Douze contaminants émergents (composés pharmaceutiques, pesticides et hormones) ont été quantifiés et extraits de l'eau de rivières et d’échantillons d'eaux usées municipales. La séparation des solides en suspension est effectuée par filtration des échantillons d'eau. L'effet de filtration sur les concentrations de contaminants dissous a été évaluée afin de minimiser les pertes de composés cibles. Les échantillons ont été lyophilisés et ont été extraits en deux cycles par ultrasons combinés avec une étape de nettoyage sur cartouche d’extraction de type C18. La quantification a été réalisée en utilisant la spectrométrie de masse. Les recouvrements de la méthode pour tous les composés ont varié de 68 à 112% dans toutes les matrices étudiées, sauf pour le sulfaméthoxazole et le diclofénac où des recouvrements plus modestes ont été obtenus (38 à 85%). Les limites de détection pour les 12 analytes dans les sédiments et particules en suspension (SPM) de la rivière variaient de 0,7 à 9,4 ng g-1 et de 21 à 92 ng g-1, pour les échantillons SPM de station d'épuration. Tous les contaminants émergents cibles ont été détectés à des concentrations variant de 3 à 5440 ng g-1 dans les matrices étudiées, les concentrations les plus élevées ont été observées dans les échantillons SPM de stations d'épuration. Une partie importante de certains de ces contaminants est clairement associée aux sédiments de rivière ou aux particules en suspension. L’optimisation des processus de traitement de l'eau et le devenir environnemental doit absolument tenir compte de la fraction de contaminants qui liée à des particules si on espère avoir un bilan de masse raisonnable.

Selenoproteins are proteins containing selenium in the form of the 21st amino acid, selenocysteine. Selenocysteine (Sec) is directly synthesized onto its cognate tRNA (tRNA[Ser]Sec or tRNASec) and inserted into selenoproteins co-translationally with the help of various cis- and trans-acting factors. Among those factors, SecP43 has been reported to possibly play an essential role in the methylation at the 2’-hydroxylribosyl moiety in the wobble position (Um34) of Sec-tRNA[Ser]Sec and consequently reduce the expression of glutathione peroxidase 1. SecP43 also called tRNASec-associated protein has also been reported to interact in with SepSecS and tRNASec in vivo and the targeted removal of one of these proteins affected the binding of the other to the Sec-tRNASec. The initial aim of the project was to solve the structure of SecP43 by means of x-ray crystallography. Secondly, we were interested in characterizing the interaction of the latter with some of the components of the selenocysteine insertion machinery. These factors are SepSecS and tRNASec. We were able to optimize the expression and the purification of soluble form of the human homologue of SecP43 and of SepSecS by using an adapted auto-induction protocol. This was a major challenge considering that full length SecP43 has not been expressed and purify to date. We did not succeed in crystallizing SecP43. Our failure to crystallize SecP43 is probably due to the fact that it is a partially folded protein as we were able to demonstrate by SAXS (Small Angle X-ray Scattering). The SecP43 envelope calculated by SAXS displayed a rod-shape like structure. In order to enhance the stability of SecP43 required for crystallization, binding affinity studies were conducted to characterize the interaction between SecP43, tRNASec and SepSecS. We did not detect an interaction between SecP43 and tRNASec by using EMSA (Electrophoretic Mobility Shift Assay) and gel filtration. We also could not detect an interaction between SecP43 and SepSecS using a cross-linking assay. In contrast, the tRNASec/SepSecS interaction was demonstrated by EMSA and the addition of SecP43 seemed to reduce the binding affinity. Therefore, SecP43 might induce a conformational change in SepSecS in the presence of tRNASec.

