The attitude of the medieval church towards violence before the First Crusade in 1095 underwent a significant institutional evolution, from the peaceful tradition of the New Testament and the Roman persecution, through the prelate-led military campaigns of the Carolingian period and the Peace of God era. It would be superficially easy to characterize this transformation as the pragmatic and entirely secular response of a growing power to the changing world. However, such a simplification does not fully do justice to the underlying theology. While church leaders from the 5th Century to the 11th had vastly different motivations and circumstances under which to develop their responses to a variety of violent activities, the teachings of Augustine of Hippo provided a unifying theme. Augustine’s just war theology, in establishing which conflicts are acceptable in the eyes of God, focused on determining whether a proper causa belli or basis for war exists, and then whether a legitimate authority declares and leads the war. Augustine masterfully integrated aspects of the Old and New Testaments to create a lasting and compelling case for his definition of justified violence. Although at different times and places his theology has been used to support a variety of different attitudes, the profound influence of his work on the medieval church’s evolving position on violence is clear.

Nicotinic receptors are the target of nicotine in the brain. They are pentameric ion channels. The pentamer structure allows many combinations of receptors to be formed. These various subtypes exhibit specific properties determined by their subunit composition. Each brain region contains a fixed complement of nicotinic receptor subunits. The midbrain region is of particular interest because the dopaminergic neurons of the midbrain express several subtypes of nicotinic receptors, and these dopaminergic neurons are important for the rewarding effects of nicotine. The α6 nicotinic receptor subunit has garnered intense interest because it is present in dopaminergic neurons but very few other brain regions. With its specific and limited presence in the brain, targeting this subtype of nicotinic receptor may prove advantageous as a method for smoking cessation. However, we do not fully understand the trafficking and membrane localization of this receptor or its effects on dopamine release in the striatum. We hypothesized that lynx1, a known modulator of other nicotinic receptor subtypes, is important for the proper function of α6 nicotinic receptors. lynx1 has been found to act upon several classes of nicotinic receptors, such as α4β2 and α7, the two most common subtypes in the brain. To determine whether lynx1 affects α6 containing nicotinic receptors we used biochemistry, patch clamp electrophysiology, fast scan cyclic voltammetry, and mouse behavior. We found that lynx1 has effects on α6 containing nicotinic receptors, but the effects were subtle. This thesis will detail the observed effects of lynx1 on α6 nicotinic receptors.

A tese analisa a relação íntima que há entre o pragmatismo ou o conseqüencialismo e a modulação temporal dos efeitos das decisões judiciais. Nesta relação, interessa ressaltar o ponto de interseção que certamente sobressai em várias ocasiões: o argumento de cunho econômico. Tal tipo de argumento pode assumir especial relevo quando do exame da oportunidade e conveniência na tomada das decisões eminentemente políticas. No âmbito jurisdicional, no entanto, o argumento pragmático ou consequencialista de cunho econômico não deve prevalecer como fundamento das decisões judiciais, especialmente cuidando-se de matéria tributária. Os problemas que centralizam o estudo podem ser colocados através das seguintes indagações: é possível que o Supremo Tribunal Federal compute, no julgamento de certa matéria tributária, argumento como o eventual rombo de X bilhões de reais que a decisão contrária ao Fisco possa acarretar para os cofres públicos? A fundamentação de eventual decisão judicial calcada exclusiva ou predominantemente em tal argumento é legítima ou ilegítima? Que importância pode ter na tomada de decisão judicial? Quando aplicada, há parâmetros a serem seguidos? Quais? Demonstramos que a prevalência de tal argumento é inadequada na seara judicial, ou seja, deve ter peso reduzido ou periférico, servindo para corroborar ou reforçar os argumentos jurídicos que centralizam o debate submetido ao exame do Poder Judiciário de modo geral, e do Supremo Tribunal Federal, de maneira particular. Em busca de esclarecer quais os principais limites e possibilidades de tal argumento, especialmente relacionando-o à modulação temporal dos efeitos da decisão judicial, explicitamos algumas regras necessárias para a sua adequada utilização, sob pena de inconcebível subversão de variados princípios e direitos fundamentais assegurados em sede constitucional. No exame das questões submetidas à apreciação da Corte Suprema em matéria tributária, o seu parâmetro consiste na maior efetividade e concretude ao texto constitucional. A modulação temporal dos efeitos se aplica a uma decisão que, declarando a inconstitucionalidade do ato normativo, se afastaria ainda mais da vontade constitucional, caso fosse aplicado o tradicional efeito ex tunc (retroativo até o nascimento da lei). Nestas situações específicas e excepcionais se justifica aplicar a modulação, com vistas a dar maior concretude e emprestar maior eficácia à Constituição. A tese proposta, ao final, consiste na reunião das regras explicitadas no trabalho e em proposta legislativa.

