The constant search for improvements and the survival of organizations makes the strategic guidelines are deployed and executed at the operational levels. This work is the approach to critical analysis of the equipment of a chemical industry through a case study based on the classification of each equipment manufactures through qualitative and quantitative analysis on the pillars of maintenance costs, loss of production, MTBF, contribution margin, Health Safety and Environment (SHE). From this study and future data collection, along with the flow diagram show the main equipment that should be special attention. To this can be prepared an action plan with deadlines and responsible. With the results one can measure the maintenance costs, loss of production and technical availability of the plant, with future gains
Gestão de Estoques: análise e melhoria do processo de inventário físico em uma indústria siderúrgica

Accuracy in physical inventory process is essential to develop anefficient inventorycontrol. This ensures the availability of products and ensure that the information contained in the information systems areinaccordancewith the reality of inventories. The inventory management policies of the company aim ofthis study established that all the materials in their inventory must becounted, which is proving a challenge. Therefore, the aim of thiswork is to identify the critical inventories and analyze them, seeking flaws and possible improvements in the inventory count process. For this, we used the tools of quality management, such as Pareto and Cause and Effect Diagrams in an action research. The results show that the stocks of finished products are critical in volume and can be hampered by a lack of training and personnel trained in performing the process, as well as limitations in ERP used. With the actions taken against these problems, we could notice an improvement in the process, because the data collection and processing of data has become easier, besides the occurrence of errors decreased. In addition, there was an acceleration in meeting targets compared to the previous period

This work was developed in order to reduce the discrepancy found between the stock data of an item in the system and the amount found physically in stock in the company presented on this study case. Therefore, this paper makes a study on system of information that can assist in the collection, control and manipulation of data regarding the company's stock. It is very important to do the inventory management, so we can control the level of this sub-product to meet sales orders and production. In the study discussed below, we used the Six Sigma methodology to identify points that had flaws in the process and thus improve their work where the results show grater process control and provide a reliable database to the point where no further problems of lack of components in stock

The purpose of this work was to define the processes through which the cooling of thermoplastics parts occur inside the mold cavity in an injection process. The plastic materials have become more widespread in the automobile industry and, among its manufacturing processes, injection moulding develops quickly, allowing the manufacturing of quality parts in great volumes. Data was collected from the injection of Volkswagen Gol NF 23X (Gol Generation 5). Using approximated methods for calculation for the heat Exchange inside the mould, in the cooling system, the required water flow was determined to properly cool the parts. Comparing the obtained value with Project specifications, it was verified that the method, in spite of incurring in some mistakes, is efficient in determining the flow of cooling fluid and serves as a verification tool for the parameters defined on project, and can be applied to simple projects. The definition of the cooling system, in practice, is dependent on innumerable variables and each case must be approached in itself, since the parameters for one product may not be ideal for another

The cosmetic industry has presenting growth in Brazil and one way to support this demand is improving production and reducing waste. Wastes can be related to setup time, which is spent to support demand of product´s mix and volume. One of the methodologies used to reduce setup time is SMED (Single Minute Exchange of Die). This paper aims to describe and analyze the setup process in a production line in a cosmetic industrial branch, before and after the application of SMED methodology, highlighting good practices and difficulties in order to help other companies develop fast setup. The research method used was the case study. From the analysis of the implementation of the methodology on the company which is object from the case study was possible to summarize good practices, such as: the use of pillar E&T to store information about the training on the SMED methodology; creation of a multifunctional teamwork and involving different areas; creation of a “From-To” matrix. Difficulties were identified, such as: high and constant turnover of people who work on the shop floor; lack of discipline in filling the forms and by doing the setup activities that were standardized; lack of financial resources; lack of people´s availability in orer to conduct necessary meetings and trainings. Recommendations for other companies have also been proposed, such as: training of the team involved, before starting the project and a refresher training; standardization of training, forms and documentation of all steps of the project; create a multifunctional team; training after an employee, who is responsible for the setup, leaves the company

