(SPA) Introducción: los medios de fijación a la piel de un catéter venoso periférico, de un catéter venoso central o de un catéter arterial central presentan los siguientes graves inconvenientes: ser la sede de infecciones a nivel del lugar de la punción, provocar una saturación de la zona anatómica, la cual resulta muy difícil de soportar e incómodo para el paciente portador, y suponer un riesgo para el personal de enfermería cuando se usan hilos de sutura para fijar el soporte del catéter. Objetivo: por lo anteriormente expuesto, se ha detecta- do la necesidad de diseñar un dispositivo que reduzca la complejidad y aparatosidad de los sistemas conocidos y empleados en la actualidad, favoreciendo la asepsia y la movilidad del paciente. Método: el desarrollo presentado en este artículo se refie- re a un sistema mecánico de fijación, cierre y acoplamiento para catéter de perfusión intravenosa. El sistema dispone de una pieza circular de pequeño espesor con al menos dos orificios para su fijación subcutánea, centralmente de uno o más conductos para acoplar lúmenes, y de una parte cilín- drica hueca que sobresale a uno de los lados, exteriormente roscada e interiormente lisa con un tetón de posición. Para uso domiciliario se dispone de un tapón de estanqueidad con uno o más tubos de pequeño diámetro que se acoplan en los orificios para lúmenes, una ranura de posición, un tirador y un tapón roscado ciego para el cierre hermético con la parte roscada. Para uso hospitalario se dispone de un tapón intermedio con una o más entradas para lúmenes, así como de un tapón roscado hueco que permite su acopla- miento hermético con la parte roscada. Resultados: el desarrollo descrito en este artículo va a ser utilizado en dos ambientes: ambiente domiciliario y ambiente hospitalario. Además, reduce el riesgo de infec- ción y saturación de la zona anatómica de los pacientes en los que se realiza la punción cutánea en la que se fijan catéteres venosos periféricos, catéteres venosos centrales o catéteres arteriales centrales. (ENG) Introduction: skin fixing devices in peripheral, central or arterial catheters have several important drawbacks: site infection, stacking of material in the anatomical area which is very annoying for the patient and medical staff risk when fixating stitches are used. Objective: to develop a fixing device that simplifies presently used systems, favoring asepsis and motility. Methods: the device herein described is composed by a mechanical fixation, a closing system and coupling for intravenous catheters. The system has a thin circular pie- ce with at least two holes for subcutaneous fixation, one or several conducts for lumina and a hollow cylindrical part in one side, screwed exteriorly and flat inside, with an oriented protuberance. A watertight plug with one or several thin tubes that adapt to the lumina, a positional slot, a handle and a solid screwed tap for perfect closure are available for at home use. An intermediate plug with one or several lumina and a screwed hollow plug are pro- vided for in hospital use. Results: the above described device is intended to be used in two settings: in hospital and at home. It is su- pposed to reduce the risk of infection and stacking of the anatomical site where cutaneo us puncture with fixation of peripheral, central or arterial catheters is performed.

We report on spectroscopic studies of the chiral structure in phospholipid tubules formed in mixtures of alcohol and water. Synthetic phospholipids containing diacetylenic moieties in the acyl chains self-assemble into hollow, cylindrical tubules in appropriate conditions. Circular dichroism provides a direct measure of chirality of the molecular structure. We find that the CD spectra of tubules formed in mixtures of alcohol and water depends strongly on the alcohol used and the lipid concentration. The relative spectral intensity of different circular dichroism bands correlates with the number of bilayers observed using microscopy. The results provide experimental evidence that tubule formation is based on chiral packing of the lipid molecules and that interbilayer interactions are important to the tubule structure.

