The objective of this study was to verify application of two methodologies: substrate moistened with herbicide solution (SM) and immersion of seeds in herbicide solution (IH) for detecting soybean seeds genetically modified. For this, non-transgenic and transgenic soybean seeds, harvested in the 2008/2009 crop seasons were used. The treatments with substrate moistened were: SM1) 0.03% herbicide solution, at 25 ºC, with evaluation in the sixth day (hs -0.03% -25 ºC, 6th d); SM2) HS -0.03% -35 ºC, 5th d; SM3) HS -0.03% - 40 ºC, 5th d; and SM4) hs -0.06% -5 ºC, 5th d. In the methodology of immersion of seeds the following treatments were performed: IH1) seed immersion in a 0.6% herbicide solution, at 25 ºC, for 1 h, (si -0.06% -25 ºC, 1 h; IH2) si -0.06% - 35 ºC, 30 min.; IH3) si -0.06% -40 ºC, 30 min.; IH4) si -0.12% -35 ºC, 30 min.; and IH5) si -0.12% -40 ºC, 30 min. Bioassays allow detecting soybean seeds tolerant to glyphosate herbicide within five days. The seeds of non-genetically modified and genetically modified soybean cultivars may be easily distinguished through the treatments SM2 and SM4 of the moistened substrate methodology; and treatments IH3, IH4, and IH5 of seed immersion methodology. Both methodologies are easily feasible, practical, and applicable in seed analysis laboratories, once do not require special equipments.

Buffaloes and bovines are polyestrous and seasonal or annual livestock, respectively, that show reduced fertility during heat stress. To investigate whether reduced fertility is related to oocyte competence in both species, immature oocytes from buffalo and bovine heifers were collected during winter and summer and subjected to molecular analyses. In each season, heifers of both species had their follicular wave emergence synchronized with a standard protocol (Ferreira et al., 2011). Before being subjected to ovum pick up (OPU), cutaneous (CT; degrees C) and rectal (RT; degrees C) temperatures and respiratory rate (RR; breaths/min) were measured. Oocytes' RNA was extracted to evaluate the expression of target genes related to mtDNA replication/transcription (PPARGC1A, TFAM and MT-CO1), apoptosis (BAX and BCL2) and HS (HSP90AA1 and HSPA1AB). ACTB, HIST1H2AG and GAPDH were initially chosen as housekeeping genes. In buffaloes, CT (35.0 +/- 0.4 vs 23.8 +/- 0.5), RT (38.7 +/- 0.1 vs 38.0 +/- 0) and RR (21.3 +/- 1.2 vs 15.4 +/- 1.1) were higher during summer than winter. However, in bovine heifers, RT (38.7 +/- 0.1 vs 38.6 +/- 0.1) and RR (44.8 +/- 1.5 vs 40.6 +/- 1.5) were similar in both seasons, while CT (31.6 +/- 0.3 vs 30.2 +/- 0.3) was increased during summer. Reduced expression of ACTB, HIST1H2AG and GAPDH was evidenced during summer, disqualifying them as housekeeping genes. Similarly, the expression of all target genes was reduced during summer in oocytes of both species. In summary, physiological responses to heat stress seem to be more intense in buffalo than bovine heifers. However, in both species, negative effects of heat stress upon oocyte quality occur at the molecular level and affects genes related to several biological functions.

Contents The IGF system is related to embryo quality. We aim to determine the effect of the heat stress on the mRNA expression of IGF1 and IGF2, IGFR1 and IGFR2, IGFBP2 and IGFBP4, and PAPPA in in vitro production (IVP) blastocysts from Nelore and Holstein after ovum pick up (OPU) to better understand the differences between these breeds.Oocytes from four Nelore and seven Holstein were collected in six OPU sessions. Following in vitro maturation and fertilization using six Nelore or Holstein sires, embryos were divided into control (cultured at 39 degrees C) and heat stress (HS; exposed to 41 degrees C for 9h). Blastocysts were submitted to RNA extraction. The IGF1 expression was higher in blastocysts under HS in both breeds, and the expression of IGFBP2 and IGFBP4 was higher in Holstein blastocysts under HS. The high PAPPA expression and the low expression of IGFBP2 and IGFBP4 are associated with a more efficient degradation of IGFBPs, which results in greater IGF bioavailability in Nelore blastocysts and may contribute to the superior HS tolerance in Nelore, when compared to Holstein.

