The medial septum participates in the modulation of exploratory behavior triggered by novelty. Also, selective lesions of the cholinergic component of the septohippocampal system alter the habituation of rats to an elevated plus-maze without modifying anxiety indices. We investigated the effects of the intraseptal injection of the cholinergic immunotoxin 192 IgG-saporin (SAP) on the behavior of rats in an open-field. Thirty-nine male Wistar rats (weight: 194-230 g) were divided into three groups, non-injected controls and rats injected with either saline (0.5 µl) or SAP (237.5 ng/0.5 µl). Twelve days after surgery, the animals were placed in a square open-field (120 cm) and allowed to freely explore for 5 min. After the test, the rats were killed by decapitation and the septum, hippocampus and frontal cortex were removed and assayed for acetylcholinesterase activity. SAP increased acetylcholinesterase activity in the septum, hippocampus and frontal cortex and decreased the total distance run (9.15 ± 1.51 m) in comparison to controls (13.49 ± 0.91 m). The time spent in the center and at the periphery was not altered by SAP but the distance run was reduced during the first and second minutes (2.43 ± 0.36 and 1.75 ± 0.34 m) compared to controls (4.18 ± 0.26 and 3.14 ± 0.25 m). SAP-treated rats showed decreased but persistent exploration throughout the session. These results suggest that septohippocampal cholinergic mechanisms contribute to at least two critical processes, one related to the motivation to explore new environments and the other to the acquisition and storage of spatial information (i.e., spatial memory).

Pro gradu -tutkielmassa selvitettiin organisaatiokulttuurin vaikutusta tiedon jakamiseen sekä tiedon jakamisen edistämistä asiantuntijaorganisaatiossa. Tavoitteena oli kehittää toimeksiannosta Fingrid Oyj:n tiedon jakamisen käytäntöjä sekä määrittää yhtiölle tavoitekulttuurin piirteet, joihin johto voi sitoutua ja jota kohti organisaatio voi kehittyä. Tutkimus toteutettiin pääosiltaan kvalitatiivisena tutkimuksena ja tutkimusotteena oli toimintatutkimus. Tutkimuksessa perehdyttiin aikaisempiin tiedon jakamista ja organisaatiokulttuurin yhteyttä selvittäneisiin tutkimuksiin. Case-yrityksen henkilöstön näkemykset nykyisestä organisaatiokulttuurista kartoitettiin hyödyntäen Cameron & Quinnin kilpailevien arvojen mallia. Pöytätutkimuksena tutustuttiin organisaation eri dokumentteihin, strategiaan, arvoihin ja ohjeisiin. Lisäksi toteutettiin 10 kpl teemahaastatteluita yhtiön organisaatiokulttuurista ja tiedon jakamisen edistämisen keinoista. Tulosten mukaan organisaatiokulttuurilla ja tiedon jakamisella on yhteys toisiinsa. Tälle löydettiin vahvistusta aikaisempien tutkimusten lisäksi myös case-yrityksen käytänteistä. Kulttuurit, joissa vuorovaikutus on avointa ja valtasuhteet matalia ja joissa kannustetaan kollektiiviseen tekemiseen yksilösuoritusten sijasta, suosivat tiedon jakamista tiedon panttaamisen sijasta. Case-yrityksen organisaatiokulttuurin dominoiviksi piirteiksi muodostuivat hierarkinen ja ryhmäkulttuuri: hierarkisuus näkyy yrityksen toiminnan ohjauksessa runsaina ohjeina ja sääntöinä, silti yrityksen ilmapiiri on epämuodollinen, organisaatiomalli on matala ja päätöksentekojärjestelmässä valtaa on jalkautettu alaspäin. Yrityksen kulttuurin todettiin tukevan tiedon jakamisen käytänteitä. Toimintatutkimuksessa Fingrid Oyj:lle määriteltiin yhdessä ylimmän johdon kanssa tavoitekulttuurin piirteet, linjattiin miten kulttuurin tulee näkyä esimiestyössä sekä laadittiin ehdotuksia tiedon jakamisen kehittämiseksi yhtiössä.

