On the nesting biology of Pirhosigma Giordani Soika (Hymenoptera, Vespidae, Eumeninae), with special reference to the use of vegetable matter. The use of vegetable matter in nest building is not widespread among the Eumeninae, and is reported for the first time for the two species of potter wasps Pirhosigma superficiale and P. limpidum. These wasps make mostly spherical mud nests over which they attach small pieces of unmasticated plant matter. Use of plant fragments in this group of wasps is interpreted as camouflage behavior.

Observations on the biology and distribution of Uresiphita reversalis (Lepidoptera, Crambidae), a defoliator of the native tree Calia secundiflora in México. Uresiphita reversalis (Guenée, 1854) feeding on Calia secundiflora (Ortega) Yakovlev is recorded for the first time in the state of Hidalgo, Mexico. New aspects regarding the life cycle, feeding behaviour, geographical distribution and host plant damage by U. reversalis on C. secundiflora are here presented and discussed.

Adults of Cyclocephala distincta are flower visitors of Neotropical palms (Arecaceae) and commonly found in the Atlantic Forest of Pernambuco, Brazil. Males and females were collected in the wild and subjected to captive rearing and breeding. The egg hatching rate, the life cycle, longevity of immatures and adults, and oviposition parameters in captivity were analyzed. The average duration of the life cycle of C. distinctawas 108.2 days (n = 45). The egg stage lasted on average 10.9 days, and the egg-hatching rate was 73.9%. The immature stage lasted on average 93.4 days. The larvae stage exhibited negative phototaxis, and the size of their head capsules increased at a constant rate of 1.6 between instars, following Dyar's rule. The average duration of the first instar was 24.8 days (n = 88), whereas the second and third instars lasted for 17.2 (n = 76) and 40.4 (n = 74) days respectively, and survival rates were 21.6%, 86.4% and 97.4%. The pre-pupal stage was recorded, and pupal chambers were built before pupation. The average number of eggs laid per female was 15.5, the total reproductive period lasted for 3.3 days, and the total fertility was 81.2%. Adults that emerged in captivity exhibited an average longevity of 18.9 days. Adult C. distincta exhibited thanatosis behavior upon manipulation, a strategy observed for the first time in Cyclocephala.

This study aimed to evaluate the development, survival and reproductive capacity of Spodoptera eridania in four soybean cultivars. The experiment was conducted in the laboratory, in a climatic chamber at 25 °C ± 1 °C, 70 ± 10% relative humidity and 12 h photophase. The cultivars used were: FMT Tabarana, BRS/MT Pintado, FMT Tucunaré and Monsoy 8757, all conventional cultivars with medium cycles. All cultivars tested allowed the development of S. eridania. However, Monsoy 8757 was the cultivar that most affected the prolonged in the duration larval, pupal and total cycle, showed lower pupal weight as well as reduction in the intrinsic rate increase. These results contribute to the management of this species in regions of outbreaks in soybean areas.

Our understanding of eumenine nesting biology is still elusive. The use of two nesting strategies, namely renting and building, are reported concomitantly for the first time for Minixi brasilianum (de Saussure, 1875). Ecological factors such as resource availability and protection against potential enemies may play an important role in eumenine nesting biology.

A incidência do Acidente Vascular Cerebral (AVC) em Cabo Verde acompanha a tendência mundial, sendo uma das primeiras causas de morte; nos anos 2013 e 2014, é a primeira causa de morte entre as doenças do aparelho circulatório, isto justificado pelo aumento da esperança média de vida. Também tendo como influência a adoção de novos estilos e padrões de vida não saudáveis, e com o aumento dos fatores de risco que o avançar da idade acarreta. Sendo que a enfermagem como a base de qualquer organização de saúde, necessita fundamentar a sua prática em bases científicas, levando em consideração o individuo como um ser holístico. Desta forma, considerou-se pertinente desenvolver um estudo no âmbito do atendimento de urgência a um utente com Acidente Vascular Cerebral, tendo como objetivo principal identificar as dificuldades que os enfermeiros enfrentam no atendimento de urgência a um utente com AVC. No entanto para dar resposta ao estudo foram considerados outros pontos não menos importantes, tais como: aspetos utilizado no atendimento do AVC, as dificuldades e as estratégias na prestação do cuidado e as técnicas para superar os mesmos. Optou-se por um estudo do tipo fenomenológico, com base na metodologia qualitativo. Sendo que a população do estudo é constituído por 6 enfermeiros, que trabalham no serviço da urgência do Hospital Doutor Baptista de Sousa (HBS). Para que fosse possível recolher os dados, foi estabelecido como instrumento de recolha de dados um guião de entrevista semiestruturada. O tratamento dos dados foi efetuado através da análise de conteúdo. Os resultados dos dados recolhidos, foram apresentados através de quadros e daí retiradas as conclusões consideradas importantes para dar respostas aos objetivos estabelecidos. Uma das principais conclusões do estudo, realça o fato do serviço da urgência apresentar algumas dificuldades no tratamento dos utentes com AVC, devido a falta de alguns recursos materiais considerados indispensáveis, para o diagnóstico diferencial do AVC e assim a tomada de decisões para um tratamento adequado do mesmo. Sendo que, a enfermagem tem um papel pivô no atendimento dos pacientes com Acidente Vascular Cerebral, são esses profissionais que estabelecem o contato entre esses utentes e o resto da equipa multidisciplinar.