Delamination or crack propagation between plies is a critical issue for structural composites. In viewing this issue and the large application of woven fabrics in structural applications, especially the ones that requires high drapeability to be preformed in a RTM mold cavity such as the asymmetric ones, e.g HS series, this research aimed in dynamically testing the carbon fiber 5HS/RTM6 epoxy composites under opening mode using DCB set up in order to investigate the crack growth rate behavior in an irregular surface produced by the fabric waviness. The evaluation of the energy involved in each crack increment was based on the Irwin-Kies equation using compliance beam theory. The tests were conducted at constant stress ratio of R=0.1 with displacement control, frequency of 10 Hz, in accordance to ASTM E647-00 for measurement of crack growth rate. The results showed large scatter when compared to unidirectional carbon fiber composites due to damage accumulation at the fill tows.

Since the beginning, some pattern recognition techniques have faced the problem of high computational burden for dataset learning. Among the most widely used techniques, we may highlight Support Vector Machines (SVM), which have obtained very promising results for data classification. However, this classifier requires an expensive training phase, which is dominated by a parameter optimization that aims to make SVM less prone to errors over the training set. In this paper, we model the problem of finding such parameters as a metaheuristic-based optimization task, which is performed through Harmony Search (HS) and some of its variants. The experimental results have showen the robustness of HS-based approaches for such task in comparison against with an exhaustive (grid) search, and also a Particle Swarm Optimization-based implementation.

Diuron (3-(3,4-Dichlorophenyl)-1,1-dimethylurea) is a substituted urea herbicide widely used on agricultural crops such as soy, cotton and sugar cane. In a previous long-term study this herbicide exerted carcinogenic activity on the urinary bladder and renal pelvis mucosa of Wistar rats and breast of mice. Also, it was shown to be carcinogenic to the mice skin in a initiation-promotion assay. In 1997, the northamerican EPA evaluated Diuron as a “known/likely” carcinogen for humans (USEPA, 2004). In a previous study developed at this laboratory, male Wistar rats treated with Diuron 2500 ppm during 20 weeks presented increased indices of cell proliferation and incidences of simple urothelial hyperplasia (HS) in the urinary bladder. Under scanning electron microscopy (SEM) severe urothelial necrosis and hyperplasia were observed. However, in that study the urinary bladders of animals exposed to lower doses of Diuron were not examined under SEM. Therefore, the possible dose-response influence of Diuron on the urothelium under SEM is not known. The present study aimed to analyze under SEM the urinary bladder of male Wistar rats exposed to 125 ppm, 500 ppm and 2500 ppm doses of Diuron through diet during 20 weeks and to compare to the previous histological findings in the same material. Under SEM, 125 ppm and 2500 ppm groups presented significantly (p<0,05) increased incidences of simple hyperplasia, i.e., 7/10 and 8/10 respectively, compared to control group and the 500 ppm group The sensitivity of SEM was higher since it detected a 45% incidence of hyperplasiaswhile the histological analysis found only 27%. Considering SEM as the gold-standard, histology showed a 44% sensitivity, 86.4% specificity, a positive predictive value of 72,7% and negative predictive value of 65,5% and accuracy of 67,5%. AC Biosusceptometry (ACB) is a low-cost biomagnetic tool that has been successfully applied on pharmaceutical research to evaluate performance of solid dosage forms. The aim of this work was to evaluate the Horn & Shunck method to access tablet disintegration. To evaluate the HS results was record on video a test with a objet moving in a rail with a constant velocity. The desintegration was recorded on video and ACB, which used have seven pairs of detection coils and a pair of excitation coils to mensure the magnetic ux variation. The signals were ampli ed and digitalized to create images, which were restored by Wiener lter, while the video images are converted to gray scale, both are normalized and binarized and had the optical ow estimation calculated by Horn & Schunck (HS) algorithm. All signals and images are processed and developed algorithm on Matlab. During the tests the ve tablets (500mg ferrite, 375mg excipients, compression 10 to 50 kN) were on a becker between of the ACB system and of the video system, and only touching the surface of the water. With all OF maps calculated was realized the sum of the resultants of each, to get a disintegration process resultant for each compression. Whit that was possible observed the disintegration behaves. For the compression force study the HS components of each sequence was sum, take mean and normalized for sequence's max modulo, therefore can be observed a high growing on less compression tablets. We can conclude the HS algorithm is viable to tablets disintegration data collection and whit that was possible to create a tablets disintegration analyzes protocol, which would be useful on desintegration kinetics study
