Cutaneous melanoma (CM) is a potentially lethal form of skin cancer and its most important histopathologic factor for staging is Breslow thickness (BT). Its correct determination is fundamental for pathologists. A deeper understanding of the molecular processes guiding CM pathogenesis could improve diagnosis, treatment and prognosis. MicroRNAs (miRNAs) play a key role in CM biology. The firs aim was to investigate miRNA expression in reference to BT assessment. We found that the combined miRNA expression of miR-21-5p and miR-146a-5p above or below 1.5 was significantly associated with overall survival and successfully identified all superficially spreading melanoma (SSM) patients with relapsing suggesting that the combined assessment of these miRNAs expression could aid in SSM staging. Secondly, we focus on multiple primary melanoma (MPM) patients, which develop multiple primary melanomas in their lifetime, and represent a model of high-risk CM occurrence. We explored the miRNome of single CM and MPM: CM and MPM present several dysregulated miRNAs, including key miRNAs involved in epithelial-mesenchymal transition. A different miRNA profile was observed between 1st and 2nd melanoma from the same patient. MiRNA target analysis revealed a more differentiated and less invasive status of MPMs compared to CMs. This characterization of the miRNA regulatory network of MPMs highlights molecular features differentiating this subtype from CM. Recently, NGS experiments revealed the existence of miRNA variants (isomiRs) with different length and sequence. We identified a shorter 3’isoform as tenfold over-represented compared to the canonical form of miR-125a-5p. Target analysis revealed that miRNA shortening could change the pattern of target gene regulation. Finally, we study miRNA and isomiR dysregulation in benign nevi (BN) and CM and in CM and melanoma metastasis. The reported non-random dysregulation of specific isomiRs contributes to the understanding of the complex melanoma pathogenesis and serves as the basis for further functional studies.

Introduzione: La stiffness epato-splenica, misurata attraverso la transient elastography (TE), è stata associata con la fibrosi midollare nei pazienti con malattia mieloproliferativa (MPNs). La rigidità dei tessuti può essere valutata con la shear-wave elastography (SWE), con due tecniche: point (pSWE) e bidimensionale (2DSWE). Obiettivi dello studio sono: 1) identificare le differenze di TE fra i pazienti con MPNs, i cirrotici e volontari sani (HV); 2) valutare specifiche caratteristiche di TE in pazienti con MF, PV ed ET; 3) stabilire una correlazione con il grado di fibrosi midollare. Metodi: in questo studio monocentrico, MPN, cirrotici ed HV hanno eseguito elastometria epato-splenica con pSWE e 2DSWE. Risultati: 236 pazienti sono stati inclusi in questo studio: 64 con MF (27.1%), 33 con PV (14%), 46 con ET (19.4%), 75 HV (32%) e 18 (8%) cirrotici. Al confronto con gli HV, i pazienti con MF hanno maggiore stiffness splenica (pSWE 40.9 vs 26.3 kPa, p<0.001; 2DSWE 34.9 vs 20.1 kPa, p<0.001) ed epatica (pSWE 7.72 vs 5.52 kPa, p<0.001; 2DSWE 6.96 vs 5.01 kPa, p<0.001). Al confronto con i pazienti con PV ed ET, quelli con MF hanno maggiori valori di stiffness epatici (p<0.001) e splenici (p<0.001). In fibrosi di basso (0-1) (n=81 , 60.4%) vs alto grado (2-3) (n=42, 39.6%), sono evidenti valori di stiffness maggiori nei pazienti con fibrosi di alto grado sia per il fegato (pSWE 5.2 vs 6.65 kPa; 2DSWE 5.1 vs 6.05 kPa) che nella milza (pSWE 27.2 vs 37.9 kPa, 2DSWE 21.7 vs 30.75 kPa – p<0.001) Conclusioni: La TE distingue i pazienti con MF sia dai sani che dalle altre MPNs. Valori di TE sono significativamente associati con caratteristiche rilevanti che includono la fibrosi midollare in tutte le MPNs. I valori di stiffness epatici e splenici sono pertanto rilevanti nella diagnosi e management delle MPNs.

Le travail de recherche se focalise sur l'étude de l'adaptation de romans naturalistes pour le théâtre. Phénomène largement répandu au XIXe siècle, les adaptations pour le théâtre promettaient la gloire aux romanciers qui tentaient une carrière littéraire. Par ailleurs, ils ont marqué une poussée vers un renouveau du théâtre lui-même sous toutes les formes possibles : interprétation, scénographie, thèmes. Pour cette raison, une étude comparative de plusieurs auteurs naturalistes européens (Zola, Goncourt, Capuana, Lemonnier, Galdós, Hardy) permet de construire un cadre général et d'identifier des pratiques communes. Par ailleurs, l'analyse effectuée au cours de la recherche met également en évidence des tendances socio-littéraires significatives, emblématiques de la construction d'une société capitaliste et démocratique en Europe ouvrant la voie au XXe siècle.

The recent adoption of IFRS 9 is a highly disruptive accounting reform, with significant impacts on how and when negative news (i.e., negative adjustments to reported earnings) are recognized on the financial statements. Using a unique dataset of two major banks operating in one European country we provide evidence of a tightening of the corporate loans pricing after the IFRS 9 adoption. Furthermore, by focusing on the post reform period, we show that the tightening is driven by the new staging classification. Higher risk premiums are associated to clients with previous underperforming exposures (stage 2) and higher probability of default. We also observe that the staging classification is not affecting climate risk premiums. Our results highlight that the lenders, as expected by the regulation, change their risk appetite by charging higher spreads to discourage loan origination for clients that became too risky and expensive under the new standard.
