The effect of inoculation of Aspergillus flavus, Fusarium verticillioides, and Penicillium sp. in Dystrophic Red Latosol (DRL) and Eutroferric Red Latosol (ERL) soils with or without glucose on the total carbohydrate content and the dehydrogenase and amylase activities was studied. The fungal growth and spore production in culture medium with and without glucose were also evaluated. A completely randomized design with factorial arrangement was used. The addition of glucose in the culture medium increased the growth rate of A. flavus and Penicillium sp. but not of F. verticillioides. The number of spores increased 1.2 for F. verticillioides and 8.2 times for A. flavus in the medium with glucose, but was reduced 3.5 times for Penicillium sp. The total carbohydrates contents reduced significantly according to first and second degree equations. The consumption of total carbohydrates by A. flavus and Penicillium sp. was higher than the control or soil inoculated with F. verticillioides. The addition of glucose to soils benefited the use of carbohydrates, probably due to the stimulation of fungal growth. Dehydrogenase activity increased between 1.5 to 1.8 times (p <0.05) in soils with glucose and inoculated with the fungi (except F. verticillioides), in relation to soil without glucose. Amylase activity increased 1.3 to 1.5 times due to the addition of glucose in the soil. Increased amylase activity was observed in the DRL soil with glucose and inoculated with A. flavus and Penicillium sp. when compared to control.

At present, the Brazilian market prefers cabbage (Brassica oleracea var. capitata) of smaller size, which can be achieved by increasing population density; yet this management can alter the optimum rate of nitrogen (N), its second most required nutrient. This study was conducted in the Universidade Estadual Paulista, Jaboticabal, São Paulo, Brazil, from February to June 2004. The objective was to evaluate the effect of population density: 31 250 (DI) and 46 875 plants ha(-1) (D2) with 0, 100, 200 and 300 kg N ha-1 on the growth and production of cabbage Astrus. The experimental design consisted of randomized blocks with a 2 x 4 factorial arrangement and three replicates. Statistical analysis were a variance analysis (F test), the Tukey test for population density averages and polynomial regression for the N rates. In D1, an increase was recorded in the number of inner and outer leaves, dry matter of inner and outer leaves, the stem diameter at the insertion of the head, and the stem dry and fresh matter. The maximum size of the plant in D2 was 1.57 kg and was obtained with 300 kg N ha(-1), while in D1 it was 2.1 kg and was obtained with 244 kg N ha(-1). The optimal economic rate in 131 was 227.1 kg N ha(-1). The highest yield (72.7 t ha(-1)) was obtained with the highest N rate in D2. Smaller cabbage heads, commercially preferred, were obtained without the application of N, regardless of plant population.

Objetivou-se avaliar os efeitos da suplementação com diferentes fontes de energia e de compostos nitrogenados sobre o crescimento e produção de bacteriocinas de bactérias ácido-láticas in vitro. As incubações foram conduzidas utilizando-se fluido ruminal originado de um novilho Holandês-Zebu fistulado no rúmen. O animal foi mantido em pastagem de Brachiaria decumbens recebendo 200 g/dia de proteína bruta suplementar. Os substratos e o inóculo foram acondicionados em frascos de vidro considerando-se oito tratamentos: celulose, celulose e caseína, celulose e peptona de soja, celulose e ureia, amido, amido e caseína, amido e peptona de soja e amido e ureia. Incubações sucessivas foram conduzidas para seleção dos microrganismos em função das fontes energéticas e de compostos nitrogenados. O amido favoreceu o crescimento de bactérias ácido-láticas em comparação à celulose. A suplementação com proteína verdadeira (peptona de soja e caseína) estimulou o crescimento dessas bactérias em comparação ao controle (sem suplementação com compostos nitrogenados). A adição de ureia não estimulou o crescimento de bactérias ácido-láticas. Nenhuma atividade antimicrobiana foi detectada a partir das colônias de bactérias ácido-láticas isoladas. Fontes de proteína verdadeira incrementam a competição entre bactérias fermentadoras de carboidratos estruturais e não-estruturais.