La malaltia cardiovascular és una de les principals causes de morbimortalitat. Els factors de risc cardiovasculars són diversos. Hi ha moltes guies de prevenció clínica i escales de risc. Malgrat l’existència de guies de pràctica clínica i millor identificació dels factors de risc, persisteix l’impacte negatiu cardiovascular. L’estudi enregistra aquests esdeveniments cardiovasculars en pacients que ingressen a urgències, durant tres anys, els factors de risc, els tractaments a l’ingrés i a l’alta i l’evolució i mortalitat durant l’ingrés. Compararà la població que ens ocupa amb d’altres i els tractaments. Detectarà el seguiment de les guies i circuits de millora.

ABSTRACT We document the biology and morphology of the egg, caterpillar, and pupa of Strymon crambusa (Hewitson, 1874), a Neotropical Eumaeini. In the Cerrado, the caterpillar feeds on the inflorescences and leaves of Oxalis L. S. crambusa has four larval instars, all of which are illustrated. The density of caterpillars on plants is higher than that recorded for leaf-feeding caterpillars and other flower-feeding Eumaeini, which suggests that the species is a specialist on Oxalidaceae in the Cerrado.

In vivo exposure to chronic hypoxia is considered to be a cause of myocardial dysfunction, thereby representing a deleterious condition, but repeated aeration episodes may exert some cardioprotection. We investigated the possible role of ATP-sensitive potassium channels in these mechanisms. First, rats (n = 8/group) were exposed for 14 days to either chronic hypoxia (CH; 10% O(2)) or chronic hypoxia with one episode/day of 1-hr normoxic aeration (CH+A), with normoxia (N) as the control. Second, isolated hearts were Langendorff perfused under hypoxia (10% O(2), 30 min) and reoxygenated (94% O(2), 30 min) with or without 3 microM glibenclamide (nonselective K(+)(ATP) channel-blocker) or 100 microM diazoxide (selective mitochondrial K(+)(ATP) channel-opener). Blood gasses, hemoglobin concentration, and plasma malondialdehyde were similar in CH and CH+A and in both different from normoxic (P < 0.01), body weight gain and plasma nitrate/nitrite were higher in CH+A than CH (P < 0.01), whereas apoptosis (number of TUNEL-positive nuclei) was less in CH+A than CH (P < 0.05). During in vitro hypoxia, the efficiency (ratio of ATP production/pressure x rate product) was the same in all groups and diazoxide had no measurable effects on myocardial performance, whereas glibenclamide increased end-diastolic pressure more in N and CH than in CH+A hearts (P < 0.05). During reoxgenation, efficiency was markedly less in CH with respect to N and CH+A (P < 0.0001), and ratex pressure product remained lower in CH than N and CH+A hearts (P < 0.001), but glibenclamide or diazoxide abolished this difference. Glibenclamide, but not diazoxide, decreased vascular resistance in N and CH (P < 0.005 and < 0.001) without changes in CH+A. We hypothesize that cardioprotection in chronically hypoxic hearts derive from cell depolarization by sarcolemmal K(+)(ATP) blockade or from preservation of oxidative phosphorylation efficiency (ATP turnover/myocardial performance) by mitochondrial K(+)(ATP) opening. Therefore K(+)(ATP) channels are involved in the deleterious effects of chronic hypoxia and in the cardioprotection elicited when chronic hypoxia is interrupted with short normoxic aeration episodes.

Photodynamic therapy (PDT) is an effective clinical treatment for a number of different cancers. PDT can induce hypoxia and inflammation, pro-angiogenic side effects, which may counteract its angio-occlusive mechanism. The combination of PDT with anti-angiogenic drugs offers a possibility for improved anti-tumour outcome. We used two tumour models to test the effects of the clinically approved angiostatic tyrosine kinase inhibitors sunitinib, sorafenib and axitinib in combination with PDT, and compared these results with the effects of bevacizumab, the anti-VEGF antibody, for the improvement of PDT. Best results were obtained from the combination of PDT and low-dose axitinib or sorafenib. Molecular analysis by PCR revealed that PDT in combination with axitinib suppressed VEGFR-2 expression in tumour vasculature. Treatment with bevacizumab, although effective as monotherapy, did not improve PDT outcome. In order to test for tumour vessel normalization effects, axitinib was also applied prior to PDT. The absence of improved PDT outcome in these experiments, as well as the lack of increased oxygenation in axitinib-treated tumours, suggests that vascular normalization did not occur. The current data imply that there is a future for certain anti-angiogenic agents to further improve the efficacy of photodynamic anti-cancer therapy.

