We describe here the construction of a delivery system for stable and directed insertion of gene constructs in a permissive chromosomal site of the bacterial wilt pathogen Ralstonia solanacearum. The system consists of a collection of suicide vectors the Ralstonia chromosome (pRC) series that carry an integration element flanked by transcription terminators and two sequences of homology to the chromosome of strain GMI1000, where the integration element is inserted through a double recombination event. Unique restriction enzyme sites and a GATEWAY cassette enable cloning of any promoter::gene combination in the integration element. Variants endowed with different selectable antibiotic resistance genes and promoter::gene combinations are described. We show that the system can be readily used in GMI1000 and adapted to other R. solanacearum strains using an accessory plasmid. We prove that the pRC system can be employed to complement a deletion mutation with a single copy of the native gene, and to measure transcription of selected promoters in monocopy both in vitro and in planta. Finally, the system has been used to purify and study secretion type III effectors. These novel genetic tools will be particularly useful for the construction of recombinant bacteria that maintain inserted genes or reporter fusions in competitive situations (i.e., during plant infection).

Objective: We used demographic and clinical data to design practical classification models for prediction of neurocognitive impairment (NCI) in people with HIV infection. Methods: The study population comprised 331 HIV-infected patients with available demographic, clinical, and neurocognitive data collected using a comprehensive battery of neuropsychological tests. Classification and regression trees (CART) were developed to btain detailed and reliable models to predict NCI. Following a practical clinical approach, NCI was considered the main variable for study outcomes, and analyses were performed separately in treatment-naïve and treatment-experienced patients. Results: The study sample comprised 52 treatment-naïve and 279 experienced patients. In the first group, the variables identified as better predictors of NCI were CD4 cell count and age (correct classification [CC]: 79.6%, 3 final nodes). In treatment-experienced patients, the variables most closely related to NCI were years of education, nadir CD4 cell count, central nervous system penetration-effectiveness score, age, employment status, and confounding comorbidities (CC: 82.1%, 7 final nodes). In patients with an undetectable viral load and no comorbidities, we obtained a fairly accurate model in which the main variables were nadir CD4 cell count, current CD4 cell count, time on current treatment, and past highest viral load (CC: 88%, 6 final nodes). Conclusion: Practical classification models to predict NCI in HIV infection can be obtained using demographic and clinical variables. An approach based on CART analyses may facilitate screening for HIV-associated neurocognitive disorders and complement clinical information about risk and protective factors for NCI in HIV-infected patients.

The complement system is a major effector of innate immunity that has been involved in stroke brain damage. Complement activation occurs through the classical, alternative and lectin pathways. The latter is initiated by mannose-binding lectin (MBL) and MBL-associated serine proteases (MASPs). Here we investigated whether the lectin pathway contributes to stroke outcome in mice and humans.

