Clostridium difficile is a gram positive, spore former, anaerobic bacterium that is able to cause infection and disease, with symptoms ranging from mild diarrhea to pseudomembranous colitis, toxic megacolon, sepsis and death. In the last decade new strains have emerged that caused outbreaks of increased disease severity and higher recurrence, morbidity and mortality rates, and C. difficile is now considered both a main nosocomial pathogen associated with antibiotic therapy as well as a major concern in the community.(...)

A vancomicina é um membro da família dos antibióticos glicopeptídicos considerado de último recurso no tratamento de infecções causadas por bactérias Gram-positivas. A fim de encontrar novos agentes para combater a resistência bacteriana é importante compreender os detalhes do mecanismo de acção de antibióticos glicopeptídicos. Estes antibióticos evitam a formação de peptidoglicano (PGN), o principal componente da parede celular bacteriana, o qual é constituído por uma cadeia alternada de N-acetil-glucosamina (GlucNAc) e ácido N-acetil-murâmico (MurNAc) ligados entre si poruma ligação glicosídica β(1→4), e estas cadeias ligam-se entre si por pequenas cadeias peptídicas. Está bem estabelecido que a substituição do último aminoácido da cadeia peptídica ligada à unidade MurNAc altera a interacção das bactérias com os antibióticos glicopéptidicos. Até agora, os estudos de interacção foram limitados ao uso de dipéptidos e tripéptidos N-protegidos. De modo a clarificar a forma como as diferentes composições da unidade peptídica e da unidade de carbohidratodos muropéptidos bacterianos afectam o reconhecimento pela vancomicina, desenvolvemos neste estudo a síntese de uma pequena biblioteca de muropeptidos e péptidos para futuros testes de interação molecular. A unidade MurNAc foi sintetizada por duas vias diferentes, envolvendo diferentes grupos protectores do grupo amina, o acetilo e o aliloxicarbonilo. O derivado de MurNAc 8 foi preparado em 7 passos, com 3% de rendimento pela via envolvendo a GlucNAc. Para a síntese das cadeias peptídicas utilizou-se a técnica de síntese de péptidos em fase sólida (SPFS). Os péptidos e muropéptidos foram sintetizados utilizando como terminal de D-Ala, Gly e D-Ser por SPFS com sucesso. Os estudos preliminares de interacção entre a vancomicina e a unidade MurNAc sugerem uma possível alteração conformacional da vancomicina na presença de MurNAc, pelo que de futuro as interações entre estas duas estruturas e os muropéptidos sintetizados deverão ser investigadas.

Neste trabalho estudou-se a composição e actividade antioxidante de chás verdes e pretos (8 provenientes dos Açores, e 11 de outras origens), 4 chás vermelhos e 6 tisanas de ervas. A composição mineral das folhas de chá e das correspondentes infusões de 5 e 15 minutos foi avaliada no que se refere aos macroelementos (K, Ca, Mg e Na) com valor nutricional e ao alumínio (Al), tendo-se verificado que as suas concentrações dependem do tipo de chá e do tempo de infusão. A actividade antioxidante dos chás e tisanas foi avaliada pela reacção de Folin Ciocalteu, a actividade antiradicalar (DPPH) e actividade redutora férrica (FRAP). Estas actividades aumentaram ou diminuiram com o tempo de infusão e dependendo do tipo de chá e do teste específico o que indica diferentes cinéticas de extracção e degradação de compostos específicos. Os compostos fenólicos dos chás foram quantificados por HPLC-DAD e agrupados nas famílias de catequinas, metilxantinas, ácidos hidroxibenzóicos e hidroxicinâmicos, teoflavinas, flavonas, flavonóis, e seus derivados glicosilados e/ou acilados. A cinética de extracção destes compostos foi avaliada em dois chás dos Açores para tempos de infusão entre 5 min e 60 min. Testaram-se diferenças significativas na composição dos chás e correlações entre composição e actividade antioxidante. A valorização de resíduos dos chás Gorreana como matérias-primas para a produção de suplementos nutracêuticos foi estudada efectuando a extracção de dois chás dos Açores e de dois resíduos de chá utilizando água, etanol acetona e suas misturas. Os resultados mostraram que a polaridade do solvente afectou significativamente o teor de polifenóis, actividade antioxidante e actividade antimicrobiana dos extractos. Os extractos de chá verde inibiram o crescimento de bactérias gram-positivas (Staphylococcus aureus e Staphylococcus aureus resistentes à meticilina), e observou-se uma correlacção forte entre teor de polifenóis e as actividades antioxidante e antimicrobiana de extractos de chá.

