Abstract: In order to understand better the pathological aspects and spread of Pasteurella multocida type A as the primary cause of pneumonia in pigs, was made an experiment with intranasal inoculation of different concentrations of inocula [Group (G1): 108 Colony Forming Units (CFU)/ml; G2: 107 CFU/ml; G3: 106 CFU/ml and G4: 105 CFU/ml], using two pigs per group. The pigs were obtained from a high health status herd. Pigs were monitored clinically for 4 days and subsequently necropsied. All pigs had clinical signs and lesions associated with respiratory disease. Dyspnoea and hyperthermia were the main clinical signs observed. Suppurative cranioventral bronchopneumonia, in some cases associated with necrosuppurative pleuropneumonia, fibrinous pericarditis and pleuritic, were the most frequent types of lesion found. The disease evolved with septicaemia, characterized by septic infarctions in the liver and spleen, with the detection of P. multocida type A. In this study, P. multocida type A strain #11246 was the primary agent of fibrinous pleuritis and suppurative cranioventral bronchopneumonia, pericarditis and septicaemia in the pigs. All concentrations of inoculum used (105-108 CFU/ml) were able to produce clinical and pathological changes of pneumonia, pleuritis, pericarditis and septicemia in challenged animals.

A região do trópico úmido amazônico ocupa 350 milhões de hectares, constituindo a maior área de fronteira agrícola do planeta. A pecuária como exploração do uso da terra é a principal forma de ocupação e de desmatamento na região, apesar da baixa produtividade e das conseqüências ambientais. O declínio da produtividade da pastagem está associado, entre outras, ao manejo inadequado e à elevada infestação de plantas daninhas herbáceas e arbustivas, anuais ou perenes, geralmente denominadas "juquira" na região. Se essas plantas não forem controladas adequadamente, podem levar à degradação da pastagem, resultando na completa perda de produtividade e no posterior abandono da área. Com o objetivo de identificar as plantas daninhas mais importantes que ocorrem em área de pastagem de baixa produtividade no município de Terra Alta, PA (0º 58" S e 47º 52" W. Gr.), foi efetuado um levantamento florístico em 0,33 ha de área cultivada com capim-quicuio-da-amazônia (Brachiaria humidicola). Foram registrados 4.700 indivíduos, distribuídos em 17 famílias botânicas, representadas por 36 espécies vegetais. Destacaram-se as famílias Poaceae, Rubiaceae, Fabaceae, Asteraceae e Solanaceae, sendo encontrado em cada uma delas cinco, cinco, quatro, três e três espécies, respectivamente. As plantas daninhas consideradas de maior importância na comunidade vegetal foram: Borreria verticillata, Hyptis atrorubens, Rolandra argentea, Desmodium canum, Panicum pilosum, Davilla rugosa, Imperata brasiliensis, Paspalum maritimum, Vernonia scorpioides e Vismia guianensis.

A integração de certas práticas de manejo ao controle químico de plantas daninhas pode permitir a redução da dose do herbicida a ser aplicado, sem alterar significativamente os níveis de controle das plantas daninhas e rendimento de grãos de soja. Dessa forma, o objetivo do presente trabalho foi de avaliar a redução da dose recomendada da mistura formulada dos herbicidas fluazifop-p-butyl + fomesafen aplicados em condições de pós-emergência das palntas daninhas, associada à variação no espaçamento entre linhas da cultura de soja, sobre o controle de papuã (Brachiaria plantaginea) e o rendimento final de grãos da cultura. O experimento foi realizado na EEA/UFRGS, em Eldorado do Sul-RS, na estação de crescimento de 1997/98. O cultivar de soja testado foi o BR 16, em semeadura direta. Como tratamentos, utilizaram-se três espaçamentos entre linhas (20, 40 e 60 cm) e cinco doses do herbicida [fluazifop-p-butyl (200 g L-1) + fomesafen (250 g L-1)], aplicado na dose recomendada comercialmente de 1,0 L ha-1 (100%) e com reduções da dose (75, 50 e 25% da dose recomendada), mais a testemunha. Avaliou-se visualmente o controle de papuã aos 8 e 17 dias após as aplicações do herbicida, bem como a fitomassa seca de papuã e o rendimento de grãos da cultura. Os resultados evidenciaram que, para infestação moderada de papuã, o espaçamento entre linhas não tem efeito sobre o seu controle quando o herbicida é utilizado na dose recomendada. Para aplicações tardias, o espaçamento de 40 cm permite reduzir a dose do herbicida em até 50% sem afetar o controle de papuã e em 75% sem diminuir o rendimento de grãos da soja. A redução na dose do herbicida está condicionada à época de aplicação e ao espaçamento entre linhas; com espaçamentos reduzidos, é importante que as aplicações sejam feitas precocemente, a fim de garantir que quantidade suficiente de produto atinja as plantas daninhas.

