Context The bush dog (Speothos venaticus) is difficult to observe, capture, and study. To date, indirect evidence and opportunistic field observations have been the primary sources of information about the species' ecology. Field data are urgently needed to clarify the species' ecological requirements, behaviour and movement patterns. Aims The present study uses 13 months of telemetry data from a group of bush dogs to begin to address questions about area requirements, habitat preferences and movement patterns of this difficult-to-study species. Methods We tracked a group of bush dogs (two adults, one juvenile, four young) in an area of intact and altered Cerrado (woodlandsavanna biome) in eastern Mato Grosso, Brazil (Nova Xavantina District). Key results The group had a total home range of 140km2 (fixed kernel 95%), with smaller seasonal 'subareas' (areas used for 12 months before moving to another area, with repetition of some areas over time) and demonstrated a preference for native habitats. Conclusions The bush dog's home range is greater than that of other canids of the same size, even correcting for group size. Patterns of seasonal movement are also different from what has been observed in other South American canids. Implications From our observations in the Brazilian savanna, bush dogs need large tracks of native habitat for their long-term persistence. Although the present study is based on a single pack, it is highly relevant for bush dog conservation because it provides novel information on the species' spatial requirements and habitat preferences. © 2012 CSIRO.

Brazil adopted SIRGAS2000 in 2005. This adoption called for the provision of the relationships between SIRGAS2000 and the previous reference frames used for positioning, mapping and GIS, namely, the Córrego Alegre (CA) and the South American Datum of 1969 (SAD 69). Two programs were designed for this purpose. The first one, TCGeo, provided the relationships based on three-translation Similarity Transformation parameters. TCGeo was replaced in December 2008, by ProGriD. ProGriD offers, besides the same similarity transformation as TCGeo, a set of transformations based on modelling the distortions of the networks used in the various realizations of CA and SAD 69. The distortion models are represented by a grid in which each node contains a transformation value in terms of difference in latitude and in longitude. The grid follows the same specifications of the NTv2 grid, which has been used in other countries, such as Canada, USA and Australia. This article aims to understand how the participation and performance of the state of Bahia at the I Campeonato Brasileiro de Seleções (I Brazilian Soccer Championship), also known as the Torneio do Centenário (Centenary Championship) in 1922 represented an opportunity for the state to claim a centrality in the formation of a national identity associated with soccer. The tournament was conceived as a draft for the formation of a national soccer team that would participate in the VI Campeonato Sul Americano (VI South American Championship) in Brazil. In addition, the event also came to be regarded as a celebration of the fi rst centenary of the Independence, a date that has encouraged the country to refl ect upon its national identity, as well as to reconsider its insertion in modernity. Bahia did well in the tournament, ensuring the second position. As a result, the local press went on to claim the participation of their athletes in the national team and to criticize the disregard of the states of Rio de Janeiro and São Paulo towards the Northern region of the country. The local press would also challenge the policies of the Confederação Brasileira de Desportos (Brazilian Sports Confederation), the CBD, claiming that they favored the Southern states. Moreover, for the local elites the success of Bahia in the tournament represented the strength and greatness of the state, which gave them the right to claim a greater space and role in the destiny of the country. Finally, the performance of Bahia and its consequences have allowed us to question the limits and peculiarities of the formation of a national identity through the sport, at a moment when the country sought to rethink their identity historically marked by disputes and regional tensions.

