Various compositions of synthetic calcium phosphates (CaP) have been proposed and their use has considerably increased over the past decades. Besides differences in physico-chemical properties, resorption and osseointegration, artificial CaP bone graft might differ in their resistance against biofilm formation. We investigated standardised cylinders of 5 different CaP bone grafts (cyclOS, chronOS (both β-TCP (tricalcium phosphate)), dicalcium phosphate (DCP), calcium-deficient hydroxyapatite (CDHA) and α-TCP). Various physico-chemical characterisations e.g., geometrical density, porosity, and specific surface area were investigated. Biofilm formation was carried out in tryptic soy broth (TSB) and human serum (SE) using Staphylococcus aureus (ATCC 29213) and S. epidermidis RP62A (ATCC 35984). The amount of biofilm was analysed by an established protocol using sonication and microcalorimetry. Physico-chemical characterisation showed marked differences concerning macro- and micropore size, specific surface area and porosity accessible to bacteria between the 5 scaffolds. Biofilm formation was found on all scaffolds and was comparable for α-TCP, chronOS, CDHA and DCP at corresponding time points when the scaffolds were incubated with the same germ and/or growth media, but much lower for cyclOS. This is peculiar because cyclOS had an intermediate porosity, mean pore size, specific surface area, and porosity accessible to bacteria. Our results suggest that biofilm formation is not influenced by a single physico-chemical parameter alone but is a multi-step process influenced by several factors in parallel. Transfer from in vitro data to clinical situations is difficult; thus, advocating the use of cyclOS scaffolds over the four other CaP bone grafts in clinical situations with a high risk of infection cannot be clearly supported based on our data.

PURPOSE: In the radiopharmaceutical therapy approach to the fight against cancer, in particular when it comes to translating laboratory results to the clinical setting, modeling has served as an invaluable tool for guidance and for understanding the processes operating at the cellular level and how these relate to macroscopic observables. Tumor control probability (TCP) is the dosimetric end point quantity of choice which relates to experimental and clinical data: it requires knowledge of individual cellular absorbed doses since it depends on the assessment of the treatment's ability to kill each and every cell. Macroscopic tumors, seen in both clinical and experimental studies, contain too many cells to be modeled individually in Monte Carlo simulation; yet, in particular for low ratios of decays to cells, a cell-based model that does not smooth away statistical considerations associated with low activity is a necessity. The authors present here an adaptation of the simple sphere-based model from which cellular level dosimetry for macroscopic tumors and their end point quantities, such as TCP, may be extrapolated more reliably. METHODS: Ten homogenous spheres representing tumors of different sizes were constructed in GEANT4. The radionuclide 131I was randomly allowed to decay for each model size and for seven different ratios of number of decays to number of cells, N(r): 1000, 500, 200, 100, 50, 20, and 10 decays per cell. The deposited energy was collected in radial bins and divided by the bin mass to obtain the average bin absorbed dose. To simulate a cellular model, the number of cells present in each bin was calculated and an absorbed dose attributed to each cell equal to the bin average absorbed dose with a randomly determined adjustment based on a Gaussian probability distribution with a width equal to the statistical uncertainty consistent with the ratio of decays to cells, i.e., equal to Nr-1/2. From dose volume histograms the surviving fraction of cells, equivalent uniform dose (EUD), and TCP for the different scenarios were calculated. Comparably sized spherical models containing individual spherical cells (15 microm diameter) in hexagonal lattices were constructed, and Monte Carlo simulations were executed for all the same previous scenarios. The dosimetric quantities were calculated and compared to the adjusted simple sphere model results. The model was then applied to the Bortezomib-induced enzyme-targeted radiotherapy (BETR) strategy of targeting Epstein-Barr virus (EBV)-expressing cancers. RESULTS: The TCP values were comparable to within 2% between the adjusted simple sphere and full cellular models. Additionally, models were generated for a nonuniform distribution of activity, and results were compared between the adjusted spherical and cellular models with similar comparability. The TCP values from the experimental macroscopic tumor results were consistent with the experimental observations for BETR-treated 1 g EBV-expressing lymphoma tumors in mice. CONCLUSIONS: The adjusted spherical model presented here provides more accurate TCP values than simple spheres, on par with full cellular Monte Carlo simulations while maintaining the simplicity of the simple sphere model. This model provides a basis for complementing and understanding laboratory and clinical results pertaining to radiopharmaceutical therapy.