Wastewater-based epidemiology consists in acquiring relevant information about the lifestyle and health status of the population through the analysis of wastewater samples collected at the influent of a wastewater treatment plant. Whilst being a very young discipline, it has experienced an astonishing development since its firs application in 2005. The possibility to gather community-wide information about drug use has been among the major field of application. The wide resonance of the first results sparked the interest of scientists from various disciplines. Since then, research has broadened in innumerable directions. Although being praised as a revolutionary approach, there was a need to critically assess its added value, with regard to the existing indicators used to monitor illicit drug use. The main, and explicit, objective of this research was to evaluate the added value of wastewater-based epidemiology with regards to two particular, although interconnected, dimensions of illicit drug use. The first is related to trying to understand the added value of the discipline from an epidemiological, or societal, perspective. In other terms, to evaluate if and how it completes our current vision about the extent of illicit drug use at the population level, and if it can guide the planning of future prevention measures and drug policies. The second dimension is the criminal one, with a particular focus on the networks which develop around the large demand in illicit drugs. The goal here was to assess if wastewater-based epidemiology, combined to indicators stemming from the epidemiological dimension, could provide additional clues about the structure of drug distribution networks and the size of their market. This research had also an implicit objective, which focused on initiating the path of wastewater- based epidemiology at the Ecole des Sciences Criminelles of the University of Lausanne. This consisted in gathering the necessary knowledge about the collection, preparation, and analysis of wastewater samples and, most importantly, to understand how to interpret the acquired data and produce useful information. In the first phase of this research, it was possible to determine that ammonium loads, measured directly in the wastewater stream, could be used to monitor the dynamics of the population served by the wastewater treatment plant. Furthermore, it was shown that on the long term, the population did not have a substantial impact on consumption patterns measured through wastewater analysis. Focussing on methadone, for which precise prescription data was available, it was possible to show that reliable consumption estimates could be obtained via wastewater analysis. This allowed to validate the selected sampling strategy, which was then used to monitor the consumption of heroin, through the measurement of morphine. The latter, in combination to prescription and sales data, provided estimates of heroin consumption in line with other indicators. These results, combined to epidemiological data, highlighted the good correspondence between measurements and expectations and, furthermore, suggested that the dark figure of heroin users evading harm-reduction programs, which would thus not be measured by conventional indicators, is likely limited. In the third part, which consisted in a collaborative study aiming at extensively investigating geographical differences in drug use, wastewater analysis was shown to be a useful complement to existing indicators. In particular for stigmatised drugs, such as cocaine and heroin, it allowed to decipher the complex picture derived from surveys and crime statistics. Globally, it provided relevant information to better understand the drug market, both from an epidemiological and repressive perspective. The fourth part focused on cannabis and on the potential of combining wastewater and survey data to overcome some of their respective limitations. Using a hierarchical inference model, it was possible to refine current estimates of cannabis prevalence in the metropolitan area of Lausanne. Wastewater results suggested that the actual prevalence is substantially higher compared to existing figures, thus supporting the common belief that surveys tend to underestimate cannabis use. Whilst being affected by several biases, the information collected through surveys allowed to overcome some of the limitations linked to the analysis of cannabis markers in wastewater (i.e., stability and limited excretion data). These findings highlighted the importance and utility of combining wastewater-based epidemiology to existing indicators about drug use. Similarly, the fifth part of the research was centred on assessing the potential uses of wastewater-based epidemiology from a law enforcement perspective. Through three concrete examples, it was shown that results from wastewater analysis can be used to produce highly relevant intelligence, allowing drug enforcement to assess the structure and operations of drug distribution networks and, ultimately, guide their decisions at the tactical and/or operational level. Finally, the potential to implement wastewater-based epidemiology to monitor the use of harmful, prohibited and counterfeit pharmaceuticals was illustrated through the analysis of sibutramine, and its urinary metabolite, in wastewater samples. The results of this research have highlighted that wastewater-based epidemiology is a useful and powerful approach with numerous scopes. Faced with the complexity of measuring a hidden phenomenon like illicit drug use, it is a major addition to the panoply of existing indicators. -- L'épidémiologie basée sur l'analyse des eaux usées (ou, selon sa définition anglaise, « wastewater-based epidemiology ») consiste en l'acquisition d'informations portant sur le mode de vie et l'état de santé d'une population via l'analyse d'échantillons d'eaux usées récoltés à l'entrée des stations d'épuration. Bien qu'il s'agisse d'une discipline récente, elle a vécu des développements importants depuis sa première mise en oeuvre en 2005, notamment dans le domaine de l'analyse des résidus de stupéfiants. Suite aux retombées médiatiques des premiers résultats de ces analyses de métabolites dans les eaux usées, de nombreux scientifiques provenant de différentes disciplines ont rejoint les rangs de cette nouvelle discipline en développant plusieurs axes de recherche distincts. Bien que reconnu pour son coté objectif et révolutionnaire, il était nécessaire d'évaluer sa valeur ajoutée en regard des indicateurs couramment utilisés pour mesurer la consommation de stupéfiants. En se focalisant sur deux dimensions spécifiques de la consommation de stupéfiants, l'objectif principal de cette recherche était focalisé sur l'évaluation de la valeur ajoutée de l'épidémiologie basée sur l'analyse des eaux usées. La première dimension abordée était celle épidémiologique ou sociétale. En d'autres termes, il s'agissait de comprendre si et comment l'analyse des eaux usées permettait de compléter la vision actuelle sur la problématique, ainsi que déterminer son utilité dans la planification des mesures préventives et des politiques en matière de stupéfiants actuelles et futures. La seconde dimension abordée était celle criminelle, en particulier, l'étude des réseaux qui se développent autour du trafic de produits stupéfiants. L'objectif était de déterminer si cette nouvelle approche combinée aux indicateurs conventionnels, fournissait de nouveaux indices quant à la structure et l'organisation des réseaux de distribution ainsi que sur les dimensions du marché. Cette recherche avait aussi un objectif implicite, développer et d'évaluer la mise en place de l'épidémiologie basée sur l'analyse des eaux usées. En particulier, il s'agissait d'acquérir les connaissances nécessaires quant à la manière de collecter, traiter et analyser des échantillons d'eaux usées, mais surtout, de comprendre comment interpréter les données afin d'en extraire les informations les plus pertinentes. Dans la première phase de cette recherche, il y pu être mis en évidence que les charges en ammonium, mesurées directement dans les eaux usées permettait de suivre la dynamique des mouvements de la population contributrice aux eaux usées de la station d'épuration de la zone étudiée. De plus, il a pu être démontré que, sur le long terme, les mouvements de la population n'avaient pas d'influence substantielle sur le pattern de consommation mesuré dans les eaux usées. En se focalisant sur la méthadone, une substance pour laquelle des données précises sur le nombre de prescriptions étaient disponibles, il a pu être démontré que des estimations exactes sur la consommation pouvaient être tirées de l'analyse des eaux usées. Ceci a permis de valider la stratégie d'échantillonnage adoptée, qui, par le bais de la morphine, a ensuite été utilisée pour suivre la consommation d'héroïne. Combinée aux données de vente et de prescription, l'analyse de la morphine a permis d'obtenir des estimations sur la consommation d'héroïne en accord avec des indicateurs conventionnels. Ces résultats, combinés aux données épidémiologiques ont permis de montrer une bonne adéquation entre les projections des deux approches et ainsi démontrer que le chiffre noir des consommateurs qui échappent aux mesures de réduction de risque, et qui ne seraient donc pas mesurés par ces indicateurs, est vraisemblablement limité. La troisième partie du travail a été réalisée dans le cadre d'une étude collaborative qui avait pour but d'investiguer la valeur ajoutée de l'analyse des eaux usées à mettre en évidence des différences géographiques dans la consommation de stupéfiants. En particulier pour des substances stigmatisées, telles la cocaïne et l'héroïne, l'approche a permis d'objectiver et de préciser la vision obtenue avec les indicateurs traditionnels du type sondages ou les statistiques policières. Globalement, l'analyse des eaux usées s'est montrée être un outil très utile pour mieux comprendre le marché des stupéfiants, à la fois sous l'angle épidémiologique et répressif. La quatrième partie du travail était focalisée sur la problématique du cannabis ainsi que sur le potentiel de combiner l'analyse des eaux usées aux données de sondage afin de surmonter, en partie, leurs limitations. En utilisant un modèle d'inférence hiérarchique, il a été possible d'affiner les actuelles estimations sur la prévalence de l'utilisation de cannabis dans la zone métropolitaine de la ville de Lausanne. Les résultats ont démontré que celle-ci est plus haute que ce que l'on s'attendait, confirmant ainsi l'hypothèse que les sondages ont tendance à sous-estimer la consommation de cannabis. Bien que biaisés, les données récoltées par les sondages ont permis de surmonter certaines des limitations liées à l'analyse des marqueurs du cannabis dans les eaux usées (i.e., stabilité et manque de données sur l'excrétion). Ces résultats mettent en évidence l'importance et l'utilité de combiner les résultats de l'analyse des eaux usées aux indicateurs existants. De la même façon, la cinquième partie du travail était centrée sur l'apport de l'analyse des eaux usées du point de vue de la police. Au travers de trois exemples, l'utilisation de l'indicateur pour produire du renseignement concernant la structure et les activités des réseaux de distribution de stupéfiants, ainsi que pour guider les choix stratégiques et opérationnels de la police, a été mise en évidence. Dans la dernière partie, la possibilité d'utiliser cette approche pour suivre la consommation de produits pharmaceutiques dangereux, interdits ou contrefaits, a été démontrée par l'analyse dans les eaux usées de la sibutramine et ses métabolites. Les résultats de cette recherche ont mis en évidence que l'épidémiologie par l'analyse des eaux usées est une approche pertinente et puissante, ayant de nombreux domaines d'application. Face à la complexité de mesurer un phénomène caché comme la consommation de stupéfiants, la valeur ajoutée de cette approche a ainsi pu être démontrée.