Part one of this thesis consists of two sections. In the first section the fluorine chemical shift of a single crystal CaF_2 has been measured as a function of external pressure up to 4 kilobar at room temperature using multiple pulse NMR techniques. The pressure dependence of the shift is found to be -1.7 ± 1 ppm/kbar, while a theoretical calculation using an overlap model predicts a shift of -0.46 ppm/kbar. In the second section a separation of the chemical shift tensor into physically meaningful "geometrical" and "chemical" contributions is presented and a comparison of the proposed model calculations with recently reported data on hydroxyl proton chemical shift tensors demonstrates, that for this system, the geometrical portion accounts for the qualitative features of the measured tensors.
Part two of the thesis consists of a study of fluoride ion motion in β-PbF_2 doped with NaF by measurement of the ^(19)F transverse relaxation time (T_2), spin lattice relaxation time (T_1) and the spin lattice relaxation time in the rotating frame (T_(1r)). Measurements over the temperature range of -50°C to 160°C lead to activation energies for T_1, T_(1r) and T_2 of 0.205 ± 0.01, 0.29 + 0.02 and 0.27 ± 0.01 ev/ion, and a T_(1r) minimum at 56°C yields a correlation time of 0.74 μsec. Pressure dependence of T_1 and T_2 yields activation volumes of <0.2 cm^3/g-mole and 1.76 ± 0.05 cm^3/g-mole respectively. These data along with the measured magnetic field independence of T_1 suggest that the measured T_1's are not caused by ^(19)F motion, but by thermally excited carriers.
Part three of the thesis consists of a study of two samples of Th_4H_(15), prepared under different conditions but both having the proper ratio of H/Th (to within 1%). The structure of the Th_4H_(15) as suggested by X-ray measurements is confirmed through a moment analysis of the rigid lattice line shape. T_1 and T_2 measurements above 390 K furnish activation energies of 16.3 ± 1.2 kcal/mole and 18.0 ± 3.0 kcal/mole, respectively. Below 350 K, T_(1r) measurements furnish an activation energy of 10.9 ± 0.7 kcal/mole, indicating most probably more than a single mechanism for proton motion. A time-temperature hysteresis effect of the proton motion was found in one of the two samples and is strongly indicative of a phase change. T_1 at room temperature and below is dominated by relaxation due to conduction electrons with the product T_1T being 180 ± 10 K-sec. Using multiple pulse techniques to greatly reduce homonuclear dipolar broadening, a temperature-dependent line shift was observed, and the chemical shift anisotropy is estimated to be less than 16 ppm.

The authors have been studying the control of phytoplankton biomass in several Australian reservoirs. To manipulate fish communities in order to reduce phytoplankton biomass, one needs a thorough understanding of processes in the plankton-associated food webs. In contrast to the situation found in lakes of the northern hemisphere, the planktivorous fish of Australian reservoirs are relatively small and they may deplete small rather than large zooplankters, so that animals as large as the adults of Daphnia carinata may avoid predation. This would result in promotion of Daphnia, particularly if fish eliminate its smaller competitors. The aim of biomanipulation should be the establishment and maintenance of a proper ratio of planktivores/piscivores, adequate for water quality requirements. Successful selection of the appropriate ratio for a given reservoir will depend on the extent of our understanding of its food web interactions. For practical application of biomanipulation in management, further development of the food web theory under Australian conditions is needed.

