The aim of this study was to assess the desiccation tolerance and DNA integrity in Eugenia pleurantha seeds dehydrated to different moisture contents (MCs). Seeds extracted from mature fruits were dried in silica gel and evaluated at every five percentual points of decrease from the initial MC (35.5%, fresh weight basis). The effects of dehydration on seeds were verified through germination tests and DNA integrity assessment. Undried seeds achieved 87% germination, value reduced to 36% after being dried to 9.8% MC. When dried slightly more, to 7.4% MC, seeds were no longer able to germinate, suggesting an intermediate behavior in relation to desiccation tolerance. DNA degradation was observed in seeds with 7.4% MC, which might have contributed to the loss of seed germination.

This study was aimed at evaluating the desiccation sensitivity in seeds of the tree Tapirira obtusa (Benth.) J. D. Mitchell collected from three different environments and subjected to two distinct drying speeds. Seeds were collected from a rocky area, in the "Cerrado", and in a riparian forest area, in the region of municipality of Lavras, State of Minas Gerais. The seeds were subjected to drying with magnesium chloride (slow drying) or silica gel (fast drying), into closed environment, until moisture contents of 40%, 30%, 20% and 10%, considering as control, the percentage of germination at the initial moisture content in each environment, which varied from 47% to 50%. Percentages of germination and normal seedlings as well as germination speed index were assessed. For the three environments studied, there was no effect of slow drying on seed germination. Seeds from area of Cerrado, however, have shown a slight reduction on germination when subjected to fast drying. Oppositely, seeds from rocky area had germination increased when subjected to fast drying. Seeds from riparian forest area had no reduction on germination percentage, independent of drying speed. Results suggest that seeds of T. obtusa are not sensitive to desiccation.

Korkean IP-luokituksen ohutlevykotelointi on haastava kokonaisuus. Koteloinnin tiivistys ruiskutettavalla polyureapinnoitteella edellyttää usean osa-alueen samanaikaista hallintaa. Kotelo on alusta alkaen suunniteltava pinnoitettavaksi, sillä pinnoitus asettaa lukuisia vaatimuksia ja rajoituksia esimerkiksi käytettäville muodoille, rakenteille ja liittämismenetelmille. Polyurea on elastomeeri, josta valmistettua pinnoitetta voidaan levittää erityisellä ruiskutuslaitteistolla. Polyureapinnoite sallii kotelon asentamisen vaikeisiin olosuhteisiin, sillä se kestää kemikaaleja, kulutusta ja iskuja sekä tarjoaa tiiveyden lisäksi korroosiosuojan koteloinnille. Pinnoitteen ominaisuuksia, kuten kovuutta, elastisuutta ja kemikaalien sekä UV-säteilyn kestoa voidaan räätälöidä käyttökohteen mukaan. Polyurepinnoitteeella pinnoitettavat pinnat on pyrittävä pitämään mahdollisimman yksinkertaisina, mikä tarkoittaa käytännössä kaikenlaisten kohoumien, ulkonevien osien, reikien ja muiden epäjatkuvuuskohtien välttämistä. Kaikki epäjatkuvuuskohdat vaativat erityishuomiota pinnoituksen aikana, sillä epäjatkuvuuskohtien onnistunut pinnoitus vaatii ruiskutusta useasta suunnasta, mikä lisää virhemahdollisuuksia ja siten vaarantaa koteloinnin tiiveyden. Liittämismenetelmät ovat yksinkertaisten muotojen ohella avainasemassa pinnoituksen onnistumisen kannalta. Menetelmistä tulee suosia sellaisia, joiden pinnoitettavaan pintaan aiheuttama epäjatkuvuuskohta on mahdollisimman vähäinen. Tällaisia menetelmiä ovat esimerkiksi vastuspistehitsaus ja puristeruuvi.

