The plant Crassula helmsii (Kirk) Cochayne, was likely to become widely distributed and to dominate many damp and wet areas of nature reserves, recreational waters and agricultural drainage of Britain. The aim of this report was to study Australian Swamp Stonecrop in its natural habitat where it is in balance with its environment. This contrasts with its rapid and widespread distribution in the U.K. where its growth interferes with the use of fisheries and amenity lakes but also reduces the value of nature reserves and sites of special scientific interest by suppressing native flora. It was proposed to observe its growth at a variety of sites over its natural distribution and to include some environmental factors, e.g. water-level, water-chemistry (nutrients, acidity and alkalinity), frost-tolerance, salinity, with the help of portable sensors, locally-available services or data. 8 weeks of travel in Australia allowed time to study the plant in its natural habitat including the coastal areas of the southern half of the continent i.e . Western Australia, South Australia, New South Wales, Victoria, Tasmania and southern Queensland. The overall objective was to determine the environmental range by visits to selected sites of Crassula helmsii over its geographic range.

I. The binding of the intercalating dye ethidium bromide to closed circular SV 40 DNA causes an unwinding of the duplex structure and a simultaneous and quantitatively equivalent unwinding of the superhelices. The buoyant densities and sedimentation velocities of both intact (I) and singly nicked (II) SV 40 DNAs were measured as a function of free dye concentration. The buoyant density data were used to determine the binding isotherms over a dye concentration range extending from 0 to 600 µg/m1 in 5.8 M CsCl. At high dye concentrations all of the binding sites in II, but not in I, are saturated. At free dye concentrations less than 5.4 µg/ml, I has a greater affinity for dye than II. At a critical amount of dye bound I and II have equal affinities, and at higher dye concentration I has a lower affinity than II. The number of superhelical turns, τ, present in I is calculated at each dye concentration using Fuller and Waring's (1964) estimate of the angle of duplex unwinding per intercalation. The results reveal that SV 40 DNA I contains about -13 superhelical turns in concentrated salt solutions.
The free energy of superhelix formation is calculated as a function of τ from a consideration of the effect of the superhelical turns upon the binding isotherm of ethidium bromide to SV 40 DNA I. The value of the free energy is about 100 kcal/mole DNA in the native molecule. The free energy estimates are used to calculate the pitch and radius of the superhelix as a function of the number of superhelical turns. The pitch and radius of the native I superhelix are 430 Å and 135 Å, respectively.
A buoyant density method for the isolation and detection of closed circular DNA is described. The method is based upon the reduced binding of the intercalating dye, ethidium bromide, by closed circular DNA. In an application of this method it is found that HeLa cells contain in addition to closed circular mitochondrial DNA of mean length 4.81 microns, a heterogeneous group of smaller DNA molecules which vary in size from 0.2 to 3.5 microns and a paucidisperse group of multiples of the mitochondrial length.
II. The general theory is presented for the sedimentation equilibrium of a macromolecule in a concentrated binary solvent in the presence of an additional reacting small molecule. Equations are derived for the calculation of the buoyant density of the complex and for the determination of the binding isotherm of the reagent to the macrospecies. The standard buoyant density, a thermodynamic function, is defined and the density gradients which characterize the four component system are derived. The theory is applied to the specific cases of the binding of ethidium bromide to SV 40 DNA and of the binding of mercury and silver to DNA.

The biomass of the phytoplankton and its composition is one of the most important factors in water quality control. Determination of the phytoplankton assemblage is usually done by microscopic analysis (Utermöhl's method). Quantitative estimations of the biovolume, by cell counting and cell size measurements, are time-consuming and normally are not done in routine water quality control. Several alternatives have been tried: computer-based image analysis, spectral fluorescence signatures, flow cytometry and pigment fingerprinting aided by high performance liquid chromatography (HPLC). The latter method is based on the fact that each major algal group of taxa contains a specific carotenoid which can be used for identification and relative quantification of the taxa in the total assemblage. This article gives a brief comparative introduction to the different techniques available and presents some recent results obtained by HPLC-based pigment fingerprinting, applied to three lakes of different trophic status. The results show that this technique yields reliable results from different lake types and is a powerful tool for studying the distribution pattern of the phytoplankton community in relation to water depth. However, some restrictions should be taken into account for the interpretation of routine data.