La plataforma ACME (Avaluació Continuada i Millora de l’Ensenyament) va ser creada l’any 1998 per un grup de professors del departament d’Informàtica i Matemàtica Aplicada. L’ACME es va concebre com una plataforma d’e-learning, és a dir, un sistema que mitjançant l’ús d’Internet afavorís l’aprenentatge, permeten la interactivitat entre l’alumne i el professor. La creació de la plataforma ACME tenia com a objectiu reduir el fracàs dels alumnes en les assignatures de matemàtiques, però degut a l’èxit que va suposar en aquestes, es va decidir incorporar la metodologia de treball ACME a altres disciplines com la programació, les bases de dades, la química, l’economia, etc. de manera que actualment es poden desenvolupar activitats ACME en moltes disciplines. Actualment l’ACME s’utilitza com a complement a les classes presencials, on el professor exposa de manera magistral els conceptes i resol algun exercici a mode d’exemple, per a que després l’alumne, utilitzant la plataforma ACME, intenti resoldre els exercicis proposats pel professor.L’objectiu d’aquest projecte és desenvolupar l’anàlisi, disseny i implementació de les modificacions necessàries a incorporar a la plataforma ACME per tal de millorar el gestor de grups, els exercicis Excel i finalment permetre el treball en grup. El projecte consta de tres parts: millorar les interfícies del professor i de l’alumne, la millora del exercicis Excel i la resolució d’exercicis en grup