Cichlids are important in the aquaculture and ornamental fish trade and are considered models for evolutionary biology. However, most studies of cichlids have investigated African species, and the South American cichlids remain poorly characterized. Studies in neotropical regions have focused almost exclusively on classical cytogenetic approaches without investigating physical chromosomal mapping of specific sequences. The aim of the present study is to investigate the genomic organization of species belonging to different tribes of the subfamily Cichlinae (Cichla monoculus, Astronotus ocellatus, Geophagus proximus, Acaronia nassa, Bujurquina peregrinabunda, Hoplarchus psittacus, Hypselecara coryphaenoides, Hypselecara temporalis, Caquetaia spectabilis, Uaru amphiacanthoides, Pterophyllum leopoldi, Pterophyllum scalare, and Symphysodon discus) and reexamine the karyotypic evolutionary patterns proposed for this group. Variations in some cytogenetic markers were observed, although no trends were found in terms of the increase, decrease, or maintenance of the basal diploid chromosome number 2n = 48 in the tribes. Several species were observed to have 18S rDNA genetic duplications, as well as multiple rDNA loci. In most of the taxa analyzed, the 5S rDNA was located in the interstitial region of a pair of homologous chromosomes, although variations from this pattern were observed. Interstitial telomere sites were also observed and appear to be involved in chromosomal rearrangement events and the accumulation of repeat-rich satellite DNA sequences. Our data demonstrated the karyotypic diversity that exists among neotropical cichlids, suggesting that most of this diversity is due to the repetitive sequences present in heterochromatic regions and that repeat sequences have greatly influenced the karyotypic evolution of these fishes. © 2012 Springer Science+Business Media B.V.

In the neighborhood of the city of Boqueirão do Leão (State of Rio Grande do Sul, Brazil) a set of three big-sized tunnels has been found. One of the tunnels is only partially filled with sand and accessible along its entire length. It is horizontal, slightly sinuous, 36 m long, up to 4.2 m wide and up to 2.0 m high. The surface morphology of the walls is composed of anthropogenic marks, speleothems, black incrustations and traces like digging scratches and smoothed surfaces. The 2nd tunnel has its entrance blocked by sand and sandstone cobbles, but the end of the tunnel is only partially clogged and therefore accessible. This accessible portion is 12 m long, 3 m wide and 1.5 m high. The 3rd tunnel is completely filled and collapsed and is nowadays only indicated by concave roof features at its end. The general features of the tunnel system and the analysis of the surface morphology of the walls of the accessible portions permit to conclude that the tunnels were produced by ground sloths of the Cenozoic South American megafauna. The size of the tunnels suggests that its excavation was gradually carried out by successive generations of sloth herds, and not by a single individual animal. The primary function of the tunnels probably was not protection from predators, which had easy access to structures of this size, but to shelter during a drier climate. However, it is not yet possible to relate the tunnels to a specific ground sloth genus, a task that depends on the discovery of better-preserved tunnel systems. © 2013 by the Sociedade Brasileira de Paleontologia.

Paracoccidoides brasiliensis adhesion to lung epithelial cells is considered an essential event for the establishment of infection and different proteins participate in this process. One of these proteins is a 30 kDa adhesin, pI 4.9 that was described as a laminin ligand in previous studies, and it was more highly expressed in more virulent P. brasiliensis isolates. This protein may contribute to the virulence of this important fungal pathogen. Using Edman degradation and mass spectrometry analysis, this 30 kDa adhesin was identified as a 14-3-3 protein. These proteins are a conserved group of small acidic proteins involved in a variety of processes in eukaryotic organisms. However, the exact function of these proteins in some processes remains unknown. Thus, the goal of the present study was to characterize the role of this protein during the interaction between the fungus and its host. To achieve this goal, we cloned, expressed the 14-3-3 protein in a heterologous system and determined its subcellular localization in in vitro and in vivo infection models. Immunocytochemical analysis revealed the ubiquitous distribution of this protein in the yeast form of P. brasiliensis, with some concentration in the cytoplasm. Additionally, this 14-3-3 protein was also present in P. brasiliensis cells at the sites of infection in C57BL/6 mice intratracheally infected with P. brasiliensis yeast cells for 72 h (acute infections) and 30 days (chronic infection). An apparent increase in the levels of the 14-3-3 protein in the cell wall of the fungus was also noted during the interaction between P. brasiliensis and A549 cells, suggesting that this protein may be involved in host-parasite interactions, since inhibition assays with the protein and this antibody decreased P. brasiliensis adhesion to A549 epithelial cells. Our data may lead to a better understanding of P. brasiliensis interactions with host tissues and paracoccidioidomycosis pathogenesis. © 2013 Silva et al.