De nombreuses études ont bien démontré que l’activation du système rénine-angiotensine (RAS) joue un rôle important dans le développement de l’hypertension et de la néphropathie diabétique (DN). La découverte de l’enzyme de conversion de l’angiotensine-2 (ACE2) et l’identification du récepteur MAS, spécifique pour l’angiotensine 1-7 (Ang 1-7), ont permis d’identifier deux nouveaux membres du RAS. L’axe ACE2/Ang 1-7/MAS contrebalance les effets de l’axe ACE/Ang II/AT1. Plusieurs évidences impliquent la contribution du RAS intrarénal dans la DN. Des études réalisées dans notre laboratoire avec des souris transgéniques surexprimant l’angiotensinogène de rat dans les cellules de leurs tubules proximaux rénaux (RPTCs) ont permis de démontrer l’importance du RAS intrarénal dans l’induction de l’hypertension et les dommages rénaux. Nous avons également observé que l’expression rénale de l’ACE2 et les niveaux urinaires d’ANG 1-7 sont plus faibles chez les souris Akita (diabète de type 1) et qu’un traitement avec des bloqueurs du RAS permet de normaliser l’expression de l’ACE2 et de prévenir le développement de l’hypertension dans le modèle des souris Akita. Dans un milieu diabétique, à la fois la glycémie et l’angiotensine II (Ang II) peuvent induire la génération des espèces réactives de l’oxygène (ROS), contribuant ainsi aux dommages rénaux. Afin d’explorer la relation entre les ROS, ACE2 et la DN, nous avons créé des souris Akita transgéniques surexprimant la catalase (Cat) dans les RPTCs, en croisant des souris Akita diabétique de type 1 à notre modèle de souris transgéniques surexprimant la Cat de rat dans les RPTCs. Dans une seconde étude, des souris Akita ont été traitées avec l’Ang 1-7 ou une combinaison d’Ang 1-7 et de son antagoniste, A779, afin d’étudier la relation entre l’action de l’Ang 1-7, l’hypertension systolique (sHTN), le stress oxydatif, les dommages rénaux, ACE2 et l’expression du récepteur Mas. Nos résultats ont montré que la surexpression de Cat atténue le stress oxydatif rénal; prévient l’hypertension, améliore le taux de filtration glomérulaire, l’albuminurie, l’hypertrophie rénale, la fibrose tubulo-interstitielle et l’apoptose tubulaire; et supprime l’expression des gènes profibrotiques et proapoptotiques dans les RPTCs des souris Akita Cat-Tg lorsque comparées aux souris Akita. De plus, la surexpression de Cat dans les RPTC des souris Akita normalise l’expression rénale de l’ACE2 et les niveaux urinaires d’Ang 1-7. D’autre part, l’administration d’Ang 1-7 prévient l’hypertension systémique, normalise le ratio albumine/créatinine urinaire et atténue l’hyperfiltration glomérulaire des souris Akita, sans affecter la glycémie sanguine. De plus, le traitement avec l’Ang 1-7 atténue aussi le stress oxydatif et l’expression de la NADPH oxydase, Agt, ACE, TGF-β1 (transforming growth factor-β1) et collagène IV, tout en augmentant l’expression de l’ACE2 et du récepteur Mas dans les reins des souris Akita. Ces effets sont renversés par la co-admininstration d’A779. Ces résultats démontrent que la surexpression de Cat prévient l’hypertension et la progression de la néphropathie, en plus de mettre en lumière l’importance du stress oxydatif intrarénal et l’expression de l’ACE2 comme facteurs contribuant à l’hypertension et les dommages rénaux observés dans le diabète. En outre, nos données suggèrent que l’Ang 1-7 joue un rôle protecteur dans l’hypertension et les dommages aux RPTC dans le diabète, principalement en réduisant les voies de signalisations du stress oxydatif dans les reins et en normalisant l’expression de l’ACE2 et du récepteur Mas. Nos résultats indiquent aussi que l’Ang 1-7 pourrait agir comme un agent thérapeutique potentiel dans le traitement de l’hypertension systémique et les dommages rénaux observés dans le diabète. En conséquence, l’Ang 1-7 est responsable du rôle protecteur de l’ACE2 dans l’hypertension et la DN.

Diabetes Mellitus is a metabolic disorder associated with insulin deficiency, which not.only affects the carbohydrate metabolism but also is associated with various central and peripheral complications. Chronic hyperglycemia during diabetes mellitus is a major initiator of diabetic microvascular complications like retinopathy, neuropathy, The central nervous system (CNS) neurotransmitters play an important role in the regulation of glucose homeostasis. These neurotransmitters mediate rapid intracellular communications not only within the central nervous system but also in the peripheral tissues. They exert their function through receptors present in both neuronal and non neuronal cell surface that trigger second messenger signaling pathways. Dopamine is a neurotransmitter that has been implicated in various central neuronal degenerative disorders like Parkinson's disease and behavioral diseases like Schizophrenia. Dopamine is synthesised from tyrosine, stored in vesicles in axon terminals and released when the neuron is depolarised. Dopamine interacts with specific membrane receptors to produce its effect. Dopamine plays an important role both centrally and peripherally. The recent identification of five dopamine receptor subtypes provides a basis for understanding dopamine's central and peripheral actions . Dopamine receptors are classified into two major groups : DA D1 like and DA D2 like. Dopamine D1 like receptors consists of DA D1 and DA D5 receptors . Dopamine D2 like receptors consists of DA D2, DA D3 and DA D4 receptors. Stimulation of the DA D1 receptor gives rise to increased production of cAMP. Dopamine D2 receptors inhibit cAMP production, but activate the inositol phosphate second messenger system . Impairment of central dopamine neurotransmission causes muscle rigidity, hormonal regulation , thought disorder and cocaine addiction. Peripheral dopamine receptors mediate changes in blood flow, glomerular filtration rate, sodium excretion and catecholamine release. The dopamine D2 receptors increased in the corpus striatum and cerebral cortex but decreased in the hypothalamus and brain stem indicating their involvement in regulating insulin secretion. Dopamine D2 receptor which has a stimulatory effecton insulin secretion decreased in the pancreatic islets during diabetes. Our in vitro studies confirmed the stimulatory role of dopamine D2 receptors in stimulation of glucose induced insulin secretion. A detailed study at the molecular level on the mechanisms involved in the role of dopamine in insulin secretion, its functional modification could lead to therapeutic interventions that will have immense clinical importance.