Differences in parasite transmission intensity influence the process of acquisition of host immunity to Plasmodium falciparum malaria and ultimately, the rate of malaria related morbidity and mortality. Potential vaccines being designed to complement current intervention efforts therefore need to be evaluated against different malaria endemicity backgrounds. The associations between antibody responses to the chimeric merozoite surface protein 1 block 2 hybrid (MSP1 hybrid), glutamate-rich protein region 2 (GLURP R2) and the peptide AS202.11, and the risk of malaria were assessed in children living in malaria hyperendemic (Burkina Faso, n = 354) and hypo-endemic (Ghana, n = 209) areas. Using the same reagent lots and standardized protocols for both study sites, immunoglobulin (Ig) M, IgG and IgG sub-class levels to each antigen were measured by ELISA in plasma from the children (aged 6-72 months). Associations between antibody levels and risk of malaria were assessed using Cox regression models adjusting for covariates. There was a significant association between GLURP R2 IgG3 and reduced risk of malaria after adjusting age of children in both the Burkinabe (hazard ratio 0.82; 95 % CI 0.74-0.91, p < 0.0001) and the Ghanaian (HR 0.48; 95 % CI 0.25-0.91, p = 0.02) cohorts. MSP1 hybrid IgM was associated (HR 0.85; 95 % CI 0.73-0.98, p = 0.02) with reduced risk of malaria in Burkina Faso cohort while IgG against AS202.11 in the Ghanaian children was associated with increased risk of malaria (HR 1.29; 95 % CI 1.01-1.65, p = 0.04). These findings support further development of GLURP R2 and MSP1 block 2 hybrid, perhaps as a fusion vaccine antigen targeting malaria blood stage that can be deployed in areas of varying transmission intensity.

PFAPA syndrome represents the most common cause of recurrent fever in children in European populations, and it is characterized by recurrent episodes of high fever, pharyngitis, cervical adenitis, and aphthous stomatitis. Many possible causative factors have been explored so far, including infectious agents, immunologic mechanisms and genetic predisposition, but the exact etiology remains unclear. Recent findings demonstrate a dysregulation of different components of innate immunity during PFAPA flares, such as monocytes, neutrophils, complement, and pro-inflammatory cytokines, especially IL-1β, suggesting an inflammasome-mediated innate immune system activation and supporting the hypothesis of an autoinflammatory disease. Moreover, in contrast with previous considerations, the strong familial clustering suggests a potential genetic origin rather than a sporadic disease. In addition, the presence of variants in inflammasome-related genes, mostly in NLRP3 and MEFV, suggests a possible role of inflammasome-composing genes in PFAPA pathogenesis. However, none of these variants seem to be relevant, alone, to its etiology, indicating a high genetic heterogeneity as well as an oligogenic or polygenic genetic background.

We have designed and validated a novel generic platform for production of tetravalent IgG1-like chimeric bispecific Abs. The VH-CH1-hinge domains of mAb2 are fused through a peptidic linker to the N terminus of mAb1 H chain, and paired mutations at the CH1-CL interface mAb1 are introduced that force the correct pairing of the two different free L chains. Two different sets of these CH1-CL interface mutations, called CR3 and MUT4, were designed and tested, and prototypic bispecific Abs directed against CD5 and HLA-DR were produced (CD5xDR). Two different hinge sequences between mAb1 and mAb2 were also tested in the CD5xDR-CR3 or -MUT4 background, leading to bispecific Ab (BsAbs) with a more rigid or flexible structure. All four Abs produced bound with good specificity and affinity to CD5 and HLA-DR present either on the same target or on different cells. Indeed, the BsAbs were able to efficiently redirect killing of HLA-DR(+) leukemic cells by human CD5(+) cytokine-induced killer T cells. Finally, all BsAbs had a functional Fc, as shown by their capacity to activate human complement and NK cells and to mediate phagocytosis. CD5xDR-CR3 was chosen as the best format because it had overall the highest functional activity and was very stable in vitro in both neutral buffer and in serum. In vivo, CD5xDR-CR3 was shown to have significant therapeutic activity in a xenograft model of human leukemia.

