This paper examines the life cycle GHG emissions from existing UK pulverized coal power plants. The life cycle of the electricity Generation plant includes construction, operation and decommissioning. The operation phase is extended to upstream and downstream processes. Upstream processes include the mining and transport of coal including methane leakage and the production and transport of limestone and ammonia, which are necessary for flue gas clean up. Downstream processes, on the other hand, include waste disposal and the recovery of land used for surface mining. The methodology used is material based process analysis that allows calculation of the total emissions for each process involved. A simple model for predicting the energy and material requirements of the power plant is developed. Preliminary calculations reveal that for a typical UK coal fired plant, the life cycle emissions amount to 990 g CO2-e/kWh of electricity generated, which compares well with previous UK studies. The majority of these emissions result from direct fuel combustion (882 g/kWh 89%) with methane leakage from mining operations accounting for 60% of indirect emissions. In total, mining operations (including methane leakage) account for 67.4% of indirect emissions, while limestone and other material production and transport account for 31.5%. The methodology developed is also applied to a typical IGCC power plant. It is found that IGCC life cycle emissions are 15% less than those from PC power plants. Furthermore, upon investigating the influence of power plant parameters on life cycle emissions, it is determined that, while the effect of changing the load factor is negligible, increasing efficiency from 35% to 38% can reduce emissions by 7.6%. The current study is funded by the UK National Environment Research Council (NERC) and is undertaken as part of the UK Carbon Capture and Storage Consortium (UKCCSC). Future work will investigate the life cycle emissions from other power generation technologies with and without carbon capture and storage. The current paper reveals that it might be possible that, when CCS is employed. the emissions during generation decrease to a level where the emissions from upstream processes (i.e. coal production and transport) become dominant, and so, the life cycle efficiency of the CCS system can be significantly reduced. The location of coal, coal composition and mining method are important in determining the overall impacts. In addition to studying the net emissions from CCS systems, future work will also investigate the feasibility and technoeconomics of these systems as a means of carbon abatement.

Metabolic stable isotope labeling is increasingly employed for accurate protein (and metabolite) quantitation using mass spectrometry (MS). It provides sample-specific isotopologues that can be used to facilitate comparative analysis of two or more samples. Stable Isotope Labeling by Amino acids in Cell culture (SILAC) has been used for almost a decade in proteomic research and analytical software solutions have been established that provide an easy and integrated workflow for elucidating sample abundance ratios for most MS data formats. While SILAC is a discrete labeling method using specific amino acids, global metabolic stable isotope labeling using isotopes such as (15)N labels the entire element content of the sample, i.e. for (15)N the entire peptide backbone in addition to all nitrogen-containing side chains. Although global metabolic labeling can deliver advantages with regard to isotope incorporation and costs, the requirements for data analysis are more demanding because, for instance for polypeptides, the mass difference introduced by the label depends on the amino acid composition. Consequently, there has been less progress on the automation of the data processing and mining steps for this type of protein quantitation. Here, we present a new integrated software solution for the quantitative analysis of protein expression in differential samples and show the benefits of high-resolution MS data in quantitative proteomic analyses.

Geotechnical systems, such as landfills, mine tailings storage facilities (TSFs), slopes, and levees, are required to perform safely throughout their service life, which can span from decades for levees to “in perpetuity” for TSFs. The conventional design practice by geotechnical engineers for these systems utilizes the as-built material properties to predict its performance throughout the required service life. The implicit assumption in this design methodology is that the soil properties are stable through time. This is counter to long-term field observations of these systems, particularly where ecological processes such as plant, animal, biological, and geochemical activity are present. Plant roots can densify soil and/or increase hydraulic conductivity, burrowing animals can increase seepage, biological activity can strengthen soil, geochemical processes can increase stiffness, etc. The engineering soil properties naturally change as a stable ecological system is gradually established following initial construction, and these changes alter system performance. This paper presents an integrated perspective and new approach to this issue, considering ecological, geotechnical, and mining demands and constraints. A series of data sets and case histories are utilized to examine these issues and to propose a more integrated design approach, and consideration is given to future opportunities to manage engineered landscapes as ecological systems. We conclude that soil scientists and restoration ecologists must be engaged in initial project design and geotechnical engineers must be active in long-term management during the facility’s service life. For near-surface geotechnical structures in particular, this requires an interdisciplinary perspective and the embracing of soil as a living ecological system rather than an inert construction material.