A remoção de plantas aquáticas tem sido utilizada como opção ao controle químico e biológico, em razão de restrições ambientais em algumas regiões brasileiras. O objetivo deste trabalho foi desenvolver um modelo para análise econômica e operacional da remoção mecânica de plantas aquáticas, visando realizar estudo econômico comparativo com o controle químico. A operação foi estudada num reservatório de uma usina de bombeamento em Barra do Piraí-RJ. O sistema consiste de retroescavadeiras instaladas em balsas, usadas para cortar as plantas e liberá-las no fluxo de água. Antes da tomada d'água existe uma barreira flutuante que intercepta as plantas, as quais são removidas por um guindaste fixo nas margens. As plantas são armazenadas por algum tempo e depois descartadas. Existe, ainda, um sistema de limpeza das grades da tomada d'água. Dados do volume total de plantas descartadas foram coletados durante 14 meses, assim como foi avaliado o volume de biomassa produzido por área das principais espécies infestantes. A empreiteira que administra o serviço forneceu planilhas de custos e outro parâmetros operacionais. Um modelo foi desenvolvido para calcular custos por hectare de plantas removidas. Os resultados mostraram custo médio mensal de US$ 17.780,28 por hectare. Apesar do alto custo, o sistema de remoção demonstrou capacidade de controlar apenas 4,1% da área infestada no reservatório, na época da coleta dos dados. Simulando dados de uma aplicação de glyphosate, o controle químico custaria apenas 0,23% do custo da remoção. Análises de sensibilidade mostraram que a compactação das plantas para transporte, o volume de plantas produzidas por área e o custo do transporte são os parâmetros principais para a otimização.

O objetivo deste trabalho foi avaliar os efeitos da palha de cana-de-açúcar resultante da colheita sem queima e da mistura comercial dos herbicidas trifloxysulfuron sodium + ametrina, na emergência e no acúmulo de biomassa seca de nove espécies de plantas daninhas. Os tratamentos foram distribuídos em esquema fatorial 3 x 3, sendo três quantidades de palha resultantes da colheita da cana-de-açúcar sem queima (0, 10 e 15 t ha-1), e três doses da mistura formulada dos herbicidas trifloxysulfuron sodium + ametrina (0, 1,75 e 2,00 kg ha-1), aplicados em condições de pós-emergência das plantas daninhas. As densidades das plantas daninhas foram avaliadas aos 15, 60 e 90 dias após a aplicação (DAA) da mistura dos herbicidas, e a biomassa seca das plantas daninhas, aos 90 DAA. A presença de palha sobre o solo suprimiu as densidades das populações de Brachiaria plantaginea, Digitaria horizontalis, Panicum maximum, Sida glaziovii e Amaranthus hybridus a níveis de infestação considerados satisfatórios de controle. A palhada também reduziu as populações de Senna obtusifolia, Ipomoea hederifolia, I. grandifolia e I. nil, porém em níveis insatisfatórios de controle. A mistura de herbicidas nas doses estudadas controlou todas as espécies de plantas daninhas avaliadas. Na presença da palha, o controle das plantas de S. obtusifolia, I. nil, I. hederifolia e I. grandifolia somente foi satisfatório quando foi aplicada a mistura de herbicidas. Para o controle de P. maximum, houve vantagem na integração dos dois métodos de controle, em relação à aplicação isolada da menor quantidade de palha estudada ou à aplicação da mistura de herbicidas.

