The rambutan (Nephelium lappaceum) is an exotic fruit with great market potential in Brazil. However, there are few available informations about plants with potential for cultivation, because great morphologic variation is observed among plants and for consequence, little uniformity in the orchards and in the fruits. This research had for objective to evaluate the genetic diversity of a collection of rambutan plants obtained by seeds through morfo-chemical analyses of plants and fruits and by fAFLP molecular markers, to indication of promising materials to be used in new plantings of the culture in the São Paulo State. Was verified that both markers, morphologic and molecular, were efficient in the distinction of varieties, showing the presence of genetic variability among the plants of this study. Was also verified that the materials A51 and B03 presented a larger group of desirable characteristics for new cultivations of the fruitful.

The lychee (Litchi chinensis Sonnerat), of the family Sapindaceae, is a highly valued fruit throughout the world. It is native to southern China, but it is cultivated in many parts of the world. In Brazil, the cultivation of the lychee was initiated in the 1970s in the State of São Paulo and has been increasing in production even though the crop size still averages only 1.4 ha per producer. The low crop size is attributed to the high cost of seedlings and the amount of time required for the plant to bear fruit. These factors discourage planting in large areas. The purpose of this research is to study some aspects of what is vitally important for the production of lychee seedlings on a large scale, with low cost and good genetic quality. Tests were performed using rootstocks from seeds of lychee and longan (Euphoria longan Lam.). Scions for grafting were taken from plants of lychee 'Bengal' in the collection of fruit species from FCAV-UNESP, Campus of Jaboticabal, Brazil. The experiments were started in September 2007 and the grafting process was performed in each month of the year finishing in August 2008. During this period the influence of these criteria were evaluated: 1) time of the year; 2) species of rootstock; 3) percentage of grafting success; 4) stem height at grafting point; 5) stem diameter at grafting point. Therefore, through statistical analysis we obtained significant differences in relation to the rootstock used, the months of the year in which the grafting was performed and the interaction between rootstock and the month in which the grafting was performed.

The objective of this work was to evaluate the chemical alterations of the substrate in the cultivation of pepper in coconut husk fiber, in a protected environment. Initially, 160 pepper plants ('Eppo') were divided into four blocks, where two pots per block were analyzed every 21 days after transplanting. The cultivation of pepper was carried out in plastic pots of 13 L, containing coconut husk fiber, and placed in double rows with a spacing of 0.5×0.8 m between single rows and 1.10 m between double rows. After removal of the plants from the pots, individual samples of substrate (approximately 1 L) were collected from each pot and dried at ambient temperature. Electrical conductivity (EC), pH, and levels of NH4 +-N, NO3 -, P and K were determined for all periods of the cultivation. These analyses were performed using the method of extraction 1:1.5 v/v. For the conditions which the experiment was conducted, there was an increase in substrate EC, as well as in the levels of nitrogen, phosphorus and potassium.

Spent Agaricus and Pleurotus substrates are mainly used as components of amendments and growing substrates, but not in sufficient quantities to solve the problem of their accumulation in mushroom producing areas, where they represent a potential pollution risk. The mushroom growing sector in Spain generates about 5105 t of spent compost, while the EU, as a whole, produces more than 3.5106 t. Among alternative management applications, it is possible to reuse these wastes in the cultivation of mushrooms, as a casing material for growing Agaricus spp. and as substrate for growing other species. In this work, the application of commercial nutritional supplements (Calprozime, Champfood and Promycel), widely used in Agaricus cultivation, is evaluated for its possible use as additive to substrates, based on spent oyster mushroom substrate (SMS), for the cultivation of Pleurotus ostreatus. Using a mixture of straw and SMS (1:1, w/w) as base material, the addition of CaSO4 (50 g kg-1) and CaCO3 (10 g kg-1) and the above supplements at 20 g kg-1 brought about a remarkable increase in production compared with the substrate without any supplement. The biological efficiencies did not differ significantly from that obtained when a commercial substrate was used as control, reaching values of 48.9 kg/100 kg substrate (dry matter) when Calprozime was used as supplement. Sporophores harvested from the supplemented substrates presented a higher dry matter content than those obtained from both commercial and non-supplemented substrates. SMS is cheap and easily available; it can be integrated into new formulations with the added advantages of lowering production costs, limiting growers' dependence on straw, and decreasing the environmental impact of its ever-growing accumulation.

