HIV-positive adolescents face a number of challenges in dealing with their disease and its treatment. In this qualitative study, twenty-nine HIV-positive adolescents aged 13 to 20 years (22 girls), who live in Switzerland, were asked, in a semi-structured interview (duration of 40-110 minutes), to describe their perceptions and experiences with the disease itself and with therapeutic adherence. While younger adolescents most often thought of their disease as fate, older adolescents usually knew that they had received it through vertical transmission, although the topic appeared to be particularly difficult to discuss for those living with their HIV-positive mothers. Based on their attending physician's assessment, 18 subjects were judged highly adherent, 4 fairly and 7 poorly adherent. High adherence appeared linked with adequate psychological adjustment and effective coping mechanisms, as well as with the discussion and adoption of explicit medication-taking strategies. The setting and organisation of health care teams should allow for ongoing discussions with HIV-positive adolescents that focus on their perceptions of their disease, how they cope with it and with the treatment, and how they could improve their adherence.

Salmonella is distributed worldwide and is a pathogen of economic and public health importance. As a multi-host pathogen with a long environmental persistence, it is a suitable model for the study of wildlife-livestock interactions. In this work, we aim to explore the spill-over of Salmonella between free-ranging wild boar and livestock in a protected natural area in NE Spain and the presence of antimicrobial resistance. Salmonella prevalence, serotypes and diversity were compared between wild boars, sympatric cattle and wild boars from cattle-free areas. The effect of age, sex, cattle presence and cattle herd size on Salmonella probability of infection in wild boars was explored by means of Generalized Linear Models and a model selection based on the Akaike’s Information Criterion. Prevalence was higher in wild boars co-habiting with cattle (35.67%, CI 95% 28.19–43.70) than in wild boar from cattle-free areas (17.54%, CI 95% 8.74–29.91). Probability of a wild boar being a Salmonella carrier increased with cattle herd size but decreased with the host age. Serotypes Meleagridis, Anatum and Othmarschen were isolated concurrently from cattle and sympatric wild boars. Apart from serotypes shared with cattle, wild boars appear to have their own serotypes, which are also found in wild boars from cattle-free areas (Enteritidis, Mikawasima, 4:b:- and 35:r:z35). Serotype richness (diversity) was higher in wild boars co-habiting with cattle, but evenness was not altered by the introduction of serotypes from cattle. The finding of a S. Mbandaka strain resistant to sulfamethoxazole, streptomycin and chloramphenicol and a S. Enteritidis strain resistant to ciprofloxacin and nalidixic acid in wild boars is cause for public health concern.

In recent years, the emergence of Staphylococcus aureus strains with reduced susceptibility to glycopeptides has raised considerable concern. We studied the efficacy of vancomycin and teicoplanin, as well as cloxacillin and cefotaxime, against the infection caused by four S. aureus strains with different glycopeptide and β-lactam susceptibilities (strains A, B, C, and D; MICs for vancomycin of 1, 2, 4, and 8 µg/ml respectively), using a modified model of mouse peritonitis. This optimized model appeared to be straightforward and reproducible, and was able to detect low differences in bacterial killing between antibiotics and also between different S. aureus strains. Bactericidal activities in peritoneal fluid for vancomycin, teicoplanin, cloxacillin, and cefotaxime decreased from -2.98, -2.36, -3.22, and -3.57 log10 cfu/ml, respectively, in infection by strain A (MICs for vancomycin and cloxacillin of 1 and 0.38 µg/ml, respectively) to -1.22, -0.65, -1.04, and +0.24 in peritonitis due to strain D (MICs for vancomycin and cloxacillin of 8 and 1,024 µg/ml). Our data confirm the superiority of β-lactams against methicillin-susceptible S. aureus and show that bactericidal activity of glycopeptides decreases significantly with slight increases in MICs; this finding suggests a reduced efficacy of glycopeptides in the treatment of serious glycopeptide-intermediate S. aureus infections