Tämän diplomityön tarkoituksena on tarjota yritykselle ratkaisu talouden seu-rantaan reaaliaikaisesti. Nykyaikaisessa maailmassa yrityksen on kehitettävä toimintaansa jatkuvasti toimintaansa pysyäkseen kilpailussa mukana. Yrityksen toiminnan kehittämisen taustalla ovat myös muuttuneet omistussuhteet ja yri-tyksen uuden pääomistajan kiinnostus seurata taloutta reaaliaikaisemmin. Diplomityössä yritykselle tarjottavat ratkaisut tuotetaan teorian ja yritykseltä saadun tiedon yhdistämisellä. Teoriaosuudessa käydään erilaiset ratkaisumallit perusteluineen läpi. Empiirisessä osassa sovelletaan teoriaosa käytäntöön. Lop-putuloksena yrityksen johdolle tarjotaan valmiita työkaluja talouden seurantaan. Työn tulosten luotettavuus perustuu kirjoittajan laajaan yrityksen tuntemukseen, sekä avoimiin keskusteluihin yrityksen ylimmän johdon kanssa. Yritysjohto an-toi työssä käyttöön kaiken mahdollisen, hyvin salaisenkin materiaalin käyttöön, työn mahdollisimman hyvän lopputuloksen saamiseksi. Pääasiallisen lopputuloksena luotiin sähköinen tuntiseurantalomake, jota nimetyt henkilöt pystyvät seuraamaan reaaliaikaisesti internetin avulla. Yritykselle mää-ritettiin olennaisimmat taloudenmittarit, joita yrityksen tulee seurata ja päivittää määritellyin aikavälein, lisäksi määrättiin vastuu alueet, kenelle asioiden päivitys kuuluu. Lisäksi luotiin avainlukujen seurantatyökalupohja, jonne on kerätty kaikki yrityksen toiminnan kannalta tärkeimmät tiedot. Työkalut ovat valmiina käyttöönotettavaksi. Lopullinen luodun ohjelman hyödyntäminen ja käyttöönot-to tapahtuu myöhemmin.

Brazilian scientific output exhibited a 4-fold increase in the last two decades because of the stability of the investment in research and development activities and of changes in the policies of the main funding agencies. Most of this production is concentrated in public universities and research institutes located in the richest part of the country. Among all areas of knowledge, the most productive are Health and Biological Sciences. During the 1998-2002 period these areas presented heterogeneous growth ranging from 4.5% (Pharmacology) to 191% (Psychiatry), with a median growth rate of 47.2%. In order to identify and rank the 20 most prolific institutions in these areas, searches were made in three databases (DataCAPES, ISI and MEDLINE) which permitted the identification of 109,507 original articles produced by the 592 Graduate Programs in Health and Biological Sciences offered by 118 public universities and research institutes. The 20 most productive centers, ranked according to the total number of ISI-indexed articles published during the 1998-2003 period, produced 78.7% of the papers in these areas and are strongly concentrated in the Southern part of the country, mainly in São Paulo State.

The goal of the present study was to investigate the role of thigmotaxis (the tendency to remain close to vertical surfaces) in rat exploratory behavior in an open-field. Thigmotaxis was investigated in a parametric way, using 24 experimentally adult naive male Wistar rats (210-230 g). Exploratory behavior was studied in an open-field (N = 12) in 5-min sessions and behavior was analyzed in terms of where it occurred: in areas surrounded by two, one, or no walls. Another group of rats (N = 12) was studied in an open-field with blocks placed near two of the corners so as to make these corner areas surrounded by three walls. The floor of the open-fields was divided into 20-cm squares in order to locate the exact place of occurrence of each behavior. The following behaviors were recorded: entries into the squares, rearings, and groomings. In both types of open-field the rats chose to remain longer in the squares surrounded by the largest possible number of walls. In one of the open-fields, the mean time (seconds) spent in squares surrounded by two walls was longer than the time spent in squares surrounded by one or no walls (37.2, 7.7, and 1.8 s, respectively). In the other open-field, the mean time spent in squares surrounded by three walls was longer than the time spent in squares surrounded by two, one or no walls (41.7, 20.4, 7.0, and 2.6 s, respectively). Other measures presented a similar profile. These results indicate that rats are sensitive to the number of walls in an environment and prefer to remain close to them.

