In 2001, a weather and climate monitoring network was established along the temperature and aridity gradient between the sub-humid Moroccan High Atlas Mountains and the former end lake of the Middle Drâa in a pre-Saharan environment. The highest Automated Weather Stations (AWS) was installed just below the M'Goun summit at 3850 m, the lowest station Lac Iriki was at 450 m. This network of 13 AWS stations was funded and maintained by the German IMPETUS (BMBF Grant 01LW06001A, North Rhine-Westphalia Grant 313-21200200) project and since 2011 five stations were further maintained by the GERMAN DFG Fennec project (FI 786/3-1), this way some stations of the AWS network provided data for almost 12 years from 2001-2012. Standard meteorological variables such as temperature, humidity, and wind were measured at an altitude of 2 m above ground. Other meteorological variables comprise precipitation, station pressure, solar irradiance, soil temperature at different depths and for high mountain station snow water equivalent. The stations produced data summaries for 5-minute-precipitation-data, 10- or 15-minute-data and a daily summary of all other variables. This network is a unique resource of multi-year weather data in the remote semi-arid to arid mountain region of the Saharan flank of the Atlas Mountains. The network is described in Schulz et al. (2010) and its further continuation until 2012 is briefly discussed in Redl et al. (2015, doi:10.1175/MWR-D-15-0223.1) and Redl et al. (2016, doi:10.1002/2015JD024443).

Os produtos provenientes do mar têm um importante papel a nível socioeconómico, gastronómico e no legado cultural das comunidades piscatórias e costeiras de Portugal. O Percebe Pollicipes pollicipes é na Península Ibérica o recurso biológico do intertidal mais explorado pelo ser humano, devido à sua enorme popularidade e consequente procura, fazendo em algumas ocasiões disparar o preço de mercado até 150€/kg, gerando na sobreexploração dos stocks existentes. No entanto, na área marinha protegida da Reserva Natural da Berlenga, a apanha do percebe é fortemente regulada, tendo-se tornado em Portugal num bom exemplo da gestão de recursos marinhos. Com o intuito de prevenir fraudes, adulteração alimentar ou quaisquer outras práticas que possam induzir o consumidor em erro a Comissão Europeia declara que, o consumidor tem o direito de receber informação correcta acerca dos produtos que adquire, para além de definir regras para a correcta aplicação destas regras. Métodos analíticos que possibilitem identificar a origem do percebe, tornam-se deste modo importantes ferramantas no desenvolvimento de um selo de Denominação de Origem Protegida (DOP) e na gestão comercial do produto. Deste modo, investigou-se se o Percebe possui diferenças específicas de cada local de captura, através da forma da unha (CS), da composição microquímica da unha (EM) e do perfil de ácidos gordos (FA). A análise foi efectuada em indivíduos recolhidos em 3 locais na Reserva Natural das Berlengas e 7 ao longo de 300 km da costa Portuguesa. Em cada indivíduo analisou-se a forma da unha (CS) através da morfometria geométrica, a composição microquímica da unha (EM) através de ICP-MS e o perfil de ácidos gordos do músculo através de GC-FID. A análise das funções discriminantes (DFA) quer para a EM quer para a FA em separado obteve um elevado sucesso de reclassificação (77,6 % e 99% respectivamente, através de validação cruzada), enquanto que EM combinado com FA permitiu um sucesso de reclassificação de 100 %. A análise discriminante baseada apenas na CS, demonstrou um baixo sucesso (29,6 %) .Estes resultados demonstram que a composição microquímica da unha e o perfil de ácidos gordos do músculo de Percebe, poderá ser uma ferramenta de elevada importância, na determinação da origem do Percebe. Esta abordagem poderá ser utilizada para identificar a origem dos percebes comercializados, bem como ajudar no desenvolvimento de um selo DOP, aumentando ao mesmo tempo o valor potencial dos recursos biológicos provenientes de áreas marinhas protegidas em Portugal.