[ES]En este trabajo se hace un análisis y una comparación de las diferentes formas estructurales que puede adoptar una marquesina de aparcamiento, principalmente estructuras hechas en madera y acero. Para la realización del citado análisis se han valorado las ventajas y desventajas de los materiales, funcionalidad, mantenimiento, estética, dificultad de ejecución y economicidad, realizándose asimismo un diseño de marquesina utilizando uno u otro de los materiales citados. Para la ejecución de la marquesina, se formula un diseño que cumplimente los condicionantes de funcionalidad del elemento. A su vez, se analiza, parte por parte, todo el sistema estructural, empezando por determinar las acciones que van a aparecer y deberán ser transmitidas al terreno. Una vez obtenidos los esfuerzos que se han de transmitir, se formula una hipótesis de calidad del terreno de cimentación, con el fin de determinar las características de la cimentación. Previamente se realiza un predimensionado del sistema estructural, para cuyo cálculo se ha utilizado el programa informático Tricalc. Obtenidas las dimensiones exactas y los detalles constructivos para cada una de las opciones de materiales a analizar (madera y acero), se evalúa económicamente cada diseño obtenido, para determinar la solución más económica y rentable desde el punto de vista, por ejemplo del montaje y desmontaje, así como por el mantenimiento. Además de las funciones estructurales, también se analiza la adaptación al entorno de la madera y el acero.

A indústria do petróleo é um dos setores com maior número de sistemas produtivos empregando alta tecnologia. O Brasil é mundialmente renomado como um líder na extração de petróleo, em águas profundas e ultraprofundas. Dentro da cadeia produtiva, grande parte do petróleo e do gás produzido é escoado através de dutos flexíveis que conectam os poços de produção com as plataformas. Existem dois segmentos dessas linhas que recebem diferentes denominações de acordo com o seu local de aplicação. Quando estão apoiadas sobre o fundo do mar, em condição de serviço estático, são denominados flowlines e quando se elevam do fundo do mar até a plataforma, em condição de serviço dinâmico, são denominados risers. Os tubos projetados para aplicações dinâmicas são dotados de bends stiffeners, componentes com formato cônico e, em geral, de base uretânica que têm a função de fornecer uma transição de rigidez suave entre a estrutura dos tubos flexíveis e a extremamente rígida, à plataforma, não permitindo que este componente infrinja seu raio mínimo de operação. A adequada compreensão dos enrijecedores de curvatura e do material empregado em sua fabricação vem se tornando cada vez mais importante na indústria devido à sua crescente utilização, bem como à ocorrência de falhas que vem sendo constatada nos últimos anos. Este trabalho abordou a variação das propriedades mecânicas de poliuretanos pela ação da hidrólise, calor e pela ação dos raios-UV por envelhecimento acelerado, assim como variação de massa, considerando que esses materiais são projetados para uma vida útil superior a vinte anos para trabalhos imersos em meio aquoso.

X-ray phase imaging with illumination by a partially coherent source with a setup similar to in-line holography is considered. Using the optical transform function, we consider the effects of partial coherence on this x-ray phase imaging for a weak phase object. The optimal contrast and the resolution of phase imaging are analyzed. As the coherence decreases, the imaging contrast and the optimal contrast frequency decrease, and the resolution degrades. It is shown that this contrast-enhanced phase-imaging method can be regarded as a linear bandpass filter and that the bandwidth and the image contrast are changeable. The frequency property of the imaging system can be improved if an incoherent x-ray source with the proper shape is used. (C) 1999 Optical Society of America.