One of the global targets for non-communicable diseases is to halt, by 2025, the rise in the age-standardised adult prevalence of diabetes at its 2010 levels. We aimed to estimate worldwide trends in diabetes, how likely it is for countries to achieve the global target, and how changes in prevalence, together with population growth and ageing, are affecting the number of adults with diabetes. We pooled data from population-based studies that had collected data on diabetes through measurement of its biomarkers. We used a Bayesian hierarchical model to estimate trends in diabetes prevalence-defined as fasting plasma glucose of 7.0 mmol/L or higher, or history of diagnosis with diabetes, or use of insulin or oral hypoglycaemic drugs-in 200 countries and territories in 21 regions, by sex and from 1980 to 2014. We also calculated the posterior probability of meeting the global diabetes target if post-2000 trends continue. We used data from 751 studies including 4,372,000 adults from 146 of the 200 countries we make estimates for. Global age-standardised diabetes prevalence increased from 4.3% (95% credible interval 2.4-7.0) in 1980 to 9.0% (7.2-11.1) in 2014 in men, and from 5.0% (2.9-7.9) to 7.9% (6.4-9.7) in women. The number of adults with diabetes in the world increased from 108 million in 1980 to 422 million in 2014 (28.5% due to the rise in prevalence, 39.7% due to population growth and ageing, and 31.8% due to interaction of these two factors). Age-standardised adult diabetes prevalence in 2014 was lowest in northwestern Europe, and highest in Polynesia and Micronesia, at nearly 25%, followed by Melanesia and the Middle East and north Africa. Between 1980 and 2014 there was little change in age-standardised diabetes prevalence in adult women in continental western Europe, although crude prevalence rose because of ageing of the population. By contrast, age-standardised adult prevalence rose by 15 percentage points in men and women in Polynesia and Micronesia. In 2014, American Samoa had the highest national prevalence of diabetes (>30% in both sexes), with age-standardised adult prevalence also higher than 25% in some other islands in Polynesia and Micronesia. If post-2000 trends continue, the probability of meeting the global target of halting the rise in the prevalence of diabetes by 2025 at the 2010 level worldwide is lower than 1% for men and is 1% for women. Only nine countries for men and 29 countries for women, mostly in western Europe, have a 50% or higher probability of meeting the global target. Since 1980, age-standardised diabetes prevalence in adults has increased, or at best remained unchanged, in every country. Together with population growth and ageing, this rise has led to a near quadrupling of the number of adults with diabetes worldwide. The burden of diabetes, both in terms of prevalence and number of adults affected, has increased faster in low-income and middle-income countries than in high-income countries. Wellcome Trust.

The study consists is the application of zeolites NaX, NaY and A as builder in detergent formulations to eliminate the hardness of water. Therefore, the adsorption of ions Ca+2 and Mg+2 were evaluated, and the effect of the cleaning action of the surfactant sodium dodecil sulfate (SDS) through tests of detergency. The experiments were conducted in bath system (with shaking) and quantification of metals was performed by atomic absorption spectrometry. Zeolite A showed the best results for adsorption of Ca++ and Mg++ with retention rates of around 90 and 70% respectively and acted positively on the action of cleaning the surfactant SDS.

O presente trabalho teve como objetivo o desenvolvimento de um método analÃtico simples, rápido e eficiente para a determinação de corticosteróides (cortisona, corticosterona, acetato de hidrocortisona e acetato de dexametasona) em amostras sangüÃneas de ratos empregando um sistema isocrático de cromatografia lÃquida de alta eficiência (CLAE) com detecção ultravioleta. O método envolveu a injeção direta da amostra sangüÃnea em uma coluna cromatográfica com superfÃcie interna de fase reversa (ISRP-C18), empregando a fase móvel composta por tampão fosfato pH 4,0: acetonitrila (65:35 v/v). A detecção dos analitos foi obtida, através de um detector de ultravioleta de comprimento de onda variável (Varian Modelo 2550) ajustado em 240 nm e um integrador SP 4400 Chromaject (Varian Associates, Inc, Sunnyvale, CA, USA). A extração do analito resultou em valores de recuperação entre 90% e 108%, e coeficiente de variação entre 1,1% a 2,5%. Os limites de detecção e quantificação do método foram de 0,02 e 0,04 µg mL-1, respectivamente. Assim, o método analÃtico proposto possibilitou a injeção direta (on-line) da amostra sem tratamento prévio apresentando também várias vantagens, tais como: rapidez, exatidão, precisão e especificidade.