Background - Specific language impairment (SLI) is a common neurodevelopmental disorder, observed in 5–10 % of children. Family and twin studies suggest a strong genetic component, but relatively few candidate genes have been reported to date. A recent genome-wide association study (GWAS) described the first statistically significant association specifically for a SLI cohort between a missense variant (rs4280164) in the NOP9 gene and language-related phenotypes under a parent-of-origin model. Replications of these findings are particularly challenging because the availability of parental DNA is required. Methods - We used two independent family-based cohorts characterised with reading- and language-related traits: a longitudinal cohort (n = 106 informative families) including children with language and reading difficulties and a nuclear family cohort (n = 264 families) selected for dyslexia. Results - We observed association with language-related measures when modelling for parent-of-origin effects at the NOP9 locus in both cohorts: minimum P = 0.001 for phonological awareness with a paternal effect in the first cohort and minimum P = 0.0004 for irregular word reading with a maternal effect in the second cohort. Allelic and parental trends were not consistent when compared to the original study. Conclusions - A parent-of-origin effect at this locus was detected in both cohorts, albeit with different trends. These findings contribute in interpreting the original GWAS report and support further investigations of the NOP9 locus and its role in language-related traits. A systematic evaluation of parent-of-origin effects in genetic association studies has the potential to reveal novel mechanisms underlying complex traits.

Oil polluted and not oil polluted soils (crude oil hydrocarbons contents: 20-92500 mg/kg dry soil mass) under natural grass and forest vegetation and in a bog in the Russian tundra were compared in their principal soil ecological parameters, the oil content and the microbial indicators. CFE biomass-C, dehydrogenase and arylsulfatase activity were enhanced with the occurrence of crude oil. Using these parameters for purposes of controlling remediation and recultivation success it is not possible to distinguish bctween promotion of microbial activity by oil carbon or soil organic carbon (SOC). For this reason we think that these parameters are not appropriate to indicate a soil damage by an oil impact. In contrast the metabolie quotient (qC02), calculated as the ratio between soil basal respiration and the SIR biomass-C was adequate to indicate a high crude oil contamination in soil. Also, the ß-glucosidase activity (parameter ß-GL/SOC) was correlated negatively with oil in soil. The indication of a soil damage by using the stress parameter qCO, or the specific enzyme activities (activity/SOC) minimizes the promotion effect of the recent SOC content on microbial parameters. Both biomass methods (SIR, CFE) have technical problems in application for crude oil-contaminated and subarctic soils. CFE does not reflect the low C_mic level of the cold tundra soils. We recommend to test every method for its suitability before any data collection in series as well as application for cold soils and the application of ecophysiological ratios as R_mic/C_mic, C_mic/SOC or enzymatic activity/SOC instead of absolute data.

During the 14th expedition of the research vessel "Meteor" from the 2nd of July to the 7th of August 1968 continously recording instruments for measuring the CO2 partial pressure of seawater and atmospheric CO2 were developped by the Meteorological Institute, University of Frankfurt/M. During the Faroer expedition instrumental constants, such as relative and absolute accuracy, inertia and solvent power were tested. The performance of discontinous analyses of water samples was adopted to shipboard conditiones and correction factors depending on water volume, depth of sampling and water temperature were measured. After having computed average values of the continous records (atmosp. CO2 content, CO2 partial pressure, water temperature) geographical distribution, diurnal variation and dependence of diurnal averages were tested. At four different locations CO2 partial pressure was measured in various depths. During the voyage from the Faroer islands to Helgoland the measured concentrations of atmospheric CO2 content and CO2 partial pressure were tested with respect to a correlation of the geographical latitude.