ADSL is becoming the standard form of residential and small-business broadband Internet access due to, primarily, its low deployment cost. These ADSL residential lines are often deployed with 802.11 Access Points (AP) that providewireless connectivity. Given the density of ADSL deployment, it is often possible for a residential wireless client to be in range of several other APs, belonging to neighbors, with ADSL connectivity. While the ADSL technology has showed evident limits in terms of capacity (with speeds ranging 1-10 Mbps), the short-range wireless communication can guarantee a muchhigher capacity (up to 20 Mbps). Furthermore, the ADSL links in the neighborhood are generally under-utilized, since ADSL subscribers do not connect 100% of the time. Therefore, it is possible for a wireless client to simultaneously connect to several APs in range and effectively aggregate their available ADSL bandwidth.In this paper, we introduce ClubADSL, a wireless client that can simultaneously connect to several APs in range on different frequencies and aggregate both their downlink and uplink capacity. ClubADSL is a software that runs locally on the client-side, and it requires neither modification to the existing Internet infrastructure, nor any hardware/protocol upgradesto the 802.11 local area network. We show the feasibility of ClubADSL in seamlessly transmitting TCP traffic, and validate its implementation both in controlled scenarios and with current applications over real ADSL lines. In particular we show that a ClubADSL client can greatly benefit from the aggregated download bandwidth in the case of server-client applications such as video streaming, but can also take advantage of the increased upload bandwidth greatly reducing download times with incentive-based P2P applications such as BitTorrent.

The antiretroviral protein TRIM5alpha is known to have evolved different restriction capacities against various retroviruses, driven by positive Darwinian selection. However, how these different specificities have evolved in the primate lineages is not fully understood. Here we used ancestral protein resurrection to estimate the evolution of antiviral restriction specificities of TRIM5alpha on the primate lineage leading to humans. We used TRIM5alpha coding sequences from 24 primates for the reconstruction of ancestral TRIM5alpha sequences using maximum-likelihood and Bayesian approaches. Ancestral sequences were transduced into HeLa and CRFK cells. Stable cell lines were generated and used to test restriction of a panel of extant retroviruses (human immunodeficiency virus type 1 [HIV-1] and HIV-2, simian immunodeficiency virus [SIV] variants SIV(mac) and SIV(agm), and murine leukemia virus [MLV] variants N-MLV and B-MLV). The resurrected TRIM5alpha variant from the common ancestor of Old World primates (Old World monkeys and apes, approximately 25 million years before present) was effective against present day HIV-1. In contrast to the HIV-1 restriction pattern, we show that the restriction efficacy against other retroviruses, such as a murine oncoretrovirus (N-MLV), is higher for more recent resurrected hominoid variants. Ancestral TRIM5alpha variants have generally limited efficacy against HIV-2, SIV(agm), and SIV(mac). Our study sheds new light on the evolution of the intrinsic antiviral defense machinery and illustrates the utility of functional evolutionary reconstruction for characterizing recently emerged protein differences.