The endocannabinoid system is involved in the control of many physiological functions, including the control of emotional states. In rodents, previous exposure to an open field increases the anxiety-like behavior in the elevated plus-maze. Anxiolytic-like effects of pharmacological compounds that increase endocannabinoid levels have been well documented. However, these effects are more evident in animals with high anxiety levels. Several studies have described characteristic inverted U-shaped dose-response effects of drugs that modulate the endocannabinoid levels. However, there are no studies showing the effects of different doses of exogenous anandamide, an endocannabinoid, in animal models of anxiety. Thus, in the present study, we determined the dose-response effects of exogenous anandamide at doses of 0.01, 0.1, and 1.0 mg/kg in C57BL/6 mice (N = 10/group) sequentially submitted to the open field and elevated plus-maze. Anandamide was diluted in 0.9% saline, ethyl alcohol, Emulphor® (18:1:1) and administered ip (0.1 mL/10 g body weight); control animals received the same volume of anandamide vehicle. Anandamide at the dose of 0.1 mg/kg (but not of 0.01 or 1 mg/kg) increased (P < 0.05) the time spent and the distance covered in the central zone of the open field, as well as the exploration of the open arms of the elevated plus-maze. Thus, exogenous anandamide, like pharmacological compounds that increase endocannabinoid levels, promoted a characteristic inverted U-shaped dose-response effect in animal models of anxiety. Furthermore, anandamide (0.1 mg/kg) induced an anxiolytic-like effect in the elevated plus-maze (P < 0.05) after exposing the animals to the open field test.

The generation of bradykinin (BK; Arg-Pro-Pro-Gly-Phe-Ser-Pro-Phe-Arg) in blood and kallidin (Lys-BK) in tissues by the action of the kallikrein-kinin system has received little attention in non-mammalian vertebrates. In mammals, kallidin can be generated by the coronary endothelium and myocytes in response to ischemia, mediating cardioprotective events. The plasma of birds lacks two key components of the kallikrein-kinin system: the low molecular weight kininogen and a prekallikrein activator analogous to mammalian factor XII, but treatment with bovine plasma kallikrein generates ornitho-kinin [Thr6,Leu8]-BK. The possible cardioprotective effect of ornitho-kinin infusion was investigated in an anesthetized, open-chest chicken model of acute coronary occlusion. A branch of the left main coronary artery was reversibly ligated to produce ischemia followed by reperfusion, after which the degree of myocardial necrosis (infarct size as a percent of area at risk) was assessed by tetrazolium staining. The iv injection of a low dose of ornitho-kinin (4 µg/kg) reduced mean arterial pressure from 88 ± 12 to 42 ± 7 mmHg and increased heart rate from 335 ± 38 to 402 ± 45 bpm (N = 5). The size of the infarct was reduced by pretreatment with ornitho-kinin (500 µg/kg infused over a period of 5 min) from 35 ± 3 to 10 ± 2% of the area at risk. These results suggest that the physiological role of the kallikrein-kinin system is preserved in this animal model in spite of the absence of two key components, i.e., low molecular weight kininogen and factor XII.

Occupational therapy (OT) is a profession concerned with promoting health and well-being through occupation, by enabling handicapped people to participate in the activities of everyday life. OT is part of the clinical rehabilitation of progressive genetic neurodegenerative diseases such as spinocerebellar ataxias; however, its effects have never been determined in these diseases. Our aim was to investigate the effect of OT on both physical disabilities and depressive symptoms of spinocerebellar ataxia type 3 (SCA3) patients. Genomically diagnosed SCA3 patients older than 18 years were invited to participate in the study. Disability, as evaluated by functional independence measurement and Barthel incapacitation score, Hamilton Rating Scale for Depression, and World Health Organization Quality of Life questionnaire (WHOQOL-BREF), was determined at baseline and after 3 and 6 months of treatment. Twenty-six patients agreed to participate in the study. All were treated because OT prevents blinding of a control group. Fifteen sessions of rehabilitative OT were applied over a period of 6 months. Difficult access to food, clothing, personal hygiene, and leisure were some of the main disabilities focused by these patients. After this treatment, disability scores and quality of life were stable, and the Hamilton scores for depression improved. Since no medication was started up to 6 months before or during OT, this improvement was related to our intervention. No association was found between these endpoints and a CAG tract of the MJD1 gene (CAGn), age, age of onset, or neurological scores at baseline (Spearman test). Although the possibly temporary stabilization of the downhill disabilities as an effect of OT remains to be established, its clear effect on depressive symptoms confirms the recommendation of OT to any patient with SCA3 or spinocerebellar ataxia.