In the process of riverine forest restoration, increasing emphasis has been given to the study of herbaceous and shrub species. However, for many of these we still lack basic knowledge, such as reproduction biology. Therefore, the objective of this study was to characterize the production, seed germination and reproductive capacity of Tibouchina clavata (Pers.) Wurdack., a species of wide distribution in wetlands. Fruits were collected from different mother trees located on the southern coast of São Paulo state and, after processing, were subjected to germination under constant temperatures of 25 and 30°C, and alternated temperatures of 25-30°C in the first essay, and at 15, 20, 25, 30, 35, 40°C in another essay, and at 25 °C with presence or absence of light. 10 plants were marked and, in each one, the number of fruits per plant and seed number per fruit were determined to characterize their reproductive capacity. The species germinates over a wide temperature range (15 to 35 °C), and the optimum temperature was in the range of 20 to 30 °C. Seed didn't germinate at 40 °C and could be characterized as being of absolute positive photoblastism. In general, there were over 1600 seeds per fruit and 85.000 seeds per plant. The high seed yield, coupled with good germination percentage demonstrated that this is a species with high reproductive potential.

The shelf life of the organic passion fruit pulp, both fresh and pasteurized at 70C and 90C and stored under refrigeration, was evaluated. The heat treatment did not affect the sensory and physicochemical characteristics of the pasteurized pulps when compared with the fresh pulp, except for the ascorbic acid content. The pulps were also microbiologically safe. The pulps pasteurized at 70 and 90C were suitable for consumption for a minimum shelf-life period of 207 days of storage under refrigeration and for the fresh pulp it was attributed a shelf-life period of 60 to 90 days. The pulp pasteurized at 70C showed higher acceptance scores for all the attributes and purchase intention scores, suggesting a more stable behavior and higher sensory quality. Practical Applications: This work intended to evaluate the influence of the minimum pasteurization on the sensory acceptance and microbiological and physicochemical characteristics of the organic passion fruit pulp stored under refrigeration, with the aim to identify the shelf life. Heat treatment is one of the processes used for food preservation. Lower pasteurization temperature than that used by the Brazilian industries and storage under refrigeration showed to be appropriate for passion fruit pulp quality. In this way, the microbiological, physicochemical and sensory features of passion fruit were preserved. This work can be used as a reference for passion fruit pasteurization, which is able to increase the shelf life of this fruit while preserving its desirable original features. A better understanding of the differences between the levels of nutrients, depending on the type of pruning used in the cultivation of the guava tree, may allow a more adequate understanding of the physiological processes of this fruit. The analysis of flowers is a tool that can be used to assist in assessing the nutritional status of crops, especially perennials. We evaluated the effects of different types of pruning on nutrient concentrations in flowers and fruit, at different developmental stages and in different parts of the fruit. The study was carried out in Vista Alegre do Alto, in orchards of guava variety Paluma. Flowers and fruit were collected in orchards, one under heavy pruning and the other with continuous pruning. The fruit were collected in two stages (two millimeters length and mature) and divided into basal part and apex, with the top toward the stalk. Flowers were collected in the same orchards as the fruits, sampling the basal part and apex of the flowers. F tests were performed and, when necessary, the Scott-Knott test at α= 5%. Overall, there were nutritional differences among flowers and fruits in relation to the type of pruning employed; drastic pruning provided higher levels of nutrients compared with continuous pruning. In relation to the portion of the samples, especially for fruit, there were differences between the apex and base, as well as between different stages of fruit collection.