The morphologically undivided ventricle of the heart in non-crocodilian reptiles permits the mixing of oxygen-rich blood returning from the lungs and oxygen-poor blood from the systemic circulation. A possible functional significance for this intra-cardiac shunt has been debated for almost a century. Unilateral left vagotomy rendered the single effective pulmonary artery of the South American rattlesnake, Crotalus durissus, unable to adjust the magnitude of blood flow to the lung. The higher constant perfusion of the lung circulation and the incapability of adjusting the right-left shunt in left-denervated snakes persisted over time, providing a unique model for investigation of the long-term consequences of cardiac shunting in a squamate. Oxygen uptake recorded at rest and during spontaneous and forced activity was not affected by removing control of the cardiac shunt. Furthermore, metabolic rate and energetic balance during the post-prandial metabolic increment, plus the food conversion efficiency and growth rate, were all similarly unaffected. These results show that control of cardiac shunting is not associated with a clear functional advantage in adjusting metabolic rate, effectiveness of digestion or growth rates. © 2013. Moenkhausia is one of the most speciose genera in Characidae, currently composed of 75 nominal species of small fishes distributed across South American hydrographic basins, primarily the Amazon and Guyanas. Despite the large number of described species, studies involving a substantial number of its species designed to better understand their relationships and putative monophyly are still lacking. In this study, we analysed a large number of species of Moenkhausia to test the monophyly of the genus based on the phylogenetic analysis of DNA sequences of two mitochondrial and three nuclear genes. The in-group included 29 species of Moenkhausia, and the out-group was composed of representatives of Characidae and other members of Characiformes. All species of Moenkhausia belong to the same clade (Clade C); however, they appear distributed in five monophyletic groups along with other different genera, which means that Moenkhausia is polyphyletic and indicates the necessity of an extensive revision of the group. © 2013 Blackwell Verlag GmbH.

Cancer pain is an important clinical problem and may not respond satisfactorily to the current analgesic therapy. We have characterized a novel and potent analgesic peptide, crotalphine, from the venom of the South American rattlesnake Crotalus durissus terrificus. In the present work, the antinociceptive effect of crotalphine was evaluated in a rat model of cancer pain induced by intraplantar injection of Walker 256 carcinoma cells. Intraplantar injection of tumor cells caused the development of hyperalgesia and allodynia, detected on day 5 after tumor cell inoculation. Crotalphine (6 μg/kg), administered p.o., blocked both phenomena. The antinociceptive effect was detected 1 h after treatment and lasted for up to 48 h. Intraplantar injection of nor-binaltorphimine (50 g/paw), a selective antagonist of κ-opioid receptors, antagonized the antinociceptive effect of the peptide, whereas N,N-diallyl-Tyr-Aib-Phe-Leu (ICI 174,864, 10 μg/paw), a selective antagonist of δ-opioid receptors, partially reversed this effect. On the other hand, D-Phe-Cys-Tyr-D-Trp-Orn-Thr-Pen-Thr amide (CTOP, 20 g/paw), an antagonist of μ-opioid receptors, did not modify crotalphine-induced antinociception. These data indicate that crotalphine induces a potent and long lasting opioid-mediated antinociception in cancer pain. © 2013 Elsevier Inc.

Phylogeographic studies provide an important framework for investigating the mechanisms operating during the earliest stages of speciation, as reproductive barriers can be examined among divergent lineages in a geographic context. We investigated the evolution of early stages of intrinsic postmating isolation among different populations and lineages of Epidendrum denticulatum, a Neotropical orchid distributed across different biomes in South America. We estimated genetic diversity and structure for both nuclear and plastid markers, using a haplotype network, differentiation tests, Bayesian assignment analysis, and divergence time estimates of the main lineages. Reproductive barriers among divergent lineages were examined by analyzing seed viability following reciprocal crossing experiments. Strong plastid phylogeographic structure was found, indicating that E. denticulatum was restricted to multiple refuges during South American forest expansion events. In contrast, significant phylogeographic structure was not found for nuclear markers, suggesting higher gene flow by pollen than by seeds. Large asymmetries in seed set were observed among different plastid genetic groups, suggesting the presence of polymorphic genic incompatibilities associated with cytonuclear interactions. Our results confirm the importance of phylogeographic studies associated with reproductive isolation experiments and suggest an important role for outbreeding depression during the early stages of lineage diversification. © 2013 The Society for the Study of Evolution.