A simple High Performance Liquid Chromatograph (HPLC) method has been developed to identify benamyl (methyl 1- (butylcarbamoyl)-2-benzimidazole carbamate) and MBC (methyl 2-benzimidazole carbamat~ residues on apple leaves without cleanup. Sample leaves are freeze dried in a Mason jar and residues are then extracted by tumbling them in chloroform containing 5,000 microgram per milliliter of n-propyl isocyanate (PIC) at 10 C. To the extract, n-butyl isocyanate (BIC) was added at 5,000 microgram per milliliter and 20 microliter of this mixture injected onto the HPLC system. Separation is accomplished by the use of a Brownlee LiChrosorb silica gel column with a guard column and' operated with a mixed mobile phase consisting of chloroform and hexane (4:1) saturated with water. MBC, a degradation product of benomyl is identified if present as methyl l-(npropyl carbamoyl)-2-benzimidazole carbamate (MBC-n-PIC). Both benomyl and MBC-n-PIC can be detected with aKUltraviolet (UV) detector (280nm) at a concentration as low as 0.2 microgram per milliliter in apple leaves. The fate of benomyl on apple foliage after spray application of benomyl (Ben late 50 per cent wettable powder) was investigated by the method thus described. Benomyl quickly dissipated during the first 3-7 days, but the dissipatio'n sltowed down thereafter. In contrast, the concentration of MBC in leaves gradually increased after repeated applications of Benlate.

This study examined how perturbation-evoked compensatory arm reactions in individuals with Parkinson’s disease (PD) are influenced by explicit verbal instruction. Ten individuals with PD and 15 older adults without PD responded to surface translations with or without specific instruction to reach for and grasp the handrail. Electromyographic (EMG) and kinematic recordings were taken from the reaching arm. Results showed that individuals with and without PD benefitted similarly from explicit instruction. Explicit instruction resulted in earlier (p=0.005) and larger (p<0.001) medial deltoid EMG responses in comparison to no specific instructions. Compensatory arm reactions also occurred with a higher peak medio-lateral wrist velocity (p<0.001) and higher peak shoulder abduction angular velocity (p<0.001) with explicit instruction. Explicit instruction positively influenced compensatory arm reactions in individuals with and without PD. Future research is needed to determine whether the benefits of instruction persist over time and translate to a loss of balance in real life.

La présence d’un récepteur de type RXR a récemment été rapporté chez la pensée de mer, Renilla koellikeri, de même que chez d’autres anthozoaires, et le NO semble jouer des différents rôles physiologiques, chez plusieurs cnidaires. L’acide rétinoïque (AR) et le monoxyde d’azote (NO) sont connus pour leur implication dans l’induction de la croissance des neurites chez les vertébrés ainsi que chez les invertébrés. Mais jusqu’à présent, aucun rôle de ces agents n’a encore été identifié chez ce phylum ancien des invertébrés. Dans le but de montrer que ces agents morphogénétiques ont un rôle dans le développement neuronal chez ces ancêtres des métazoaires bilatéraux, nous avons utilisé des cultures primaires de cellules du cnidaire anthozoaire Renilla koellikeri (pensée de mer), doté d’un système nerveux des plus primitif. Nous avons trouvé que les deux types d’acide rétinoïque, 9-cis et 11-trans, induisent une prolifération cellulaire dose-dépendante en fonction du temps dans les boîtes de pétri enduites de polylysine. Les cultures cellulaires exposées à l’acide rétinoïque dans les boîtes sans polylysine montrent une différenciation en des cellules épithéliales. D’autre part, le NO induit exclusivement une différenciation neuronale dans les boîtes enduites de polylysine. Aucun autre type de cellules subit un différenciation en présence de NO et la densité des cellules dédifférenciées a diminué. Les prolongements des neurones différenciés semblent s’enchevêtrer et former un réseau neuronal assez dense. L’ensemble de ces observations suggère que l’acide rétinoïque, contrairement à NO, est associé à l’activité mitotique, et que l’acide rétinoïque et le NO sont impliqués différemment dans la spécification cellulaire, respectivement épithéliale et neuronale, chez la pensée de mer. Le type d’action déclenchée, qu’il soit la mitogénèse ou la différenciation (épithéliale ou neuronale), varie alors selon l’état d’adhésion des cellules au substrat. Comme les données moléculaires et paléontologiques rapprochent les cnidaires, telle la pensée de mer, des ancêtres des eumétazoaires, nos résultats suggèrent que le rôle morphogénétique de l’acide rétinoïque et du NO est enraciné dans l’ancêtre commun de tous les métazoaires.