Foram estimados os coeficientes de herdabilidade e a mudança genética para peso à desmama (PD), peso ao sobreano (PS), ganho de peso do nascimento à desmama (GND), ganho de peso da desmama ao sobreano (GDS), perímetro escrotal (PE) e idade ao primeiro parto (IPP) em animais da raça Nelore. Foram utilizados dados de 128.148 animais nascidos entre 1984 e 2006. Os componentes de variância foram estimados pelo método da máxima verossimilhança restrita, e os valores genéticos foram preditos por modelos mistos aplicando-se modelo animal bicaracterística, incluindo peso à desmama em todas as análises. As tendências genéticas foram estimadas pela regressão dos valores genéticos sobre o ano de nascimento dos animais. Os coeficientes de herdabilidade do efeito direto estimados foram de 0,23 (0,07) (PD); 0,24 (0,02) (PS); 0,21 (0,01) (GND); 0,23 (0,01) (GDS); 0,46 (0,02) (PE) e 0,15 (0,01) (IPP). As tendências genéticas diretas estimadas foram de 0,171 (0,01); 0,219 (0,02); 0,186 (0,03) e 0,224 (0,02) kg/ano para PD, PS, GND e GDS, respectivamente, o que representa incrementos de 0,10; 0,08; 0,13 e 0,22% nas médias das mesmas características ao ano, respectivamente. Para o PE e a IPP no período de 1984 a 1995, as tendências genéticas foram nulas, com valores de 0,011 (0,03) cm/ano e -0,003 (0,06) dias/ano, respectivamente. No segundo período considerado (1996 a 2006), as tendências genéticas para PE e IPP foram de 0,069 (0,01) cm/ano e -3,024 (0,04) dias/ano, respectivamente, indicando melhorias consideráveis em tais características. Esses valores sugerem que características produtivas e reprodutivas, quando utilizadas como critério de seleção, proporcionam progresso genético no rebanho, sendo indicadas para seleção de animais da raça Nelore.

Aluminum (Al3+) toxicity is a major limiting factor to crop productivity in acid soils. The effects of aluminum on root and shoot growth of physic nut (Jatropha curcas L.) young plants and, the uptake and distribution of phosphorus, calcium, magnesium and aluminum in the roots and shoots were investigated in the present study. Plants were grown in 2.5L pots in a greenhouse. After fourteen days of adaptation to nutrient solution, plants were exposed to Al concentrations of 0, 370, 740, 1,100 and 1,480 mu mol L-1, corresponding to an active Al3+ solution of 13.3, 35.3, 90.0, 153.3 and 220.7 mu mol L-1, respectively. The dry matter partitioning between roots, stems and leaves, and the concentrations of P, Ca, Mg and Al in plant tissue, were measured after 75 days exposure to Al. The increasing level of Al3+ activity in solution progressively decreased the growth of the shoot and root of physic nut plants, and at the two highest active Al3+ levels, plants showed morphological abnormalities typical of the toxicity caused by this metal. Higher Al3+ activity reduced P concentrations in leaves and Ca and Mg in leaves and roots of physic nut, demonstrating the effect of Al on the uptake, transport and use of these nutrients by plants. The Al accumulated preferentially in the roots of physic nut, whereas only a small amount was transported to shoots.

eIF5A is a highly conserved putative eukaryotic translation initiation factor that has been implicated in translation initiation, nucleocytoplasmic transport, mRNA decay, and cell proliferation, but with no precise function assigned so far. We have previously shown that high-copy PKCI suppresses the phenotype of tif51A-1, a temperature-sensitive mutant of eIF5A in S. cerevisiae. Here, in an attempt to further understand how Pkc1 functionally interacts with eIF-5A, it was determined that PKCI suppression of tif51A-1 is independent of the cell integrity MAP kinase cascade. Furthermore, two new suppressor genes, ZDS1 and GIC1, were identified. We demonstrated that ZDS1 and ZDS2 are necessary for PKC1, but not for GIC1 suppression. Moreover, high-copy GIC1 also suppresses the growth defect of a PKCI mutant (stt1), suggesting the existence of a Pkc1-Zds1-Gic1 pathway. Consistent with the function of Gic1 in actin organization, the tif51A-1 strain shows an actin polarity defect that is partially recovered by overexpression of Pkc1 and Zds1 as well as Gic1. Additionally, PCL1 and BNI1, important regulators of yeast cell polarity, also suppress tif51A-1 temperature sensitiviiy Taken together, these data strongly Support the correlated involvement of Pkc1 and eIF5A in establishing actin polarity, which is essential for bud formation and G1/S transition in S. cerevisiae.