Ocean acidification will likely have negative impacts on invertebrates producing skeletons composed of calcium carbonate. Skeletal solubility is partly controlled by the incorporation of "foreign" ions (e.g. magnesium) into the crystal lattice of these skeletal structures, a process that is sensitive to a variety of biological and environmental factors. Here we explore effects of life stage, oceanographic region of origin, and changes in the partial pressure of carbon dioxide in seawater (pCO2) on trace elemental composition in the purple sea urchin (Strongylocentrotus purpuratus). We show that, similar to other urchin taxa, adult purple sea urchins have the ability to precipitate skeleton composed of a range of biominerals spanning low- to high-Mg calcites. Mg / Ca and Sr / Ca ratios were substantially lower in adult spines compared to adult tests. On the other hand, trace elemental composition was invariant among adults collected from four oceanographically distinct regions spanning a range of carbonate chemistry conditions (Oregon, Northern California, Central California, and Southern California). Skeletons of newly settled juvenile urchins that originated from adults from the four regions exhibited intermediate Mg / Ca and Sr / Ca between adult spine and test endmembers, indicating that skeleton precipitated during early life stages is more soluble than adult spines and less soluble than adult tests. Mean skeletal Mg / Ca or Sr / Ca of juvenile skeleton did not vary with source region when larvae were reared under present-day, global-average seawater carbonate conditions (400 µatm; pHT = 8.02 ± 0.03 1 SD; Omega calcite = 3.3 ± 0.2 1 SD). However, when reared under elevated pCO2 (900 µatm; pHT = 7.73 ± 0.03; Omega calcite = 1.8 ± 0.1), skeletal Sr / Ca in juveniles exhibited increased variance across the four regions. Although larvae from the northern populations (Oregon, Northern California, Central California) did not exhibit differences in Mg or Sr incorporation under elevated pCO2 (Sr / Ca = 2.10 ± 0.06 mmol/mol; Mg / Ca = 67.4 ± 3.9 mmol/mol), juveniles of Southern California origin partitioned ~8% more Sr into their skeletons when exposed to higher pCO2 (Sr / Ca = 2.26 ± 0.08 vs. 2.09 ± 0.005 mmol/mol 1 SD). Together these results suggest that the diversity of carbonate minerologies present across different skeletal structures and life stages in purple sea urchins does not translate into an equivalent geochemical plasticity of response associated with geographic variation or temporal shifts in seawater properties. Rather, composition of S. purpuratus skeleton precipitated during both early and adult life history stages appears relatively robust to spatial gradients and predicted future changes in carbonate chemistry. An exception to this trend may arise during early life stages, where certain populations of purple sea urchins may alter skeletal mineral precipitation rates and composition beyond a given pCO2 threshold. This potential for geochemical plasticity during early development in contrast to adult stage geochemical resilience adds to the growing body of evidence that ocean acidification can have differing effects across organismal life stages.

Black band disease (BBD) is the oldest recognised disease associated with scleractinian corals. However, despite this, few BBD surveys have been conducted in the Indonesian archipelago, one of the world’s hot spots for coral diversity. In this study, we show that BBD was recorded in the reefs of Kepulauan Seribu, Indonesia, at the time of surveying. The disease was found to mainly infect corals of the genus Montipora. In some instances, upwards of 177 colonies (31.64%) were found to be infected at specific sites. Prevalence of the disease ranged from 0.31% to 31.64% of Montipora sp. colonies throughout the archipelago. Although BBD was found at all sites, lower frequencies were
associated with sites closest to the mainland (17.99 km), as well as those that were furthest away (63.65 km). Despite there being no linear relationship between distance from major population centers and BBD incidence, high incidences of this disease were associated with sites characterized by higher
levels of light intensity. Furthermore, surveys revealed that outbreaks peaked during the transitional period between the dry and rainy seasons. Therefore, we suggest that future surveys for disease prevalence in this region of Indonesia should focus on these transitory periods.

The intestinal tract of schistosomes opens at the mouth and leads into the foregut or oesophageal region that is lined with syncytium continuous with the apical cytoplasm of the tegument. The oesophagus is surrounded by a specialised gland, the oesophageal gland. This gland releases materials into the lumen of the oesophagus and the region is thought to initiate the lysis of erythrocytes and neutralisation of immune effectors of the host. The oesophageal region is present in the early invasive schistosomulum, a stage potentially targetable by anti-schistosome vaccines. We used a 44k oligonucleotide microarray to identify highly up-regulated genes in microdissected frozen sections of the oesophageal gland of male worms of S. mansoni. We show that 122 genes were up-regulated 2-fold or higher in the oesophageal gland compared with a whole male worm tissue control. The enriched genes included several associated with lipid metabolism and transmembrane transport as well as some micro-exon genes. Since the oesophageal gland is important in the initiation of digestion and the fact that it develops early after invasion of the mammalian host, further study of selected highly up-regulated functionally important genes in this tissue may reveal new anti-schistosome intervention targets for schistosomiasis control.