The present investigations have considerably enhanced the existing knowledge on the biology and distribution/availability pattern of D.incarnatus in the Malippuram region. The species occurs in good concentration during October - March/April, and disappears from the area during late premonsoon and monsoon months. Recolonising the area in September, it grows fast in the subsequent months. The life span of the species is estimated to be about an year. Studies on the reproductive biology of the species have revealed that there are two spawning peaks, the major peak in February - March and minor peak, in December. The salinity regime of the area influences the reproductive activity. These observations form the original contribution in the thesis. The information on variation in water content, protein,glycogen and lipid levels in relation to reproductive cycle has helped to a better understanding of the gametogenic activity and spawning of the species. Similarly, the findings on salinity tolerance and filtration rate have shown that small sized clams exhibit greater tolerance range than larger clams, and grow at a faster rate with active metabolism. It is hoped that these information would considerably add to the present knowledge of the basic facts which are relevant to the improvement and management of the clam fishery of this region.

The thesis entitled "Sol-Gel Alumina Nano Composites for Functional Applications" investigate sol-gel methods of synthesis of alumina nanocomposites special reference to alumina-aluminium titanate and alumina-lanthanum phosphate composites. The functional properties such as thermal expansion coefficient and thermal shock resistance, machinability of composites as well as thermal protection are highlighted in addition to novel approach in synthesis of composites.A general introduction of alumina matrix composites materials, followed by brief coverage of alumina-aluminium titanate and alumina-lanthanum phosphate composites is highlight of the first chapter. The second chapter deals with the sol-gel synthesis of aluminium titanate and alumina-aluminium titanate composite. The synthesis of machinable substrate, based on alumina and lanthanum phosphate forms the basis of the third chapter. The fourth chapter describes the sol-gel coating of mullite on SiC substrate for the possible gas filtration application.

In India industrial pollution has become a subject of increasing concern.Incidents of industrial pollution have been reported from many parts of the country. Cochin, the collection site of the present study, being the industrial capital of Kerela is also a harbour, is vulnerable to pollution by trace metal contaminants. In the recent times, pollutants of greatest concern in the aquatic environment are those which are persistent such as toxic heavy metals and the chlorinated hydrocarbons which include insecticides and pesticides.The animals collected from the clam bed situated on the northern side af Cochin bermouth are subject to wide fluctuations in salinity both seasonal and tidal. also; salinity is considered as an important parameter influencing the.-physiological functioning of an organism. Hence, the salinity tolerance of the animal is worked out. Considering the potential vulnerability of Cochin backwaters to heavy metal pollution, the impact of heavy metal copper (II) on the bivalve Sunetta sripta was conceived. Static bioassays were conducted for the determination of the sublethal concentrations of the metal as a preliminary step towards the toxicity studies. Oxygen consumption and filtration rate which are considered as reliable sublethal toxicity indices were employed for investigating the toxic effects of the metal. Bioaccumulation, a physiological phenomenon which can be of importance from the public health point of view, and also in the assessment of environmental quality is also dealt with.