Les tumeurs solides sont infiltrées par des cellules immunes (TIIC) dont la nature, la fonction et la composition varient d’un patient à l'autre. Ces cellules inflammatoires influencent l'invasion tumorale en contrôlant la croissance et le potentiel métastatique d’une tumeur. Ainsi, il est proposé d’utiliser cette infiltration comme outil diagnostic et pronostic de routine. Certaines cellules sont bien connues pour jouer un rôle important dans le contrôle de la progression tumorale, comme c’est le cas des lymphocytes T cytotoxiques CD8+ alors que d’autres possèdent un rôle contradictoire. Étant donné la dépendance des tumeurs sur l’équilibre entre ces différentes cellules, il est important d’identifier les fonctions précises des cellules immunes au sein de la tumeur. De nombreuses études sont réalisées afin d’identifier des marqueurs descriptifs du phénotype et la fonction des cellules immunes dans la tumeur. Ce projet de doctorat se divise en deux parties : 1- Identifier la méthode de désagrégation des tissus tumoraux altérant le moins la biologie des TIIC pour leur caractérisation. 2- Caractériser l’expression de la molécule d’adhérence CD146 dans les TIIC et en identifier l’origine. L’identification de marqueurs pour la caractérisation phénotypique et fonctionnelle des TIIC a été réalisée, entre autres, par la détection de protéines exprimées par la cellule. Dans la première partie de ce projet, nous avons démontré que les méthodes utilisées pour désagréger les tissus tumoraux dans le but d’isoler les TIIC induisent des changements dans la biologie de ces cellules ce qui peut fausser les conclusions qui en dérivent. Nous avons donc comparé l'impact de trois méthodes de désagrégation : une dissociation mécanique utilisant la MédimachineTM et deux digestions enzymatiques utilisant une collagénase de type I seule ou combinée à de la collagénase de type IV et de la DNase I de type II. Nous nous sommes intéressés à l'effet de ces méthodes sur des paramètres tels que la viabilité cellulaire, l’altération des protéines de surface et la capacité des cellules à proliférer. Nous avons démontré que ces méthodes affectent la viabilité des cellules de manière comparable, alors que la détection de certaines protéines de surface et la capacité de proliférer est réduite/inhibée par les traitements enzymatiques. Nous concluons qu’une méthode mécanique utilisant la MédimachineTM est mieux adaptée à la caractérisation des TIIC afin de conserver leurs propriétés. Dans la deuxième partie de notre projet, nous avons adapté cette méthode à la caractérisation des TIIC. Nous avons porté une attention particulière à la molécule d’adhérence CD146 dont l’implication dans la migration des cellules immunes à travers l’endothélium vers les sites d’inflammation est de plus en plus étudiée dans les maladies autoimmunes. Nous avons mis en évidence une augmentation des proportions de cellules immunes exprimant CD146 dans les tumeurs comparativement au sang de patients de cancers. Cette expression est induite par les cellules tumorales tout en étant accrue par la nécrose de celles-ci. Nous démontrons que ces cellules sont majoritairement des lymphocytes T CD4+ présentant un profil immunosuppressif. En conclusion, nos résultats suggèrent que CD146 participe à la mise en place du contexte immunitaire dans la tumeur et augmente la capacité de migration des lymphocytes T CD4+. L’induction par les cellules tumorales de cette molécule d’adhérence dans les cellules suppressives pourrait contribuer aux mécanismes immunorégulateurs mis en place par la tumeur. CD146 pourrait être un marqueur d’intérêt pour l’identification des cellules immunosuppressives et pour le développement de nouvelles thérapies.

The objective of this study is to generate the knowledge base to achieve the recovery of iron, vanadium and titanium cvalues from multi- metal waste chloride liquors originating from ilmenite mineral beneficiation industries through selective separation and value added material development

The objective of this study is to understand the reasons for the enhancement in aerosol optical depth (AOD) over the Arabian Sea observed during June, July and August. During these months, high values of AOD are found over the sea beyond 10◦ N and adjacent regions. The Arabian Sea is bounded by the lands of Asia and Africa on its three sides. So the region is influenced by transported aerosols from the surroundings as well as aerosols of local origin (marine aerosols). During the summer monsoon season in India, strong surface winds with velocities around 15 m s−1 are experienced over most parts of the Arabian Sea. These winds are capable of increasing sea spray activity, thereby enhancing the production of marine aerosols. The strong winds increase the contribution of marine aerosols over the region to about 60% of the total aerosol content. The main components of marine aerosols include sea salt and sulphate particles. The remaining part of the aerosol particles comes from the western and northern land masses around the sea, of which the main component is transported dust particles. This transport is observed at higher altitudes starting from 600 m. At low levels, the transport occurs mainly from the Indian Ocean and the Arabian Sea itself, indicating the predominance of marine aerosols at these levels. The major portion of the total aerosol loading was contributed by coarse-mode particles during the period of study. But in the winter season, the concentration of coarse-mode aerosols is found to be less. From the analysis, it is concluded that the increase in marine aerosols and dust particles transported from nearby deserts results in an increase in aerosol content over the Arabian Sea during June, July and August.