This thesis analyses the influence of qualitative and quantitative herbage production on seasonal rangelands, and of herd and pasture use strategies on feed intake, body mass development and reproductive performance of sheep and goats in the Altai mountain region of Bulgan county (soum) in Khovd province (aimag). This westernmost county of Mongolia is characterized by a very poor road network and thus very difficult access to regional and national markets. The thesis explores in this localized context the current rural development, the economic settings and political measures that affect the traditional extensive livestock husbandry system and its importance for rural livelihoods. Livestock management practices still follow the traditional transhumant mode, fully relying on natural pasture. This renders animal feeding very vulnerable to the highly variable climatic conditions which is one of many reasons for gradually declining quantity and quality of pasture vegetation. Small ruminants, and especially goats, are the main important species securing economic viability of their owners’ livelihood, and they are well adapted to the harsh continental climate and the present low input management practices. It is likely that small ruminants will keep their vital role for the rural community in the future, since the weak local infrastructure and slow market developments currently do not allow many income diversification options. Since the profitability of a single animal is low, animal numbers tend to increase, whereas herd management does not change. Possibilities to improve the current livestock management and thus herders’ livelihoods in an environmentally, economically and socially sustainable manner are simulated through bio-economic modelling and the implications are discussed at the regional and national scale. To increase the welfare of the local population, a substantial infrastructural and market development is needed, which needs to be accompanied by suitable pasture management schemes and policies

G6PC3 is a widely expressed isoform of glucose-6-phosphatase, found in many foetal and adult tissues. Mutations in this gene cause developmental abnormalities and severe neutropenia due to abolition of glucose recycling between the cytoplasm and endoplasmic reticulum. Low G6PC3 expression as a result of promoter polymorphisms or dysregulation could produce similar outcomes. Here we investigated the regulation of human G6PC3 promoter activity. HeLa and H4IIE cells were transiently transfected with G6PC3 promoter coupled to the firefly luciferase gene, and promoter activity was measured by dual luciferase assay. Activity was highest in a 453 bp segment of the G6PC3 promoter, from − 455 to − 3 relative to the transcriptional start site. This promoter was unresponsive to glucostatic hormones. Its activity increased significantly between 1 and 5.5 mM glucose, and was not elevated further by glucose concentrations up to 25 mM. Pyruvate increased its activity, but β-hydroxybutyrate and sodium acetate did not. Promoter activity was reduced by inhibitors of hexokinase, glyceraldehyde phosphate dehydrogenase and the oxidative branch of the pentose phosphate pathway, but not by a transketolase inhibitor. Deletion of two adjacent Enhancer-boxes (− 274 to − 279 and − 299 to − 304) reduced promoter activity and abolished the glucose effect, suggesting they could function as a glucose response element. Deletion of an additional downstream 140 bp (− 140 to − 306) restored activity, but not the glucose response, suggesting the presence of repressor elements in this region. 5-Aminoimidazole-4-carboxamide 1-β-d-ribofuranoside (AICAR) reduced promoter activity, showing dependence on AMP-kinase. Regulation of the G6PC3 promoter is thus radically different to that of the hepatic isoform, G6PC. It is sensitive to carbohydrate, but not to fatty acid metabolites, and at much lower physiological concentrations. Based on these findings, we speculate that reduced G6PC3 expression could occur during hypoglycemic episodes in vivo, which are common in utero and in the postnatal period. If such episodes lower G6PC3 expression they could place the foetus or infant at risk of impaired immune function and development, and this possibility requires further examination both in vitro and in vivo.

Remote Sensing has been used for decades, and more and more applications are added to its repertoire. With this study we aim to show the use of Remote Sensing in the field of vegetation recovery monitoring in burned areas and the added value of data with a high spatial resolution. This was done by analysing both Landsat 7 and 8 scenes, after the forest fire of summer 2012 in the parish of Calde, in the central region of Portugal, as well as an orthophoto produced with images acquired by an unmanned aerial vehicle.