The effects of nitrogen availability on growth and photosynthesis were followed in plants of sunflower (Helianthus annuus L., var. CATISSOL-01) grown in the greenhouse under natural photoperiod. The sunflower plants were grown in vermiculite under two contrasting nitrogen supply, with nitrogen supplied as ammonium nitrate. Higher nitrogen concentration resulted in higher shoot dry matter production per plant and the effect was apparent from 29 days after sowing (DAS). The difference in dry matter production was mainly attributed to the effect of nitrogen on leaf production and on individual leaf dry matter. The specific leaf weight (SLW) was not affected by the nitrogen supply. The photosynthetic CO2 assimilation (A) of the target leaves was remarkably improved by high nitrogen nutrition. However, irrespective of nitrogen supply, the decline in photosynthetic CO2 assimilation occurred before the end of leaf growth. Although nitrogen did not change significantly stomatal conductance (gs), high-N grown plants had lower intercellular CO2 concentration (C-i) when compared with low-N grown plants. Transpiration rate (E) was increased in high-N grown plants only at the beginning of leaf growth. However, this not resulted in lower intrinsic water use efficiency (WUE). (C) 2004 Elsevier B.V.. The fatigue crack behavior in metals and alloys under constant amplitude test conditions is usually described by relationships between the crack growth rate da/dN and the stress intensity factor range Delta K. In the present work, an enhanced two-parameter exponential equation of fatigue crack growth was introduced in order to describe sub-critical crack propagation behavior of Al 2524-T3 alloy, commonly used in aircraft engineering applications. It was demonstrated that besides adequately correlating the load ratio effects, the exponential model also accounts for the slight deviations from linearity shown by the experimental curves. A comparison with Elber, Kujawski and "Unified Approach" models allowed for verifying the better performance, when confronted to the other tested models, presented by the exponential model. The different potential of initiated and non-initiated urinary bladder mucosa (UBM) to develop neoplasia was quantitatively evaluated in the male Wistar rat. Initiation of carcinogenesis was accomplished with N-butyl-N- (4-hydroxybutyl) -nitrosamine (BBN). Stimuli for cell proliferation and apoptosis were obtained by exposure followed by withdrawal of 3% Uracil in the diet. The proliferation index (PI) was estimated in UBM immunostained for the proliferating nuclear cell antigen (PCNA). The apoptotic index (AI) and the density of papillary/nodular hyperplasia (PNH) were estimated in hematoxilin-eosin stained sections. PNH was the main proliferative response to the mechanical irritation by uracil, irrespective of previous initiation with BBN. Uracil exposure induced higher PI and PNH density in the initiated rats. After uracil withdrawal, there was a significant increase of the AI in both uracil-treated groups, which correlated well to the respective PNH density. However, at the end of the experiment, PNH incidence and density were significantly higher in the BBN-initiated mucosa, which also presented 18% incidence of papillomas and 27% of carcinomas. Therefore, under prolonged uracil calculi trauma, the UBM of BBN-initiated Wistar rats gives rise to epithelial proliferative lesions that progress to neoplasia through acquired resistance to apoptosis. The growth hormone receptor (GHR) is the cell surface receptor for growth hormone (GH) and is required for GH to carry out its effects on target tissues. The objectives of the present study were to estimate the allele and genotype frequencies of the GHR/Alu I gene polymorphism located in the regulatory region in beef cattle belonging to different genetic groups and to determine associations between this polymorphism and growth and carcass traits. Genotyping was performed on 384 animals, including 79 Nellore (Zebu), 30 Canchim (5/8 Charolais+3/8 Zebu), 30 Simmental X Nellore crossbred and 245 Angus x Nellore crossbred cattle. Alleles Alu I(+), Alu I(-) and Alu I(N)-null allele-were evidenced for the GHR/Alu I polymorphism and the frequency of the Alu I(N) allele was significantly higher than the frequency of the Alu I(+) and Alu I(-) alleles in all genetic groups. Genotype Alu I(N/N) of the GHRIAlu I predominated in Nellore animals, while the Alu I(N/+) and Alu I(N/-) predominated in the other genetic groups. In the association studies, traits of interest were analyzed using the General Linear Model (GLM) procedure of the SAS program and least squares means of the genotypes were compared by the Tukey test. Significant associations (P < 0.05) were observed between the Alu I(N/N) genotype of the GHRIAlu I polymorphism and lower weight gain and body weight at slaughter, although a confounding between genotypes and genetic groups may have occurred.