In this thesis certain important aspects of heavy metal toxicity have been worked out. Recent studies have clearly shown that when experimental media contained more than one heavy metals, such metals could conspicuously influence the toxic reaction of the animals both in terms of quantity and nature. The experimental results available on individual metal toxicity show that, in majority of such results, unrealistically high concentrations of dissolved metals are involved. A remarkable number of factors have been shown to influence metal toxicity such as various environmental factors particularly temperature and salinity, the condition of the organism and the ability of some of the marine organisms to adapt to metallic contamination. Further, some of the more sensitive functions like embryonic and larval development, growth and fecundity, oxygen utilization and the function of various enzymes are found to be demonstrably sensitive in the presence of heavy metals. However, some of the above functions could be compensated for by adaptive process. If it is assumed that the presence of a single metal in higher concentrations could affect the life function of marine animals, more than one metal in the experimental media should manifest such effects in a greater scale. Commonly known as synergism or more than additivity, majority of heavy metals bring about synergistic reaction .The work presented in this thesis comprises lethal and sublethal toxicities of different salt forms of copper and silver on the brown mussel Perna indica. during the present investigation sublethal concentrations of copper and silver in their dent effects on survival, oxygen consumption, filtration, accumulation and depuration on Perna indica. The results are presented under different sections to make the presentation meaningful .

The most common and conventional method for removing turbidity from water is by coagulating with alum or iron salts, and settling the precipitate in suitably designed clarifiers followed by filtration. But the sludge produced is bulky, difficult to dewater and accumulates in the dumping grounds causing environmental problems. Synthetic polymers such as polyacrylamide and polyethyleneoxide have been investigated for their ability to remove turbidity. They overcome many of the disadvantages of conventional methods, but are cost—effective only when rapid flocculation and reduction in sludge volume are demanded. Considering the aforementioned situation, it was felt that more easily available and eco-friendly materials must be developed for removing turbidity from water. The results of our studies in this direction are presented in this thesis. The thesis comprises of nine chapters, with a common bibliography at the end. Chapter 1 gives an introduction to the nature of turbidity and colour usually present in water. Chapter 2 discusses the nature and availability of the principal material used in these studies, namely chitosan. Chapters 3 to 8, which deal with the actual experimental work, are further subdivided into (a) introduction, (b) materials and methods, (c) results and discussion and (d) conclusions. Chapter 9 summarises the entire work so as to put the results and conclusions into proper perspective.

The present study has yielded a great deal of information on nutrition of pearl oyster larvae. T he formulae presented may be used effectively and with advantage in improving the larval rearing system with specific reference to nutritional aspects. It is also hoped that this is the first comprehensive study on pearl oyster larval nutrition would stimulate further detailed investigations on many of the other finer aspects of tropical bivalve larval nutrition.