The catalytic function of extended-spectrum β-lactamases can result in high degrees of bacterial resistance to β-lactamic antimicrobials and in the emergence of ESBL among the members of Enterobacteriaceae family, especially Klebsiella pneumoniae and Escherichia coli. This occurs due to the dissemination and emergence of new variants of these enzymes caused by the high utilization of antibiotics like broad-spectrum cephalosporins. The ESBL are β-lactamases capable of conferring bacterial resistance to the penicillins, 1st, 2nd and 3rd generation cephalosporins, and aztreonam (but not cephamycins and carbapenems) through the hydrolysis of these antibiotics. In view of this phenomenon, the exact screening and detection of the producers of ESBL are essential for the appropriate selection of the antimicrobial therapy. The purposes of this study were to evaluate the best antimicrobial for the selection of ESBL producers and to determine the best method for the detection of such microorganisms. We evaluated 200 sequential bacterial samples including the species Klebsiella pneumoniae (56.5%), Escherichia coli (34%), Proteus mirabilis (8.5%) and Klebsiella oxytoca (1%), previously characterized as ESBL producers between February and September 2008 in the Laboratory of Microbiology, Botucatu Medical School - UNESP, Botucatu, São Paulo State, Brazil. To select the ESBL-producer bacteria, we used the disks recommended by CLSI 2008, aztreonam (ATM), cefpodoxime (CPD), ceftriaxone (CRO), cefotaxime (CTX) and ceftazidime (CAZ), besides cefepime (FEP). ESBL production was confirmed by three methods: double disk screening, ESBL Etest®, and Vitek® automated system. This work presents a project of Action Search in a consumption industry of the sector of personal care, health and beauty. Following the implementation steps of Autonomous Maintenance Pillar of TPM methodology, stands for Total Productive Maintenance, this work aims to ensure the advancement of some machines in a production line of the company to the next steps of the methodology implementation, demonstrating the results achieved by the TPM. In the company in question, the TPM has been implemented in the past but lost strength over the years and some of its concepts were abandoned, producing then a drop in the equipments efficiency, increased wastes and breaks in the processes, as well as loss in product quality. Then, the need arose to restart the implementation process from the beginning, to strictly follow all the steps of the methodology, ensuring increased efficiency of equipment and processes. Through training and a changing in the company culture, it was possible a joint effort between Operation and Maintenance in order to enhance the knowledge of the operators on their machines. Initially, it was developed a general cleanliness program of equipments so that it could be possible to find the anomalies in the process. Subsequently, operators and maintainers were trained to detect anomalies, enabling equipments to work under their basic conditions of operation and subsequently building provisional standards of equipment cleaning, lubrication and inspection. Through the improvement presented by some indicators such as OEE, wastes, bankruptcies and unavailability, it was proved the importance and the positive effects of TPM implementation in manufacturing

Aquatic ecosystems are suffering many impacts caused by human activities resulting from the activities occurring around them. With technological progress observed in recent years, this environment has received large amounts of chemicals from industries, agriculture and urban area that affect the aquatic biota. Among these sources of contamination, the oil industry has contributed to the pollution of aquatic environments with both effluents as produced water well as oil spills and their derivatives having toxicity to various organisms. With all the environmental issues has increased concern about water quality and has been used ecotoxicological tests with aquatic organisms to ecosystems to assess the toxicity of chemicals present in the water. In this context the microcrustacea Daphnia similis stands out as a freshwater organism very representative of the aquatic fauna of rivers and high sensitivity to environmental impacts. Thus, the present study aimed to evaluate the lethal toxicity of crude oil and produced water on this microcrustacea. The results showed that the microcrustacea presented high sensitivity to contaminants primarily crude oil. There was also the influence of environmental variables pH and temperature on the survival of organisms