Abstract This thesis investigates the pathogenicity of a Chlamydia-related bacterium: Waddlia chondrophila, which is suspected to cause abortion in bovines as well as miscarriages in humans. Macrophages represent the first line of defense of the innate immunity against invading pathogens, we thus studied the interaction between W. chondrophila and human macrophages. We observed that W. chondrophila multiplied very efficiently inside monocyte-derived macrophages. Diagnostic tools to detect obligate intracellular bacteria are lacking so we developed a Waddlia specific real-time PCR based on the 16S rRNA encoding gene. Since W. chondrophila could be involved in human miscarriage, we analyzed samples from women having miscarriage or uneventful pregnancy. Waddlia serologies results confirmed an association between the presence of W. chondrophila antibodies and miscarriage (23.2% versus 14.6% in the control group, p-value 0.044). As W. chondrophila's pathogenicity was suggested, we studied its antibiotic susceptibility. W. chondrophila revealed to be susceptible to macrolides and tetracylines but resistant to beta-lactams and fluoroquinolones. This resistance to fluoroquinolones could be explained by mutations in the quinolone resistance determining region of the gyrase (GyrA) and topoisomerase IV (ParC) encoding genes. In conclusion, this thesis allowed to precise the role of W. chondrophila in human miscarriage. However, more studies will be necessary to fully understand the pathogenesis of W. chondrophila in adverse pregnancy outcomes. Résumé Le but de cette thèse a été d'étudier la pathogénicité d'une bactérie apparentée aux Chlamydia: Waddlia chondrophila. Les macrophages représentant la première ligne de défense du système immunitaire inné contre les pathogènes, nous avons d'abord étudié l'interaction entre W. chondrophila and les macrophages humains. Nous avons pu observer que W. chondrophila résistait aux effecteurs microbicides des macrophages et se multipliait efficacement au sein de ces cellules. Au vu du manque d'outils diagnostiques pour détecter cette bactérie intracellulaire obligatoire, nous avons également développé une PCR en temps réel, spécifique pour Waddlia, basée sur le gène de l'ARN ribosomal 16S. Cette PCR a été utilisée dans les différents projets afin de détecter la présence de W. chondrophila. W. chondrophila étant suspectée de pouvoir causer des fausses couches chez la femme, nous avons analysé des échantillons provenant de femmes ayant souffert de fausse couche, ainsi que, comme contrôles, des femmes ayant eu une grossesse normale. Les sérologies ont révélé une association entre la présence d'anticorps dirigés contre Waddlia et la fausse couche (23.2% versus 14.6% chez les contrôles, p-value=0.044). La présence de la bactérie a aussi été détectée par PCR et immunohistochimie dans plusieurs échantillons. L'implication de W. chondrophila dans la fausse couche se précisant, nous avons étudié sa susceptibilité aux antibiotiques. W. chondrophila s'est révélée sensible aux macrolides et aux tetracyclines mais résistante aux beta-lactames et aux quinolones. Cette résistance aux quinolones peut être expliquée par la présence de mutations dans le QRDR (région déterminant la résistance aux quinolones) des gènes gyrA et parC. En conclusion, cette thèse a permis de préciser l'implication de W. chondrophila dans la fausse couche. Des études complémentaires seront cependant nécessaires pour confirmer et préciser le rôle exact de W. chondrophila dans les problèmes obstétricaux.

In superficial venous insufficiency, surgery remains the treatment of choice. Endovenous therapies are a minimal invasive alternative, whose long-term results are not demonstrated yet. In the treatment of abdominal aortic aneurysm, endovascular repair (EVAR) and laparoscopic approach are comparatively studied with open repair, to define their precise indications. In occlusive arterial disease, endovascular treatment offers inferior results in term of durability and patency, however with a decrease in morbidity and mortality.