Työ aloitettiin perehtymällä Metro Ethernet verkon eriosiin kuten Ethernet-verkkoon, MultiProtocol Label Switching verkkoon ja näiden verkkojen eriosiin. Työssä siis tarkasteltiin lähemmin Ethernet verkon toimintaa sekä sen tarjoamia standardeja kaistannopeuteen, kaapeli materiaaleihin ja kehysrakenteeseen. Tarkasteltiin myös MultiProtocol Label Swit-ching verkon rakennetta, pakettien reititys perusteita pohjautuen Ethernet verkon kehysrakenteeseen, Internet Protocol over MultiProtocol Label Switching tekniikkaa sekä näihin liittyviä alakäsitteitä kuten palvelunlaatu, virtuaaliyhteys, MAC-osoite jne. Kun työssä oli selvitetty Metro Ethernet verkon taustat ja perusteet voitiin siirtyä tutkimaan Metro Area Network ja Metro Ethernet Networks verkkojen tarkempaa rakennetta ja niiden mahdollisuuksia. Koska työn aiheena on Metro Ethernet-verkko, tulee myös tutkia Metro Ethernet Forum järjestöä jossa päätetään kaikki Metro Ethernet-verkkoon liittyvät asiat ja mm. MEF standardit. MEF standardeja on 16 kappaletta ja ne määrittelevät mm. Metro Ethernet verkon palvelut ja verkon arkkitehtuurin. Viimeisenä tutkimusalueena oli kolmen eri verkkolaitteen tutkiminen ja jo valmiina olevien verkkolaitteiden mahdollisuuksien kartoitus. Laitteiden tutkinnassa perehdyttiin Metro Et-hernet-verkko mahdollisuuksiin jokaisen laitevalmistajan tarjoamalla Internet Protocol over MultiProtocol Label Switching tekniikalla. Lopuksi tutkittiin vielä verkkosuunnitelman poh-jan luomista sekä itse suunnitelmaa, aikaisempien tutkimus tulosten pohjalle. Tutkimuksen tuloksena saatiin Metro Ethernet verkon ja käyttökelpoisten verkkolaitteiden tutkimusraportti ja käyttökelpoinen Metro Ethernet-verkkosuunnitelma jota voi käyttää mahdollisesti suoraan pohjana tai ainakin vertailu pohjana käytännön toteutuksessa.

O presente estudo analisou a relação entre percepção de estresse, sintomas depressivos e autoestima em idosos com e sem queixa subjetiva de comprometimento de memória. Foram incluídos 204 idosos (104 sem e 100 com queixa de memória) avaliados a partir do instrumento Memory Assessment Complain Questionnaire (MAC-Q). O protocolo de estudo incluiu a Escala de Estresse Percebido (EEP), a Escala de Depressão Geriátrica (GDS) e a Escala de Autoestima de Rosenberg (EAE). Os idosos com queixa de comprometimento apresentaram escores significativamente maiores na EEP e GDS e menores na EAE (p < 0.001). Foi observada correlação negativa entre o escore do MAC-Q e EPP (p < 0.001) e EAE (p = 0.01). A análise de regressão multivariada identificou somente o estresse como fator preditor da queixa subjetiva de memória. Esses dados sugerem que a percepção de estresse e os sintomas depressivos estão associados com a queixa de memória em idosos.

Genome-wide scans of genetic differentiation between hybridizing taxa can identify genome regions with unusual rates of introgression. Regions of high differentiation might represent barriers to gene flow, while regions of low differentiation might indicate adaptive introgression-the spread of selectively beneficial alleles between reproductively isolated genetic backgrounds. Here we conduct a scan for unusual patterns of differentiation in a mosaic hybrid zone between two mussel species, Mytilus edulis and M. galloprovincialis. One outlying locus, mac-1, showed a characteristic footprint of local introgression, with abnormally high frequency of edulis-derived alleles in a patch of M. galloprovincialis enclosed within the mosaic zone, but low frequencies outside of the zone. Further analysis of DNA sequences showed that almost all of the edulis allelic diversity had introgressed into the M. galloprovincialis background in this patch. We then used a variety of approaches to test the hypothesis that there had been adaptive introgression at mac-1. Simulations and model fitting with maximum-likelihood and approximate Bayesian computation approaches suggested that adaptive introgression could generate a "soft sweep," which was qualitatively consistent with our data. Although the migration rate required was high, it was compatible with the functioning of an effective barrier to gene flow as revealed by demographic inferences. As such, adaptive introgression could explain both the reduced intraspecific differentiation around mac-1 and the high diversity of introgressed alleles, although a localized change in barrier strength may also be invoked. Together, our results emphasize the need to account for the complex history of secondary contacts in interpreting outlier loci.