A comprehensive biostratinomic study was carried out with abundant stems from the Lower Permian Motuca Formation of the intracratonic Parnaíba Basin, central-north Brazil. The fossils represent a rare tropical to subtropical paleofloristic record in north Gondwana. Tree ferns dominate the assemblages (mainly Tietea, secondarily Psaronius), followed by gymnosperms, sphenophytes, other ferns and rare lycophytes. They are silica-permineralized, commonly reach 4 m length (exceptionally more than 10 m), lie loosely on the ground or are embedded in the original sandstone or siltstone matrix, and attract particular attention because of their frequent parallel attitudes. Many tree fern stems present the original straight cylindrical to slightly conical forms, other are somewhat flattened, and the gymnosperm stems are usually more irregular. Measurements of stem orientations and dimensions were made in three sites approximately aligned in a W-E direction in a distance of 27.3 km at the conservation unit Tocantins Fossil Trees Natural Monument In the eastern site, rose diagrams for 54 stems indicate a relatively narrow azimuthal range to SE. These stems commonly present attached basal bulbous root mantles and thin cylindrical sandstone envelopes, which sometimes hold, almost adjacent to the lateral stem surface, permineralized fern pinnae and other small plant fragments. In the more central site, 82 measured stems are preferentially oriented in the SW-NE direction, the proportion of gymnosperms is higher and cross-stratification sets of sandstones indicate paleocurrents mainly to NE and secondarily to SE. In the western site, most of the 42 measured stems lie in E-W positions. The predominantly sandy succession, where the fossil stems are best represented, evidences a braided fluvial system under semiarid conditions. The low plant diversity, some xeromorphic features and the supposedly almost syndepositional silica impregnation of the plants are coherent with marked dry seasons. Thick mudstones and some coquinites below and above the sandy interval may represent lacustrine facies formed in probably more humid conditions. The taphonomic history of the preserved plants began with exceptional storms that caused fast-flowing high water in channels and far into the floodplains. In the eastern site region, many tree ferns only fell, thus sometimes covering and protecting plant litter and leaves from further fragmentation. Assemblages of the central and western sites suggest that the trees were uprooted and transported in suspension (floating) parallel to the flow. Heavier ends of stems (according to their form or because of attached basal bulbous root mantle or large apical fronds) were oriented to upstream because of inertial forces. During falling water stage, the stems were stranded on riverbanks, usually maintaining the previous transport orientation, and were slightly buried. The perpendicular or oblique positions of some stems may have been caused by interference with other stems or shallow bars. Rare observed stems were apparently waterlogged before the final depositional process and transported as bedload. The differences of interpreted channel orientations between the three sites are expected in a braided fluvial system, considering the very low gradients of the basin and the work scale in the order of tens of kilometers. The mean direction of the drainage probably was to east and the flows apparently became weaker downstream. This study seems to provide reliable data for paleocurrent interpretations, especially considering areas with scarce preserved sedimentary structures. © 2013 Elsevier Ltd.