A criticism of a recent article published in this journal, claiming to have reached a classical description of the Stern-Gerlach phenomenon, is presented here. The author of the article, among other mistakes, wrongly writes the total energy of each silver atom and, moreover, presents a nonsensical equation, from which his results and the conclusion of his article are derived.

Neutral trehalase from Neurospora crassa was expressed in Escherichia coli as a polypeptide of similar to 84 kDa in agreement with the theoretical size calculated from the corresponding cDNA. The recombinant neutral trehalase, purified by affinity chromatography exhibited a specific activity of 80-150 mU/mg protein. Optima of pH and temperature were 7.0 and 30 degrees C, respectively. The enzyme was absolutely specific for trehalose, and was quite sensitive to incubation at 40 degrees C. The recombinant enzyme was totally dependent on calcium, and was inhibited by ATP, copper, silver, aluminium and cobalt. K(M) was 42 mM, and V(max) was 30.6 nmol of glucose/min. The recombinant protein was phosphorylated by cAMP-dependent protein kinase, but not significantly activated. Immunoblotting with polyclonal antiserum prepared against the recombinant protein showed that neutral trehalase protein levels increased during exponential phase of N. crassa growth and dropped at the stationary phase. This is the first report of a neutral trehalase produced in E. coli with similar biochemical properties described for fungi native neutral trehalases, including calcium-dependence. Este trabalho analisou 2770 empresas nos BRICs entre 1995 e 2008 para identificar quais os setores mais atrativos para investimento conforme a relação risco-retorno, geração de valor e a tradição da Organização Industrial (IO). Os resultados reforçam os de Fama e French (1992), Mohanram (2005) e Goldszmidt, Brito e Vasconcelos (2007), porém divergem de World Bank (2008) quanto à China. Constatou-se que os setores mais atrativos na perspectiva de risco-retorno sobre o patrimônio líquido seriam óleo & gás na Rússia e mineração no Brasil, Índia e China, enquanto aqueles com menor atratividade seriam os setores têxtil, motores, máquinas & ferramentas e telecomunicações no Brasil.

Nos últimos anos, a indústria elétrica brasileira passou, basicamente, por duas reestruturações setoriais, implantadas segundo políticas e ideologias divergentes, mas que convergiram para o mesmo objetivo final: a busca por investimentos privados para dar suporte à expansão do sistema. Esse é o grande desafio do novo modelo institucional de 2004, que surge em um ambiente conturbado, de forte percepção de riscos e incertezas. A própria instabilidade regulatória é a grande responsável pelo recuo e ponderação na hora de investir. Contudo, a pergunta que se faz na presente pesquisa objetiva identificar os fatores motivadores da nova reestruturação setorial. Só assim, pode-se correlacionar os fatos do passado com a recente reforma, identificando possíveis deficiências e fatores críticos de sucesso. Para tanto, o presente estudo recorreu à literatura existente, assim como a entrevistas semi-estruturadas com especialistas do setor elétrico brasileiro. Os dados foram analisados qualitativamente, por meio de categorias estabelecidas a posteriori, buscando-se aceitar ou refutar suposições. As evidências sugerem que, apesar de a nova reforma institucional ter realizado avanços, ao introduzir mecanismos absorvidos, principalmente, a partir das experiências regulatórias dos últimos anos, dificuldades ainda permanecem e devem ser enfrentadas, especialmente no que tange à mitigação dos riscos ambientais e regulatórios, este último, percebido com maior intensidade a partir de 2004. Sendo assim, cabe ao governo federal, representado pelo Ministério de Minas e Energia (MME), conseguir atrair investimentos e proporcionar estabilidade regulatória, a fim de afastar o “fantasma” do racionamento.