A successful bone tissue engineering strategy entails producing bone-scaffold constructs with adequate mechanical properties. Apart from the mechanical properties of the scaffold itself, the forming bone inside the scaffold also adds to the strength of the construct. In this study, we investigated the role of in vivo cyclic loading on mechanical properties of a bone scaffold. We implanted PLA/β-TCP scaffolds in the distal femur of six rats, applied external cyclic loading on the right leg, and kept the left leg as a control. We monitored bone formation at 7 time points over 35 weeks using time-lapsed micro-computed tomography (CT) imaging. The images were then used to construct micro-finite element models of bone-scaffold constructs, with which we estimated the stiffness for each sample at all time points. We found that loading increased the stiffness by 60% at 35 weeks. The increase of stiffness was correlated to an increase in bone volume fraction of 18% in the loaded scaffold compared to control scaffold. These changes in volume fraction and related stiffness in the bone scaffold are regulated by two independent processes, bone formation and bone resorption. Using time-lapsed micro-CT imaging and a newly-developed longitudinal image registration technique, we observed that mechanical stimulation increases the bone formation rate during 4-10 weeks, and decreases the bone resorption rate during 9-18 weeks post-operatively. For the first time, we report that in vivo cyclic loading increases mechanical properties of the scaffold by increasing the bone formation rate and decreasing the bone resorption rate.

A bibliographic revision on the fisheries research in the Canary, Madeira, and the Azores Islands related to stock assessment of the striped soldier shrimp (Plesionika edwardsii) (Crustacea, Decapoda, Pandalidae) was made. Biological information –reproduction, growth, mortality, recruitment– of the target species, selective technological developments –multiple semi-floating shrimp traps–, and know-how gained by the ICCM can give support to elaborate and execute an ICCM-INDP joint project, in order to explore and assess its insular stocks in the Capeverdian deepwaters (150-300 m). In the frame of the UE Trans-national Cooperation Programme MAC (2007- 2013), the next coming call for proposals seems to be an adequate source to fund it. This project should include some activities for the valorisation and promotion of this new shellfish product, as well as for scientific-technical and social divulgation, and for giving advice to the decision-makers. Lastly, with the aim of developing a new fishing activity in the Cape Verdes on a responsible and sustainable basis, authors have scheduled a second study phase. This should consist of a process of technology transfer, followed by an experimental fishing pilot project including a study on biological and commercial viability with the participation of the Capeverdian fishing sector.

Esta dissertação descreve os resultados das medições observadas em um dos Laboratórios de uma Operadora de Telecomunicações (LOP), onde foram avaliados e analisados alguns requisitos de QoS em redes de pacotes IP (Internet Protocol). Essas medições foram feitas no âmbito do objetivo desta dissertação que é avaliar formas de prover serviços VoIP (Voice over Internet Protocol) em redes de pacotes conforme a recomendação do padrão FRF.12. Essa rede é assim, uma rede de link de 512kbps que também provê serviços VoIP compartilhados, concorrentemente com dados e serviços multimídia. Dos ítens analisados destacam-se: Análise de Codecs; QoS (Quality Of Service) Diffserv; Compressão de cabeçalho RTP (Real Time Protocol) - cRTP; Fragmentação com intercalação - LFI; Comportamento da Rede em situações diversas; a adequação do software free Multi Generator (MGEN) de geração - medição - coleta de dados, em redes. A análise foi, essencialmente, em enlace Frame Relay nos CPE (Customer Premise Equipment), passando pelo Backbone IP VPN / MPLS Multicast, pois o Frame Relay Fórum v12 (FRF.12) dá suporte à intercalação de voz entre os pacotes de dados. O FRF.12 é indispensável, pois este esta dissertação tem como objetivo realizar um conjuntos de testes e medidas que avaliam a aplicação dos serviços VoIP em links de baixa capacidades com trafego de dados compartilhados. Para oferecer esse serviço e de qualidade é necessário fragmentar e intercalar frames de voz entre os pacotes de dados usando o FRF.12. Depois do estudo teórico das recomendações, normas de padronização internacional e dos fabricantes, foram realizados testes que resultam na validação prática de toda a teoria outrora analisada através de testes específicos que comprovam em definitivo a viabilidade das aplicações VoIP em uma rede de enlace de baixa velocidade. Feitos esses testes chegou-se a conclusão de que em determinados casos não se revela necessário nem preocupante o aumento da banda para se puder prover determinados serviços. Na sequência dos testes foram também avaliados o desempenho, a ocupação da banda e a eficácia dos equipamentos - softwares. Da bancada dos testes e medições, provou-se o seguinte: que de fato consegue-se melhor otimização da banda ao realizar compressão do cabeçalho cRTP; que de fato a fragmentação de pacote FTP (File Transfer Protocol) com intercalação de pacotes VoIP faz reduzir o delay e jitter1 para as aplicações de tempo real; que de fato a habilitação de QoS Intserv provê classificação e faz diferenciação dos tráfegos, e que o CODEC G729 apresenta melhor adequação em lidar com aplicações VoIP em routers2 CISCO, disponível em CRT (Centro de referência Tecnológica) de uma LOP.