ADP ribosylation factor (ARF) is thought to play a critical role in recruiting coatomer (COPI) to Golgi membranes to drive transport vesicle budding. Yeast strains harboring mutant COPI proteins exhibit defects in retrograde Golgi to endoplasmic reticulum protein transport and striking cargo-selective defects in anterograde endoplasmic reticulum to Golgi protein transport. To determine whether arf mutants exhibit similar phenotypes, the anterograde transport kinetics of multiple cargo proteins were examined in arf mutant cells, and, surprisingly, both COPI-dependent and COPI-independent cargo proteins exhibited comparable defects. Retrograde dilysine-mediated transport also appeared to be inefficient in the arf mutants, and coatomer mutants with no detectable anterograde transport defect exhibited a synthetic growth defect when combined with arf1Δ, supporting a role for ARF in retrograde transport. Remarkably, we found that early and medial Golgi glycosyltransferases localized to abnormally large ring-shaped structures. The endocytic marker FM4–64 also stained similar, but generally larger ring-shaped structures en route from the plasma membrane to the vacuole in arf mutants. Brefeldin A similarly perturbed endosome morphology and also inhibited transport of FM4–64 from endosomal structures to the vacuole. Electron microscopy of arf mutant cells revealed the presence of what appear to be hollow spheres of interconnected membrane tubules which likely correspond to the fluorescent ring structures. Together, these observations indicate that organelle morphology is significantly more affected than transport in the arf mutants, suggesting a fundamental role for ARF in regulating membrane dynamics. Possible mechanisms for producing this dramatic morphological change in intracellular organelles and its relation to the function of ARF in coat assembly are discussed.

A number of pathogenic, Gram-negative bacteria are able to secrete specific proteins across three membranes: the inner and outer bacterial membrane and the eukaryotic plasma membrane. In the pathogen Yersinia enterocolitica, the primary structure of the secreted proteins as well as of the components of the secretion machinery, both plasmid-encoded, is known. However, the mechanism of protein translocation is largely unknown. Here we show that Y. enterocolitica polymerizes a 6-kDa protein of the secretion machinery into needles that are able to puncture the eukaryotic plasma membrane. These needles form a conduit for the transport of specific proteins from the bacterial to the eukaryotic cytoplasm, where they exert their cytotoxic activity. In negatively stained electron micrographs, the isolated needles were 60–80 nm long and 6–7 nm wide and contained a hollow center of about 2 nm. Our data indicate that it is the polymerization of the 6-kDa protein into these needles that provides the force to perforate the eukaryotic plasma membrane.

Cell differentiation, tissue formation, and organogenesis are fundamental patterns during the development of multicellular animals from the dividing cells of fertilized eggs. Hence, the complete morphogenesis of any developing organism of the animal kingdom is based on a complex series of interactions that is always associated with the development of a blastula, a one-layered hollow sphere. Here we document an alternative pathway of differentiation, organogenesis, and morphogenesis occurring in an adult protochordate colonial organism. In this system, any minute fragment of peripheral blood vessel containing a limited number of blood cells isolated from Botrylloides, a colonial sea squirt, has the potential to give rise to a fully functional organism possessing all three embryonic layers. Regeneration probably results from a small number of totipotent stem cells circulating in the blood system. The developmental process starts from disorganized, chaotic masses of blood cells. At first an opaque cell mass is formed. Through intensive cell divisions, a hollow, blastula-like structure results, which may produce a whole organism within a short period of a week. This regenerative power of the protochordates may be compared with some of the characteristics associated with the formation of mammalian embryonal carcinomous bodies. It may also serve as an in vivo model system for studying morphogenesis and differentiation by shedding more light on the controversy of the "stem cell" vs. the "dedifferentiation" theories of regeneration and pattern formation.

The aim of this work is to provide an analytical method based on experimental measurements in order to obtain the prismatic film deformation for different curvatures of Hollow Cylindrical Prismatic Light Guides (CPLG). To conform cylindrical guides is necessary bend the film to guide the light, changes induced by curving the film give rise to deformation shifts. Light losses affected by deformation has been experimentally evaluated and numerically analyzed. The effect of deformation in prism angle is specially increased for CPLG of curvatures higher than 20 m-1. An experimental method for accurate transmittance measurements related to bending is presented.

