The fully relaxed single-bond torsional potentials and orientation-related rotational potentials of 2,2'-bithiophene (BT) under the interaction of an external electric field (EF) constructed by point charges have been evaluated with semi-empirical AMI and PM3 calculations. The torsional potentials are sensitive to both EF strength and direction. While the EF is parallel to the molecular long axis, the torsional barrier around C-x-C-x' bond obviously rises with increasing the EF strength, whereas the relative energies of syn and anti minima show a slight change. The interaction between the EF and the induced dipole moment has been proposed to elucidate this observation. On the other hand, the relative energy difference between the syn and anti minima shows an obvious change, while the EF is perpendicular to the molecular long axis. This feature has been ascribed to the interaction between the EF and the permanent dipole moment of BT. Furthermore, conformational and orientational analyses in two dimensions have been carried out by changing the torsional and rotational angles in the different EF. The conformation and orientation of a gas-phase BT in the EF are governed by both the above factors.

Copolymers based on monomers phenolphthalein (PP)/4,4'-thiodiphenol (Bis-T)/4,4'-dichlorodiphenylsulfone (DCDPS) were prepared by a route involving the toluene, N-methyl-2-pyrrolidone and anhydrous potassium carbonate synthesis. The range of optimum reaction temperature was between 185 and 195 degrees C. The copolymers were characterized by C-13 NMR, differential scanning calorimetry (DSC) and torsion braid analysis. It was found that all of the copolymers were random and homogeneous and their glass transition temperatures (T-g) decreased linearly with an increase of Bis-T contents in the copolymers. The thermal stability determined by thermogravimetry analysis in air atmosphere indicated that the copolymer had better resistance to thermo-oxidative degradation. Dynamic mechanical measurement showed that (PP/Bis-T) PES copolymers containing 0-50 mol% of Bis-T components had two secondary relaxations. (C) 1998 Elsevier Science Ltd. All rights reserved.

The crystal structure of poly(ether ketone ketone) (PEKK) is predicted by using Cerius2 software according to the wide angle X-ray diffraction (WAXD) experiment result. The predicted structure has a planar zigzag chain conformation between ether oxygen and ketone carbons in an orthorhombic lattice. Average zigzag angle is 126 degrees and average torsion angle is 30.32 degrees. The WAXD powder pattern calculated from the crystal packing model is in good agreement with the experiment result.

The ontogenetic development of the digestive enzymes amylase, lipase, trypsin, and alkaline phosphatase and the effect of starvation in miiuy croaker Miichthys miiuy larvae were studied. The activities of these enzymes were detected prior to exogenous feeding, but their developmental patterns differed remarkably. Trypsin activity continuously increased from 2 days after hatching (dah), peaked on 20 dah, and decreased to 25 dah at weaning. Alkaline phosphatase activity oscillated at low levels within a small range after the first feeding on 3 dah. In contrast, amylase and lipase activities followed the general developmental pattern that has been characterized in fish larvae, with a succession of increases or decreases. Amylase, lipase, and trypsin activities generally started to increase or decrease at transitions from endogenous to exogenous feeding or diet changes, suggesting that these enzymatic activities can be modulated by feeding modes. The activities of all the enzymes remained stable from 25 dah onwards, coinciding with the formation of gastric glands and pyloric caecum. These results imply that specific activities of these enzymes underwent changes due to morphological and physiological modifications or diet shift during larval development but that they became stable after the development of the digestive organs and associated glands was fully completed and the organs/glands functioned. Trypsin and alkaline phosphatase were more sensitive to starvation than amylase and lipase because delayed feeding up to 2 days after mouth opening was able to adversely affect their activities. Enzyme activities did not significantly differ among feeding groups during endogenous feeding; however, all activities were remarkably reduced when delayed feeding was within 3 days after mouth opening. Initiation of larvae feeding should occur within 2 days after mouth opening so that good growth and survival can be obtained in the culture.

