We describe an adaptation of the rolling circle amplification (RCA) reporter system for the detection of protein Ags, termed “immunoRCA.” In immunoRCA, an oligonucleotide primer is covalently attached to an Ab; thus, in the presence of circular DNA, DNA polymerase, and nucleotides, amplification results in a long DNA molecule containing hundreds of copies of the circular DNA sequence that remain attached to the Ab and that can be detected in a variety of ways. Using immunoRCA, analytes were detected at sensitivities exceeding those of conventional enzyme immunoassays in ELISA and microparticle formats. The signal amplification afforded by immunoRCA also enabled immunoassays to be carried out in microspot and microarray formats with exquisite sensitivity. When Ags are present at concentrations down to fM levels, specifically bound Abs can be scored by counting discrete fluorescent signals arising from individual Ag–Ab complexes. Multiplex immunoRCA also was demonstrated by accurately quantifying Ags mixed in different ratios in a two-color, single-molecule-counting assay on a glass slide. ImmunoRCA thus combines high sensitivity and a very wide dynamic range with an unprecedented capability for single molecule detection. This Ag-detection method is of general applicability and is extendable to multiplexed immunoassays that employ a battery of different Abs, each labeled with a unique oligonucleotide primer, that can be discriminated by a color-coded visualization system. ImmunoRCA-profiling based on the simultaneous quantitation of multiple Ags should expand the power of immunoassays by exploiting the increased information content of ratio-based expression analysis.

Facile modification of oligodeoxyribonucleotides is required for efficient immobilization to a pre-activated glass surface. This report presents an oligodeoxyribonucleotide which contains a hairpin stem–loop structure with multiple phosphorothioate moieties in the loop. These moieties are used to anchor the oligo to glass slides that are pre-activated with bromoacetamidopropylsilane. The efficiency of the attachment reaction was improved by increasing the number of phosphorothioates in the loop, as shown in the remarkable enhancement of template hybridization and single base extension through catalysis by DNA polymerase. The loop and stem presumably serve as lateral spacers between neighboring oligodeoxyribonucleotides and as a linker arm between the glass surface and the single-stranded sequence of interest. The oligodeoxyribonucleotides of this hairpin stem–loop architecture with multiple phosphorothioate moieties have broad application in DNA chip-based gene analysis.

In order to support the structural genomic initiatives, both by rapidly classifying newly determined structures and by suggesting suitable targets for structure determination, we have recently developed several new protocols for classifying structures in the CATH domain database (http://www.biochem.ucl.ac.uk/bsm/cath). These aim to increase the speed of classification of new structures using fast algorithms for structure comparison (GRATH) and to improve the sensitivity in recognising distant structural relatives by incorporating sequence information from relatives in the genomes (DomainFinder). In order to ensure the integrity of the database given the expected increase in data, the CATH Protein Family Database (CATH-PFDB), which currently includes 25 320 structural domains and a further 160 000 sequence relatives has now been installed in a relational ORACLE database. This was essential for developing more rigorous validation procedures and for allowing efficient querying of the database, particularly for genome analysis. The associated Dictionary of Homologous Superfamilies [Bray,J.E., Todd,A.E., Pearl,F.M.G., Thornton,J.M. and Orengo,C.A. (2000) Protein Eng., 13, 153–165], which provides multiple structural alignments and functional information to assist in assigning new relatives, has also been expanded recently and now includes information for 903 homologous superfamilies. In order to improve coverage of known structures, preliminary classification levels are now provided for new structures at interim stages in the classification protocol. Since a large proportion of new structures can be rapidly classified using profile-based sequence analysis [e.g. PSI-BLAST: Altschul,S.F., Madden,T.L., Schaffer,A.A., Zhang,J., Zhang,Z., Miller,W. and Lipman,D.J. (1997) Nucleic Acids Res., 25, 3389–3402], this provides preliminary classification for easily recognisable homologues, which in the latest release of CATH (version 1.7) represented nearly three-quarters of the non-identical structures.

The CluSTr (Clusters of SWISS-PROT and TrEMBL proteins) database offers an automatic classification of SWISS-PROT and TrEMBL proteins into groups of related proteins. The clustering is based on analysis of all pairwise comparisons between protein sequences. Analysis has been carried out for different levels of protein similarity, yielding a hierarchical organisation of clusters. The database provides links to InterPro, which integrates information on protein families, domains and functional sites from PROSITE, PRINTS, Pfam and ProDom. Links to the InterPro graphical interface allow users to see at a glance whether proteins from the cluster share particular functional sites. CluSTr also provides cross-references to HSSP and PDB. The database is available for querying and browsing at http://www.ebi.ac.uk/clustr.