Apesar de ser considerado um combustível sustentável, o etanol, produzido a partir da cana de açúcar, deixa um passivo de grandes proporções durante seu processo produtivo, a vinhaça, que vem sendo depositada nas próprias lavouras de cana de açúcar. É gerada na proporção de 12 litros para cada litro de etanol produzido em média, sendo rica em diversos nutrientes, os quais podem ser aproveitados para diversos fins como, por exemplo, meio de cultivo para microalgas. A presente pesquisa avaliou em uma primeira etapa a clarificação da vinhaça por um processo de coagulação com auxílio de um polímero catiônico, seguida de uma etapa de microfiltração tangencial em filtro de fibras ocas, o que permitiu uma redução superior a 77% para a cor aparente, de 99% para a turbidez e de 20% para a DQO, facilitando a utilização deste efluente para o cultivo de microalgas. Numa segunda etapa, foi avaliado o cultivo da microalga Chlorella vulgaris, em escala de bancada e operação em batelada, em meio preparado a partir da diluição da vinhaça em água de poço profundo, obtendo um aumento na biomassa produzida, determinado em termos de clorofila-a, em concentrações de vinhaça inferiores a 7,5% utilizando inóculo da ordem de 106 indivíduos. Tais dados permitiram a realização de ensaios de cultivo em escala contínua, com fotobiorreatores em escala piloto, gerando assim a biomassa utilizada nas próximas fases do estudo, que avaliaram a separação da biomassa gerada pelo processo de flotação por ar dissolvido. Os ensaios inicialmente realizados em escala de bancada e operados em batelada permitiram identificar as condições ótimas de operação, as quais foram então avaliadas em um flotador operando em fluxo contínuo. Tal flotador permitiu a obtenção de um lodo com teor de sólidos superior a 2%, o qual foi submetido à um processo final de desaguamento por centrifugação. Os ensaios de desaguamento, permitiram verificar que a utilização do mesmo polímero utilizado na etapa de clarificação permite a obtenção de um lodo mais estável, quando comparado com a não utilização de produto químico, na dosagem de polímero catiônico de 6 g.kg-1. A conclusão deste trabalho permitiu verificar a possibilidade de utilização da vinhaça como meio de cultivo de microalgas, reduzindo assim um dos impactos causados pela produção de etanol. Além disso foi possível verificar o potencial da FAD, para o espessamento de biomassa produzido em fotobiorreatores.

A lamotrigina (LTG) é um fármaco pertencente à classe das feniltriazinas utilizado no tratamento de crises epilépticas generalizadas e focais e no tratamento adjunto da epilepsia refratária. Devido à alta variabilidade interindividual, às interações medicamentosas e aos efeitos adversos apresentados durante a administração da LTG, a monitorização terapêutica nos pacientes que fazem uso deste fármaco é necessária para ajuste de dose individual e evitar os efeitos adversos. Assim, o objetivo deste trabalho foi a avaliação de duas técnicas de microextração: a microextração em fase líquida com fibras ocas (HF-LPME) e a microextração líquido-líquido dispersiva (DLLME) para análise da lamotrigina em amostras de plasma de pacientes epilépticos. Primeiramente foram definidas as condições eletroforéticas: foi utilizado um capilar de sílica fundida de 75 ?m de diâmetro interno e 50 cm de comprimento efetivo. O eletrólito de corrida (BGE) foi composto por ácido 2-morfolinoetanosulfônico (MES), na concentração de 130 mmol L-1 e pH 5,0. As análises foram realizadas à temperatura de 20°C e tensão de 15 kV. A amostra foi injetada hidrodinamicamente (0,5 psi por 10 s) e a detecção foi feita em 214 nm. Nestas condições a LTG e o padrão interno (PI), lidocaína, puderam ser analisados em menos de 7 minutos. A HF-LPME foi avaliada no modo de 3 fases, usando 500 ?L de plasma e 3,5 mL de solução fosfato de sódio 50 mmol L-1 pH 9,0 como fase doadora. O solvente utilizado para impregnar a fibra foi o 1-octanol. Como fase aceptora foram utilizados 60 ?L de solução de ácido clorídrico pH 4,0. Para avaliação da DLLME, foi necessária uma etapa de pré-tratamento da amostra (500 ?L de plasma) com 1 mL de acetonitrila. Após isto, 1,3 mL do sobrenadante foram adicionados a 4 mL de solução fosfato de sódio 50 mmol L-1 pH 9,0 e 120 ?L de clorofórmio (solvente extrator) foram injetados nesta amostra aquosa e 165 ?L de fase sedimentada foram recuperados. As características de desempenho analítico para ambos os métodos foram avaliadas, sendo obtida linearidade na faixa de concentração plasmática de 1-20 ?g/mL e limite inferior de quantificação (LIQ) de 1 ?g mL-1. Os ensaios de precisão e exatidão apresentaram valores de acordo com os guias oficiais. Além disso, os métodos foram seletivos, não apresentaram efeito residual e as amostras foram estáveis. Os valores de recuperação foram de 54,3 e 23% para HF-LPME e DLLME, respectivamente. Os métodos validados foram aplicados com sucesso em amostras de plasma de pacientes epilépticos em tratamento com a LTG. Além disso, as duas técnicas foram comparadas e a HF-LPME apresentou vantagens em relação à DLLME, mostrando ser uma técnica promissora para análise de matrizes complexas, com reduzido consumo de solvente orgânico e possibilidade de automação.

