Aim: Current analysis characterizes the effluent from bullfrog-rearing ponds during the grow-out phase; Methods: Temperature, pH, dissolved oxygen, electric conductivity, turbidity, total phosphorus, N-NH3, N-NO3, BOD5 and COD and the number of thermotolerant coliforms (Escherichia coli) of the inlet and outlet water of the ponds were analyzed twice a week. Assay consisted of a completely randomized experimental design with two treatments (inlet and outlet water) and six repetitions in a split-plot, coupled to collection over time as subplot; Results: All variables were significantly different (p < 0.05) between treatments and over time (p < 0.05). Average rates of temperature, pH and dissolved oxygen levels of the supply water were higher when compared to those of the effluent. The other variables such as conductivity, turbidity, total phosphorus, ammonia, nitrate, biological oxygen demand, chemical oxygen demand and E. coli were higher in the effluent when compared to rates in the supply water; Conclusions: The management during grow-out phase caused the deterioration of the water quality, with increasing levels of dissolved nutrients and the number of thermotolerant coliform. Ammonia and phosphorus levels in the effluent, caused by waste food, skin and feces, accelerate the eutrophication process of the receiving water body. Further studies on effluent treatment are required.

The aim of this study was to evaluate the effects of yeast cell wall extract (YCW) in dry diet on the fecal microbiota, concentration of short-chain fatty acids (SCFA) and on the odor reduction of cats feces. We used 20 animals of both sexes, randomly assigned to four treatments and five repetitions totaling 20 experimental units: 1) dry commercial diet (control); 2) control + 0.2%, 3) control + 0.4%, and 4) control + 0.6% of YCW in dry matter. Enterobacteriaceae and lactic acid bacteria, fecal concentration of acetic, propionic and butyric acids, ammonia nitrogen and sensory panel were performed. There were no significant differences (p> 0.05) for bacterial counts and the concentration of SCFA and ammonia, but in sensory panel a reduction in the odor of feces could be noted with the use of 0.2% of YCW. We concluded that the addition of up to 0.6% YCW had no effect on the microbiology and the concentration of fatty acids, but there is potential for its use as an additive because of the improvement in the odor of feces. However, further studies are needed to understand the mechanisms of action and the effects of prebiotics for domestic cats.

Fish farms' water quality management is analyzed with regard to the management employed and the different trophic states are compared within the system during the dry and rainy seasons. Six sites were marked two in the water supply (P1 and P2), and four within the fish farm (P3 to P6). Whereas sites P1 and P2 (water supply) were characterized as oligotrophic, the others were mesotrophic and eutrotrophic sites. Environmental variables, mainly nutrients, conductivity, COD, BOD5 and TSS tended to increase as from P3 due to management and fertilization. Greater impact has been registered in the fish farm under analysis for variables COD, ammonia, total phosphorus and TSS during the discharge and pond emptying period. Frequent monitoring of water quality should be undertaken in fish breeding and plankton production ponds, especially in those close to P3 and P4. Removal of sediment in decantation lake or P5 is also recommended to decrease nutrient concentrations, especially phosphorus, accumulated on the bottom soil.

Gastric ulcer is an excoriated area of the gastric mucosa. It is among the predominant gastrointestinal chronic diseases. The essential oils represent an important part of the traditional pharmacopoeia in many countries and have been successfully used for gastroprotection and ulcer healing. Thus, this review presents the experimental activity of essential oils traditionally used in the gastric ulcer prevention and treatment, considering their families, part of the plant studied, bioassays, and their mechanisms of gastroprotection and ulcer healing, with the aim of stimulating novel studies in the search for a new phytomedicine to treat gastric diseases. © 2012 The Authors Fundamental and Clinical Pharmacology © 2012 Société Française de Pharmacologie et de Thérapeutique.