This work consists of the implementation of the steps Define, Measure , Analyze , Improve and Control ( DMAIC ) to develop a Six Sigma project in an industry the food industry. The objective was to demonstrate a potential for reducing the occurrence of crushed cans in potting milk powder industry in a White Belt project. The food industry accounts for about 9 % of Brazil's gross domestic product ( GDP ), generating thousands of jobs . Among the major sectors of the food industry is the manufacturing sector of milk , occupies approximately 10 % of the total turnover of the food industry . Brazil is considered today one of the eight largest producers of milk powder in the world. The milk powder is packed , mostly for aluminum cans that are lined internally with varnishes and other materials to protect the milk of metals from aluminum. When the cans are dented food protection is compromised and may lead ingestion causing dis-eases such as botulism. Aiming to solve the problem of dented cans methodology was used as a case study with a quantitative approach through the DMAIC method. Some quality tools used in each step of the project as brainstorming , cause and effect diagram , flowchart , ef-fort and impact matrix, 5W1H , among other Pareto diagram is presented . A survey about the disposal of cans in the company verifying a mean loss and, from this histor-ical , a goal loss was calculated was performed . With the target set we calculated the annual saving design . During application of DMAIC was found that the highest rate of loss occurred in transportation between the factory and the factory that fills cans milk . Several actions were taken to resolve problems that resulted in dented cans and the first two months of phase control it was found that the smaller losses calculated target resulting in a saving for the company. This work is an action research conducted in an industry of consumer goods, presenting a new model of inventory management in the company's processors. This replacement of the inventory management of the company stemmed from the need to reduce the large number of deviations in the product write off in stock, thereby generating a low accuracy and reliability of data on inventories of processors shown by the company's ERP system. Spending on inventory adjustments could thus be reduced with the implementation of the new model, thus generating a cost savings for the company and thus increasing their competitive potential in the market. In the old system adopted by the company, write off raw material inventory was done automatically by the system for customized transactions by the company. However, since the implementation of ERP in the company, the automatic write off based on historical consumption of each product were made in many cases at random, generating a lot of mistakes. The new management system has replaced the automatic system by manual at the time of the return of the processed product in the company, thus creating a control which lots and quantities were consumed in the processing and making the stock shown by the ERP reliable and accurate

Uma coleção de 50 amostras de Staphylococcus aureus e 50 de estafilococos coagulase-negativa (ECN) isoladas de recém-nascidos (RN) da unidade neonatal do Hospital das Clínicas da Faculdade de Medicina de Botucatu foram estudadas quanto à produção de biofilme. Para isto foi utilizada a técnica de PCR na detecção dos genes icaA, icaC e icaD e os métodos fenotípicos de aderência em placa de poliestireno, aderência em tubo de borossilicato e método do Agar Congo Vermelho (CRA). Dos 50 S. aureus estudados, 100% foram positivos para a produção de biofilme pela PCR, 98% pelo método do tubo, 100% pelo método da placa de poliestireno e 98% pelo CRA. Já das 50 amostras de ECN, 94% foram positivas pela PCR, 76% pelo método do tubo, 82% pelo teste da placa e 74% pelo CRA. Feita a comparação dos métodos utilizados, tendo por referência o padrão-ouro (PCR), foi possível observar que o método que melhor se correlacionou com o padrão-ouro, foi o método da aderência em placa de poliestireno, apresentando melhor sensibilidade e especificidade para ambas as espécies

In today's competitive environment of automakers, it is essential to obtain the highest efficiency of the production process. This paper presents a study in a pre-assembly of brake pipes and fuel of a vehicle where the value stream maps and information (VSM – Value Stream Mapping) were designed in order to improve the process by reducing the Lead Time Production of a product, reducing waste and decrease time between processes. This work can be divided into three stages, the first building the VSM of the initial state, the second VSM of the proposed state and finally the VSM than was actually performed and to present the gains were achieved effectively. The proposed VSM would lead to a gain of 54% in lead time and 61% in processing time, since the VSM implemented had gains of 47% in lead time and 48% in processing time even without major investments as originally proposed. Concluding that even without big investment, using the techniques of lean manufacturing is possible to achieve high levels of process efficiency