Salmonella enterica serovars are Gram-negative facultative intracellular bacterial pathogens that infect a wide variety of animals. Salmonella infections are common in humans, causing usually typhoid fever and gastrointestinal diseases. Salmonella enterica serovar Typhimurium (S. Typhimurium), which is a leading cause of human gastroenteritis, has been extensively used to study the molecular pathogenesis of Salmonella, because of the availability of sophisticated genetic tools, and of suitable animal and tissue culture models mimicking different aspects of Salmonella infections.(...)

RESUMO: Clostridium difficile é presentemente a principal causa de doença gastrointestinal associada à utilização de antibióticos em adultos. C. difficile é uma bactéria Gram-positiva, obrigatoriamente anaeróbica, capaz de formar endósporos. Tem-se verificado um aumento dos casos de doença associada a C. difficile com sintomas mais severos, elevadas taxas de morbilidade, mortalidade e recorrência, em parte, devido à emergência de estirpes mais virulentas, mas também devido à má gestão do uso de antibióticos. C. difficile produz duas toxinas, TcdA e TcdB, que são os principais fatores de virulência e responsáveis pelos sintomas da doença. Estas são codificadas a partir do Locus de Patogenicidade (PaLoc) que codifica ainda para um regulador positivo, TcdR, uma holina, TcdE, e um regulador negativo, TcdC. Os esporos resistentes ao oxigénio são essenciais para a transmissão do organismo e recorrência da doença. A expressão dos genes do PaLoc ocorre em células vegetativas, no final da fase de crescimento exponencial, e em células em esporulação. Neste trabalho construímos dois mutantes de eliminação em fase dos genes tcdR e tcdE. Mostrámos que a auto-regulação do gene tcdR não é significativa. No entanto, tcdR é sempre necessário para a expressão dos genes presentes no PaLoc. Trabalho anterior mostrou que, com a exceção de tcdC, os demais genes do PaLoc são expressos no pré-esporo. Mostrámos aqui que TcdA é detectada à superfície do esporo maduro e que a eliminação do tcdE não influencia a acumulação de TcdA no meio de cultura ou em associação às células ou ao esporo. Estas observações têm consequências para o nosso entendimento do processo infecioso: sugeremque o esporo possa ser também um veículo para a entrega da toxina nos estágios iniciais da infecção, que TcdA possa ser libertada durante a germinação do esporo, e que o esporo possa utilizar o mesmo receptor reconhecido por TcdA para a ligação à mucosa do cólon.---------------------------ABSTRACT: Clostridium difficile is currently the major cause of antibiotic-associated gastrointestinal diseases in adults. This is a Gram-positive bacterium, endospore-forming and an obligate anaerobe that colonizes the gastrointestinal tract. Recent years have seen a rise in C. difficile associated disease (CDAD) cases, associated with more severe disease symptoms, higher rates of morbidity, mortality and recurrence, which were mostly caused due to the emergence of “hypervirulent” strains but also due to changing patterns of antibiotics use. C. difficile produces two potent toxins, TcdA and TcdB, which are the main virulence factors and the responsible for the disease symptoms. These are codified from a Pathogenicity Locus (PaLoc), composed also by the positive regulator, TcdR, the holin-like protein, TcdE, and a negative regulator, TcdC. Besides the toxins, the oxygen-resistant spores are also essential for transmission of the organism through diarrhea; moreover, spores can accumulate in the environment or in the host, which will cause disease recurrence. The expression of the PaLoc genes occurs in vegetative cells, at the end of the exponential growth phase, and in sporulating cells. In this work, we constructed two in-frame deletion mutants of tcdR and tcdE. We showed that the positive auto regulation of tcdR is not significant. However, tcdR is always necessary for the expression of the PaLoc genes. A previous work showed that, except tcdC, all the PaLoc genes are expressed in the forespore. Here, we detected TcdA at the spore surface. Furthermore, we showed that the in-frame deletion of tcdE does not affect the accumulation of TcdA in the culture medium or in association with cells or spores. This data was important for us to conclude about the infeccious process: it suggests that the spore may be the vehicle for the delivery of TcdA in early stages of infection, that TcdA may be released during spores germination and that this spore may use the same receptor recognized by TcdA to bind to the colonic mucosa.