Studies to determine suitable levels of intensification are essential for developing sustainable aquaculture. The objective of this study was to evaluate the quality of effluents discharged from ponds stocked with 10 (D10), 20 (D20), 40 (D40), and 80 (D80) postlarvae of Macrobrachium amazonicum/m2. Intake and effluent water samples were taken throughout a 5.5-mo grow-out cycle. In that study, twelve 0.01-ha earthen ponds were stocked postlarvae with 0.01g. Average water exchange rate was 15%/d; water was discharged from the bottom of the ponds. Prawns were fed a commercial feed with 38% crude protein according to their biomass (3-10%) and the concentration of dissolved oxygen (DO). In our research, temperature, turbidity, total suspended solids, conductivity, DO, pH, biochemical oxygen demand (BOD), chemical oxygen demand (COD), N-ammonia, N-nitrite, N-nitrate, N-Kjeldahl nitrogen, total phosphorus, and soluble orthophosphate were measured every 15d throughout the experiment in the early morning (0630 to 0730h). Turbidity was lower in D10 than in D20 and D40 and total phosphorus was higher in D80 than in D10 and D20. An analysis of principal components comparing treatments and intake water showed three groups: intake, D10 and a cluster of D20, D40, and D80. On the basis of the water characteristics found in our study it appears that the farming of M. amazonicum is likely to have a low environmental impact, at least up to a stocking density of 80prawns/m2. © by the World Aquaculture Society 2013.

The goal of this study was to evaluate the effects of different soybean oil availabilities on the intake and partial and total digestibility of dry matter (DM) and nutrients, rumen fermentation parameters, efficiency of microbial synthesis, and the rumen microbiota of crossbred beef heifers. Nine crossbred heifers fitted with rumen and duodenal cannulae were evaluated in a triple 3 × 3 Latin square design with three treatments and three periods in three simultaneous repetitions. Heifers approximately 18 months old, with mean initial and final body weights of 316.3±28.8 and 362.6±34.4 kg, respectively, were fed a diet containing 600. g/kg of corn silage and 400. g/kg concentrate with a 58.0. g/kg fat content in the total diet. The sources of lipids included soybean grain, rumen-protected fat, and soybean oil. The statistical analyses were conducted using PROC MIXED from SAS, and the means were compared using Tukey's test (P<0.05). Dietary lipid sources did not affect nutrient intake (P>0.05). Except the apparent digestibility of organic matter (P=0.024), the apparent digestibility of the other nutrients did not differ among the treatment groups. Regarding body nitrogen retention, the soybean grain treatment was more effective than the rumen-protected fat treatment (P=0.045); however, the soybean oil treatment did not differ from the other two treatments. In relation to the efficiency of microbial protein synthesis (g. N/kg of organic matter apparently digested in the rumen corrected for microbial organic matter), the soybean oil and soybean grain treatments were more efficient than the rumen-protected fat treatment (P=0.001). Animals fed rumen-protected fat had larger numbers of protozoa (P<0.001) and fungi (P<0.001) than those supplemented with soybean grain and soybean oil. The dietary lipid sources did not affect pH, the molar concentration of propionate and total volatile fatty acids (P>0.05), whereas the concentrations of ammonia nitrogen and acetate were higher in animals fed with rumen-protected fat than in those submitted to the other treatments. The use of different soybean oil availabilities did not affect nutrient intake; however, treatments with soybean oil and soybean grain were more efficient regarding nutrient intake than rumen-protected fat because they reduced the numbers of fungi and protozoa and consequently improved the efficiency of microbial protein synthesis. © 2013 Elsevier B.V.