The σD values of nitrated cellulose from a variety of trees covering a wide geographic range have been measured. These measurements have been used to ascertain which factors are likely to cause σD variations in cellulose C-H hydrogen.
It is found that a primary source of tree σD variation is the σD variation of the environmental precipitation. Superimposed on this are isotopic variations caused by the transpiration of the leaf water incorporated by the tree. The magnitude of this transpiration effect appears to be related to relative humidity.
Within a single tree, it is found that the hydrogen isotope variations which occur for a ring sequence in one radial direction may not be exactly the same as those which occur in a different direction. Such heterogeneities appear most likely to occur in trees with asymmetric ring patterns that contain reaction wood. In the absence of reaction wood such heterogeneities do not seem to occur. Thus, hydrogen isotope analyses of tree ring sequences should be performed on trees which do not contain reaction wood.
Comparisons of tree σD variations with variations in local climate are performed on two levels: spatial and temporal. It is found that the σD values of 20 North American trees from a wide geographic range are reasonably well-correlated with the corresponding average annual temperature. The correlation is similar to that observed for a comparison of the σD values of annual precipitation of 11 North American sites with annual temperature. However, it appears that this correlation is significantly disrupted by trees which grew on poorly drained sites such as those in stagnant marshes. Therefore, site selection may be important in choosing trees for climatic interpretation of σD values, although proper sites do not seem to be uncommon.
The measurement of σD values in 5-year samples from the tree ring sequences of 13 trees from 11 North American sites reveals a variety of relationships with local climate. As it was for the spatial σD vs climate comparison, site selection is also apparently important for temporal tree σD vs climate comparisons. Again, it seems that poorly-drained sites are to be avoided. For nine trees from different "well-behaved" sites, it was found that the local climatic variable best related to the σD variations was not the same for all sites.
Two of these trees showed a strong negative correlation with the amount of local summer precipitation. Consideration of factors likely to influence the isotopic composition of summer rain suggests that rainfall intensity may be important. The higher the intensity, the lower the σD value. Such an effect might explain the negative correlation of σD vs summer precipitation amount for these two trees. A third tree also exhibited a strong correlation with summer climate, but in this instance it was a positive correlation of σD with summer temperature.
The remaining six trees exhibited the best correlation between σD values and local annual climate. However, in none of these six cases was it annual temperature that was the most important variable. In fact annual temperature commonly showed no relationship at all with tree σD values. Instead, it was found that a simple mass balance model incorporating two basic assumptions yielded parameters which produced the best relationships with tree σD values. First, it was assumed that the σD values of these six trees reflected the σD values of annual precipitation incorporated by these trees. Second, it was assumed that the σD value of the annual precipitation was a weighted average of two seasonal isotopic components: summer and winter. Mass balance equations derived from these assumptions yielded combinations of variables that commonly showed a relationship with tree σD values where none had previously been discerned.
It was found for these "well-behaved" trees that not all sample intervals in a σD vs local climate plot fell along a well-defined trend. These departures from the local σD VS climate norm were defined as "anomalous". Some of these anomalous intervals were common to trees from different locales. When such widespread commonalty of an anomalous interval occurred, it was observed that the interval corresponded to an interval in which drought had existed in the North American Great Plains.
Consequently, there appears to be a combination of both local and large scale climatic information in the σD variations of tree cellulose C-H hydrogen.

With one weak probe field and two strong pumping fields, superluminal optical solitons are formed in a lifetime-broadened four-level tripod atomic medium. With proper parameters, both dark and bright solitons can occur in the highly resonant medium. The corresponding group velocity of the solitons can be superluminal. Meanwhile, the conditions for superluminal solitons occurrence are given.

We present an experimental scheme of a cold atom space clock with a movable cavity. By using a single microwave cavity, we find that the clock has a significant advantage, i.e. the longitudinal cavity phase shift is eliminated. A theoretical analysis has been carried out in terms of the relation between the atomic transition probability and the velocity of the moving cavity by taking into account the velocity distribution of cold atoms. The requirements for the microwave power and its stability for atomic pi/2 excitation at different moving velocities of the cavity lead to the determination of the proper working parameters of the rubidium clock in frequency accuracy 10(-17). Finally, the mechanical stability for the scheme is analysed and the ways of solving the possible mechanical instability of the device are proposed.

With one weak probe field and two strong pumping fields, the possibility of producing superluminal optical solitons is discussed in a lifetime-broadened inverted-Y atomic medium with proper parameters. As the group velocity of the solitons is larger than c, its occurrence can be controlled by modulating the intensities and the detunings of lasers.