Saproxylic insect communities inhabiting tree hollow microhabitats correspond with large food webs which simultaneously are constituted by multiple types of plant-animal and animal-animal interactions, according to the use of trophic resources (wood- and insect-dependent sub-networks), or to trophic habits or interaction types (xylophagous, saprophagous, xylomycetophagous, predators and commensals). We quantitatively assessed which properties of specialised networks were present in a complex networks involving different interacting types such as saproxylic community, and how they can be organised in trophic food webs. The architecture, interacting patterns and food web composition were evaluated along sub-networks, analysing their implications to network robustness from random and directed extinction simulations. A structure of large and cohesive modules with weakly connected nodes was observed throughout saproxylic sub-networks, composing the main food webs constituting this community. Insect-dependent sub-networks were more modular than wood-dependent sub-networks. Wood-dependent sub-networks presented higher species degree, connectance, links, linkage density, interaction strength, and were less specialised and more aggregated than insect-dependent sub-networks. These attributes defined high network robustness in wood-dependent sub-networks. Finally, our results emphasise the relevance of modularity, differences among interacting types and interrelations among them in modelling the structure of saproxylic communities and in determining their stability.

La principal aportación del trabajo es la formulación de una alternativa que facilita la determinación experimental del factor de pérdidas y, en consecuencia, mejorar las predicciones de aislamiento a ruido aéreo para vidrios con una o más capas intermedias sea cual fuere su naturaleza. Además, se realiza una revisión de las normativas relacionadas con los ensayos de los parámetros mecánicos necesarios para la caracterizar los vidrios, centrándonos en los monolíticos y los laminados. En efecto, uno de los problemas que se plantea en el contexto de la acústica de la edificación actualmente es el de cumplir con los requisitos de aislamiento acústico a ruido aéreo en fachada que exige el vigente Código Técnico de la Edificación (CTE). Como sabemos, en la fachada podemos distinguir la parte ciega y la parte hueca. La parte más débil en lo concerniente a aislamiento a ruido aéreo es la hueca (compuesta por el vidrio, carpintería y otros elementos). Si la carpintería es la adecuada, la superficie de vidrio se convierte en el elemento limitante. El Catálogo de Elementos Constructivos (CEC) del CTE, la propia norma UNE-EN 12758:2011, así como algunos, cada vez más, fabricantes ofrecen datos del aislamiento de vidrios simples, vidrios laminados y vidrios dobles. En el caso de vidrios laminados, estos datos se limitan únicamente en los que tienen un sola lámina intermedia y, además, no acústicos. Podemos hablar, por tanto, de una laguna de información en este sentido. Para obtener predicciones fiables de aislamiento acústico a ruido aéreo de particiones multicapa, como pueden ser los vidrios laminados es necesario disponer de las características mecánicas y una de las más relevantes es el factor de pérdidas.

The choice of sampling methods to survey saproxylic beetles is a key aspect to assessing conservation strategies for one of the most endangered assemblages in Europe. We evaluated the efficiency of three sampling methods: baited tube traps (TT), window traps in front of a hollow opening (WT), and emergence traps covering tree hollows (ET) to study richness and diversity of saproxylic beetle assemblages at species and family levels in Mediterranean woodlands. We also examined trap efficiency to report ecological diversity, and changes in the relative richness and abundance of species forming trophic guilds: xylophagous, saprophagous/saproxylophagous, xylomycetophagous, predators and commensals. WT and ET were similarly effective in reporting species richness and diversity at species and family levels, and provided an accurate profile of both the flying active and hollow-linked saproxylic beetle assemblages. WT and ET were the most complementary methods, together reporting more than 90 % of richness and diversity at both species and family levels. Diversity, richness and abundance of guilds were better characterized by ET, which indicates higher efficiency in outlining the ecological community of saproxylics that inhabit tree hollows. TT were the least effective method at both taxonomic levels, sampling a biased portion of the beetle assemblage attracted to trapping principles, however they could be used as a specific method for families such as Bostrichiidae, Biphyllidae, Melyridae, Mycetophagidae or Curculionidae Scolytinae species. Finally, ET and WT combination allows a better characterization of saproxylic assemblages in Mediterranean woodland, by recording species with different biology and linked to different microhabitat types.