Os resíduos vegetais de uma cultura de cobertura de outono/inverno podem interferir na infestação das plantas daninhas das culturas de verão subseqüentes. Dessa forma, com o objetivo de avaliar o efeito da palha de híbridos de sorgo (Sorghum bicolor), associada ao uso do herbicida imazamox, no controle de plantas daninhas na cultura da soja (Glycine max), cv. 'Conquista', em sucessão, foi conduzido no ano agrícola 2000/2001 um experimento em Uberlândia-MG. Foram utilizados quatro tipos de cobertura: três provenientes de resíduos culturais de híbridos de sorgo (Sara, DKB 860 e Ambar) e uma sem restos vegetais (anteriormente sob pousio). Aos 24 dias após aplicação do herbicida, o controle das espécies Leonotis nepetifolia, Alternanthera tenella, Amaranthus hibridus, A. retroflexus, A. spinosus, Ipomoea grandifolia, Commelina benghalensis e Nicandra physaloides foi mais eficaz nas palhas dos híbridos Sara e Ambar, na ausência de imazamox, com porcentagens de controle de 40 e 41%, respectivamente. Quando associada a 15 g ha-1 de imazamox, a palha do Ambar resultou em melhor controle dessas espécies de plantas daninhas, com controle de 76%; e a 30 g ha-1 a palha do DKB 860 foi a mais eficaz, promovendo 85% de controle. Sem cobertura do solo, com 30 g ha-1 de imazamox obteve-se controle de 62,5%, e 47,5% com metade da dosagem. Os resultados indicaram variabilidade de controle em relação ao híbrido de sorgo estudado e a possibilidade de redução de dosagens do herbicida imazamox quando associado aos resíduos vegetais de sorgo.

Com o objetivo de colaborar na delimitação taxonômica de Aechmea, subgênero Lamprococcus, foram estudadas as folhas de 25 táxons morfologicamente relacionados entre si (13 de Aechmea, três de Ronnbergia E. Morren & André, quatro de Araeococcus Brongn. e cinco de Lymania Read). Apesar das folhas serem todas hipoestomáticas e possuírem mesofilo dorsiventral portador de hipoderme, elas exibem características que podem ser utilizadas na delimitação de cada representante. Essas características incluem: posição dos estômatos na superfície foliar, presença ou não de oclusão nas câmaras subestomáticas, ocorrência ou não de grupos de fibras dispersos no mesofilo, natureza da parede das células que compõem as bainhas e extensões dos feixes vasculares e morfologia das células dos diafragmas, que interrompem os canais de aeração. A distribuição desses caracteres, nos diferentes táxons, permitiu elaborar uma chave de identificação, com base apenas na estrutura foliar.

The purpose of the present study was to validate the quantitative culture and cellularity of bronchoalveolar lavage (BAL) for the diagnosis of ventilator-associated pneumonia (VAP). A prospective validation test trial was carried out between 1992 and 1997 in a general adult intensive care unit of a teaching hospital. Thirty-seven patients on mechanical ventilation with suspected VAP who died at most three days after a BAL diagnostic procedure were submitted to a postmortem lung biopsy. BAL effluent was submitted to Gram staining, quantitative culture and cellularity count. Postmortem lung tissue quantitative culture and histopathological findings were considered to be the gold standard exams for VAP diagnosis. According to these criteria, 20 patients (54%) were diagnosed as having VAP and 17 (46%) as not having the condition. Quantitative culture of BAL effluent showed 90% sensitivity (18/20), 94.1% specificity (16/17), 94.7% positive predictive value and 88.8% negative predictive value. Fever and leukocytosis were useless for VAP diagnosis. Gram staining of BAL effluent was negative in 94.1% of the patients without VAP (16/17). Regarding the total cellularity of BAL, a cut-off point of 400,000 cells/ml showed a specificity of 94.1% (16/17), and a cut-off point of 50% of BAL neutrophils showed a sensitivity of 90% (19/20). In conclusion, BAL quantitative culture, Gram staining and cellularity might be useful in the diagnostic investigation of VAP.

The present study compares the performance of stochastic and fuzzy models for the analysis of the relationship between clinical signs and diagnosis. Data obtained for 153 children concerning diagnosis (pneumonia, other non-pneumonia diseases, absence of disease) and seven clinical signs were divided into two samples, one for analysis and other for validation. The former was used to derive relations by multi-discriminant analysis (MDA) and by fuzzy max-min compositions (fuzzy), and the latter was used to assess the predictions drawn from each type of relation. MDA and fuzzy were closely similar in terms of prediction, with correct allocation of 75.7 to 78.3% of patients in the validation sample, and displaying only a single instance of disagreement: a patient with low level of toxemia was mistaken as not diseased by MDA and correctly taken as somehow ill by fuzzy. Concerning relations, each method provided different information, each revealing different aspects of the relations between clinical signs and diagnoses. Both methods agreed on pointing X-ray, dyspnea, and auscultation as better related with pneumonia, but only fuzzy was able to detect relations of heart rate, body temperature, toxemia and respiratory rate with pneumonia. Moreover, only fuzzy was able to detect a relationship between heart rate and absence of disease, which allowed the detection of six malnourished children whose diagnoses as healthy are, indeed, disputable. The conclusion is that even though fuzzy sets theory might not improve prediction, it certainly does enhance clinical knowledge since it detects relationships not visible to stochastic models.