Signals emanating from CD40 play crucial roles in B-cell function. To identify molecules that transduce CD40 signalings, we have used the yeast two-hybrid system to done cDNAs encoding proteins that bind the cytoplasmic tail of CD40. A cDNA encoding a putative signal transducer protein, designated TRAF5, has been molecularly cloned. TRAF5 has a tumor necrosis factor receptor-associated factor (TRAF) domain in its carboxyl terminus and is most homologous to TRAF3, also known as CRAF1, CD40bp, or LAP-1, a previously identified CD40-associated factor. The amino terminus has a RING finger domain, a cluster of zinc fingers and a coiled-coil domain, which are also present in other members of the TRAF family protein except for TRAF1. In vitro binding assays revealed that TRAF5 associates with the cytoplasmic tail of CD40, but not with the cytoplasmic tail of tumor receptor factor receptor type 2, which associates with TRAF2. Based on analysis of the association between TRAF5 and various CD40 mutants, residues 230-269 of CD40 are required for the association with TRAF5. In contrast to TRAF3, overexpression of TRAF5 activates transcription factor nuclear factor kappa B. Furthermore, amino-terminally truncated forms of TRAF5 suppress the CD40-mediated induction of CD23 expression, as is the case with TRAF3. These results suggest that TRAF5 and TRAF3 could be involved in both common and distinct signaling pathways emanating from CD40.

The three-dimensional structure of murine mitochondrial carbonic anhydrase V has been determined and refined at 2.45-A resolution (crystallographic R factor = 0.187). Significant structural differences unique to the active site of carbonic anhydrase V are responsible for differences in the mechanism of catalytic proton transfer as compared with other carbonic anhydrase isozymes. In the prototypical isozyme, carbonic anhydrase II, catalytic proton transfer occurs via the shuttle group His-64; carbonic anhydrase V has Tyr-64, which is not an efficient proton shuttle due in part to the bulky adjacent side chain of Phe-65. Based on analysis of the structure of carbonic anhydrase V, we speculate that Tyr-131 may participate in proton transfer due to its proximity to zinc-bound solvent, its solvent accessibility, and its electrostatic environment in the protein structure. Finally, the design of isozyme-specific inhibitors is discussed in view of the complex between carbonic anhydrase V and acetazolamide, a transition-state analogue. Such inhibitors may be physiologically important in the regulation of blood glucose levels.

Cell-cell adhesion in zonula adherens and desmosomal junctions is mediated by cadherins, and recent crystal structures of the first domain from murine N-cadherin provide a plausible molecular basis for this adhesive action. A structure-based sequence analysis of this adhesive domain indicates that its fold is common to all extracellular cadherin domains. The cadherin folding topology is also shown to be similar to immunoglobulin-like domains and to other Greek-key beta-sandwich structures, as diverse as domains from plant cytochromes, bacterial cellulases, and eukaryotic transcription factors. Sequence similarities between cadherins and these other molecules are very low, however, and intron patterns are also different. On balance, independent origins for a favorable folding topology seem more likely than evolutionary divergence from an ancestor common to cadherins and immunoglobulins.

Trabajo que tiene por objetivo observar y describir el panorama de la investigación bibliométrica en la Argentina en el período comprendido entre los años 1984 y 2012, a partir del análisis de las publicaciones de autores de instituciones argentinas localizadas en buscadores web, repositorios temáticos y bases de datos regionales e internacionales. Interpreta las formas que revisten los ítems estudiados enfocando la atención en el volumen y evolución de la producción, tipo de literatura, idioma, temas y unidades de análisis. Calcula el índice de coautoría y tasas de colaboración nacional e internacional. Identifica los autores más productivos y las instituciones de afiliación más frecuentes. Determina la existencia de algunos grupos de investigación, caracterizando sus temáticas de investigación, las revistas donde publican y los congresos más frecuentes en los que participan

Este trabalho tem por finalidade mostrar a aplicação e a utilização de um aeromodelo elétrico de asa fixa, também conhecido como veículo aéreo não tripulado (VANT), com controle manual ou automático, para coleta de dados e imagens em propriedades rurais, com a premissa de auxiliar os gestores no processo de gestão e tomada de decisão. A metodologia utilizada para a realização das coletas foi feita por meio de voos programados em dias e condições diferentes, para verificação e análise de desempenho do aeromodelo. Os resultados obtidos com os voos foram acima do esperado, gerando excelentes imagens e dados confiáveis. Sendo assim, pôde-se concluir que a utilização de VANTs, em coletas de dados e imagens em propriedades rurais foi satisfatória e auxiliou os gestores no processo de gerenciamento e rotacionamento de animais no pasto, uma vez que as imagens permitiram uma boa visualização e o aeromodelo desenvolvido cumpriu o seu objetivo com bom desempenho e agilidade.