Foram realizados dois experimentos, com objetivo de avaliar a suscetibilidade de Brachiaria subquadripara e Brachiaria mutica a diferentes herbicidas aplicados em pósemergência. Os herbicidas e doses testados foram: glyphosate (Rodeo) a 2.400, 3.360 e 4.320 g e.a. ha-1 com Aterbane a 0,5% v/v; glyphosate a 2.400, 3.360 e 4.320 g e.a. ha-1 com Silwet a 0,1% v/v; imazapyr (Arsenal) a 750 e 1.500 g e.a. ha-1; e diquat (Reward) a 400 e 800 g i.a. ha-1 com aplicação seqüencial. Manteve-se uma testemunha sem aplicação de herbicidas. As parcelas experimentais foram constituÃdas de caixas d'água de 60 x 60 x 45 cm, com 55 L de solo. A aplicação foi realizada quando as plantas se encontravam a 45 e 65 cm de altura para B. subquadripara e B. mutica, respectivamente. Foi utilizado um pulverizador costal, a pressão constante de CO2 (2 bars), pontas 110.02 XR, com um consumo de calda de 200 L ha-1. O controle foi avaliado visualmente, através de escala percentual de notas, além de se avaliar a massa seca das plantas. O herbicida glyphosate nas doses de 3.360 e 4.320 g e.a. ha-1, independentemente do adjuvante testado, proporcionou controle excelente para as duas espécies, porém as parcelas que receberam glyphosate a 2.400 g e.a. ha-1 apresentaram controle apenas satisfatório. O herbicida imazapyr nas doses de 750 e 1.500 g e.a. ha-1 proporcionou controle eficiente para B. subquadripara e excelente para B. mutica. Já o herbicida diquat, apesar de duas aplicações e independentemente da dose utilizada, mostrou-se ineficiente no controle das duas espécies.

Objetivou-se com este trabalho avaliar os efeitos do 2,4-D sobre o crescimento das plantas, a produção de massa seca e verde e a produtividade de grãos do milheto. O experimento foi realizado no perÃodo de março a junho de 2006, em Rio Verde-GO, em um Latossolo Vermelho eutroférrico. O milheto (cultivar ADR 500) foi semeado manualmente em área cultivada sob sistema de plantio direto, em espaçamento de 0,45 m, distribuindo-se 12 sementes por metro. Utilizou-se o delineamento de blocos casualizados, em esquema fatorial 4 x 4, sendo avaliadas quatro doses de 2,4-D (0, 335, 670 e 1.005 g ha-1) aplicadas em quatro épocas [10 dias após a emergência das plantas de milheto (DAE) (3 folhas); 20 DAE (5 a 6 folhas expandidas); 30 DAE (inÃcio de emissão da inflorescência); e 40 DAE (florescimento pleno)]. Para evitar a interferência das plantas daninhas na cultura, esta foi capinada manualmente, sempre que necessário. Não se observou nenhum sinal de intoxicação das plantas de milheto pelo 2,4-D aos 15 dias após a aplicação, independentemente da dose ou época de aplicação do herbicida. Todavia, as maiores doses de 2,4-D, em qualquer época de aplicação, provocaram menor acúmulo de massa verde e seca das plantas de milheto, quando se avaliaram os resultados no ponto de rolagem da cultura. O 2,4-D, independentemente da dose utilizada ou época de aplicação, não influenciou a produtividade de grãos do milheto.
Anxiolytic-like effects of 4-phenyl-2-trichloromethyl-3H-1,5-benzodiazepine hydrogen sulfate in mice