In the past years a considerable amount of research has been carried out on the development of theory of mind (ToM) reporting that language and ToM performance are correlated. However it is less clear what aspect of language is related with ToM development. Some studies show a greater influence of semantics aspects, whereas others suggest that sentential complement sentences, a specific aspect of syntax, are crucial for false belief understanding (FB). Yet others argue that pragmatic abilities correlate with FB understanding. The aim of this study is investigate, on the one side, the development of ToM between the ages of 6 and 8 and on the other side, the relationship between some aspects of language and the ToM improvement. Several ToM tasks were administrated to evaluate children’s performance at two different ages (60 participants aged 6 and 8 years) and four tasks to assess language abilities. The results show a significant increase in the understanding of the mind between the ages of 6 and 8. Furthermore, results reveal a significant relationship between syntax and tasks that require FB understanding, while pragmatics is more strongly associated with tasks that involve more complex socio-cognitive understandings

Disseny d’una planta de producció de microalgues com a complement de depuració d’aigües residuals actuant com a tractament terciari i basant-se en l’aprofitament dels nutrients sortints de la depuració d’aigües convencionals, donant un segon ús al CO2 per tal de fer créixer les microalgues i utilitzant la biomassa resultant com a combustible. La principal millora seria el reciclatge dels nutrients i la reutilització del CO2 en biomassa aprofitable

Des de la comunitat científica s’ha posat de manifest la necessitat d’avaluar els programes d’intervenció per a la millora de la parentalitat, especialment en el context de la infància i adolescència en risc. En aquesta recerca es presenten els resultats de l’avaluació del programa Límits aportant evidències d’eficàcia i efectivitat, que permeten identificar i descriure les seves fortaleses, així com també aquells elements que poden ser objecte de millora. El programa Límits és una proposta estructurada d’intervenció preventiva de caire selectiu amb grups de famílies de joves d’entre 14 i 18 anys que passen pel circuit de la justícia juvenil. El programa ha estat funcionant des del 2007, en diverses edicions, a tot el territori català aplegant una experiència i trajectòria remarcable que ha incidit en 351 persones i 245 famílies. El programa consta de vuit sessions en grup d’entre dotze i quinze participants, dinamitzat per dos monitors, en les que es treballen diferents estratègies a partir de l’abordatge de continguts relacionats amb les relacions i els lligams, la comunicació, els conflictes, l’establiment de normes i la disciplina. Per dur a terme l’avaluació del programa Límits s’ha partit d’un disseny quasi-experimental, pretest–postest aplicat a un grup programa (GP) i a grup col·laborador (GC) i de manera diferida (dos mesos després de la seva finalització) només al GP per constatar si s’han assolit els resultats previstos. Quan s’analitza l’evolució del GP s’observen millores en les habilitats d’autocontrol de les emocions, el reforç positiu i les relacions familiars. Tanmateix, des del punt de vista de les famílies no s’han observat canvis significatius en la millora de la percepció d’auto eficàcia del rol parental respecte el GC abans i després del programa. La percepció majoritària és que els objectius que planteja el programa són excessivament ambiciosos i en conseqüència, difícilment assolibles. Els responsables consideren que, en alguns casos, el perfil de famílies derivades al programa no és el més adequat perquè presenten problemàtiques molt més agudes a les desitjables per aquest tipus d’intervenció. Com a punts forts, es destaca que el programa és útil als ulls dels responsables i tècnics perquè genera una presa de consciència del problema i de la necessitat de canvi per part de les famílies. Els participants diuen transferir a la vida real algunes habilitats apreses especialment la comunicació i millora de les relacions familiars. Els tècnics aplicadors coincideixen amb la percepció de les famílies tot i declarar percentatges lleugerament inferiors. També és vist com un complement professional a la seva tasca que ajuda a fer un millor seguiment dels casos, i permet seguir treballant amb posterioritat amb les famílies.