Interleukin 2 (IL2) is the primary growth hormone used by mature T cells and this lymphokine plays an important role in the magnification of cell-mediated immune responses. Under normal circumstances its expression is limited to antigen-activated type 1 helper T cells (TH1) and the ability to transcribe this gene is often regarded as evidence for commitment to this developmental lineage. There is, however, abundant evidence than many non-TH1 T cells, under appropriate conditions, possess the ability to express this gene. Of paramount interest in the study of T-cell development is the mechanisms by which differentiating thymocytes are endowed with particular combinations of cell surface proteins and response repertoires. For example, why do most helper T cells express the CD4 differentiation antigen?
As a first step in understanding these developmental processes the gene encoding IL2 was isolated from a mouse genomic library by probing with a conspecific IL2 cDNA. The sequence of the 5' flanking region from + 1 to -2800 was determined and compared to the previously reported human sequence. Extensive identity exists between +1 and -580 (86%) and sites previously shown to be crucial for the proper expression of the human gene are well conserved in both sequence location in the mouse counterpart.
Transient expression assays were used to evaluate the contribution of various genomic sequences to high-level gene expression mediated by a cloned IL2 promoter fragment. Differing lengths of 5' flanking DNA, all terminating in the 5' untranslated region, were linked to a reporter gene, bacterial chloramphenicol acetyltransferase (CAT) and enzyme activity was measured after introduction into IL2-producing cell lines. No CAT was ever detected without stimulation of the recipient cells. A cloned promoter fragment containing only 321 bp of upstream DNA was expressed well in both Jurkat and EL4.El cells. Addition of intragenic or downstream DNA to these 5' IL2-CAT constructs showed that no obvious regulatory regions resided there. However, increasing the extent of 5' DNA from -321 to -2800 revealed several positive and negative regulatory elements. One negative region that was well characterized resided between -750 and -1000 and consisted almost exclusively of alternating purine and pyrimidines. There is no sequence resembling this in the human gene now, but there is evidence that there may have once been.
No region, when deleted, could relax either the stringent induction-dependence on cell-type specificity displayed by this promoter. Reagents that modulated endogenous IL2 expression, such as cAMP, cyclosporin A, and IL1, affected expression of the 5' IL2-CAT constructs also. For a given reagent, expression from all expressible constructs was suppressed or enhanced to the same extent. This suggests that these modulators affect IL2 expression through perturbation of a central inductive signal rather than by summation of the effects of discrete, independently regulated, negative and positive transcription factors.

The molecular inputs necessary for cell behavior are vital to our understanding of development and disease. Proper cell behavior is necessary for processes ranging from creating one’s face (neural crest migration) to spreading cancer from one tissue to another (invasive metastatic cancers). Identifying the genes and tissues involved in cell behavior not only increases our understanding of biology but also has the potential to create targeted therapies in diseases hallmarked by aberrant cell behavior.
A well-characterized model system is key to determining the molecular and spatial inputs necessary for cell behavior. In this work I present the C. elegans uterine seam cell (utse) as an ideal model for studying cell outgrowth and shape change. The utse is an H-shaped cell within the hermaphrodite uterus that functions in attaching the uterus to the body wall. Over L4 larval stage, the utse grows bidirectionally along the anterior-posterior axis, changing from an ellipsoidal shape to an elongated H-shape. Spatially, the utse requires the presence of the uterine toroid cells, sex muscles, and the anchor cell nucleus in order to properly grow outward. Several gene families are involved in utse development, including Trio, Nav, Rab GTPases, Arp2/3, as well as 54 other genes found from a candidate RNAi screen. The utse can be used as a model system for studying metastatic cancer. Meprin proteases are involved in promoting invasiveness of metastatic cancers and the meprin-likw genes nas-21, nas-22, and toh-1 act similarly within the utse. Studying nas-21 activity has also led to the discovery of novel upstream inhibitors and activators as well as targets of nas-21, some of which have been characterized to affect meprin activity. This illustrates that the utse can be used as an in vivo model for learning more about meprins, as well as various other proteins involved in metastasis.

This thesis consists of two independent chapters. The first chapter deals with universal algebra. It is shown, in von Neumann-Bernays-Gӧdel set theory, that free images of partial algebras exist in arbitrary varieties. It follows from this, as set-complete Boolean algebras form a variety, that there exist free set-complete Boolean algebras on any class of generators. This appears to contradict a well-known result of A. Hales and H. Gaifman, stating that there is no complete Boolean algebra on any infinite set of generators. However, it does not, as the algebras constructed in this chapter are allowed to be proper classes. The second chapter deals with positive elementary inductions. It is shown that, in any reasonable structure ᶆ, the inductive closure ordinal of ᶆ is admissible, by showing it is equal to an ordinal measuring the saturation of ᶆ. This is also used to show that non-recursively saturated models of the theories ACF, RCF, and DCF have inductive closure ordinals greater than ω.