海水经济鱼类的养殖在我国已经形成第四次海水养殖浪潮,经济效益显著,有力地推动了我国海水养殖的产业结构调整和可持续发展。然而在海水养殖发展过程中也存在着诸多问题,尤其是早期发育阶段的高死亡率,严重制约了我国海水养殖产业的稳定和健康发展。 海水鱼类养殖的关键为高质量,高存活率苗种的生产和培育,由于鱼类种类繁多,生物多样性丰富,对应实际的繁育技术,尤其是新品种的开发,必须要做出相应的调整。这就要求我们必须对每一种鱼类早期发育有所了解,并将形态和组织上的数据用于指导生产。 本文通过显微观察和组织学研究,主要描述和研究了我国北方三种重要的海水经济鱼类(条斑星鲽、杂交鲆、条石鲷)的早期发育生物学,并结合实际生产进一步阐明关键期的产生原因,机理以及采用相应的对策。具体结果如下: 1.条斑星鲽:作为冷温性鲆鲽鱼类,条斑星鲽早期发育过程的特征主要有: ① 条斑星鲽受精卵无油球,卵子呈半浮性;不同步卵裂现象提前,发生在第三次卵裂;卵裂期裂球大小差异大。孵化过程较长,在水温8 ± 0.3℃,盐度33的条件下,经9 d孵化。条斑星鲽胚胎发育的不同时期对温度的敏感性不同,其中原肠期对温度比较敏感。 ②在8-10℃,盐度33的条件下,8-9 dph开口摄食。且开口时,其吻前端出现有一点状黑褐色素,构成了条斑星鲽仔鱼“开口期”的重要标志。卵黄囊于消失。在后期仔鱼末期,背鳍和臀鳍上形成特有的黑褐色条斑带。 ③杯状细胞首先出现在咽腔后部和食道前段,胃腺和幽门盲囊出现于29 dph,变态期始于30dph。在条斑星鲽早期发育过程中,观察到其直肠粘膜层细胞质出现大量嗜伊红颗粒,为仔鱼肠道上皮吸收的蛋白质。 ④首先淋巴化的免疫器官是头肾,然后是胸腺和脾脏,这与大部分硬骨鱼类不同。条斑星鲽除头肾和脾脏外,胸腺实质也形成MMCs。其中以脾脏形成MMCs最为丰富,形态多样。 2. 杂交鲆:为同属的牙鲆和夏鲆间的远缘杂交种,其发育过程的特点为: ① 在温度为15.4~16.0℃,杂交鲆胚胎从受精到孵化所需的时间为76 h左右,胚孔关闭前期,胚胎先出现视囊及克氏囊,而后形成体节。孵出前胚体在卵膜内环绕不到1周。 ② 孵化后消失。杂交鲆群体变态间隔长(34-60 dph),且变态高峰期出现的冠状幼鳍不明显(与母本牙鲆相比),数量为7-8根。 ③组织学观察发现,其消化系统中胃腺出现较晚,且胃腺发育过程缓慢(与母本牙鲆相比)。甲状腺滤泡增生不明显,颜色较浅,数量较少。杂交鲆在早期发育过程中,并没有出现鳔原基。 3. 条石鲷作为岩礁性的暖水性鱼类,早期发育过程也较为特殊,包括外形以及内部的器官结构。主要特点有: ① 受精卵:受精卵卵黄上具有龟裂结构,为鱼卵的分类特征之一。 ② 初孵仔鱼:初孵仔鱼背鳍膜上的黑色素,从体背面向背鳍膜边缘移动,到3dph仔鱼基本消失,此为本种仔鱼发育所特有的特点。 ③ 后期仔鱼和稚鱼:肠道肌肉层加厚明显,仔稚鱼胃肠排空率急剧上升,死亡率增加,通过改善常规的投饵方式部分解决了这个死亡高峰的问题。在幼鱼初期,牙齿融合为骨喙,为石鲷科鱼类的特征。 ④胸腺上皮分泌细胞:类似的现象同样在虹鳟鱼中发现,但是虹鳟鱼胸腺上皮分泌细胞不如条石鲷的丰富,同样也不如条石鲷的排列整齐,而是零星分布在胸腺上皮与咽腔接触的表面。除了正常的造血器官—脾脏和头肾外,肝脏、胰腺和鳔等多种组织等也出现MMCs,此现象在硬骨鱼类不多见,一般发生在软骨鱼类。