IDOKI SCF Technologies S.L. is a technology-based company, set up on September 2006 in Derio (Biscay) with the main scope of developing extraction and purification processes based on the use of supercritical fluid extraction technology (SFE) in food processing, extraction of natural products and the production of personal care products. IDOKI¿s researchers have been working on many different R&D projects so far, most of them using this technology. However, the optimization of a SFE method for the different matrices cannot be performed unless we have an analytical method for the characterisation of the extracts obtained in each experiment. The analytical methods are also essential for the quality control of the raw materials that are going to be used and also for the final product. This PhD thesis was born to tackle this problem and therefore, it is based on the development of different analytical methods for the characterisation of the extracts and products. The projects that we could include in this thesis were the following: the extraction propolis, the recovery of agroindustrial residues (soy and wine) and the dealcoholisation of wine.On the one hand, for the extraction of propolis, several UV-Vis spectroscopic methods were used in order to measure the antioxidant capacity and the total polyphenol and flavonoid content of the extracts. A SFC method was also developed in order to measure more specific phenolic compounds. On the other hand, for the recovery of agroindustrial residues UV-Vis spectroscopy was used to determine the total polyphenol content and two SFC methods were developed to analyse different phenolic compounds. Extraction methods such as MAE, FUSE and rotary agitation were also evaluated for the characterisation of the raw materials.Finally, for the dealcoholisation of wine, the development of a SBSE-TD-GC-MS and DHS-TD-GC-MS methods for the analysis of aromas and a NIR spectroscopic method for the determination of ethanol content with the help of chemometrics was necessary. Most of these methods are typically used in IDOKI¿s lab as routine analyses apart from others not included in this PhD thesis.

We propose to utilize the leading pulse of a petawatt class laser to create a conic plasma channel in the dense plasmas. This plasma channel could serve as a natural cone to guide the main pulse to the cone tip, as behaves similarly to the physical Au cone. We estimate that the leading pulse of a petawatt laser could create a natural cone with cone tip only about 100 mu m away from the edge of compressed core plasma. The natural cone formation should be compatible for a good uniform compression and efficient fast heating of the imploded fuel.

Neste estudo nós fornecemos os primeiros dados acerca dos parâmetros da comunidade de anuros de folhiço de uma floresta no estado do Paraná, sul do Brasil, incluindo informações sobre riqueza de espécies, densidades específicas e biomassa. Nosso estudo foi realizado na Reserva Natural Salto Morato entre julho de 2009 e abril de 2010. Para amostrar a comunidade de anuros de folhiço usamos 40 parcelas de 4 x 4 m em cada estação do ano (inverno, primavera, verão e outono), totalizando 2.560 m2 de chão de floresta amostrados. Nós amostramos um total de 96 anuros habitando o chão da floresta, pertencentes a sete espécies: Brachycephalus hermogenesi, Ischnocnema guentheri, Haddadus binotatus, Leptodactylus gr. marmoratus, Physalaemus spiniger, Proceratophrys boiei e Rhinella abei. A densidade total de anuros vivendo no chão da floresta foi de 3,73 ind/100m2, sendo I. guentheri (1,37 ind/100m2) a espécie mais numerosa e R. abei (0,19 ind/100m2), a mais rara. A estimativa da biomassa total na comunidade de anuros de folhiço foi de 3,290g. A temperatura foi um fator ambiental significativo para a abundância de anuros de folhiço, enquanto a umidade não foi importante na estruturação da comunidade na área estudada. A abundância, riqueza e densidade variaram consistentemente entre as quatro estações do ano amostradas, com os maiores valores ocorrendo nos meses mais quentes da primavera e verão. Esse estudo aumenta a distribuição geográfica de Brachycephalus hermogenesi.