The pharmacological effects of 4-phenyl-2-trichloromethyl-3H-1,5-benzodiazepine hydrogen sulfate (PTMB), a novel synthetic benzodiazepine, were examined in mice. In the elevated plus-maze test of anxiety, 0.3-1 mg/kg diazepam ip (F(3,53) = 3.78; P<0.05) and 1-10 mg/kg PTMB ip increased (F(5,98) = 3.26; P<0.01), whereas 2 mg/kg picrotoxin ip decreased (F(3,59) = 8.32; P<0.001) the proportion of time spent in the open arms, consistent with an anxiolytic action of both benzodiazepines, and an anxiogenic role for picrotoxin. In the holeboard, 1.0 mg/kg diazepam ip increased (F(3,54) = 2.78; P<0.05) and 2 mg/kg picrotoxin ip decreased (F(3,59) = 4.69; P<0.01) locomotor activity. Rotarod assessment revealed that 1 mg/kg diazepam ip and 3, 10 and 30 mg/kg PTMB ip produced significant motor incoordination compared to vehicle control (F(4,70) = 7.6; P<0.001). These data suggest that the recently synthesized PTMB compound possesses anxiolytic activity and produces motor incoordination similar to those observed with diazepam.

Hepatitis C, a worldwide viral infection, is an important health problem in Brazil. The virus causes chronic infection, provoking B lymphocyte dysfunction, as represented by cryoglobulinemia, non-organ-specific autoantibody production, and non-Hodgkin's lymphoma. The aim of this research was to screen for the presence of antiphospholipid autoantibodies in 109 Brazilian hepatitis C virus carriers without clinical history of antiphospholipid syndrome. Forty healthy individuals were used as the control group. IgA, IgG, and IgM antibodies against cardiolipin and 46;2-glycoprotein I were measured with an enzyme-linked immunosorbent assay, using a cut-off point of either 20 UPL or 20 SBU. While 24 (22.0%) hepatitis C carriers had moderate titers of IgM anticardiolipin antibodies (median, 22.5 MPL; 95%CI: 21.5-25.4 MPL), only three carriers (<3%) had IgG anticardiolipin antibodies (median, 23 GPL; 95%CI: 20.5-25.5 GPL). Furthermore, IgA anticardiolipin antibodies were not detected in these individuals. Male gender and IgM anticardiolipin seropositivity were associated in the hepatitis C group (P = 0.0004). IgA anti- 46;2-glycoprotein-I antibodies were detected in 29 of 109 (27.0%) hepatitis C carriers (median, 41 SAU; 95%CI: 52.7-103.9 SAU). Twenty patients (18.0%) had IgM anti- 46;2-glycoprotein I antibodies (median, 27.6 SMU; 95%CI: 23.3-70.3 SMU), while two patients had IgG antibodies against this protein (titers, 33 and 78 SGU). Antiphospholipid antibodies were detected in only one healthy individual, who was seropositive for IgM anticardiolipin. We concluded that Brazilian individuals chronically infected with hepatitis C virus present a significant production of antiphospholipid antibodies, mainly IgA anti- 46;2-glycoprotein I antibodies, which are not associated with clinical manifestations of antiphospholipid syndrome.

We evaluated the expression of glial fibrillary acidic protein (GFAP), glutamine synthetase (GS), ionized calcium binding adaptor protein-1 (Iba-1), and ferritin in rats after single or repeated lipopolysaccharide (LPS) treatment, which is known to induce endotoxin tolerance and glial activation. Male Wistar rats (200-250 g) received ip injections of LPS (100 µg/kg) or saline for 6 days: 6 saline (N = 5), 5 saline + 1 LPS (N = 6) and 6 LPS (N = 6). After the sixth injection, the rats were perfused and the brains were collected for immunohistochemistry. After a single LPS dose, the number of GFAP-positive cells increased in the hypothalamic arcuate nucleus (ARC; 1 LPS: 35.6 ± 1.4 vs control: 23.1 ± 2.5) and hippocampus (1 LPS: 165.0 ± 3.0 vs control: 137.5 ± 2.5), and interestingly, 6 LPS injections further increased GFAP expression in these regions (ARC = 52.5 ± 4.3; hippocampus = 182.2 ± 4.1). We found a higher GS expression only in the hippocampus of the 6 LPS injections group (56.6 ± 0.8 vs 46.7 ± 1.9). Ferritin-positive cells increased similarly in the hippocampus of rats treated with a single (49.2 ± 1.7 vs 28.1 ± 1.9) or repeated (47.6 ± 1.1 vs 28.1 ± 1.9) LPS dose. Single LPS enhanced Iba-1 in the paraventricular nucleus (PVN: 92.8 ± 4.1 vs 65.2 ± 2.2) and hippocampus (99.4 ± 4.4 vs 73.8 ± 2.1), but had no effect in the retrochiasmatic nucleus (RCA) and ARC. Interestingly, 6 LPS increased the Iba-1 expression in these hypothalamic and hippocampal regions (RCA: 57.8 ± 4.6 vs 36.6 ± 2.2; ARC: 62.4 ± 6.0 vs 37.0 ± 2.2; PVN: 100.7 ± 4.4 vs 65.2 ± 2.2; hippocampus: 123.0 ± 3.8 vs 73.8 ± 2.1). The results suggest that repeated LPS treatment stimulates the expression of glial activation markers, protecting neuronal activity during prolonged inflammatory challenges.

