Background: The aim of this study is to characterize and evaluate the host response caused by three different models of experimental periodontitis in mice.Methods: C57BL/6 wild-type female mice were distributed into six experimental groups and sacrificed at 7, 15, and 30 days after the induction of periodontal disease: 1) group C: no treatment control group; 2) group L: periodontal disease induced by ligature; 3) group G-Pg: oral gavage with Porphyromonas gingivalis (Pg); 4) group G-PgFn: oral gavage with Fusobacterium nucleatum + Pg; 5) group I-Pg: heat-killed Pg injected into the palatal mucosa between the molars; and 6) group I-V: phosphatebuffered saline injected into the palatal mucosa. The samples were used to analyze the immune-inflammatory process in the gingival tissue via descriptive histologic and real-time polymerase chain reaction analyses. The alveolar bone loss was evaluated using microcomputed tomography. The data were analyzed using the Kruskal-Wallis test, followed by a post hoc Dunn test and analysis of variance, followed by a Tukey test using a 5% significance level.Results: Only the ligature model displayed significant alveolar bone loss in the initial period (7 days), which was maintained with time. The group injected with heat-killed Pg displayed significant alveolar bone loss starting from day 15, which continued to progress with time (P < 0.05). A significant increase (P < 0.05) in the gene expression of proinflammatory cytokines (interleukin-6 and -1b) and proteins involved in osteoclastogenesis (receptor activator of nuclear factor-kB ligand and osteoprotegerin) was observed in the ligature group on day 7.Conclusion: The ligature and injection of heat-killed Pg models were the most representative of periodontal disease in humans, whereas the oral gavage models were not effective at inducing the disease under the experimental conditions.

To assess the importance of the leucine residues in positions 262 and 265 of the angiotensin AT, receptor for signaling pathways and receptor expression and regulation, we compared the properties of CHO cells transfected with the wild type or the L262D or L265D receptor point mutants. It was found that the two mutants significantly increased the basal intracellular cyclic AMP (cAMP) formation in an agonist-independent mode. The morphology transformation of CHO cells was correlated with the increased cAMP formation, since forskolin, a direct activator of adenylate cyclase mimicked this effect on WT-expressing CHO cells. DNA synthesis was found to be inhibited in these cell lines, indicating that cAMP may also have determined the inhibitory effect on cell growth, in addition to the cell transformation from a tumorigenic to a non-tumorigenic phenotype. However a role for an increased Ca2(+) influx induced by the mutants in non-stimulated cells cannot be ruled out since this ion also was shown to cause transformed cells to regain the morphology and growth regulation. Aim: To demonstrate the magnitude and direction of skeletal changes in the maxilla and mandible during and after the use of bionator, as well as their rotations. Methods: Partial superimposition on the maxilla and mandible on the metallic implants and total superimposition on the cranial base were performed at three periods, T1 before bionator therapy, T2 after bionator therapy, and T3 5.68 years after T2. Results: There was total clockwise maxillary rotation and counterclockwise mandibular rotation, in the North American technique, throughout the study period, as well as extensive remodeling on the condylar region, especially in vertical direction and on the gonial region in horizontal direction. Conclusions: The total maxillary rotation seemed to be significantly affected by therapy than the mandible. There was a clear change in the direction of condylar remodeling compared to the period of bionator therapy and posterior bionator therapy. Considering the entire study period, it was observed that intra-matrix rotation of the maxilla and mandible masked their total rotation, causing minimum changes in the matrix rotation.

The study aimed to assess the degree of dental crowding correction on the lower anterior region of patients treated with T4k functional appliance using Little’s irregularity index. Twenty caucasian patients of both gender (10 female and 10 male) were selected. They featured malocclusion Class I and II in mixed dentition, with chronological age between 5.7 and 11 years. The treatment lasted from 1 year and 11 months up to 3 years and 11 months. Lower anterior crowding was measured using Little’s irregularity index. Measurements were obtained on study models achieved before and after functional orthopedic therapy, using a digital caliper in millimeters and placed parallel to the occlusal plane. Overjet and overbite measurements were also performed using a caliper. Results demonstrated that the therapy provided significant decrease in overjet (average = 1.55 mm) and also in the irregularity index (average = 1.23 mm). There was a nonsignificant raise in overbite. There was no relation between the studied variables and the treatment period or even with the patient’s age in the beginning of the therapy. The treatment with T4k resulted on reduction and improvement of dental crowding.

The aim of this study was to evaluate the vertical and horizontal dentoskeletal changes induced by the Klammt Open Elastic Activator in the treatment of the Class II Division 1 malocclusion. The sample of 34 children was divided into two groups of 17 subjects each (ten girls, seven boys), matched by age and gender and with an initial mean age of 8.5 years. The data was analyzed using a Student's t-test for intragroup and intergroup comparison. The results showed that the appliance promoted dentoalveolar restriction of the maxilar growth within normal anteroposterior and vertical growth of the maxillary apical base; increment in the vertical displacement the mandibular symphysis associated with normal horizontal growth of the mandible; palatal tipping of the upper incisors; restriction of the anterior migration of the upper molars; greater eruption pattern and normal anterior displacement of the lower incisors and molars. It was concluded that Klammt appliance induce changes that are predominantly horizontal dentoalveolar in upper arch and vertical dentoalveolar in lower arch and vertical skeletal changes in the mandible.

The deficiency of data in the literature took us to evaluate the Bionator of Balters appliance in the alterations of the dimensions and the relationship of the dental archs in children with malocclusion Class II, division 1 of Angle. The experimental group was constituted by 36 pairs cast Caucasians patients, aged between 7 years and 10 months - 11 years and 8 months, being 10 females and 8 males. The Levene´s test showed statistical evidences of likeness among the groups. Statistical analysis was preceded and showed significant alterations (p < 0,005) in the variable indicatives of maxillary first molars' distance, overjet, upper arch total length, upper arch anterior length, right molar relationship, left molar relationship, right canine relationship and left canine relationship. On the other hand, there wasn't significant alteration related to the lower arch and maxillary intercanines distance. The Balters' Bionator appliance had a favorable effect in the improvement of the correction of the malocclusion in Class II (foremost in molars and canines relationship) and transversal increase of the upper arch, mainly in the posterior area of arch.