A partir da publicação, em 1994, de um relatório que afirmava que a presença de óleos aromáticos pesados, com altos teores de compostos policíclicos, em formulações de borrachas estaria relacionada ao desenvolvimento de doenças como o câncer, associado ao fato da crescente conscientização ambiental na fabricação de produtos elastoméricos, foi iniciado um estudo sobre a substituição do óleo aromático de origem do petróleo por óleos vegetais de menor risco. Nessa Dissertação de Mestrado foi proposta a substituição do óleo aromático derivado do petróleo por óleos vegetais como o de tungue, palma e linhaça, em composições de borracha natural (NR) contendo diferentes teores do copolímero de butadieno-estireno (0, 10 e 50 phr) e diferentes tipos de carga (negro de fumo N330 e negro de fumo N375). As composições obtidas foram avaliadas quanto a propriedades reométricas, mecânicas e fractográficas. Foi observado que não houve variação significativa na viscosidade Mooney, na resistência à tração, no alongamento na ruptura e na dureza Shore das composições de NR SBR1502 quando o óleo aromático foi substituído pelos óleos vegetais, quando diferentes negros de fumo foram utilizados como carga reforçante e quando foram utilizados teores crescentes de SBR1502. Por outro lado, a resistência ao rasgamento das composições de NR SBR1502 apresentaram melhores resultados quando o negro de fumo N375 foi utilizado. A adição de teores crescentes de SR 1502 levou à diminuição dos valores de rasgo. A natureza do óleo utilizado influenciou esse resultado e melhores valores foram obtidos com o óleo de tungue. Os resultados obtidos foram corroborados pela morfologia do material, avaliadas por microscopia eletrônica de varredura

In this paper, the evolution of the gradient force pattern, focal shift, and focal switch induced by a three-portion pure phase-shifting apodizer is numerically investigated in detail. The results show that the proposed apodizer may induce tunable gradient force on the particles in the focal region, focal shift, and focal switch. By adjusting the geometrical parameters of the phase-shifting apodizer, multiple traps may occur with changeable distance between them, and the shape of the optical trap also evolves evidently. More interestingly, for certain geometrical parameters of the proposed apodizer, by changing the phase shift of inner annular portion, the considerable focal shift may occur with focal switch accompanying, which is discussed to show that this kind of apodizer may be a very promising method of transporting trapped particles. © 2005 Elsevier GmbH. En este proyecto se describirá como construir un modelo predictivo de tipo gradient boosting para predecir el número de ventas online de un producto X del cual solo sabremos su número de identificación, teniendo en cuenta las campañas publicitarias y las características tanto cualitativas y cuantitativas de éste. Para ello se utilizarán y se explicarán las diferentes técnicas utilizadas, como son: la técnica de la validación cruzada y el Blending. El objetivo del proyecto es implementar el modelo así como explicar con exactitud cada técnica y herramienta utilizada y obtener un resultado válido para la competición propuesta en Kaggle con el nombre de Online Product Sales.

As espécies de corais invasores, Tubastraea tagusensis e T. coccinea foram acidentalmente introduzidos no Brasil através de plataformas de petróleo. O rápido crescimento e estágio reprodutivo, competição contra espécies nativas, defesas químicas contra predadores e competidores naturais e uso amplo em diferentes substratos utilizados contribuem para o sucesso e expansão de Tubastraea spp. na costa brasileira. O presente estudo teve dois objetivos principais: 1) investigar uma metodologia que resulte em uma maior eficiência e custo-benefício nos processsos de monitoramento dos corais invasores Tubastraea spp. no litoral brasileiro; 2) mapear a distribuição geográfica, caracterizar as populações e estudar o efeito da inserção dos corais na comunidade bêntica de costões rochosos do litoral norte do estado de São Paulo (LNSP). O primeiro avaliou quatro metodologias, comparando o método do censo visual, e outras três metodologias que utilizam fotografia e filmagem. O método do censo visual mostrou ser mais eficiente na obtenção dos resultados quando comparado com os outros métodos, principalmente para identificar pequenos organismos. Contudo, seu tempo em campo e seus custos foram maiores. O segundo utilizou o método visual para estudar o efeito da inserção dos corais invasores na comunidade local do LNSP. Ainda, foi realizado um monitoramento espacial semi-quantitativo em larga escala para caracterizar a distribuição espacial dos corais invasores; transectos com quadrados amostrais foram usados para estimar a densidade de Tubastraea ao longo da profundidade, e transectos e arcos graduados empregados para estimar a ocorrência de colônias em diferentes inclinações do substrato, no LNSP. Os corais invasores estão aumentado sua distribuição, causando diversos impactos nas comunidades e nos organismos nativos. T. tagusensis é comumente encontrado dominando diversos costões rochosos, com uma densidade maior em ambientes mais profundos e com maior ocorência em substratos de inclinções verticais e negativas no LNSP. A erradicação e/ou controle do coral invasor é recomendado no litoral brasileiro, principalmente onde as populações estão isoladas ou ainda são pequenas.