The growth of Fe–Ni based amorphous nanocolumns has been studied using atomic force microscopy. The root mean square roughness of the film surface increased with the deposition time but showed a little change at higher deposition time. It was found that the separation between the nanostructures increased sharply during the initial stages of growth and the change was less pronounced at higher deposition time. During the initial stages of the column growth, a roughening process due to self shadowing is dominant and, as the deposition time increases, a smoothening mechanism takes place due to the surface diffusion of adatoms

To study the behaviour of beam-to-column composite connection more sophisticated finite element models is required, since component model has some severe limitations. In this research a generic finite element model for composite beam-to-column joint with welded connections is developed using current state of the art local modelling. Applying mechanically consistent scaling method, it can provide the constitutive relationship for a plane rectangular macro element with beam-type boundaries. Then, this defined macro element, which preserves local behaviour and allows for the transfer of five independent states between local and global models, can be implemented in high-accuracy frame analysis with the possibility of limit state checks. In order that macro element for scaling method can be used in practical manner, a generic geometry program as a new idea proposed in this study is also developed for this finite element model. With generic programming a set of global geometric variables can be input to generate a specific instance of the connection without much effort. The proposed finite element model generated by this generic programming is validated against testing results from University of Kaiserslautern. Finally, two illustrative examples for applying this macro element approach are presented. In the first example how to obtain the constitutive relationships of macro element is demonstrated. With certain assumptions for typical composite frame the constitutive relationships can be represented by bilinear laws for the macro bending and shear states that are then coupled by a two-dimensional surface law with yield and failure surfaces. In second example a scaling concept that combines sophisticated local models with a frame analysis using a macro element approach is presented as a practical application of this numerical model.

High aspect ratio polymeric micro-patterns are ubiquitous in many fields ranging from sensors, actuators, optics, fluidics and medical. Second generation PDMS molds are replicated against first generation silicon molds created by deep reactive ion etching. In order to ensure successful demolding, the silicon molds are coated with a thin layer of C[subscript 4]F[subscript 8] plasma polymer to reduce the adhesion force. Peel force and demolding status are used to determine if delamination is successful. Response surface method is employed to provide insights on how changes in coil power, passivating time and gas flow conditions affect plasma polymerization of C[subscript 4]F[subscript 8].

In this paper, a new methodology for predicting fluid free surface shape using Model Order Reduction (MOR) is presented. Proper Orthogonal Decomposition combined with a linear interpolation procedure for its coefficient is applied to a problem involving bubble dynamics near to a free surface. A model is developed to accurately and efficiently capture the variation of the free surface shape with different bubble parameters. In addition, a systematic approach is developed within the MOR framework to find the best initial locations and pressures for a set of bubbles beneath the quiescent free surface such that the resultant free surface attained is close to a desired shape. Predictions of the free surface in two-dimensions and three-dimensions are presented.

A wind-tunnel study was conducted to investigate ventilation of scalars from urban-like geometries at neighbourhood scale by exploring two different geometries a uniform height roughness and a non-uniform height roughness, both with an equal plan and frontal density of λ p = λ f = 25%. In both configurations a sub-unit of the idealized urban surface was coated with a thin layer of naphthalene to represent area sources. The naphthalene sublimation method was used to measure directly total area-averaged transport of scalars out of the complex geometries. At the same time, naphthalene vapour concentrations controlled by the turbulent fluxes were detected using a fast Flame Ionisation Detection (FID) technique. This paper describes the novel use of a naphthalene coated surface as an area source in dispersion studies. Particular emphasis was also given to testing whether the concentration measurements were independent of Reynolds number. For low wind speeds, transfer from the naphthalene surface is determined by a combination of forced and natural convection. Compared with a propane point source release, a 25% higher free stream velocity was needed for the naphthalene area source to yield Reynolds-number-independent concentration fields. Ventilation transfer coefficients w T /U derived from the naphthalene sublimation method showed that, whilst there was enhanced vertical momentum exchange due to obstacle height variability, advection was reduced and dispersion from the source area was not enhanced. Thus, the height variability of a canopy is an important parameter when generalising urban dispersion. Fine resolution concentration measurements in the canopy showed the effect of height variability on dispersion at street scale. Rapid vertical transport in the wake of individual high-rise obstacles was found to generate elevated point-like sources. A Gaussian plume model was used to analyse differences in the downstream plumes. Intensified lateral and vertical plume spread and plume dilution with height was found for the non-uniform height roughness