Zusammenfassung (deutsch) Seit den 1980iger Jahren wächst die Bedeutung der sog. Bildschaffenden Methoden für die Bestimmung der Qualität ökologischer Produkte. Zu diesen Methoden gehört die Biokristallisation, Steigbild und Rundfilter-Chromatographie. Die Ergebnisse dieser Methoden sind Bilder, die anhand definierter Kriterien ausgewertet werden. Bei der Biokristallisation sind es mehr oder weniger geordnete Kristallisationen auf einer Glasplatte, bei dem Steigbild zweidimensionale Strukturen auf Chromatographiepapier. In der Vergangenheit wurden die Bilder von Spezialisten ausgewertet, die nach einer längeren Schulung produktspezifische Kriterien entwickelt hatten. Im Gegensatz zur Dünnschicht-Chromatographie, wo der einzelne Stoff von der Matrix separiert wird, ist das Ziel beim Steigbild, Strukturen der möglichst ganzen Probe zu erzeugen. Die Methode wurde von Kolisko in den 1929iger Jahren entwickelt, wobei eine Kombination aus Chromatographieprozess und Metallkomplexreaktionen genutzt wurde. Die Firma WALA entwickelte die Methode für die Kontrolle ihrer Produkte und setze Silbernitrat und Eisensulfat ein. Bisher wurde die Methode qualitativ beschreibend ausgewertet, wobei einzelne Bildelemente und deren Interaktion beschrieben wurden. Deshalb musste für die vorliegende Arbeit Auswertungsmethoden entwickelt werden, mit denen auch eine statistische Bearbeitung der Ergebnisse möglich ist (nominale Unterscheidung von proben anhand der Bilder). Die Methode wurde bisher in einer Reihe von Studien eingesetzt (u.a. die Unterscheidung von Produktionsweisen). Obwohl die Bilder nur qualitativ ausgewertet wurden, konnten geschulte Prüfpersonen Proben aus verschiedenen Anbausystemen anhand der Bilder trennen. Die Ergebnisse wurden aber nicht so dokumentiert, dass sie den Erfordernissen internationaler Standardnormen für Laboratorien genügten. Deshalb mussten für diese Arbeit zunächst die Prozeduren dokumentiert und eine systematische Untersuchung zu den Einflussgrößen durchgeführt werden. Dazu wurde die visuelle Bildauswertung entwickelt und standardisiert. Die visuelle Bildauswertung basiert auf morphologischen Kriterien der Bilder von den untersuchten Weizen- und Möhrenproben. Ein Panel aus geschulten Personen entwickelte dann die Kriterien und legte sie anhand von Referenzbildern fest. Die Bilder der vorliegenden Arbeit wurden mit der einfach beschreibenden Prüfung ausgewertet, wie sie aus der sensorischen Prüfung von Lebensmitteln übernommen werden konnte. Mit geschulten und ungeschulten Prüfpersonen wurden Weizenproben und verschiedene Möhrensäfte mit der sog. Dreiecksprüfung ausgewertet (von ISO 4120). Alle Laborprozeduren wurden dokumentiert. Mit der Anwendung dieser Prozeduren wurden Vergleichsversuche mit Laboren in Dänemark und Holland (BRAD, LBI) durchgeführt. Die Ergebnisse waren sowohl für Weizen- als auch für Möhrenproben vergleichbar, wobei alle drei Labore zwischen jeweils zwei Proben unterscheiden konnten. Die systematische Untersuchung zu den Einflussgrößen zeigte, dass das Unterscheidungsvermögen der Methode vor allem von den klimatischen Bedingungen während der Steigphasen beeinflusst wird. Auch die Präkonditionierung der Papiere hat einen großen Einfluss, während die Wasserqualität (ultra-filtriert, de-ionisiert, destilliert) eine untergeordnete Bedeutung hat. Für Weizen- und Möhrenproben wurde sowohl die Wiederholbarkeit als auch die Reproduzierbarkeit getestet. Die Unterschiede in den Bildern der verschiedenen Proben waren dabei immer größer als die Variation durch Proben- und Bildwiederholung und das Labor. Die so charakterisierte Methode wurde auf kodierte Proben von definierten Feldversuchen und auf Marktproben (Paarvergleich von Anbausystemen ökologisch und konventionell) angewandt, wobei als Ergebnis mehr als 90% der Proben mit der einfach beschreibenden Prüfung anhand der Bilder unterschieden werden konnten. Die Auswertung mit der Dreiecksprüfung zeigte, dass sowohl Sorten und Verarbeitungsschritte (Saft) als auch Anbauweisen signifikant getrennt wurden. Darüber hinaus wurde die Methode auch erfolgreich auf Apfelproben angewandt. Weitere Untersuchungen müssen zeigen, ob sich das Potential der Methode, verschiedene Fragen wie die Authentizitätsprüfung von Lebensmitteln verifizieren lassen.

This study was conducted to evaluate the effects of feeding molasses or maize grain with agro-processing by-products on yield and quality of meat from Tanzania shorthorn zebu (TSZ) cattle. Forty five steers aged 2.5 to 3.0 years with 200 +/- 5.4 kg body weight were allocated into five dietary treatments namely hominy feed with molasses (HFMO), rice polishing with molasses (RPMO), hominy feed with maize meal (HFMM), rice polishing with maize meal (RPMM) and maize meal with molasses (MMMO). Ad libitum amount of each dietary treatment and hay were offered to nine steers for 90 days. Cooking loss (CL) and Warner Bratzler shear force (WBSF) values were determined on M. longissimus thoracis et lumborum aged for 3, 6, 9 and 12 days. Steers fed on HFMO diet had higher (P < 0.05) nutrient intake (86.39 MJ/d energy; 867 g/d CP), weight gain (919 g/d) and half carcass weight (75.8 kg) than those fed other diets. Meat of steers from all diets was tender with average WBSF values of 47.9 N cm^(−2). The CL (22.0 +/- 0.61%) and WBSF (53.4 +/- 0.70 N cm^(−2)) were highest in meat aged for 3 days followed by 6, 9 and 12 days. WBSF values for meat aged for 9 and 12 days from steers fed HFMO and RPMM diets were similar and lower than those on other dietary treatments x aging periods. Overall, molasses and hominy feed can be used to replace maize meal in feedlot finishing diets to spare its use in animal feeds.

The work presented in this PhD thesis includes various partial studies aimed at developing a decision support system for membrane bioreactor integrated control. The decision support systems (DSS) have as a main goal to facilitate the operation of complex processes due to the multiple variables that are processed. For this reason, the research used has focused on aspects related to nutrient removal, and on the development of indicators or sensors capable of facilitating, automating and controlling the filtration process in an integrated way with the biological processes that taking place. Work has also been done on the design, development, implementation and validation of tools based on the knowledge made available by the automatic control and the supervision of the MBRs