Because the superficial lymphatics in the lungs are distributed in the subpleural, interlobular and peribroncovascular interstitium, lymphatic impairment may occur in the lungs of patients with idiopathic interstitial pneumonias (IIPs) and increase their severity. We investigated the distribution of lymphatics in different remodeling stages of IIPs by immunohistochemistry using the D2-40 antibody. Pulmonary tissue was obtained from 69 patients with acute interstitial pneumonia/diffuse alveolar damage (AIP/DAD, N = 24), cryptogenic organizing pneumonia/organizing pneumonia (COP/OP, N = 6), nonspecific interstitial pneumonia (NSIP/NSIP, N = 20), and idiopathic pulmonary fibrosis/usual interstitial pneumonia (IPF/UIP, N = 19). D2-40+ lymphatic in the lesions was quantitatively determined and associated with remodeling stage score. We observed an increase in the D2-40+ percent from DAD (6.66 ± 1.11) to UIP (23.45 ± 5.24, P = 0.008) with the advanced process of remodeling stage of the lesions. Kaplan-Meier survival curves showed a better survival for patients with higher lymphatic D2-40+ expression than 9.3%. Lymphatic impairment occurs in the lungs of IIPs and its severity increases according to remodeling stage. The results suggest that disruption of the superficial lymphatics may impair alveolar clearance, delay organ repair and cause severe disease progress mainly in patients with AIP/DAD. Therefore, lymphatic distribution may serve as a surrogate marker for the identification of patients at greatest risk for death due to IIPs.

Ventilator-associated pneumonia (VAP) remains one of the major causes of infection in the intensive care unit (ICU) and is associated with the length of hospital stay, duration of mechanical ventilation, and use of broad-spectrum antibiotics. We compared the frequency of VAP 10 months prior to (pre-intervention group) and 13 months after (post-intervention group) initiation of the use of a heat and moisture exchanger (HME) filter. This is a study with prospective before-and-after design performed in the ICU in a tertiary university hospital. Three hundred and fourteen patients were admitted to the ICU under mechanical ventilation, 168 of whom were included in group HH (heated humidifier) and 146 in group HME. The frequency of VAP per 1000 ventilator-days was similar for both the HH and HME groups (18.7 vs 17.4, respectively; P = 0.97). Duration of mechanical ventilation (11 vs 12 days, respectively; P = 0.48) and length of ICU stay (11 vs 12 days, respectively; P = 0.39) did not differ between the HH and HME groups. The chance of developing VAP was higher in patients with a longer ICU stay and longer duration of mechanical ventilation. This finding was similar when adjusted for the use of HME. The use of HME in intensive care did not reduce the incidence of VAP, the duration of mechanical ventilation, or the length of stay in the ICU in the study population.

Idiopathic interstitial pneumonias include complex diseases that have a strong interaction between genetic makeup and environmental factors. However, in many cases, no infectious agent can be demonstrated, and these clinical diseases rapidly progress to death. Theoretically, idiopathic interstitial pneumonias could be caused by the Epstein-Barr virus, cytomegalovirus, adenovirus, hepatitis C virus, respiratory syncytial virus, and herpesvirus, which may be present in such small amounts or such configuration that routine histopathological analysis or viral culture techniques cannot detect them. To test the hypothesis that immunohistochemistry provides more accurate results than the mere histological demonstration of viral inclusions, this method was applied to 37 open lung biopsies obtained from patients with idiopathic interstitial pneumonias. As a result, immunohistochemistry detected measles virus and cytomegalovirus in diffuse alveolar damage-related histological patterns of acute exacerbation of idiopathic pulmonary fibrosis and nonspecific interstitial pneumonia in 38 and 10% of the cases, respectively. Alveolar epithelium infection by cytomegalovirus was observed in 25% of organizing pneumonia patterns. These findings were coincident with nuclear cytopathic effects but without demonstration of cytomegalovirus inclusions. These data indicate that diffuse alveolar damage-related cytomegalovirus or measles virus infections enhance lung injury, and a direct involvement of these viruses in diffuse alveolar damage-related histological patterns is likely. Immunohistochemistry was more sensitive than the histological demonstration of cytomegalovirus or measles virus inclusions. We concluded that all patients with diffuse alveolar damage-related histological patterns should be investigated for cytomegalovirus and measles virus using sensitive immunohistochemistry in conjunction with routine procedures.