Inúmeros estudos vêm sendo realizados com o objetivo de compreender o comportamento das proteÃnas do Concentrado Protéico de Soro (CPS). A capacidade destas proteÃnas em formar géis estáveis a temperaturas entre 70ºC e 90ºC, é uma propriedade funcional importante para a confecção de vários produtos alimentÃcios, tais como: produtos de padaria, cárneos, texturizados e lácteos. A concentração protéica, pH, composição iônica e temperatura podem ser controladas para obter um gel com as caracterÃsticas desejadas. Neste trabalho foi investigado o efeito dos Ãons metálicos bivalentes Ca++ e Mg++, na propriedade de dureza de géis de CPS induzidos termicamente. Dispersões protéicas a 6; 7 e 8% de proteÃna do CPS a pH 6,3 com 0; 7,5; 15; 30 e 75mm de CaCl2 ou MgCl2 foram aquecidas a 75ºC por 45 min, resfriadas a 4ºC por 12 horas, e os géis avaliados em um texturômetro TA-XT2. Os géis formados com maiores concentrações do sal adicionado obtiveram maiores valores de dureza na faixa estudada. Nas concentrações de 15 a 75mM os valores de dureza dos géis com a adição de CaCl2 foram significativamente maiores (p<0,05) do que com adição de MgCl2. Na concentração de 8% de proteÃnas do CPS ambos os tratamentos atingem o maior valor de dureza (p<0,05) a 30mM do sal adicionado. O cálcio iônico mostrou ter um importante papel na formação do gel de proteÃnas do soro.

Este trabalho teve por objetivos avaliar o efeito da temperatura e do estresse hÃdrico na germinação das sementes e na formação de plântulas normais de Amburana acreana. Foram realizados dois ensaios. No primeiro, as sementes foram colocadas para germinar nas temperaturas de 20, 25, 30, 35 e 40°C. No segundo, as sementes foram dispostas em substrato umedecido com soluções de polietilenoglicol 6000 nos seguintes potenciais hÃdricos: zero (testemunha), -0,2, -0,4, -0,8, -1,0 e -1,2MPa, a 30°C. A germinação das sementes e a formação de plântulas normais foi avaliada durante 30 dias, em cinco repetições de 20 sementes, sendo consideradas germinadas aquelas que apresentaram emissão de raiz primária com no mÃnimo 5mm de comprimento e curvatura geotrópica positiva. Os melhores resultados, no primeiro ensaio, foram encontrados nas temperaturas de 30 e 35°C, com maiores valores de porcentagens de germinação, sendo os tratamentos onde o processo germinativo ocorreu em menor tempo. A porcentagem de germinação, no segundo ensaio, diminuiu à medida que se reduziu a disponibilidade de água no substrato, a partir de -0,4MPa, e foi nula entre -1,0 e -1,2MPa. A formação de plântulas foi reduzida já no potencial -0,2MPa, sendo totalmente inibida entre -0,4 e -0,8MPa. Conclui-se que a temperatura de 30°C é mais recomendada para a condução do teste de germinação por propiciar melhores condições para o processo germinativo, com maior porcentagem de plântulas normais e menores porcentagens de plântulas anormais e de sementes deterioradas. A redução na disponibilidade hÃdrica diminui a germinação das sementes e a formação de plântulas de cerejeira e os limites para germinação ocorrem na faixa de zero a -1,0MPa e, para formação de plântulas, de zero a -0,4MPa.