Plant Growth Promoting Rhizobacteria (PGPR) has been used as a biofertilizer, bringing benefits to agriculture as Phosphorus Solubilizing Bacteria (PSB), indole-acetic acid (IAA) producers, and with other activites. The goal of this report was the identification of PGPR from soils under sugarcane crops by 16S rRNA sequencing, and the evaluation of the ability of phosphorus solubilizing and IAA production by biological assays. The isolates of this work were obtained from three areas of sugarcane crop from São Paulo State, Brazil. All isolates came from rhizosphere soil, and in a total of 60 isolates just 10 have showed high ability in phosphorus solubilizing. The selection of PSB may be done by phenotypic and/or genotypic characterization. Among ten isolates Enterobacter sp. (FJ890899), Entrobacter homaechei subsp. verschuerennii (FJ890998), Burkholderia sp. (FJ890895), and Labrys portucalensis (FJ890891) were able to IAA production. © 2006-2012 Asian Research Publishing Network (ARPN).

The purpose of this in vitro study was to quantify the alterations on human root dentin permeability after exposure to different acid fruit juices and to evaluate the effect of toothbrushing with electric or sonic toothbrush after acid exposure. The root dentin of 50 extracted third molars was exposed with a high speed bur. Crowns were sectioned above the cementoenamel junction and root fragments were used to prepare dentin specimens. Specimens were randomly assigned to 5 groups according to the fruit juice (kiwifruit, starfruit, green apple, pineapple and acerolla). Each specimen was connected to a hydraulic pressure apparatus to measure root dentin permeability using fluid filtration method after the following sequential steps: I) conditioning with 37% phosphoric acid for 30 s, II) root scaling, III) exposure to acid fruit juices for 5 min and IV) electric or sonic toothbrushing without dentifrice for 3 min. Data were analyzed statistically by the Wilcoxon and Mann-Whitney tests at 5% significance level. All fruit juices promoted a significant increase of dentin permeability while toothbrushing decreased it significantly (p<0.05). It may be concluded that all acid fruit juices increased root dentin permeability, while toothbrushing without dentifrice after acid exposure decreased the permeability. The toothbrush mechanism (electric or sonic) had no influence on the decrease of root dentin permeability.

There is much information on the adequate soil water matrix potential for returning irrigation in conventional soil tillage systems; however there is not enough information on the best soil water matrix potential for no-tillage production systems. This work aimed to study the effect of five irrigation levels on yield, soluble solids, and fruit acidity of the Hypeel tomato, cultivated for industrial processing and cropped on no-tillage system in Yellow Latosol. The experiment was carried out on a randomized block design with five treatments and four replications. The treatments consisted in different soil water matrix potentials for returning irrigation (m at 13cm of soil depth): -15, -30, -45, -60 and -75 kPa. The results obtained showed that the soil water matrix potential of -28.5 kPa resulted in maximum yields for tomato crop; whereas the value of -50.8 kPa resulted in the maximum fruit soluble solids concentration.

Predicting and mapping productivity areas allows crop producers to improve their planning of agricultural activities. The primary aims of this work were the identification and mapping of specific management areas allowing coffee bean quality to be predicted from soil attributes and their relationships to relief. The study area was located in the Southeast of the Minas Gerais state, Brazil. A grid containing a total of 145 uniformly spaced nodes 50 m apart was established over an area of 31. 7 ha from which samples were collected at depths of 0. 00-0. 20 m in order to determine physical and chemical attributes of the soil. These data were analysed in conjunction with plant attributes including production, proportion of beans retained by different sieves and drink quality. The results of principal component analysis (PCA) in combination with geostatistical data showed the attributes clay content and available iron to be the best choices for identifying four crop production environments. Environment A, which exhibited high clay and available iron contents, and low pH and base saturation, was that providing the highest yield (30. 4l ha-1) and best coffee beverage quality (61 sacks ha-1). Based on the results, we believe that multivariate analysis, geostatistics and the soil-relief relationships contained in the digital elevation model (DEM) can be effectively used in combination for the hybrid mapping of areas of varying suitability for coffee production. © 2012 Springer Science+Business Media New York.