Phosphorus is a macronutrient essential to life which comes from phosphate rock, a non-renewable resource. Sewage sludge from wastewater treatment plants (WWTP) is a secondary resource rich in phosphorus that can be valorized. However, organic compounds are detected in sewage sludge, due to its non-polar and hydrophobic character, being considered an environmental risk. The present dissertation aims to study the efficiency of the electrodialytic process (ED) when applied to sewage sludge aiming phosphorus recovery and organic contaminants removal. Four organic compounds were analyzed: 17α-ethynylestradiol (EE2), bisphenol A (BPA), caffeine (Caf) and oxybenzone (MBPh). The experiments took place in an ED cell with two compartments and an anion exchange membrane, with the sludge in the cathode compartment. The experiments were carried out for three days with spiked sewage sludge (six assays). One control experiment was done without current, three experiments were carried out applying a constant current of 50, 75, and 100 mA and two experiments were carried out applying sequential currents: 50 mA, 75 mA and 100 mA and the opposite (100-75-50 mA). A qualitative and quantitative analysis of microorganisms existing in the samples was also done. At the end, the pH increased in the sewage sludge favoring phosphorus recovery. In terms of phosphorus, the highest recovery was achieved in the experiment run with 100 mA, where 70.3±2.0% of total phosphorus was recovered in the electrolyte. Generally, compounds degradation was favored by the current. Caf and MBPh achieved degradation percentages of 96.2±0.2% and 84.8±1.3%, respectively, in 100 mA assay. EE2 (83.1±1.7%) and BPA (91.8±4.6%) degradations were favored by 50 mA current. A total of 35 taxa from four different groups were identified, totalizing between 81,600-273,000 individuals per gram of initial sludges. After ED, microbial community population decreased between 47-98%. Arcella gibbosa represented 61% of the total observed organisms and revealed to be more tolerant to medium changes.

An assessment of sewage workers' exposure to airborne cultivable bacteria, fungi and inhaled endotoxins was performed at 11 sewage treatment plants. We sampled the enclosed and unenclosed treatment areas in each plant and evaluated the influence of seasons (summer and winter) on bioaerosol levels. We also measured personal exposure to endotoxins of workers during special operation where a higher risk of bioaerosol inhalation was assumed. Results show that only fungi are present in significantly higher concentrations in summer than in winter (2331 +/- 858 versus 329 +/- 95 CFU m(-3)). We also found that there are significantly more bacteria in the enclosed area, near the particle grids for incoming water, than in the unenclosed area near the aeration basins (9455 +/- 2661 versus 2435 +/- 985 CFU m(-3) in summer and 11 081 +/- 2299 versus 2002 +/- 839 CFU m(-3) in winter). All bioaerosols were frequently above the recommended values of occupational exposure. Workers carrying out special tasks such as cleaning tanks were exposed to very high levels of endotoxins (up to 500 EU m(-3)) compared to routine work. The species composition and concentration of airborne Gram-negative bacteria were also studied. A broad spectrum of different species within the Pseudomonadaceae and the Enterobacteriaceae families were predominant in nearly all plants investigated. [Authors]

Cellular responses to LPS, the major lipid component of the outer membrane of Gram-negative bacteria, are enhanced markedly by the LPS-binding protein (LBP), a plasma protein that transfers LPS to the cell surface CD14 present on cells of the myeloid lineage. LBP has been shown previously to potentiate the host response to LPS. However, experiments performed in mice with a disruption of the LBP gene have yielded discordant results. Whereas one study showed that LBP knockout mice were resistant to endotoxemia, another study did not confirm an important role for LBP in the response of mice challenged in vivo with low doses of LPS. Consequently, we generated rat mAbs to murine LBP to investigate further the contribution of LBP in experimental endotoxemia. Three classes of mAbs were obtained. Class 1 mAbs blocked the binding of LPS to LBP; class 2 mAbs blocked the binding of LPS/LBP complexes to CD14; class 3 mAbs bound LBP but did not suppress LBP activity. In vivo, class 1 and class 2 mAbs suppressed LPS-induced TNF production and protected mice from lethal endotoxemia. These results show that the neutralization of LBP accomplished by blocking either the binding of LPS to LBP or the binding of LPS/LBP complexes to CD14 protects the host from LPS-induced toxicity, confirming that LBP is a critical component of innate immunity.

Un nuevo método, el conteo de ovocitos hidratados, se aplica por primera vez para determinar la fecundidad de la caballa peruana (Scomber Japonicus peruanus) . En febrero de 1985, la fecundidad relativa media fue 278 huevos por gramo de hembra y la fecundidad parcial media de la muestra fue 78,174 huevos por hembra. Se muestra que la caballa del Perú es un desovador múltiple.