Els objectius del projecte són: realitzar un intèrpret de comandes en VAL3 que rebi les ordres a través d’una connexió TCP/IP; realitzar una toolbox de Matlab per enviar diferents ordres mitjançant una connexió TCP/IP; adquirir i processar mitjançant Matlab imatges de la càmera en temps real i detectar la posició d’objectes artificials mitjançant la segmentació per color i dissenyar i realitzar una aplicació amb Matlab que reculli peces detectades amb la càmera. L’abast del projecte inclou: l’estudi del llenguatge de programació VAL3 i disseny de l’ intèrpret de comandes, l’estudi de les llibreries de Matlab per comunicació mitjançant TCP/IP, per l’adquisició d’imatges, pel processament d’imatges i per la programació en C; el disseny de la aplicació recol·lectora de peces i la implementació de: un intèrpret de comandes en VAL3, la toolbox pel control del robot STAUBLI en Matlab i la aplicació recol·lectora de peces mitjançant el processament d’imatges en temps real també en Matlab

Pour lever les contraintes multiples auxquelles le pays est confronté, et développer les différents secteurs de l’agriculture et de la pêche, le gouvernement a adopté plusieurs programmes qui sont à différents stades de mise oeuvre, notamment: (i) le Programme national pour l'environnement, (ii) le Programme national de lutte contre la désertification, (iii) le Programme national de lutte contre la pauvreté, (iv) le Plan d'action forestier national, (v) le Plan directeur des ressources hydriques, (vi) le programme national de sécurité alimentaire, (vii) le Plan directeur pour l'élevage, et (viii) le plan de développement de l’horticulture. Bien que pertinents individuellement, les différents programmes nationaux ne sont point reliés entre eux par une stratégie cohérente de développement agricole au sens large, à moyen et long terme. Une assistance avait été apportée par la FAO au Cap-Vert en 19961 pour appuyer le GEP dans la formulation d'une stratégie sectorielle, en vue de la préparation du 4ème plan national de développement pour la période 1997-2000. Les nouvelles autorités nationales, en place en 2001, voudraient disposer d’une stratégie de développement agricole de long terme, allant au-delà des programmes nationaux de développement élaborés tous les 4 ans, au gré des mandats politiques. Par ailleurs, plusieurs partenaires au développement du Cap-Vert ont également indiqué au Gouvernement que leur nouveau cadre de coopération orienté vers l’aide budgétaire, doit disposer d’un cadre stratégique cohérent à long terme, et d’un plan d'action opérationnel indiquant les actions à entreprendre et les moyens humains, institutionnels, et financiers nécessaires à leur mise en oeuvre. La stratégie de développement agricole, au sens large, s’appuie sur des politiques, initiatives et dynamiques en cours, parmi lesquelles : la vision stratégique de développement à moyen termes du Gouvernement, présentée dans les Grandes Options du Plan 2002-2005 (GOP), le document intérimaire de stratégie de réduction de la pauvreté (DSRP) du pays, indiquant la sécurité alimentaire et la garantie de la cohésion sociale en milieu rural comme des axes stratégiques importants, le programme national de sécurité alimentaire du Cap-Vert, avec une orientation marquée pour la réduction de la pauvreté le PANA II en cours d’élaboration, le document UNDAF2 pour le Cap-Vert élaboré en septembre 2002, avec quatre axes d’intervention prioritaire et cinq dimensions transversales. la dimension genre, pour favoriser la participation effective de toutes les couches de la société (notamment les femmes) au processus de développement socio-économique du pays, particulièrement dans les politiques, stratégies et programmes du MAAP en cohérence avec la politique de décentralisation du Gouvernement. les dynamiques d’intégration sous-régionale en cours dans le cadre de la CEDEAO3, et du CILSS4, et continentales dans le cadre du NEPAD5.