Este trabalho tem como objetivo estudar as modificações introduzidas, ao longo de sucessivas versões, nos modelos empíricos do programa computacional FRAPCON utilizado para a simulação do comportamento sob irradiação de varetas combustíveis de Reatores a Água Leve Pressurizada (Pressurized Water Reactor - PWR) em regime de estado estacionário e sob condições de alta queima. No estudo, foram analisados os modelos empíricos utilizados pelo FRAPCON e que são apresentados em sua documentação oficial. Um estudo bibliográfico foi conduzido sobre os efeitos da alta queima em combustíveis nucleares visando melhorar o entendimento dos modelos utilizados pelo FRAPCON nestas condições. Foram feitas simulações do comportamento sob irradiação de uma vareta combustível típica de um reator PWR utilizando as versões 3.3, 3.4 e 3.5 do FRAPCON. Os resultados apresentados pelas diferentes versões do programa foram comparados entre si de forma a verificar as consequências das mudanças de modelo nos parâmetros de saída do programa. Foi possível observar que as modificações introduzidas trouxeram diferenças significativas nos resultados de parâmetros térmicos e mecânicos da vareta combustível, principalmente quando se evoluiu da versão FRAPCON-3.3 para a versão FRAPCON-3.5. Nessa ultima versão, obteve-se menores temperaturas na vareta combustível, menores tensões e deformações no revestimento, menor espessura da camada de oxido formada no revestimento a altas queimas na vareta combustível.

Neste trabalho, foi investigado o efeito do tamanho do abrasivo e do pH do meio na resistência ao desgaste abrasivo do aço H-13 com matriz martensítica e do aço Hadfield com matriz austenítica. Ensaios de abrasão foram realizados utilizando o equipamento roda de borracha a úmido, variando o tamanho do abrasivo entre 0,15 e 2,40 mm e o pH do meio entre 5,5 e 12,8. As microestruturas dos materiais estudados foram analisadas utilizando microscopia óptica, as superfícies de desgaste e as partículas de desgaste foram analisadas em microscópio eletrônico de varredura. A macrodureza e a microdureza, antes e após os ensaios, foram obtidas utilizando durômetro Vickers. A topografia da região central da superfície de desgaste foi obtida utilizando Perfilometria 3D, visando obter valores de profundidade de penetração do abrasivo. Os resultados mostraram que o aço Hadfield é mais resistente do que o aço H-13 em todos os valores de pH e tamanhos de abrasivo utilizados. Para os dois materiais, a perda de massa aumenta linearmente até um tamanho crítico de abrasivo (TCA) e, após este, a mesma continua a aumentar, mas com uma intensidade menor. Para os dois materiais e para todos os tamanhos de abrasivo, o aumento do pH do meio resultou em menores perdas de massa, sendo este efeito maior para os dois menores tamanhos de abrasivo. Para maiores valores de pH, foram observadas menores profundidades de penetração do abrasivo. A microdureza da superfície de desgaste do aço H-13 sofreu um pequeno aumento com o aumento do tamanho do abrasivo enquanto que para o aço Hadfield esse aumento foi mais intenso. A análise das partículas de desgaste mostraram que, para todas as condições ensaiadas, os debris do aço H-13 tinham duas morfologias, contínuas e descontínuas enquanto que os cavacos do aço Hadfield foram sempre descontínuos. Para os dois materiais, foram observados dois micromecanismos de desgaste, sendo eles microcorte e microsulcamento. Por fim, os resultados apresentados neste trabalho sugerem que a análise de desempenho do aço Hadfield em serviço deve considerar o pH do meio bem como a granulometria do abrasivo em contato.