Purpose: Pretargeted radioimmunotherapy (PRIT) using streptavidin (SAv)-biotin technology can deliver higher therapeutic doses of radioactivity to tumors than conventional RIT. However, "endogenous" biotin can interfere with the effectiveness of this approach by blocking binding of radiolabeled biotin to SAv. We engineered a series of SAv FPs that downmodulate the affinity of SAv for biotin, while retaining high avidity for divalent DOTA-bis-biotin to circumvent this problem.Experimental Design: The single-chain variable region gene of the murine 1F5 anti-CD20 antibody was fused to the wild-type (WT) SAv gene and to mutant SAv genes, Y43A-SAv and S45A-SAv. FPs were expressed, purified, and compared in studies using athymic mice bearing Ramos lymphoma xenografts.Results: Biodistribution studies showed delivery of more radioactivity to tumors of mice pretargeted with mutant SAv FPs followed by (111)In-DOTA-bis-biotin [6.2 +/- 1.7% of the injected dose per gram (%ID/gm) of tumor 24 hours after Y43A-SAv FP and 5.6 +/- 2.2%ID/g with S45A-SAv FP] than in mice on normal diets pretargeted with WT-SAv FP (2.5 +/- 1.6%ID/g; P = 0.01). These superior biodistributions translated into superior antitumor efficacy in mice treated with mutant FPs and (90)Y-DOTA-bis-biotin [tumor volumes after 11 days: 237 +/- 66 mm(3) with Y43A-SAv, 543 +/- 320 mm(3) with S45A-SAv, 1129 +/- 322 mm(3) with WT-SAv, and 1435 +/- 212 mm(3) with control FP (P < 0.0001)].Conclusions: Genetically engineered mutant-SAv FPs and bis-biotin reagents provide an attractive alternative to current SAv-biotin PRIT methods in settings where endogenous biotin levels are high. La prospección para evaluar la biología y pesquería de Coryphaena hippurus perico o dorado, se efectuó del 10 de febrero al 1 de marzo 2010, a bordo del BIC Imarpe V. El área de extracción fue de 4°59’32,8’’ a 10°15’20,1’’S y de 79°55’16,2’’ a 84°35’04,2’’W; la captura total fue 1.500,5 kg. La CPUE varió de 0,9 a 18,2 kg/Nº Anzuelos/1000*hora efectiva de pesca. Se encontró alta selección interespecífi ca del espinel superfi cial, al capturar 87,8% de perico y 8 especies de peces, tortugas y cefalópodos. El rango de tallas para hembras fue 79 - 141 cm de LT, en machos fue 100 - 157 cm LT. El factor de condición de Fulton en machos varió de 0,30 a 0,46 y en hembras de 0,32 a 0,40. Se determinó para hembras: L∞=148,92 cm, tasa de crecimiento anual (K) 1,081, se estimó t0 = -0,076 años, el Índice de performance Ǿ= 4,38; para machos, estos parámetros fueron L∞=169,75 cm, tasa de crecimiento anual (K) de 0,893, t0 = -0,115, índice de performance Ǿ= 4,40. La curva de crecimiento en talla para hembras, indica que a un año de edad la LT es 102,4 cm y en machos 107 cm. El máximo crecimiento en peso para hembras y machos fue a 1,5 años de vida. El Índice gonadosomático (IGS) fl uctuó de 3,2 a 4,6 mostrando comportamiento heterogéneo. La proporción sexual fue de 2:1 favorable a las hembras. La fecundidad parcial fue de 324.416 ovocitos por tanda de desove. La fecundidad relativa fue 56 ovocitos/ gramo de peso corporal. El tipo de alimento fue peces, cefalópodos y crustáceos con dominancia de Exocoetus volitans (%IRI = 31,6), Argonauta spp. (%IRI = 26,4) mostrando variaciones según zonas y tallas. El consumo de alimento fue 472,7 g/ind./día correspondiente a 7,4% de su peso corporal, presentando fl uctuaciones con respecto a la talla; alimentándose en horas de la mañana, con un máximo entre las 8:00 y 9:59 horas. Se encontraron 5.762 parásitos, identifi cándose 8 especies en cuatro taxas. La prevalencia parasitaria indica que Tentacularia coryphaenae y Bathycotyle coryphaenae alcanzan el 100%, seguido por Hysterothylacium sp. 62% y Caligus coryphaenae 46%. El mayor número de parásitos se encontró en el estómago. Los peces entre 96–100 cm de LT son los más parasitados. La mayor incidencia parasitaria se encontró en la zona norte, en el tramo Paita-Salaverry. Los registros de temperatura en la columna de agua fueron de 12,7 a 27,3 °C y las ATSM de 0,2 a 4,1 °C. Por el frente oceánico se observaron comportamientos térmicos cálidos asociados a las masas de Aguas Ecuatoriales Superfi ciales y Aguas Subtropicales Superfi ciales y por la zona costera se localizaron Aguas Costeras Frías tanto en Chimbote como en Chicama con temperaturas próximas a 22,0 °C. La concentración de oxígeno disuelto superfi cial se presentó favorable para el perico.

