Purification, Characterization, and Preliminary X-Ray Diffraction Analysis of a Lactose-Specific Lectin from Cymbosema roseum Seeds

The unique carbohydrate-binding property of lectins makes them invaluable tools in biomedical research. Here, we report the purification, partial primary structure, carbohydrate affinity characterization, crystallization, and preliminary X-ray diffraction analysis of a lactose-specific lectin from Cymbosema roseum seeds (CRLII). Isolation and purification of CRLII was performed by a single step using a Sepharose-4B-lactose affinity chromatography column. The carbohydrate affinity characterization was carried using assays for hemagglutination activity and inhibition. CRLII showed hemagglutinating activity toward rabbit erythrocytes. O-glycoproteins from mucine mucopolysaccharides showed the most potent inhibition capacity at a minimum concentration of 1.2 A mu g mL(-1). Protein sequencing by mass spectrometry was obtained by the digestion of CRLII with trypsin, Glu-C, and AspN. CRLII partial protein sequence exhibits 46% similarity with the ConA-like alpha chain precursor. Suitable protein crystals were obtained with the hanging-drop vapor-diffusion method with 8% ethylene glycol, 0.1 M Tris-HCl pH 8.5, and 11% PEG 8,000. What is Consciousness? Towards a Preliminary Definition

There is little or no general agreement about what researchers should focus on when studying consciousness. The most active scientific studies often use the methods of Cognitive Neuroscience and focus mainly on vision. Other aspects and contents of consciousness, namely thoughts and emotions, are much less studied, possibly leading to a biased view of what consciousness is and how it works. In this essay we describe what we call a referential nucleus, implicit in much of consciousness research. In this context, 'consciousness' refers to (partially) reportable content experienced by living individuals. We then discuss the philosophical concept of a phenomenal world and another contemporary view that conscious experience involves, besides integration of information in the brain, participation in action-perception cycles in a natural, social and cultural environment. These views imply a need to reconceptualize 'qualia' as the conscious aspect of subjective experiences, thus stating properties of consciousness that pose serious challenges to an exclusive approach via Cognitive Neuroscience, because experimental settings oversimplify conscious experiences, narrowing them to fragments correlated with measured brain activity and behaviour In conclusion we argue that a science of consciousness requires a broad interdisciplinary range of research, including qualitative methods from the Human Sciences.

A preliminary study of precipitable water over Brazil

A preliminary study of precipitable water over Brazil is attempted. Mean values for January are presented in this paper. A regression equation connecting the surface dew-point temperature and the precipitable water was computed along the lines of studies made by Reitan (1963), and the results are presented. © 1979 Birkhäuser Verlag.

Application of the GPS system for preliminary satellite orbit determination

In this paper, we discuss a method of preliminary orbit determination for an artificial satellite based on the navigation message of the GPS constellation. Orbital elements are considered as state variables and a simple dynamic model, based on the classic two-body problem, is used. The observations are formed by range and range and range-rate with respect to four visible GPS. A discrete Kalman filter with simulated data is used as filtering technique. The data are obtained through numerical propagation (Cowell's method), which considers special perturbations for the GPS satellite constellation and a user satellite. © 1997 COSPAR. Crystallization and preliminary X-ray analysis of SIII-SPIII, a ahospholipase A2 isolated from the venom of Bothrops jararacussu

Large single crystals have been obtained of SIII-SPIII, a phospholipase A2 from the venom of Bothrops jararacussu. The crystals belong to the orthorhombic system space group C222, and diffract X-rays to a resolution of 1.9 Å. Preliminary analysis reveals the presence of one molecule in the crystallographic asymmetric unit. The crystal structure is currently being determined using molecular replacement techniques.