Mediante a crescente necessidade de aumento na oferta de energia elétrica devido à constante elevação na demanda mundial, esta dissertação avalia o desempenho de um sistema conversor de energia de ondas marítimas em energia elétrica. O sistema em análise é o de coluna de água oscilante com turbina de dupla ação instalado na costa. Utiliza-se um modelo regular de ondas como perturbação à dinâmica de uma câmara semi-submersa gerando fluxo de ar através de uma turbina à ar de dupla ação. O sistema final é não linear e com parâmetros variantes no tempo. A dissertação investiga possibilidades para o aumento do rendimento da turbina em diferentes condições de mar através do método de simulação numérica. Após a modelagem física e matemática do sistema escolhido, inicia-se a síntese de um controlador proporcional derivativo para controle da pressão de ar na turbina em torno da pressão ideal de trabalho da mesma. A análise inclui o comparativo entre os resultados do sistema com e sem controlador e a avaliação de robustez utilizando ondas com amplitude variável. O trabalho apresenta ainda propostas de otimização do sistema para trabalhar em condições similares a região de Pecém no Brasil. Pelos resultados obtidos nas simulações, conclui-se que o rendimento e a robustez do sistema podem melhorar utilizando um sistema controlado. O rendimento do sistema poderá ainda ser otimizado para a região de instalação.

Examining a team’s performance from a physical point of view their momentum might indicate unexpected turning points in defeat or success. Physicists describe this value as to require some effort to be started, but also that it is relatively easy to keep it going once a sufficient level is reached (Reed and Hughes, 2006). Unlike football, rugby, handball and many more sports, a regular volleyball match is not limited by time but by points that need to be gathered. Every minute more than one point is won by either one team or the other. That means a series of successive points enlarges the gap between the teams making it more and more difficult to catch up with the leading one. This concept of gathering momentum, or the reverse in a performance, can give the coaches, athletes and sports scientists further insights into winning and losing performances. Momentum investigations also contain dependencies between performances or questions if future performances are reliant upon past streaks. Squash and volleyball share the characteristic of being played up to a certain amount of points. Squash was examined according to the momentum of players by Hughes et al. (2006). The initial aim was to expand normative profiles of elite squash players using momentum graphs of winners and errors to explore ‘turning points’ in a performance. Dynamic systems theory has enabled the definition of perturbations in sports exhibiting rhythms (Hughes et al., 2000; McGarry et al., 2002; Murray et al., 2008), and how players and teams cause these disruptions of rhythm can inform on the way they play, these techniques also contribute to profiling methods. Together with the analysis of one’s own performance it is essential to have an understanding of your oppositions’ tactical strengths and weaknesses. By modelling the oppositions’ performance it is possible to predict certain outcomes and patterns, and therefore intervene or change tactics before the critical incident occurs. The modelling of competitive sport is an informative analytic technique as it directs the attention of the modeller to the critical aspects of data that delineate successful performance (McGarry & Franks, 1996). Using tactical performance profiles to pull out and visualise these critical aspects of performance, players can build justified and sophisticated tactical plans. The area is discussed and reviewed, critically appraising the research completed in this element of Performance Analysis.

El objetivo de estudio es conocer los tipos de saques utilizados, dependiendo el momento del set en el que se producen. Este estudio ha sido desarrollado durante el torneo Nestea Spanish Master de Vóley Playa disputado en Valencia en el año 2006. La muestra de estudio la componen 10 jugadoras que conforman 5 equipos con un total de 4 encuentros analizados que suman 331 saques analizados. El análisis de las videograbaciones se llevó a cabo con el software SportCode Pro v.8.5.2. Los saques se clasificaron dependiendo del momento en el que se produjeron, siendo la franja 1 (del punto 1 al 7), franja 2 (del punto 8 al 14) y franja 3 (del punto 15 al 21). El análisis de datos se llevo a cabo con el software SPSS v.19. La prueba Chi-cuadrado, estableció diferencias significativas entre los diferentes tipos de saques para la franja 1 y 3 (p<.05), pero no se establecieron diferencias significativas en la franja 2 para los tres tipos de saque utilizado (p>.05). Se experimenta una disminución en el uso del saque en potencia (SP) de la franja 1 (84.1%) con respecto al de la franja 3 (4.8%), mientras que el saque flotante (SF) aumenta de la franja 1 (13.5%) a la franja 3 (81%). Finalmente el saque flotante (SF) en salto aumentan de la franja 1 (2.4%) a la franja 2 (28%) y decrece de esta última a la franja 3 (14.3%).

In this paper we present a novel image processing algorithm providing good preliminary capabilities for in vitro detection of malaria. The proposed concept is based upon analysis of the temporal variation of each pixel. Changes in dark pixels mean that inter cellular activity happened, indicating the presence of the malaria parasite inside the cell. Preliminary experimental results involving analysis of red blood cells being either healthy or infected with malaria parasites, validated the potential benefit of the proposed numerical approach.