Os estudos sobre desenvolvimento, sem dúvida, se mantiveram como um dos últimos bastiões do modernismo nas ciências sociais (Rapley, 2004). Muitos dos dilemas chave em estudos contemporâneos sobre desenvolvimento se centraram nas disjuntivas entre inovação teórica, política e prática (Simon, 2003). No entanto, a discussão que envolve a relação entre desenvolvimento e mineração, que interessa neste estudo, ainda permanece acrítica dentro da literatura dominante. Segundo Graulau (2008), o tema de mineração encontra-se num vaivém entre o favoritismo e a oposição. O estudo sob o ponto de vista normativo da mineração no campo de desenvolvimento mostra a mentalidade econômica de longa data que prevalece nesse campo. No Peru as reformas neoliberais implantadas desde a década 1990 têm promovido fortemente o setor de mineração. Os investimentos nacionais e estrangeiros, o volume das exportações e impostos certamente têm influenciado favoravelmente na economia em termos macroeconômicos, obtendo quantidades consideráveis de divisas (UNCTAD, 2008). Não obstante, a grande mineração parece não ter beneficiado as comunidades envolvidas com a extração de minérios (Barrantes, 2005; Glave e Kuramoto, 2007; Zegarra; Orihuela e Paredes, 2007). A quantidade e gravidade dos conflitos que vem acontecendo evidenciam a resistência ao setor, frente à ação discursiva do Estado peruano sobre o “desenvolvimento” que assegura o que a mineração traz. Neste contexto este estudo tem como objetivo analisar as práticas discursivas das políticas de mineração peruana em relação a construção do discurso de desenvolvimento no período compreendido entre 1990-2009. Com esse objetivo, foi necessário abordar primeiramente as principais teorias sobre desenvolvimento, mineração e mineração no Peru. No que diz respeito à metodologia o presente estudo utilizou duas técnicas de análise: a Análise Crítica de Discurso, baseado no método tridimensional proposto por Fairclough (2001), para realizar a análise de três discursos de representantes da política de mineração peruana, a segunda abordagem utiliza a Análise de Conteúdo de Bardin (2009), para examinar os artigos relacionados à política de mineração entre as principais revistas especializadas do setor–Mineria e Desde Adentro. Foram utilizadas também categorias de análise constantes e convergentes ao conceito de desenvolvimento para orientar a presente pesquisa. Finalmente as conclusões sugerem que as políticas de mineração reproduzidas pelas autoridades do Estado peruano introduziram práticas discursivas sobre desenvolvimento sustentável e que essas se mantêm relacionadas com as novas ordens de discurso: Responsabilidade Social, Minerção Sustentável, Mineração moderna, Gestão ambiental.

-D-glucosidase (EC 3.2.1.21) is one of the most interesting glycosidases, especially for hydrolysis cellobiose releasing glucose, is last step degradation of cellulose. This function makes the -D-glucosidase is of great interest as a versatile industrial biocatalyst, being critical to various bio-treatment / biorefinery processes, such as bioethanol production. Hen in the report, a -D-glucosidase was extracts from protein extracted of the invertebrate marine Artemia franciscana was purified and characterized with a combination of precipitation with ammonium sulfate (0 - 30%, 30 to 50%, 50 to 80%), the fraction saturated in the range of 30 to 50% (called F-II) was applied in a molecular exclusion chromatography, in Sephacryl S-200, the fractions corresponding to the first peak of activity of -D-glucosidase were gathered and applied in a chromatography of ion exchange in Mono Q; the third peak this protein obtained chromatography, which coincides with the peak of activity of -D-glucosidase was held and applied in a gel filtration chromatography Superose 12 where the first peak protein, which has activity of -D-glucosidase was rechromatography on Superose 12. This enzyme is probably multimerica, consisting of three subunit molecular mass of 52.7 kDa (determined by SDS-PAGE) with native molecular mass of 157 kDa (determined by gel filtration chromatography on Superose 12 under the system FPLC). The enzyme was purified 44.09 times with a recovery of 1.01%. Using up p-nitrophenyl-β-D-glucopiranoside as substrate obtained a Km apparent of 0.229 mM and a Vmax of 1.109 mM.60min-1.mL-1mM. The optimum pH and optimum temperature of catalysis of the synthetic substrate were 5.0 and 45 °C, respectively. The activity of the -D-glucosidase was strongly, inhibited by silver nitrate and N- etylmaleimide, this inhibition indicates the involvement of radical sulfidrila the hydrolysis of synthetic substrate. The -D-glucosidase of Artemia franciscana presented degradativa action on celobiose, lactose and on the synthetic substrate -nitrophenyl-β-D-glucopiranoside indicating potential use of this enzyme in the industry mainly for the production of bioethanol (production of alcohol from the participating cellulose), and production hydrolysate milk (devoid of milk lactose)