本文在Bruker AM-400 NMR谱仪上,在不同温度下研究了线形脑啡肽(N-Tyr~1-Gly~2-Gly~3-Phe~4-Leu~5)在DMSO中的NMR溶液构象。由NMR测试结果,得到了NH化学位移温度梯度系数、扭转角φ、χ'约束和~1H-~1H NOE距离约束,用目标函数法计算了脑啡肽的溶液构象,分析了优势边链构象。研究结果指明了多肽骨架的柔变性且处于构象平衡中。

Ethnopharmacological relevance: A common plant used to treat several gastric disorders is Buddleja scordioides Kunth,commonly known as salvilla. Aim of thes tudy: To detect inflammatory markers,in order to evaluate the gastroprotective potential of salvilla infusions,as this could have beneficial impact on the population exposed to gastric ulcers and colitis. Materials and methods: The present work attempted infusions were prepared with B. scordioides (1% w/w) lyophilized and stored.Total phenolic content and GC–MS analysis were performed. Wistar rats were divided into five groups a negative vehicle control,an indomethacin group,and three experimental groups,named preventive,curative,and suppressive. All rats were sacrificed under deep ether anesthesia(6h)after the last oral administration of indomethacin/infusion.The rat stomachs were promptly excised,weighed,and chilled in ice-cold and 0.9%NaCl.Histological analysis,nitrites quantification and immunodetection assays were done. Results: B.scordioides infusions markedly reduced the visible hemorrhagic lesions induced byindomethacin in rat stomachs,also showed down-regulation of COX2, IL-8 and TNFα and up-regulation of COX-1with a moderate down-regulation of NFkB and lower amount of nitrites.However,this behavior was dependent on the treatment,showing most down-regulation of COX-2,TNFα and IL-8 in the curative treatment;more down-regulation of NF-kB in the preventive treatment;and more up-regulation of COX-1 for the suppressor and preventive treatments. Conclusion: The anti-inflammatory potential of B. scordioides infusions could be related with the presence of polyphenols as quercetin in the infusion and how this one is consumed.

A diabetes mellitus do tipo 2 é caracterizada pela resistência à insulina e pela disfunção das células β do pâncreas. Os péptidos gastrintestinais, “gastric inhibitory polypeptide” (GIP) e “glucagon-like peptide-1” (GLP-1), são hormonas incretinas que estimulam, maioritariamente, a produção de insulina pós-prandial. Formulações contendo GLP-1 possuem um grande potencial no tratamento desta doença. Porém, o GLP-1 é eficaz apenas quando administrado por via parentérica. Para o tratamento da diabetes mellitus tipo 2 são usados análogos do GLP‑ 1 ou miméticos da incretina os quais são eficazes por via subcutânea. The pathogenesis of diabetes mellitus type 2 includes insulin resistance and progressive β-cell dysfunction. The gastrointestinal peptides, gastric inhibitory polypeptide (GIP) and glucagon‑like peptide-1 (GLP-1), are incretin hormones which are responsible for the major part of postprandial insulin secretion. Formulations containing GLP-1 have a great potential in the treatment of diabetes mellitus type 2. Nonetheless, GLP-1 is only efficient by continuous parenteral administration. GLP-1 analogues or incretin mimetics, exendine-4, are active after subcutaneous injection and can be used in the treatment of diabetes mellitus of type 2.