To study the possibility of producing better water quality from municipal wastewater, a membrane bioreactor (MBR) pilot plant with flat sheet (FS) and hollow fiber (HF) membranes coupled with another pilot plant equipped with nanofiltration (NF)/reverse osmosis (RO) membranes were operated to treat municipal wastewater from the wastewater treatment plant (WWTP) Rincón de León, Alicante (Spain). This study was focused on improving the quality of the permeate obtained from the MBR process when complemented by NF or RO stages with respect to salinity, organic matter and nutrients. Furthermore, the removal efficiencies of 10 EMPs were evaluated, comparing the reductions achieved between the wastewater treatment by MBR (adsorption to sludge and biodegradation) and the later treatment using NF or RO (mainly size exclusion). The results showed that the high quality of water was obtained which is appropriate for reuse with salinity removal efficiencies higher than 97%, 96% for total organic carbon (TOC), 91% for nitrates View the MathML sourceNO3- and 99% for total phosphorous (TP). High removal efficiencies were obtained for the majority of the analyzed EMP compounds.

Acid pretreatment of lignocellulosic biomass, required for bioethanol production, generates large amounts of by-products, such as lignin and hydrolyzed hemicellulose fractions, which have found so far very limited applications. In this work, we demonstrate how the recovered hemicellulose hydrolysis products can be effectively utilized as a precursor for the synthesis of functional carbon materials through hydrothermal carbonization (HTC). The morphology and chemical structure of the synthesized HTC carbons are thoroughly characterized to highlight their similarities with glucose-derived HTC carbons. Furthermore, two routes for introducing porosity within the HTC carbon structure are presented: i) silica nanoparticle hard-templating, which is shown to be a viable method for the synthesis of carbonaceous hollow spheres; and ii) KOH chemical activation. The synthesized activated carbons (ACs) show an extremely high porosity (pore volume≈1.0 cm3 g−1) mostly composed of micropores (90 % of total pore volume). Because of their favorable textural properties, the ACs are further tested as electrodes for supercapacitors, yielding very promising results (300 F g−1 at 250 mA g−1) and confirming the high suitability of KOH-activated HTC carbons derived from spruce and corncob hydrolysis products as materials for electric double layer supercapacitors.

Silica sub-microtubes loaded with platinum nanoparticles have been prepared in flexible non-woven mats using co-axial electrospinning technique. A partially gelated sol made from tetraethyl orthosilicate was used as the silica precursor, and oil was used as the sacrificial template for the hollow channel generation. Platinum has been supported on the wall of the tubes just adding the metallic precursor to the sol–gel, thus obtaining the supported catalyst by one-pot method. The silica tubes have a high aspect ratio with external/internal diameters of 400/200 nm and well-dispersed platinum nanoparticles of around 2 nm. This catalyst showed a high NO conversion with very high selectivity to N2 at mild conditions in the presence of excess oxygen when using C3H6 as reducing agent. This relevant result reveals the potential of this technique to produce nanostructured catalysts onto easy to handle conformations.

The “dehesa” (grassland with scattered oak trees) is a typical Mediterranean ecosystem from west Iberian Peninsula that has resulted from the transformation of the forest by clearing and brushwood removing and the landscape is maintained mainly bulls and/or Iberian pigs. This ecosystem is characterized by the presence of old scattered trees that are considered as “keystone-structures”, which favor the presence of a wide range of biodiversity, especially those species that are wood-dependent (saproxylic insects). Saproxylics are a diversified group involved in the recycling process of nutrients in forest, and thus they are considered as a bioindicator group of the quality and conservation status of habitats, including a wide number of species under some categories of threat according the IUCN criteria. It is widely recognized the importance of studying the main factors that determine the structure and distribution of species assemblages at both spatial and temporal scales, nevertheless, the saproxylic assemblages has been poorly studied from the temporal dimension. With this study we provide knowledge about the effect of the “dehesa” heterogeneity, species seasonality and distribution on this habitat and we highlight the importance of the maintaining of traditional practices as a tool for saproxylic insect diversity and conservation.