The effects of exogenous enzymes supplementation on kibble diets for dogs formulated with soybean meal (SBM) as a substitute for poultry by-product meal (PM) was investigated on nutrient digestibility, fermentation products formation, post-prandial urea response and selected faecal bacteria counts. Two kibble diets with similar compositions were used in two trials: PM-based diet (28.9% of PM; soybean hulls as a fibre source) and SBM-based diet (29.9% of SBM). In experiment 1, the SBM diet was divided into three diets: SBM-0, without enzyme addition; SBM-1, covered after extrusion with 7500U protease/kg and 45U cellulase/kg; and SBM-2, covered with 15000U protease/kg and 90U cellulase/kg. In experiment 2, the SBM diet was divided into three diets: SBM-0; SBM-1, covered with 140U protease/kg; 8U cellulase/kg, 800U pectinase/kg, 60U phytase/kg, 40U betaglucanase/kg and 20U xylanase/kg; and SMB-2, covered with 700U protease/kg, 40U cellulase/kg, 4000U pectinase/kg, 300U phytase/kg, 200U betaglucanase/kg and 100U xylanase/kg. Each experiment followed a block design with six dogs per diet. Data were submitted to analysis of variance and means compared by orthogonal and polynomial contrasts (p<0.05). In both experiments, nutrients and energy digestibility did not differ between diets (p>0.05). SBM consumption resulted in increased faecal moisture and production (p<0.05), without effect on faecal score. Higher concentration of propionate, acetate and lactate, and lower ammonia and pH were found in the faeces of dogs fed SBM (p<0.05). Higher post-prandial urea was verified in dogs fed SBM (p<0.05). In experiment 2, the addition of enzymes increased faecal concentration of propionate, acetate and total short-chain fatty acid (p<0.05) and tended to reduce post-prandial urea concentration (p=0.06). Although with similar digestibility, SBM shows a worse utilization of absorbed amino acids than the PM. Soybean oligosaccharides can beneficially change gut fermentation product formation. Enzymes can increase the gut fermentation activity and improve the SBM proteic value. © 2013 Blackwell Verlag GmbH.

Recently, there is an interest in technologies that favour the use of coproducts for animal nutrition. The effect of adding two enzyme mixtures in diets for dogs formulated with wheat bran (WB) was evaluated. Two foods with similar compositions were formulated: negative control (NC; without WB) and test diet (25% of WB). The test diet was divided into four treatments: without enzyme (positive control), enzyme mixture 1 (ENZ1; added before extrusion β-glucanase, xylanase, cellulase, glucoamylase, phytase); enzyme mixture 2 (ENZ2; added before extrusion the ENZ1 more α-amylase); enzyme mixture 2 added after the extrusion (ENZ2ex). ENZ1 and ENZ2 were used to evaluate the enzyme effect on extruder pre-conditioner (processing additive) and ENZ2ex to evaluate the effect of enzyme supplementation for the animal. Digestibility was measured through total collection of faeces and urine. The experiment followed a randomized block design with five treatments (diets) and six dogs per diet, totalling 30 dogs (7.0 ± 1.2 years old and 11.0 ± 2.2 kg of body weight). Data were submitted to analysis of variance and means compared by Tukey's test and orthogonal contrasts (p < 0.05). Reducing sugars showed an important reduction after extrusion, suggesting the formation of carbohydrate complexes. The apparent total tract digestibility (ATTD) of dry matter, organic matter, crude protein, acid-hydrolysed fat and energy was higher in NC than in diets with WB (p < 0.001), without effects of enzyme additions. WB diets resulted in higher faecal production and concentration of short-chain fatty acids (SCFA) and reduced pH and ammonia concentration (p < 0.01), with no effect of enzyme addition. The enzyme addition did not result in improved digestibility of a diet high in non-starch polysaccharides; however, only ATTD was measured and nutrient fermentation in the large intestine may have interfered with the results obtained. In this work, experimental results are reported for a small scale cogeneration plant for power and refrigeration purposes. The plant includes a natural gas microturbine and an ammonia/water absorption chiller fired by steam. The system was tested under different turbine loads, steam pressures and chiller outlet temperatures. An evaluation based on the 1st and 2nd Laws of Thermodynamics was also performed. For the ambient temperature around 24°C and microturbine at full load, the plant is able to provide 19 kW of saturated steam at 5.3 bar (161 °C), corresponding to 9.2 kW of refrigeration at -5 °C (COP = 0.44). From a 2nd law point-of-view, it was found that there is an optimal chiller outlet temperature that maximizes the chiller exergetic efficiency. As expected, the microturbine presented the highest irreversibilities, followed by the absorption chiller and the HRSG. In order to reduce the plant exergy destruction, it is recommended a new design for the HRSG and a new insulation for the exhaust pipe. © 2013 Elsevier Ltd. All rights reserved.