Anopheles darlingi is the principal neotropical malaria vector, responsible for more than a million cases of malaria per year on the American continent. Anopheles darlingi diverged from the African and Asian malaria vectors ∼100 million years ago (mya) and successfully adapted to the New World environment. Here we present an annotated reference A. darlingi genome, sequenced from a wild population of males and females collected in the Brazilian Amazon. A total of 10 481 predicted protein-coding genes were annotated, 72% of which have their closest counterpart in Anopheles gambiae and 21% have highest similarity with other mosquito species. In spite of a long period of divergent evolution, conserved gene synteny was observed between A. darlingi and A. gambiae. More than 10 million single nucleotide polymorphisms and short indels with potential use as genetic markers were identified. Transposable elements correspond to 2.3% of the A. darlingi genome. Genes associated with hematophagy, immunity and insecticide resistance, directly involved in vectorhuman and vectorparasite interactions, were identified and discussed. This study represents the first effort to sequence the genome of a neotropical malaria vector, and opens a new window through which we can contemplate the evolutionary history of anopheline mosquitoes. It also provides valuable information that may lead to novel strategies to reduce malaria transmission on the South American continent. The soilborne fungus Rhizoctonia solani anastomosis group 3 (AG-3PT) is a globally important potato pathogen. However, little is known about the population genetic processes affecting field populations of R. solani AG-3PT, especially in the South American Colombian Andes, which is near the center of diversity of the two most common groups of cultivated potato, Solanum tuberosum and S. phureja. We analyzed the genetic structure of 15 populations of R. solani AG-3PT infecting potato in Colombia using 11 simple-sequence repeat (SSR) markers. In total, 288 different multilocus genotypes were identified among 349 fungal isolates. Clonal fractions within field populations were 7 to 33%. R ST statistics indicated a very low level of population differentiation overall, consistent with high contemporary gene flow, though moderate differentiation was found for the most distant southern populations. Genotype flow was also detected, with the most common genotype found widely distributed among field populations. All populations showed evidence of a mixed reproductive mode, including both asexual and sexual reproduction, but two populations displayed evidence of inbreeding. © 2013 The American Phytopathological Society.

Paracoccidioidomycosis is a human systemic mycosis caused by the fungus Paracoccidioides brasiliensis. The mechanisms involved in innate immune response to this fungus are not fully elucidated. Leukotrienes are known to be critical for the clearance of various microorganisms, mainly by mediating the microbicidal function of phagocytes. We investigated the involvement of leukotriene B4 in the early stages of experimental paracoccidioidomycosis, which was induced by intratracheal inoculation of the fungus in selected mouse lines. The mouse lines utilized were produced through bi-directional phenotypic selection, endowed with maximal or minimal acute inflammatory reactivity, and designated AIRmax and AIRmin, respectively. AIRmax mice were more resistant to the infection, which was demonstrated by reduced lung fungal loads. However, the two lines produced similar amounts of leukotriene B4, and pharmacological inhibition of this mediator provoked similar fungal load increases in the two lines. The lower fungal load in the AIRmax mice was associated with a more effective inflammatory response, which was characterized by enhanced recruitment and activation of phagocytic cells and an increased production of activator cytokines. This process resulted in an increased release of fungicidal molecules and a diminution of fungal load. In both lines, leukotriene production was associated with a protective response in the lung that was consequent to the effect of this eicosanoid on the influx and activation of phagocytes. © 2013 ISHAM.

Biodiesel fuel is gradually replacing petroleum-based diesel oil use. Despite the biodiesel being considered friendlier to the environment, little is known about its effects in aquatic organisms. In this work we evaluated whether biodiesel exposure can affect oxidative stress parameters and biotransformation enzymes in armored catfish (Pterygoplichthys anisitsi, Loricariidae), a South American endemic species. Thus, fish were exposed for 2 and 7d to 0.01mLL-1 and 0.1mLL-1 of pure diesel, pure biodiesel (B100) and blends of diesel with 5% (B5) and 20% (B20) biodiesel. Lipid peroxidation (malondialdehyde) levels and the activities of the enzymes glutathione S-transferase, superoxide dismutase, catalase and glutathione peroxidase were measured in liver and gills. Also, DNA damage (8-oxo-7, 8-dihydro-2'-deoxyguanosine) levels in gills and 7-ethoxyresorufin-O-deethylase activity in liver were assessed. Pure diesel, B5 and B20 blends changed most of the enzymes tested and in some cases, B5 and B20 induced a higher enzyme activity than pure diesel. Antioxidant system activation in P. anisitsi was effective to counteract reactive oxygen species effects, since DNA damage and lipid peroxidation levels were maintained at basal levels after all treatments. However, fish gills exposed to B20 and B100 presented increased lipid peroxidation. Despite biodiesel being more biodegradable fuel that emits less greenhouse gases, the increased lipid peroxidation showed that biofuel and its blends also represent hazards to aquatic biota. © 2013 Elsevier Ltd.
