This project identified a novel family of six 66-68 residue peptides from the venom of two Australian funnel-web spiders, Hadronyche sp. 20 and H. infensa: Orchid Beach (Hexathelidae: Atracinae), that appear to undergo N- and/or C-terminal post-translational modifications and conform to an ancestral protein fold. These peptides all show significant amino acid sequence homology to atracotoxin-Hvf17 (ACTX-Hvf17), a non-toxic peptide isolated from the venom of H. versuta, and a variety of AVIT family proteins including mamba intestinal toxin 1 (MIT1) and its mammalian and piscine orthologs prokineticin 1 (PK1) and prokineticin 2 PK2). These AVIT family proteins target prokineticin receptors involved in the sensitization of nociceptors and gastrointestinal smooth muscle activation. Given their sequence homology to MITI, we have named these spider venom peptides the MIT-like atracotoxin (ACTX) family. Using isolated rat stomach fundus or guinea-pia ileum organ bath preparations we have shown that the prototypical ACTX-Hvf17, at concentrations up to 1 mu M, did not stimulate smooth muscle contractility, nor did it inhibit contractions induced by human PK1 (hPK1). The peptide also lacked activity on other isolated smooth muscle preparations including rat aorta. Furthermore, a FLIPR Ca2+ flux assay using HEK293 cells expressing prokineticin receptors showed that ACTX-Hvf17 fails to activate or block hPK1 or hPK2 receptors. Therefore, while the MIT-like ACTX family appears to adopt the ancestral disulfide-directed beta-hairpin protein fold of MIT1, a motif believed to be shared by other AVIT family peptides, variations in the amino acid sequence and surface charge result in a loss of activity on prokineticin receptors. An adaptive back-propagation algorithm is studied and compared with gradient descent (standard back-propagation) for on-line learning in two-layer neural networks with an arbitrary number of hidden units. Within a statistical mechanics framework, both numerical studies and a rigorous analysis show that the adaptive back-propagation method results in faster training by breaking the symmetry between hidden units more efficiently and by providing faster convergence to optimal generalization than gradient descent.

We analyse Gallager codes by employing a simple mean-field approximation that distorts the model geometry and preserves important interactions between sites. The method naturally recovers the probability propagation decoding algorithm as a minimization of a proper free-energy. We find a thermodynamical phase transition that coincides with information theoretical upper-bounds and explain the practical code performance in terms of the free-energy landscape.

Heiner Müller has often been described as the only rightful (literary) heir and flag- bearer of Bertolt Brecht in German drama of the late twentieth century. This article first examines the critical position that Müller took towards his predecessor. The essay will then focus on the Fatzer complex which Müller considered to be by far the most important work by Brecht. Next, I will discuss the relationship between Fatzer and Müllerʼs synthesised stage version FATZER from 1978, followed by a brief assessment of the various ways in which Mu ̈ller treated Brechtʼs Fatzer material over the next two decades. Special attention will be paid to the radio play that Müller produced of his version of Fatzer during the final years of the GDR; I argue that it represents an exemplary engagement with Brecht’s fragment because, rather than aiming for a standard illusionistic radio play, Mu ̈ller’s intention is to allow his FATZER version, as it were, to speak for itself. To conclude, the essay undertakes a brief and inevitably speculative discussion of the influence of Brechtʼs unfinished drama project on Müller’s works.

A model of the cognitive process of natural language processing has been developed using the formalism of generalized nets. Following this stage-simulating model, the treatment of information inevitably includes phases, which require joint operations in two knowledge spaces – language and semantics. In order to examine and formalize the relations between the language and the semantic levels of treatment, the language is presented as an information system, conceived on the bases of human cognitive resources, semantic primitives, semantic operators and language rules and data. This approach is applied for modeling a specific grammatical rule – the secondary predication in Russian. Grammatical rules of the language space are expressed as operators in the semantic space. Examples from the linguistics domain are treated and several conclusions for the semantics of the modeled rule are made. The results of applying the information system approach to the language turn up to be consistent with the stages of treatment modeled with the generalized net.