This study aimed to evaluate the quality of 'Palmer' mangoes previously stored at low temperatures, after their transference to the environmental condition. Fruits harvested at physiological maturity were carefully transported to the Laboratory where they were selected, standardized as the color, size and absence of injuries and treated with fungicide before they were stored at 2°C (75.7% RH), 5°C (73.8% RH) e 12°C (82% RH) for 7, 14 and 21 days. At the end of each period, the fruits were transferred to environmental condition (22.9°C; 62.3% RH), where they were kept for 1, 3, 5 and 7 days, simulating the trading period, and evaluated for the occurrence of injuries and rottenness; peel and pulp color; firmness; contents of soluble solids, titratable acidity and ascorbic acid, as well as, the activities of the enzymes peroxidase, polyphenoloxidase and phenylalanine ammonia-lyase. The results indicated that 'Palmer' mangoes can be stored at 12°C for 21 days without damage to ripening, but with limitations due to the occurrence of decay. The storage at 2°C and 5°C was limited by the occurrence of injuries in the peel, but at the temperature of 2°C these symptoms were more severe and compromised the development of the characteristic color of the peel. However, the ripening of the pulp was not harmed, but this process occurred with less intensity than in mangoes maintained at 12°C.

Background: How to maintain gut health is a goal for scientists throughout the world. Therefore, microbiota management models for testing probiotics, prebiotics, and synbiotics have been developed.Methods: The SHIME® model was used to study the effect of Lactobacillus acidophilus 1014 on the fermentation pattern of the colon microbiota. Initially, an inoculum prepared from human feces was introduced into the reactor vessels and stabilized over 2-wk using a culture medium. This stabilization period was followed by a 2-wk control period during which the microbiota was monitored. The microbiota was then subjected to a 4-wk treatment period by adding 5 mL of sterile peptone water with L. acidophilus CRL1014 at the concentration of 108 CFU/mL to vessel one (the stomach compartment). Plate counts, Denaturing Gradient Gel Electrophoresis (DGGE), short-chain fatty acid (SCFA) and ammonium analyses were carried out for monitoring of the microbial community from the colon compartments.Results: A significant increase (p < 0.01) in the Lactobacillus spp. and Bifidobacterium spp. populations was observed during the treatment period. The DGGE obtained showed changes in the lactobacilli community from the colon compartments of the SHIME® reactor. The (SCFA) concentration increased (p < 0.01) during the treatment period, due mainly to significant increased levels of acetic, butyric, and propionic acids. However, ammonium concentrations decreased during the same period (p < 0.01).Conclusions: This study showed the beneficial influence of L. acidophilus CRL 1014 on microbial metabolism and lactobacilli community composition for improving human health. © 2013 Sivieri et al.; licensee BioMed Central Ltd.

The stimulation by Mg2+, Na+, K+, NH 4 +, and ATP of (Na+, K+)-ATPase activity in a gill microsomal fraction from the freshwater prawn Macrobrachium rosenbergii was examined. Immunofluorescence labeling revealed that the (Na +, K+)-ATPase α-subunit is distributed predominantly within the intralamellar septum, while Western blotting revealed a single α-subunit isoform of about 108 kDa M r. Under saturating Mg2+, Na+, and K+ concentrations, the enzyme hydrolyzed ATP, obeying cooperative kinetics with V M = 115.0 ± 2.3 U mg-1, K 0.5 = 0.10 ± 0.01 mmol L-1. Stimulation by Na+ (V M = 110.0 ± 3.3 U mg-1, K 0.5 = 1.30 ± 0.03 mmol L -1), Mg2+ (V M = 115.0 ± 4.6 U mg -1, K 0.5 = 0.96 ± 0.03 mmol L-1), NH4 + (V M = 141.0 ± 5.6 U mg -1, K 0.5 = 1.90 ± 0.04 mmol L-1), and K+ (V M = 120.0 ± 2.4 U mg-1, K M = 2.74 ± 0.08 mmol L-1) followed single saturation curves and, except for K+, exhibited site-site interaction kinetics. Ouabain inhibited ATPase activity by around 73 % with K I = 12.4 ± 1.3 mol L-1. Complementary inhibition studies suggest the presence of F0F1-, Na+-, or K +-ATPases, but not V(H+)- or Ca2+-ATPases, in the gill microsomal preparation. K+ and NH4 + synergistically stimulated enzyme activity (≈25 %), suggesting that these ions bind to different sites on the molecule. We propose a mechanism for the stimulation by both NH4 +, and K+ of the gill enzyme. © 2013 Springer Science+Business Media New York.

