. lugubris, but genetically different from all other analysed populations of this species. This population could represent a cryptic species within the Swiss National Park and has been provisionally named Formica lugubris-A2. To verify our hypothesis, we conducted a behavioural test based on the ability of ants to recognize pupae of their own species when compared to those of another species. The three red wood ant species present in the Swiss National Park (F. lugubris, F. paralugubris and F. aquilonia) and the F. lugubris-A2 population were used in our study. Results indicate that the F. lugubris-A2 population differs from other F. lugubris and from all other species in the behaviour of its workers and in the way its pupae are discriminated by other species. This is in accordance with the genetic data and strengthens our hypothesis on the existence of a new cryptic red wood ant species within the Swiss National Park.

The identification of CTL-defined tumor-associated Ags has allowed the development of new strategies for cancer immunotherapy. To potentiate the CTL responses, peptide-based vaccines require the coadministration of adjuvants. Because oligodeoxynucleotides (ODN) containing CpG motifs are strong immunostimulators, we analyzed the ability of CpG ODN to act as adjuvant of the CTL response against tumor-derived synthetic peptide in the absence or presence of IFA. Mice transgenic for a chimeric MHC class I molecule were immunized with a peptide analog of MART-1/Melan-A(26-35) in the presence of CpG ODN alone or CpG ODN emulsified in IFA. The CTL response was monitored ex vivo by tetramer staining of lymphocytes. In blood, spleen, and lymph nodes, peptide mixed with CpG ODN alone was able to elicit a stronger systemic CTL response as compared with peptide emulsified in IFA. Moreover, CpG ODN in combination with IFA further enhanced the CTL response in terms of the frequency of tetramer+CD8+ T cells ex vivo. The CTL induced in vivo against peptide analog in the presence of CpG ODN are functional, as they were able to recognize and kill melanoma cells in vitro. Overall, these results indicate that CpG ODN by itself is a good candidate adjuvant of CTL response and can also enhance the effect of classical adjuvant.

Despite advances in surgery, radio- and chemotherapy, therapeutic approaches for patients with head and neck squamous carcinoma (HNSCC) need to be improved. Immunotherapies eliciting tumor specific immune responses might constitute novel treatment options. We therefore investigated the expression and immunogenicity of two tumor-associated antigens (TAA) the receptor for hyaluronic acid mediated motility (RHAMM) and carboanhydrase IX (G250/CAIX) in HNSCC patients. Twenty-two HNSCC samples were examined for the expression of RHAMM and G250 by Western blotting and immunohistochemistry, 14/22 samples were tested for HLA-A2 expression by flow cytometry. For 8/22 samples single tumor-cell suspensions were generated, and mixed lymphocyte peptide cultures (MLPC) were performed to evaluate the frequencies of cytotoxic T cells specifically recognizing RHAMM and G250 using Tetramer staining/multi-color flow cytometry and enzyme linked immunosorbent spot (ELISPOT) assays. RHAMM and G250 were expressed in 73 and 80% of the HNSCC samples at the protein level. A co-expression of both TAAs could be detected in 60% of the patients. In 4/8 HLA-A2+ patients, 0.06-0.13% of CD8+ effector T cells recognized Tetramers for RHAMM or G250 and secreted IFNgamma and granzyme B in ELISPOT assays. RHAMM and G250 are expressed at high frequency and high protein level in HNSCCs and are recognized by cytotoxic CD8+ effector T cells. Therefore both TAAs constitute interesting targets for T cell based immunotherapies for HNSCC.

Recognition by the T-cell receptor (TCR) of immunogenic peptides (p) presented by class I major histocompatibility complexes (MHC) is the key event in the immune response against virus infected cells or tumor cells. The major determinant of T cell activation is the affinity of the TCR for the peptide-MHC complex, though kinetic parameters are also important. A study of the 2C TCR/SIYR/H-2Kb system using a binding free energy decomposition (BFED) based on the MM-GBSA approach had been performed to assess the performance of the approach on this system. The results showed that the TCR-p-MHC BFED including entropic terms provides a detailed and reliable description of the energetics of the interaction (Zoete and Michielin, 2007). Based on these results, we have developed a new approach to design sequence modifications for a TCR recognizing the human leukocyte antigen (HLA)-A2 restricted tumor epitope NY-ESO-1. NY-ESO-1 is a cancer testis antigen expressed not only in melanoma, but also on several other types of cancers. It has been observed at high frequencies in melanoma patients with unusually positive clinical outcome and, therefore, represents an interesting target for adoptive transfer with modified TCR. Sequence modifications of TCR potentially increasing the affinity for this epitope have been proposed and tested in vitro. T cells expressing some of the proposed TCR mutants showed better T cell functionality, with improved killing of peptide-loaded T2 cells and better proliferative capacity compared to the wild type TCR expressing cells. These results open the door of rational TCR design for adoptive transfer cancer therapy.

Dois métodos, utilizando permeâmetros a disco, foram usados para medir a sorvidade e a condutividade hidráulica em três horizontes de um solo de Santa Maria da Boa Vista, Pernambuco, utilizando-se potenciais de fornecimento de água de 0, -2,5, -5 e -10 cm de água. No primeiro método, a condutividade hidráulica foi obtida através de estimativas da sorvidade e do fluxo estacionário, utilizando-se apenas um permeâmetro. No segundo, as medidas da sorvidade e da condutividade hidráulica foram feitas utilizando-se valores dos fluxos estacionários, obtidos a partir de dois permeâmetros de diferentes raios. Esse segundo método apresentou resultados mais consistentes dos tempos -- gravitacional e geométrico --, e do raio característico de poros que o primeiro método. A sorvidade foi tão importante quanto a condutividade hidráulica para caracterizar o processo de infiltração. A redução brusca dos valores dos raios característicos de poros do horizonte A2/B, com mudanças no potencial de fornecimento de água, revelou a heterogeneidade da estrutura deste horizonte, permitindo identificar a influência dos volumes argilosos compactados sobre o processo de infiltração.