Unsaturated polyester based composites reinforced with curauá fiber - Preliminary studies

Curauá fiber processing characterization has been performed throughout the different processing steps. Unsaturated polyester has been used as matrix in the production of curauá reinforced composite samples. Compression molding process has been used to prepare the samples. Tensile strength, impact resistance, flexural strength, Young's modulus and elongation at break have been accessed for curauá composites in comparison with fiberglass composites. Mechanical properties were found not to attend the company's internal standards specification. However, the work has shown some alternatives to solve these problems such as the modification of equipment characteristics and resin formulation, the necessity of incorporation of a higher content of fiber and the possibility of using a new type of filler. Preliminary studies on agonistic behavior between colonies of Coptotermes havilandi (Isoptera, Rhinotermitidae) in laboratory bioassays

Intercolonial aggression is being used to delimit foraging territory in termite species of subterranean termites. The advance of the introduced pest termite Coptotermes havilandi Holmgren in the interior of São Paulo State is increasing its economic impact as well as its interspecific and intraspecific competition in Brazil. In order to evaluate the intraspecific agonism among different colonies collected in urban areas of Sȧo Paulo State were set up a series of preliminary bioassays. Different combination of nestmates from field colonies of C. havilandi of Rio Claro city showed lack of agonistic behavior. Nevertheless, encounters among individuals from São Paulo and Rio Claro cities showed agonistic behaviors. These preliminary results suggest that caution should be taken in using intercolonial aggression to delimit the foraging territory of C. havilandi colonies in São Paulo State.

Lethal photosensitization in microbiological treatment of ligature-induced peri-implantitis: a preliminary study in dogs

This pilot study evaluated, by culture testing, the effectiveness of lethal photosensitization for the microbiological treatment of peri-implantitis in dogs. Experimental peri-implantitis was induced by ligature placement for 2 months. Following ligature removal, plaque control was instituted by scrubbing with 0.12% chlorhexidine daily for 12 months. Subsequently, mucoperiosteal flaps were elevated for scaling the implant surface. Microbial samples were obtained with paper points before and after treatment of implant surfaces by means of 100 microg/ml toluidine blue O (TBO,) and were exposed, for 80 s, to light with a wavelength of 685 nm from a 50 mW GaAlAs diode laser. The mean initial and final bacterial counts were 7.22 +/- 0.20 and 6.84 +/- 0.44 CFU/ml, respectively for TVC (P < 0.0001); 6.19 +/- 0.45 and 3.14 +/- 3.29 CFU/ml for P. intermedia/nigrescens (P = 0.001); 5.98 +/- 0.38 and 1.69 +/- 2.90 CFU/ml for Fusobacterium spp. (P = 0.001); and 6.07 +/- 0.22 to 1.69 +/- 2.94 CFU/ml for beta-hemolytic Streptococcus (P = 0.0039). It may be concluded that lethal photosensitization resulted in a reduction of the bacterial count. Complete elimination of bacteria was achieved in some samples.

Preliminary core sets of measures for disease activity and damage assessment in juvenile systemic lupus erythematosus and juvenile dermatomyositis

Objective. To identify preliminary core sets of outcome variables for disease activity and damage assessment in juvenile systemic lupus erythematosus (JSLE) and juvenile dermatomyositis (JDM). Methods. Two questionnaire surveys were mailed to 267 physicians from 46 different countries asking each member to select and rank the response variables used when assessing clinical response in patients with JSLE or JDM. Next, 40 paediatric rheumatologists from 34 countries met and, using the nominal group technique, selected the domains to be included in the disease activity and damage core sets for JSLE and JDM. Results. A total of 41 response variables for JSLE and 37 response variables for JDM were selected and ranked through the questionnaire surveys. In the consensus conference, domains selected for both JSLE and JDM activity or damage core sets included the physician and parent/patient subjective assessments and a global score tool. Domains specific for JSLE activity were the immunological tests and the kidney function parameters. Concerning JDM, functional ability and muscle strength assessments were indicated for both activity and damage core sets, whereas serum muscle enzymes were included only in the activity core set. A specific paediatric domain called 'growth and development' was introduced in the disease damage core set for both diseases and the evaluation of health-related quality of life was advised in order to capture the influence of the disease on the patient lifestyle. Conclusions. We developed preliminary core sets of measures for disease activity and damage assessment in JSLE and JDM. The prospective validation of the core sets is in progress.