Desde la comunidad científica se ha puesto de manifiesto la necesidad de evaluar los programas de intervención para la mejora de la parentalidad, especialmente en el contexto de la infancia y la adolescencia en riesgo. En esta investigación se presentan los resultados de la evaluación del programa Límites aportando evidencias de eficacia y efectividad, que permiten identificar y describir sus fortalezas, así como también aquellos elementos que pueden ser objeto de mejora. El programa Límites es una propuesta estructurada de intervención preventiva de carácter selectivo, con grupos de familias de jóvenes de entre 14 y 18 años que pasan por el circuito de la justicia juvenil. El programa ha estado funcionando desde 2007 en todo el territorio catalán, y ha incidido en un total de 351 personas y 245 familias. El programa consta de ocho sesiones en grupo en las que se trabajan contenidos relacionados con las relaciones y los vínculos, la comunicación, los conflictos, el establecimiento de normas y la disciplina. Para llevar a cabo la evaluación del programa Límites se ha partido de un diseño cuasi-experimental, pretest-postest aplicado a un grupo programa (GP) y grupo colaborador (GC) y de manera diferida (dos meses después de su finalización) sólo en el GP, para constatar si se han alcanzado los resultados previstos. Cuando se analiza la evolución del GP se observan mejoras en las habilidades de autocontrol de las emociones, el refuerzo positivo y las relaciones familiares. Sin embargo, desde el punto de vista de las familias no se han observado cambios significativos en la mejora de la percepción de auto eficacia del rol parental respecto al GC antes y después del programa. La percepción mayoritaria es que los objetivos que plantea el programa son excesivamente ambiciosos y en consecuencia, difícilmente alcanzables. Los responsables consideran que, en algunos casos, el perfil de familias derivadas al programa no es el más adecuado puesto que presenten problemáticas mucho más agudas a las deseables para este tipo de intervención. Como puntos fuertes, se destaca que el programa es útil a los ojos de los responsables y técnicos porque genera una toma de conciencia del problema y de la necesidad de cambio por parte de las familias. Los participantes dicen transferir a la vida real algunas habilidades aprendidas, especialmente la comunicación y mejora de las relaciones familiares. Los técnicos aplicadores coinciden con la percepción de las familias, a pesar de declarar porcentajes ligeramente inferiores. También es visto como un complemento profesional a su tarea que ayuda a hacer un mejor seguimiento de los casos, y permite seguir trabajando con posterioridad con las familias.

This work intends to identify the reasons why people either like or dislike chemistry classes, based on a written questionnaire answered by 157 students of private and public schools. It has a main question - "Do you like to study chemistry? Why?" - and along with others that complement it, we attempt to explore both pupils' school and personal experiences which help them to increase their learning capacities. The answers have given us a very rich piece of discussing material about the like-and-dislike on the study of chemistry mainly about the social interactions in teaching not only this but all other subjects.

Lyme borreliosis is a tick-transmitted infection caused by the spirochete bacterium Borrelia burgdorferi sensu lato. The tick injects bacteria into host skin, where a first line defence, mainly the complement system, neutrophils, dendritic cells and macrophages are ready to attack foreign intruders. However, in the case of Lyme borreliosis, the original immune response in the skin is untypically mild among bacterial infections. A further untypical feature is the ability of B. burgdorferi to disseminate to distant organs, where, in some patients, symptoms appear after years after the original infection. This study aimed at uncovering some of the immune evasion mechanisms utilized by B. burgdorferi against the complement system, neutrophils and dendritic cells. B. burgdorferi was shown to inhibit chemotaxis of human neutrophils towards nformyl- methyl-leucyl-phenylalanine (fMLP). Outer surface protein B (OspB) of B. burgdorferi was shown to promote resistance to the attack of the complement system and neutrophil phagocytosis at low complement concentrations. B. burgdorferi was shown to inhibit migration of dendritic cells in vitro towards CCL19 and CCL21 and also in an in vivo model. This effect was shown to be due to the absence of CD38 on the borrelia-stimulated dendritic cell surface. A defect in p38 mitogen-activated-protein-kinase (p38) signaling was linked to defective CD38 expression. A defect in CD38 expression on B. burgdorferi-stimulated neutrophils was also observed. In this study, a number of novel immune evasion strategies utilized by B burgdorferi were chracterized. However, further studies are needed as other immune evasion mechanisms await to be uncovered.