Limitations on tissue proliferation capacity determined by telomerase/apoptosis balance have been implicated in pathogenesis of idiopathic pulmonary fibrosis. In addition, collagen V shows promise as an inductor of apoptosis. We evaluated the quantitative relationship between the telomerase/apoptosis index, collagen V synthesis, and epithelial/fibroblast replication in mice exposed to butylated hydroxytoluene (BHT) at high oxygen concentration. Two groups of mice were analyzed: 20 mice received BHT, and 10 control mice received corn oil. Telomerase expression, apoptosis, collagen I, III, and V fibers, and hydroxyproline were evaluated by immunohistochemistry, in situ detection of apoptosis, electron microscopy, immunofluorescence, and histomorphometry. Electron microscopy confirmed the presence of increased alveolar epithelial cells type 1 (AEC1) in apoptosis. Immunostaining showed increased nuclear expression of telomerase in AEC type 2 (AEC2) between normal and chronic scarring areas of usual interstitial pneumonia (UIP). Control lungs and normal areas from UIP lungs showed weak green birefringence of type I and III collagens in the alveolar wall and type V collagen in the basement membrane of alveolar capillaries. The increase in collagen V was greater than collagens I and III in scarring areas of UIP. A significant direct association was found between collagen V and AEC2 apoptosis. We concluded that telomerase, collagen V fiber density, and apoptosis evaluation in experimental UIP offers the potential to control reepithelization of alveolar septa and fibroblast proliferation. Strategies aimed at preventing high rates of collagen V synthesis, or local responses to high rates of cell apoptosis, may have a significant impact in pulmonary fibrosis.

Exposure to air pollutants is associated with hospitalizations due to pneumonia in children. We hypothesized the length of hospitalization due to pneumonia may be dependent on air pollutant concentrations. Therefore, we built a computational model using fuzzy logic tools to predict the mean time of hospitalization due to pneumonia in children living in São José dos Campos, SP, Brazil. The model was built with four inputs related to pollutant concentrations and effective temperature, and the output was related to the mean length of hospitalization. Each input had two membership functions and the output had four membership functions, generating 16 rules. The model was validated against real data, and a receiver operating characteristic (ROC) curve was constructed to evaluate model performance. The values predicted by the model were significantly correlated with real data. Sulfur dioxide and particulate matter significantly predicted the mean length of hospitalization in lags 0, 1, and 2. This model can contribute to the care provided to children with pneumonia.

Community-acquired pneumonia (CAP) is amongst the leading causes of death worldwide. As inflammatory markers, cytokines can predict outcomes, if interpreted together with clinical data and scoring systems such as CURB-65, CRB, and Acute Physiology and Chronic Health Evaluation II (APACHE II). The aim of this study was to determine the impact of inflammatory biomarkers on the early mortality of hospitalized CAP patients. Twenty-seven CAP patients needing hospitalization were enrolled for the study and samples of interleukin-1 (IL-1) and interleukin-6 (IL-6), tumor necrosis factor alpha (TNF-α), C-reactive protein (CRP), and homocystein were collected at the time of admission (day 1) as well as on the seventh day of the treatment. There was a significant reduction in the levels of IL-6 between the first and the second collections. Median IL-6 values decreased from 24 pg/mL (day 1) to 8 pg/mL (day 7) (P=0.016). The median levels of TNF-α were higher in patients: i) with acute kidney injury (AKI) (P=0.045), ii) requiring mechanical ventilation (P=0.040), iii) with short hospital stays (P=0.009), iv) admitted to the intensive care unit (ICU) (P=0.040), v) who died early (P=0.003), and vi) with worse CRB scores (P=0.013). In summary, IL-6 and TNF-α levels were associated with early mortality of CAP patients. Longer admission levels demonstrated greater likelihood of early death and overall mortality, necessity of mechanical ventilation, and AKI.