Body mass index (BMI) is related with cardiorespiratory fitness (CRF), but less is known regarding the combined relationships between BMI and body fat (BF) on CRF. Cross-sectional study included 2361 girls and 2328 boys aged 10–18 years living in the area of Lisbon, Portugal. BMI was calculated by measuring height and weight, and obesity was assessed by international criteria. BF was assessed by bioimpedance. CRF was assessed by the 20-m shuttle run and the participants were classified as normal-to-high or low-CRF level according to Fitness gram criterion-referenced standards. The prevalence of low CRF was 47 and 39% in girls and boys, respectively. The corresponding values for the prevalence of obesity were 4.8 and 5.6% (not significant) and of excess BF of 12.1 and 25.1% (P <0.001), respectively. In both sexes, BMI and BF were inversely related with CRF: r = – 0.53 and – 0.45 for BMI and % BF, respectively, in boys and the corresponding values in girls were – 0.50 and – 0.33 (all P <0.01). When compared with a participant with normal BMI and BF, the odds ratios (95% confidence interval) for low CRF were 1.94 (1.46–2.58) for a participant with normal BMI and high BF, and 6.19 (5.02–7.63) for a participant with high BMI and high BF. The prevalence of low-CRF levels is high in Portuguese youths. BF negatively influences CRF levels among children/adolescents with normal BMI.

Measurements and simulations were performed to assess workers' exposure to solvent vapors and aerosols during the waterproofing of a tiled surface. This investigation followed two recent incidents in the same company where workers experienced acute respiratory illness after spraying a stain-repellent resin containing fluorinated polymers on stone-tiled walls and floors. Because the waterproofing activity had been done for years at the tile company without encountering any exposure problems prior to these cases, it was strongly suspected that the incidents were linked to a recent change in the composition of the coating mixture. Experimental measurements and simulations indicated that the emission rate of particles smaller than 10 microm may be estimated at 0.66 mg/sec (SD 0.10) for the old resin and at 0.37 mg/sec (SD 0.04) for the new one. The measurement of the solvent emission rate from surfaces coated with the two resins indicated that shortly after spraying, the emission was in the range of 18 to 20 mg/sec x m2 and was similar for both products. Solvent and overspray emission rates were introduced in a two-zone compartment model. The results obtained in the near-field indicate significant exposure to overspray mist (7 and 34 mg/m3 for new resin) and solvent vapors (80 to 350 ppm for the new resin). It was also shown that the introduction of the new resin tended to significantly decrease the levels of solvents and particulates in the workers' breathing zone. These results strongly suggest that cases of acute respiratory illness are related to the specific toxicity of the fluorinated polymer itself. The fact that the same polymer is used in various commercial products raises concern regarding other possible occupational and domestic exposures.

Contact stains recovered at break-in crime scenes are frequently characterized by mixtures of DNA from several persons. Broad knowledge on the relative contribution of DNA left behind by different users overtime is of paramount importance. Such information might help crime investigators to robustly evaluate the possibility of detecting a specific (or known) individual's DNA profile based on the type and history of an object. To address this issue, a contact stain simulation-based protocol was designed. Fourteen volunteers either acting as first or second object's users were recruited. The first user was required to regularly handle/wear 9 different items during an 8-10-day period, whilst the second user for 5, 30 and 120 min, in three independent simulation sessions producing a total of 231 stains. Subsequently, the relative DNA profile contribution of each individual pair was investigated. Preliminary results showed a progressive increase of the percentage contribution of the second user compared to the first. Interestingly, the second user generally became the major DNA contributor when most objects were handled/worn for 120 min, Furthermore, the observation of unexpected additional alleles will then prompt the investigation of indirect DNA transfer events.