Agriculture is an activity in which there is the possibility of using lower quality water and reuse appears as an alternative for irrigation. However, the negative environmental effects of reuse must be understood. The objective of the present work was to verify the potential of groundwater contamination by nitrogen duo to drip irrigation with effluent from domestic wastewater treatment. Three crop cycles of lettuce cv. Raider were conducted, the first between October and November 2009, the second in April and May 2010 and the third one in June and July 2010. The use of effluents was compared with potable water at accumulated water depths of 98.8 mm for the first, 98.4 mm for the second and 119 mm for the third cycle. Soil percolate concentrations of nitrate, nitrite and ammonia were studied at 0.25 m and 0.50 m depths in a completely randomized design. Percolate concentrations of nitrite and ammonia in soil irrigated with effluents showed no groundwater contamination risk. Nitrate however, caused concern, albeit with similar concentrations for both effluent and potable water.

Eumelanin pigments show hydration-dependent conductivity, broad-band UV-vis absorption, and chelation of metal ions. Solution-processing of synthetic eumelanins opens new possibilities for the characterization of eumelanin in thin film form and its integration into bioelectronic devices. We investigate the effect of different synthesis routes and processing solvents on the growth, the morphology, and the chemical composition of eumelanin thin films using atomic force microscopy and X-ray photoelectron spectroscopy. We further characterize the films by transient electrical current measurements obtained at 50% to 90% relative humidity, relevant for bioelectronic applications. We show that the use of dimethyl sulfoxide is preferable over ammonia solution as processing solvent, yielding homogeneous films with surface roughnesses below 0.5 nm and a chemical composition in agreement with the eumelanin molecular structure. These eumelanin films grow in a quasi layer-by-layer mode, each layer being composed of nanoaggregates, 1-2 nm high, 10-30 nm large. The transient electrical measurements using a planar two-electrode device suggest that there are two contributions to the current, electronic and ionic, the latter being increasingly dominant at higher hydration, and point to the importance of time-dependent electrical characterization of eumelanin films. This study evaluated the ethanol addition as a strategy for start-up and acclimation of a pilot scale (1300 L) anaerobic sequencing batch biofilm reactor (AnSBBR) for the treatment of municipal landfill leachate with seasonal biodegradability variations. The treatment was carried out at ambient temperature (23.8 ± 2.1 °C) in the landfill area. In a first attempt, the leachate collected directly from landfill showed to be predominantly recalcitrant to anaerobic treatment and the acclimation was not possible. In a second attempt, adding ethanol to leachate, the reactor was successfully acclimated. After acclimation, without ethanol addition, the CODTotal influent ranged from 4970 to 13040 mg L-1 and the removal efficiencies ranged from 12.1% to 70.7%. A final test was carried out increasing the ammonia and free-ammonia concentration from 2486 mgN L-1 and 184 mgN L-1 to 4519 mgN L-1 and 634 mgN L-1, respectively, with no expressive inhibition verified. The start-up strategy was found to be feasible, providing the acclimation of the biomass in the AnSBBR, and maintaining the biomass active even when the leachate was recalcitrant. © 2013 Elsevier Ltd.