In vitro studies have shown that stimulation of alpha1-adrenoceptors (ARs) directly induces proliferation, hypertrophy, and migration of arterial smooth muscle cells and adventitial fibroblasts. In vivo studies confirmed these findings and showed that catecholamine trophic activity becomes excessive after experimental balloon injury and contributes to neointimal growth, adventitial thickening, and lumen loss. However, past studies have been limited by selectivity of pharmacological agents. The aim of this study, in which mice devoid of norepinephrine and epinephrine synthesis [dopamine beta-hydroxylase (DBH-/-)] or deficient in alpha1-AR subtypes expressed in murine carotid (alpha1B-AR-/- and alpha1D-AR-/-) were used, was to test the hypothesis that catecholamines contribute to wall hypertrophy after injury. At 3 wk after injury of wild-type mice, lumen area and carotid circumference increased significantly, and hypertrophy of media and adventitia was in excess of that needed to restore circumferential wall stress to normal. In DBH-/- and alpha1B-AR-/- mice, increases in lumen area, circumference, and hypertrophy of the media and adventitia were reduced by 50-91%, resulting in restoration of wall tension to nearly normal (DBH-/-) or normal (alpha1B-AR-/-). In contrast, in alpha1D-AR-/- mice, increases in lumen area, circumference, and wall hypertrophy were unaffected and wall thickening remained in excess of that required to return tension to normal. When examined 5 days after injury, proliferation and leukocyte infiltration were inhibited in DBH-/- mice. These studies suggest that the trophic effects of catecholamines are mediated primarily by alpha1B-ARs in mouse carotid and contribute to hypertrophic growth after vascular injury.

Objectives In this study, we have investigated the effects of cannabidiol (CBD) on myocardial dysfunction, inflammation, oxidative/nitrative stress, cell death, and interrelated signaling pathways, using a mouse model of type I diabetic cardiomyopathy and primary human cardiomyocytes exposed to high glucose. Background Cannabidiol, the most abundant nonpsychoactive constituent of Cannabis sativa (marijuana) plant, exerts anti-inflammatory effects in various disease models and alleviates pain and spasticity associated with multiple sclerosis in humans. Methods Left ventricular function was measured by the pressure-volume system. Oxidative stress, cell death, and fibrosis markers were evaluated by molecular biology/biochemical techniques, electron spin resonance spectroscopy, and flow cytometry. Results Diabetic cardiomyopathy was characterized by declined diastolic and systolic myocardial performance associated with increased oxidative-nitrative stress, nuclear factor-kappa B and mitogen-activated protein kinase (c-Jun N-terminal kinase, p-38, p38 alpha) activation, enhanced expression of adhesion molecules (intercellular adhesion molecule-1, vascular cell adhesion molecule-1), tumor necrosis factor-alpha, markers of fibrosis (transforming growth factor-beta, connective tissue growth factor, fibronectin, collagen-1, matrix metalloproteinase-2 and -9), enhanced cell death (caspase 3/7 and poly[adenosine diphosphate-ribose] polymerase activity, chromatin fragmentation, and terminal deoxynucleotidyl transferase dUTP nick end labeling), and diminished Akt phosphorylation. Remarkably, CBD attenuated myocardial dysfunction, cardiac fibrosis, oxidative/nitrative stress, inflammation, cell death, and interrelated signaling pathways. Furthermore, CBD also attenuated the high glucose-induced increased reactive oxygen species generation, nuclear factor-kappa B activation, and cell death in primary human cardiomyocytes. Conclusions Collectively, these results coupled with the excellent safety and tolerability profile of CBD in humans, strongly suggest that it may have great therapeutic potential in the treatment of diabetic complications, and perhaps other cardiovascular disorders, by attenuating oxidative/nitrative stress, inflammation, cell death and fibrosis. PURPOSE: Determine the effect of repeated intravitreal injections of ranibizumab (0.5 mg; 0.05 ml) on retrobulbar blood flow velocities (BFVs) using ultrasound imaging quantification in twenty patients with exudative age-related macular degeneration treated for 6 months. METHODS: Visual acuity (ETDRS), central macular thickness (OCT), peak-systolic, end-diastolic and mean-BFVs in central retinal (CRA), temporal posterior ciliary (TPCA) and ophthalmic (OA) arteries were measured before, 2 days, 3 weeks and 6 months after the first injection. Patients were examined monthly and received 1-5 additional injections depending on ophthalmologic examination results. RESULTS: Six months after the first injection, a significant increase in visual acuity 50.9 ± 25.9 versus 44.4 ± 21.7 (p < 0.01) and decrease in mean central macular thickness 267 ± 74 versus 377 ± 115 μm (p < 0.001) were observed compared to baseline. Although mean-BFVs decreased by 16%±3% in CRA and 20%±5% in TPCA (p < 0.001) 2 days after the first injection, no significant change was seen thereafter. Mean-BFVs in OA decreased by 19%±5% at week 3 (p < 0.001). However, the smallest number of injections (two injections) was associated with the longest time interval between the last injection and month 6 (20 weeks) and with the best return to baseline levels for mean-BFVs in CRA, suggesting that ranibizumab had reversible effects on native retinal vascular supply after its discontinuation. Moreover, a significant correlation between the number of injections and percentage of changes in mean-BFVs in CRA was observed at month 6 (R = 0.74, p < 0.001) unlike TPCA or OA. CONCLUSION: Ranibizumab could impair the native choroidal and retinal vascular networks, but its effect seems reversible after its discontinuation.