Cancer is a global problem. Despite the significant advances made in recent years, a definitively effective therapeutic has yet to be developed. Oncolytic virology has fallen back into favour for the treatment of cancer with several viruses and viral vectors currently under investigation including vesicular stomatitis virus (VSV), adenovirus vectors and herpes simplex virus (HSV) vectors. Reovirus has an advantage over many viral vectors in that its wild-type form is non-pathogenic and will selectively infect transformed cells, particularly those mutated in the Ras pathway. These advantages make Reovirus an ideal candidate as a safe and non-toxic therapeutic. The aim of the first part of this study was to determine the effect, if any, of Reovirus on cell lines derived from cancers of the gastrointestinal tract. These cancers, particularly those of the oesophagus and stomach, have extremely poor prognoses and little improvement has been seen in survival of these patients in recent years. Reovirus as a single therapy showed promising results in cell lines of oesophageal, gastric and colorectal origin. Further study of partially resistant cell lines using a combination of Reovirus and conventional therapies, either chemotherapy or radiation, showed that a multi-modal approach to therapy is possible with Reovirus and no antagonism between Reovirus and other treatments was observed. The second part of this study focused on investigating a novel use of Reovirus in an in vivo setting. Cancer vaccination or the use of vaccines in cancer therapy is gaining momentum and success has been seen both in a prophylactic approach and a therapeutic approach. A cell-based Reovirus vaccine was used in both these approaches with encouraging success. When used as a prophylactic vaccine tumour development was subsequently inhibited even upon exposure to a tumorigenic dose of cells. The use of the cell-based Reovirus vaccine as a therapeutic for established tumours showed significant delay in tumour growth and a prolongation of survival in all models. This study has proven that Reovirus is an effective therapeutic in a range of cancers and the successful use of a cell-based Reovirus vaccine leads the way for new advancements in cancer immunotherapy.

Helicobacter pylori is a gastric pathogen which infects ~50% of the global population and can lead to the development of gastritis, gastric and duodenal ulcers and carcinoma. Genome sequencing of H. pylori revealed high levels of genetic variability; this pathogen is known for its adaptability due to mechanisms including phase variation, recombination and horizontal gene transfer. Motility is essential for efficient colonisation by H. pylori. The flagellum is a complex nanomachine which has been studied in detail in E. coli and Salmonella. In H. pylori, key differences have been identified in the regulation of flagellum biogenesis, warranting further investigation. In this study, the genomes of two H. pylori strains (CCUG 17874 and P79) were sequenced and published as draft genome sequences. Comparative studies identified the potential role of restriction modification systems and the comB locus in transformation efficiency differences between these strains. Core genome analysis of 43 H. pylori strains including 17874 and P79 defined a more refined core genome for the species than previously published. Comparative analysis of the genome sequences of strains isolated from individuals suffering from H. pylori related diseases resulted in the identification of “disease-specific” genes. Structure-function analysis of the essential motility protein HP0958 was performed to elucidate its role during flagellum assembly in H. pylori. The previously reported HP0958-FliH interaction could not be substantiated in this study and appears to be a false positive. Site-directed mutagenesis confirmed that the coiled-coil domain of HP0958 is involved in the interaction with RpoN (74-284), while the Zn-finger domain is required for direct interaction with the full length flaA mRNA transcript. Complementation of a non-motile hp0958-null derivative strain of P79 with site-directed mutant alleles of hp0958 resulted in cells producing flagellar-type extrusions from non-polar positions. Thus, HP0958 may have a novel function in spatial localisation of flagella in H. pylori

BACKGROUND: Shared decision-making has become the standard of care for most medical treatments. However, little is known about physician communication practices in the decision making for unstable critically ill patients with known end-stage disease. OBJECTIVE: To describe communication practices of physicians making treatment decisions for unstable critically ill patients with end-stage cancer, using the framework of shared decision-making. DESIGN: Analysis of audiotaped encounters between physicians and a standardized patient, in a high-fidelity simulation scenario, to identify best practice communication behaviors. The simulation depicted a 78-year-old man with metastatic gastric cancer, life-threatening hypoxia, and stable preferences to avoid intensive care unit (ICU) admission and intubation. Blinded coders assessed the encounters for verbal communication behaviors associated with handling emotions and discussion of end-of-life goals. We calculated a score for skill at handling emotions (0-6) and at discussing end of life goals (0-16). SUBJECTS: Twenty-seven hospital-based physicians. RESULTS: Independent variables included physician demographics and communication behaviors. We used treatment decisions (ICU admission and initiation of palliation) as a proxy for accurate identification of patient preferences. Eight physicians admitted the patient to the ICU, and 16 initiated palliation. Physicians varied, but on average demonstrated low skill at handling emotions (mean, 0.7) and moderate skill at discussing end-of-life goals (mean, 7.4). We found that skill at discussing end-of-life goals was associated with initiation of palliation (p = 0.04). CONCLUSIONS: It is possible to analyze the decision making of physicians managing unstable critically ill patients with end-stage cancer using the framework of shared decision-making.