Brazil has vast amounts of hydric resources, whose quality has been deteriorating due to pollutant dumping. Household waste disposal is one of the main sources of water pollution, stimulating bacteria proliferation and introducing microorganisms, including those from fecal matter. Conventional water disinfection methods are a solution, but on the downside, they lead to the formation byproducts hazardous to human health. In this study, aiming to develop bactericidal filters for the disinfection of drinking water; silver nanoparticles were deposited on alumina foams through three routes: sputtering DC, dip coating and in situ chemical reduction of silver nitrate. The depositions were characterized through X-ray diffraction, scanning electron microscopy and EDS element mapping. The influence of the depositions on permeability and mechanical properties of the ceramic foams was assessed and, in sequence, a preliminary antibacterial efficiency analysis was carried out. Characterization results indicate that the chemical reduction routes were efficient in depositing homogeneously distributed silver particles and that the concentration of the metallic precursor salt affects size and morphology of the particles. The antibacterial efficiency analysis indicates that the chemical reduction filters have potential for water disinfection

Tricodinídeos são protozoários ciliados móveis com ampla distribuição mundial; são considerados um dos agentes parasitários que mais acometem peixes cultivados. No Brasil, a maioria dos tricodinídeos que parasitam importantes espécies de peixes cultivados são desconhecidos, o que requer mais estudos taxonômicos com esse grupo de parasitos. Este estudo caracteriza morfologicamente Trichodina colisae Asmat & Sultana, 2005 de pacu (Piaractus mesopotamicus) e do híbrido patinga (P. mesopotamicus × P. brachypomus) cultivados, respectivamente, no Centro-Oeste e Sudeste do Brasil. Foram feitas montagens a fresco do raspado de muco da pele, nadadeiras e brânquias, fixados com metanol e, posteriormente, impregnados com nitrato de prata e coradas com Giemsa para avaliação em microscopia óptica. O presente estudo relata não só a segunda ocorrência de T. colisae no mundo, mas também a primeira ocorrência na América do Sul.

The progress of a nation is closely linked to the energy supply that it has to develop its economic capabilities. The dependence of contemporary society for energy requires the continued expansion of the use of renewable energy, and implies coordinated action of the Democratic State in the delimitation of the best ways to make full use of energy. In periods of rapid development, countries need an increment of energy supplies superior to those of periods of regular economic growth. Energy demand generated by the condition of Brazil as an emerging country reveals the need for orderly expansion of energy supply. In reverse, lack of energy planning effectively paralyzes a country and generates incalculable losses in national socioeconomic development. The Brazilian Constitution abandoned the notion of development tied to the simple increase in the gross domestic product. The respect for the environment, sovereignty, national development, and especially the constant and growing supply of energy, promotes the advancement of national economic agents, and quenches the simple accounting increase in energy supply. Constitutional principles condition the rational use of energy potentials, in ensuring adequate supply of energy for the entire national territory. The Brazilian Ministry of Mines and Energy, through its agencies, government offices and companies, establishes and formulates policies and guidelines for energy in Brazil, playing an important role in national energy planning. National development is enhanced by the good performance of the state agencies responsible for planning the energy sector

This work was conducted to evaluate biological parameters of Plutella xylostella L. reared on leaves of several cauliflower genotypes under laboratory conditions. The experiment was set in a randomized block design and arranged in a 6 x 2 factorial (genotypes x generations). Leaf disks of the cultivars Barcelona, Verona, Piracicaba Precoce, Sharon, Silver Streak, and Teresopolis Gigante were placed in Petri dishes with 12 newly-hatched larvae. Leaf disks were initially changed after the fourth day, but daily afterwards until the larvae reached the pupal stage. The same procedure was adopted for the second generation. Twenty adults of each sex were separated from each genotype to evaluate their longevity, and I 0 couples from each treatment were used to assess female fecundity. The lowest larval survival was obtained on the 'Silver Streak' (78.9%) and highest on 'Verona' (97.1%). The 'Silver Streak' and `Teresopolis Gigante' showed the lowest pupal weights (4.83 mg and 5.11 mg, respectively), as well as the lowest fecundity, 119.4 and 123.0 eggs/female, respectively, while 'Piracicaba Precoce' the highest (167.7 eggs/female). Males obtained from larvae reared on `Teresopolis Gigante' and 'Silver Streak' lived shorter (5.1 days), while the short-lived females were obtained from larvae reared on 'Barcelona' and 'Verona' (4.9 and 5.0 days). Insect development was prolonged in the second generation in all tested genotypes.
