Indivíduos obesos apresentam maior risco de morbidade e mortalidade atribuída às doenças cardiovasculares. A composição da dieta é um fator que prediz o fenótipo cardíaco em resposta a obesidade e, o tipo de ácido graxo pode afetar de forma diferencial a estrutura e a função do miocárdio. Estudos têm mostrado que a disfunção mitocondrial exerce um papel chave na patogênese da insuficiência e hipertrofia cardíaca, e as alterações mitocondriais observadas em falhas cardíacas apontam para defeitos em sítios específicos da cadeia transportadora de elétrons. Desta forma, o objetivo deste estudo foi avaliar a função contrátil ventricular em camundongos, alimentados com dieta hiperlipídica, rica em ácidos graxos poliinsaturados, buscando elucidações através da bioenergética mitocondrial. Após desmame, camundongos machos C57Bl/6 passaram a receber dieta manipulada contendo 7% (C) ou 19% (HF) de óleo de soja, até os 135 dias de idade. A ingestão alimentar e a massa corporal foram monitoradas e foi realizado teste de tolerância à glicose. No final do período experimental, os animais foram anestesiados e submetidos à avaliação da composição corporal por Absortimetria de Raios X de Dupla Energia (DXA), e em seguida, sacrificados por exsanguinação. No plasma foram determinados o perfil lipídico e a insulina. O coração, o tecido adiposo intra-abdominal e o subcutâneo foram coletados, pesados, processados para análise histomorfológica. Fibras cardíacas do ventrículo esquerdo foram utilizadas para análise da respiração mitocondrial através de oxígrafo. O coração também foi utilizado para a técnica de perfusão de coração isolado de Langendorff, e para análise da expressão de proteínas relacionadas à bioenergética de cardiomiócitos, através de Western Blotting. O índice de HOMA e de adiposidade foram calculados. O grupo HF apresentou maior adiposidade, sem alteração na ingestão alimentar. Foi observada intolerância a glicose, hiperinsulinemia e resistência à insulina, além de alterações desfavoráveis no perfil lipídico. Foi observado alteração na morfologia cardíaca e quadro de cardiomiopatia hipertrófica, refletindo em alteração hemodinâmica, determinando maior contratilidade, maior pressão ventricular e função diastólica prejudicada. Em relação à atividade mitocondrial dos cardiomiócitos foi observada menor oxidação de carboidratos (-47%) e de ácidos graxos (-60%). Porém, sem alteração na expressão de proteínas relacionadas à bioenergética de cardiomiócitos, CPT1, UCP2, GLUT1, GLUT4, AMPK e pAMPK. A partir desses resultados, concluímos que o tipo e a quantidade de ácidos graxos predizem o fenótipo cardíaco na obesidade, promovendo alteração na capacidade oxidativa mitocondrial, na morfologia e na hemodinâmica cardíaca

The efforts made to develop RNAi-based therapies have led to productive research in the field of infections in humans, such as hepatitis C virus (HCV), hepatitis B virus (HBV), human immunodeficiency virus (HIV), human cytomegalovirus (HCMV), herpetic keratitis, human papillomavirus, or influenza virus. Naked RNAi molecules are rapidly digested by nucleases in the serum, and due to their negative surface charge, entry into the cell cytoplasm is also hampered, which makes necessary the use of delivery systems to exploit the full potential of RNAi therapeutics. Lipid nanoparticles (LNP) represent one of the most widely used delivery systems for in vivo application of RNAi due to their relative safety and simplicity of production, joint with the enhanced payload and protection of encapsulated RNAs. Moreover, LNP may be functionalized to reach target cells, and they may be used to combine RNAi molecules with conventional drug substances to reduce resistance or improve efficiency. This review features the current application of LNP in RNAi mediated therapy against viral infections and aims to explore possible future lines of action in this field.