As enzimas estão presentes em todas as células vivas, onde exercem a função de catalisadores das reações que compõem as vias catabólicas e anabólicas do metabolismo celular. Esses biocatalisadores são moléculas de proteínas e seu poder catalítico está associado à conformação nativa, que depende de condições específicas de pH, temperatura e força iônica do meio. Os micro-organismos são bastante atrativos para a indústria, pois possibilitam a produção de enzimas por processos fermentativos em larga escala com regularidade necessária e simplicidade na requisição nutricional. Assim, embora alguns biocatalisadores sejam extraídos de tecidos animais e vegetais, as enzimas industriais são, em sua maior parte, obtidas a partir de micro-organismos. Este trabalho teve como objetivo a produção das enzimas lipase e β-glucanase a partir dos fungos Aspergillus niger e Trichoderma reesei, respectivamente, em diferentes meios de cultura, para determinar as condições de maior produção da enzima em questão. As enzimas produzidas em agitador orbital foram obtidas a partir da filtração do produto da fermentação, precipitação com sulfato de amônio e liofilização. Após a produção e precipitação a atividade das enzimas e a concentração de proteínas foram quantificadas, os parâmetros cinéticos foram determinados frente a diferentes pHs, temperaturas e força iônica do meio. A lipase apresentou melhor atividade a 30°C e em pH 6,0. A presença dos íons Mg2+ e Zn2+ levaram a um aumento na atividade da enzima. A β-glucanase apresentou maiores atividades quando submetidas a 37°C e pH 5,0. Os íons Mg2+, Cu2+ e Ca2+ induziram melhor a atividade enzimática.

A prática da reescrita é um dos métodos mais eficazes para aprender a dar sentido aos textos produzidos, a escrever textos coesos e coerentes, a não cometer erros de gramática, a se familiarizar com a ortografia da língua e, consequentemente, cometer cada vez menos erros desse tipo. Além disso, a reescrita permite chegar a textos com argumentação bem construída, a sair de textos de senso comum, a deixar de lado a linguagem infantilizada, a escrever para um leitor que não apenas o professor e a se acostumar com as características próprias de cada tipo de texto. Foram realizadas análises de textos escritos por estudantes do 2º ano do Ensino Médio a partir dos pressupostos teórico-metodológicos referentes ao paradigma indiciário, paradigma este que permite análises do tipo qualitativa, desenvolvido no âmbito das ciências humanas. A partir do olhar de detetive proposto pelo paradigma indiciário foi possível indicar os erros cometidos pelos estudantes, fazê-los refletir sobre os próprios textos e melhorá-los através da reescrita. Os resultados obtidos com as refacções mostram que é este um método eficiente de se trabalhar em sala de aula, pois essa prática permite que o aluno se volte para os seus erros de forma a corrigi-los. Além disso, é possível concluir que essa prática constante tende sempre a trazer resultados positivos, visto que quantas mais forem as reescritas, melhor o texto se torna. Nesse sentido, a utilização do paradigma indiciário é muito pertinente por permitir uma análise minuciosa, mais atenta aos erros cometidos e às análises dos mesmos

Kaposi´s sarcoma associated herpesvirus (KSHV) or human herpesvirus 8 (HHV-8) is a gammaherpesvirus essential for the development of all forms of Kaposi´s sarcoma (KS). The KSHV’s life cycle is basically divided into latent and lytic phases, which have distinct viral gene expression profiles. Some important oncogenic products of KSHV are expressed during the lytic phase, including the viral K1 protein. As an effect of interfer-ence with intracellular signaling, K1 expression increases proliferation and survival of KSHV-infected cells. Due to its high level of genetic variability compared to other re-gions of the viral genome, the K1-encoding ORF (ORF-K1) is commonly evaluated for KSHV genotyping. It remains unclear whether different viral genotypes have particular biological effects that might modify the KSHV oncogenicity. The present study aimed to contribute to the establishment of an experimental in vitro model for evaluation of the K1 protein from common KSHV genotypes. Recombinant expression vectors with the ORF-K1 from KSHV genotypes A, B and C were prepared by genetic cloning. The recombi-nant vectors pKSHVOK1 obtained by cloning were sequenced for structural validation. After that, HEK293 cell line was transfected with the recombinant vectors, and proteins were extracted for expression analysis by Western blot technique, for K1 functional vali-dation. Results showed that ORF-K1 vectors containing KSHV ORF-K1 from the A, B and C genotypes were produced and structurally validated by DNA sequencing. The K1 expression at the protein level was also confirmed by immunoblots using an antibody for FLAG detection, an epitope from the vector that binds to K1. Based on presented re-sults, it´s possible to conclude that the recombinant vectors will be able to be used in future studies of K1 protein biological properties from distinct KSHV genotypes