The physiological state of yeast cells changes during culture growth as a consequence of environmental changes (nutrient limitations, pH and metabolic products). Cultures that grow exponentially are heterogeneous cell populations made up of cells regulated by different metabolic and/or genetic control systems. The strain of baker's yeast selected by plating commercial compressed yeast was used for the production of glycerol-3- phosphate dehydrogenase. Glycerol-3-phosphate dehydrogenase (GPD) has been widely used in the enzyme assays with diverse compounds of industrial interest, such as glycerol or glycerol phosphate, as well as a number of important bioanalytical applications. Each cell state determines the level of key enzymes (genetic control), fluxes through metabolic pathways (metabolic control), cell morphology and size. The present study was carried out to determine the effects of environmental conditions and carbon source on GPD production from baker's yeast. Glucose, glycerol, galactose and ethanol were used as carbon sources. Glycerol and ethanol assimilations required agitation, which was dependent on the medium volume in the fermentation flask for the greatest accumulation of intracellular GPD. Enzyme synthesis was also affected by the initial pH of the medium and inoculum size. The fermentation time required for a high level of enzyme formation decreased with the inoculum size. The greatest amount of enzyme (0.45 U/ml) was obtained with an initial pH of 4.5 in the medium containing ethanol or glycerol. The final pH was maintained in YP-ethanol, but in the YP-glycerol the final pH increased to 6.9 during growth.

Chiroptera, the second largest mammalian order, presents different reproductive strategies and unique reproductive features. However, there are few reports regarding male reproductive accessory glands (RAGs) in Chiroptera. Thus, the aim of the present study was to characterise the RAGs of the exclusively neotropical bat Artibeus planirostris (Chiroptera: Phyllostomidae) macroscopically, microscopically and ultrastructurally. The RAGs were composed of a prostatic complex with two regions (ventral and dorsal) and paraurethral and bulbourethral glands, but no seminal vesicles. The ventral region had an undefined epithelium, with secretory and basal cells, and its secretions were periodic acid-Schiff (PAS) positive. The dorsal region received both deferens ducts, had a columnar pseudostratified epithelium with secretory and basal cells. There were two types of secretions from the dorsal region: one that was basophilic and another that was mixed PAS positive and PAS negative. The paraurethral glands were dispersed in the connective tissue of the urethra, whereas the bulbourethral glands were located in the penile root. Histological and ultrastructural data confirmed the prostatic nature of the ventral and dorsal regions and the holocrine nature of the ventral region, with the latter finding never having been described previously for the prostate gland. Our findings demonstrate the wide discrepancy of RAGs between A. planirostris and other mammals in terms of their composition, structure and morphology. © CSIRO 2013.

Artibeus planirostris is an endemic species of Phyllostomid bat from the Neotropical region. Some studies have indicated that it exhibits seasonal bimodal polyestry; however, others postulate that it may be able to produce young at any time during the year. Thus, the aim of this study was to evaluate the annual variations in testicular and epididymal parameters of this species in southeast Brazil and try to understand how the reproduction of this species is regulated in this environment. Sixty mature male specimens, collected between June 2009 and May 2010, were submitted to morphometric and immunohistochemical analysis. Our study showed that A. planirostris presented a continuously active pattern of spermatogenesis throughout the year, presenting spermatozoa inside its cauda epididymis in all months, but with two pronounced peaks of spermatogenic production, one in September and other in February. We propose that the males developed these two peaks in order to produce sufficient sperm for the reproduction in a harem system and to synchronize with the female reproductive cycle, which had a bimodal polyestric pattern. Control of this variation is directly linked to the expression of the androgen receptor (AR) in Sertoli cells and to serum testosterone levels, which appear to synchronize to establish these two peaks. In the months preceding the two peaks, the testis have a higher expression of the AR, which possibly stimulates the increase in PCNA, and drives a gradual increase in the testicular parameters. Taken together the results suggest that if sperm storage happens in this species, it is of short duration. © 2013 Elsevier Inc.