Depth data from archival tags on northern rock sole (Lepidopsetta polyxystra) were examined to assess whether fish used tidal currents to aid horizontal migration. Two northern rock sole, out of 115 released with archival tags in the eastern Bering Sea, were recovered 314 and 667 days after release. Both fish made periodic excursions away from the bottom during mostly night-time hours, but also during particular phases of the tide cycle. One fish that was captured and released in an area of rotary currents made vertical excursions that were correlated with tidal current direction. To test the hypothesis that the fish made vertical excursions to use tidal currents to aid migration, a hypothetical migratory path was calculated using a tide model to predict the current direction and speed during periods when the fish was off the bottom. This migration included limited movements from July through December, followed by a 200-km southern migration from January through February, then a return northward in March and April. The successful application of tidal current information to predict a horizontal migratory path not only provides evidence of selective tidal stream transport but indicates that vertical excursions were conducted primarily to assist horizontal migration.

Self-amplifying RNA or RNA replicon is a form of nucleic acid-based vaccine derived from either positive-strand or negative-strand RNA viruses. The gene sequences encoding structural proteins in these RNA viruses are replaced by mRNA encoding antigens of interest as well as by RNA polymerase for replication and transcription. This kind of vaccine has been successfully assayed with many different antigens as vaccines candidates, and has been shown to be potent in several animal species, including mice, nonhuman primates, and humans. A key challenge to realizing the broad potential of self-amplifying vaccines is the need for safe and effective delivery methods. Ideally, an RNA nanocarrier should provide protection from blood nucleases and extended blood circulation, which ultimately would increase the possibility of reaching the target tissue. The delivery system must then be internalized by the target cell and, upon receptor-mediated endocytosis, must be able to escape from the endosomal compartment into the cell cytoplasm, where the RNA machinery is located, while avoiding degradation by lysosomal enzymes. Further, delivery systems for systemic administration ought to be well tolerated upon administration. They should be safe, enabling the multiadministration treatment modalities required for improved clinical outcomes and, from a developmental point of view, production of large batches with reproducible specifications is also desirable. In this review, the concept of self-amplifying RNA vaccines and the most promising lipid-based delivery systems are discussed.

Transport phenomena still stand as one of the most challenging problems in computational physics. By exploiting the analogies between Dirac and lattice Boltzmann equations, we develop a quantum simulator based on pseudospin-boson quantum systems, which is suitable for encoding fluid dynamics transport phenomena within a lattice kinetic formalism. It is shown that both the streaming and collision processes of lattice Boltzmann dynamics can be implemented with controlled quantum operations, using a heralded quantum protocol to encode non-unitary scattering processes. The proposed simulator is amenable to realization in controlled quantum platforms, such as ion-trap quantum computers or circuit quantum electrodynamics processors.

The variability in the supply of pink shrimp (Farfantepenaeus duorarum) postlarvae and the transport mechanisms of planktonic stages were investigated with field data and simulations of transport. Postlarvae entering the nursery grounds of Florida Bay were collected for three consecutive years at channels that connect the Bay with the Gulf of Mexico, and in channels of the Middle Florida Keys that connect the southeastern margin of the Bay with the Atlantic Ocean. The influx of postlarvae in the Middle Florida Keys was low in magnitude and varied seasonally and among years. In contrast, the greater postlarval influx occurred at the northwestern border of the Bay, where there was a strong seasonal pattern with peaks in influx from July through September each year. Planktonic stages need to travel up to 150 km eastward between spawning grounds (northeast of Dry Tortugas) and nursery grounds (western Florida Bay) in about 30 days, the estimated time of planktonic development for this species. A Lagrangian trajectory model was developed to estimate the drift of planktonic stages across the SW Florida shelf. The model simulated the maximal distance traveled by planktonic stages under various assumptions of behavior. Simulation results indicated that larvae traveling with the instantaneous current and exhibiting a diel behavior travel up to 65 km and 75% of the larvae travel only 30 km. However, the eastward distance traveled increased substantially when a larval response to tides was added to the behavioral variable (distance increased to 200 km and 85% of larvae traveled 150 km). The question is, when during larval development, and where on the shallow SW Florida shelf, does the tidal response become incorporated into the behavior of pink shrimp.
