Crotalus durissus terrificus venom and its main component, crotoxin (CTX), have the ability to down-modulate the immune system. Certain mechanisms mediated by cells and soluble factors of the immune system are responsible for the elimination of pathogenic molecules to ensure the specific protection against subsequent antigen contact. Accordingly, we evaluated the immunomodulatory effects of CTX on the immune response of mice that had been previously primed by immunisation with human serum albumin (HSA). CTX inoculation after HSA immunisation, along with complete Freund`s adjuvant (CFA) or Aluminium hydroxide (Alum) immunisation, was able to suppress anti-HSA IgG1 and IgG2a antibody production. We showed that the inhibitory effects of this toxin are not mediated by necrosis or apoptosis of any lymphoid cell population. Lower proliferation of T lymphocytes from mice immunised with HSA/CFA or HSA/Alum that received the toxin was observed in comparison to the mice that were only immunised. In conclusion, CTX is able to exert potent inhibitory effects on humoural and cellular responses induced by HSA immunisation, even when injected after an innate immune response has been initiated. Angiotensin II (Ang II) exerts an acute bimodal effect on proximal tubule NHE3: while low doses stimulate the exchanger, high doses inhibit it. In the present study, we have investigated the chronic effects of Ang II on NHE3 expression and transcriptional regulation. Treatment of a tubular epithelial cell line, OKP, with Ang II 10(-11) M significantly increased NHE protein expression and mRNA levels, without evidence of bimodal effect. No change in mRNA half-life was detected, but transient transfection studies showed a significant increase in NHE3 promoter activity. Binding sites for Sp1/Egr-1 and AP2 transcription factors of the NHE3 proximal promoter were mutated and we observed that the Sp1/Egr-1 binding site integrity is necessary for Ang II stimulatory effects. Inhibition of cytochrome P450, PI3K, PKA and MAPK pathways prevented the Ang II stimulatory effect on the NHE3 promoter activity. Taking all the results together, our data reveal that chronic Ang II treatment exerts a stimulatory effect on NHE3 expression and promoter activity. The Ang II up-regulation of the NHE3 promoter activity appears to involve the Sp1/Egr-1 binding site and the interplay of several intracellular signaling pathways. (C) 2011 Elsevier Inc. Cross sections for the (6)Li(p,gamma)(7)Be, (7)Li(n,gamma)(8)Li (8)Li(n,gamma)(9)Li and (8)Li(p,gamma)(9)Be capture reactions have been investigated in the framework of the potential model. The main ingredients of the potential model are the potentials used to generate the continuum and bound-state wave functions and spectroscopic factors of the corresponding bound systems. The spectroscopic factors for the (7)Li circle times n=(8)Li(gs), (8)Li circle times n=(9)Li(gs) bound systems were obtained from a FR-DWBA analysis of neutron transfer reactions induced by (8)Li radioactive beam on a (9)Be target, while spetroscopic factor for the (8)Li circle times n=(9)Be(gs) bound system were obained from a proton transfer reaction. From the obtained capture reaction cross section, reaction rate for the (8)Li(n,gamma)(9)Li and (8)Li(p,gamma)(9)Be direct neutron and proton capture were determined and compared with other experimental and calculated values.

The crystal structures of an aspartic proteinase from Trichoderma reesei (TrAsP) and of its complex with a competitive inhibitor, pepstatin A, were solved and refined to crystallographic R-factors of 17.9% (R(free)=21.2%) at 1.70 angstrom resolution and 15.81% (R(free) = 19.2%) at 1.85 angstrom resolution, respectively. The three-dimensional structure of TrAsP is similar to structures of other members of the pepsin-like family of aspartic proteinases. Each molecule is folded in a predominantly beta-sheet bilobal structure with the N-terminal and C-terminal domains of about the same size. Structural comparison of the native structure and the TrAsP-pepstatin complex reveals that the enzyme undergoes an induced-fit, rigid-body movement upon inhibitor binding, with the N-terminal and C-terminal lobes tightly enclosing the inhibitor. Upon recognition and binding of pepstatin A, amino acid residues of the enzyme active site form a number of short hydrogen bonds to the inhibitor that may play an important role in the mechanism of catalysis and inhibition. The structures of TrAsP were used as a template for performing statistical coupling analysis of the aspartic protease family. This approach permitted, for the first time, the identification of a network of structurally linked residues putatively mediating conformational changes relevant to the function of this family of enzymes. Statistical coupling analysis reveals coevolved continuous clusters of amino acid residues that extend from the active site into the hydrophobic cores of each of the two domains and include amino acid residues from the flap regions, highlighting the importance of these parts of the protein for its enzymatic activity. The synthesis, an improved refined crystal and molecular structure re-determination, and the thermal decomposition behavior of two Zn(II) derivatives of isocinchomeronic acid (2,5-pyridinedicarboxylic acid or H(2)2,5-pydc) are presented. [Zn(2,5-pydc)(H(2)O)(3)Zn(2,5-pydc)(H(2)O)(2)](2) (1) crystallizes in the triclinic P-1 space group with a = 7.106(2), b = 11.450(2), c = 11.869(1) angstrom, alpha = 107.29(1), beta = 104.08(1), gamma = 90.32(2)degrees, and Z = 2. [Zn(2,5-pydc)(H(2)O)(2)] center dot H(2)O (2) is orthorhombic (P2(1)2(1)2(1) space group), with a = 7.342(1), b = 9.430(1), c = 13.834(2) angstrom, and Z = 4. The structures were refined to agreement R(1)-factors of 0.0315 (1) and 0.0336 (2). Complex (1) is arranged as molecular Zn(4)(2,5-pydc)(4)(H(2)O)(10) tetramers, the cages of which define channels that remain unblocked by anions. Compound (2) is polymeric with Zn(2,5-pydc)(H(2)O)(2) and Zn(2,5-pydc)(H(2)O)(3) units linked through bridging ligands. Both compounds were synthesized under mild conditions in aqueous media, without need to resort to hydrothermal media. Changing the pH from 4.51 to 5.75 suffices to direct the chemical processes toward the orthorhombic compound rather than to the triclinic one.

Successful coupling of electrochemical preconcentration (EPC) to capillary electrophoresis (CE) with contactless conductivity detection (C(4)D) is reported for the first time. The EPC-CE interface comprises a dual glassy carbon electrode (GCE) block, a spacer and an upper block with flow inlet and outlet, pseudo-reference electrode and a fitting for the CE silica column, consisting of an orifice perpendicular to the surface of a glassy carbon electrode with a bushing inside to ensure a tight press fit. The end of the capillary in contact with the GCE is slant polished, thus defining a reproducible distance from the electrode surface to the column bore. First results with EPC-CE-C(4)D are very promising, as revealed by enrichment factors of two orders of magnitude for Tl, Cu, Pb and Cd ion peak area signals. Detection limits for 10 min deposition time fall around 20 nmol L(-1) with linear calibration curves over a wide range. Besides preconcentration, easy matrix exchange between accumulation and stripping/injection favors procedures like sample cleanup and optimization of pH, ionic strength and complexing power. This was demonstrated for highly saline samples by using a low conductivity buffer for stripping/injection to improve separation and promote field-enhanced sample stacking during electromigration along the capillary. Diffusion coefficients and retardation factors of two metal cations (Cd2+ and Pb2+) were measured for a compacted Brazilian saprolitic soil derived from gneiss, aiming to assess its geoenvironmental performance as a liner for waste disposal sites. This soil occurs extensively all over the country in very thick layers, but has not been used in liners because of its hydraulic conductivity, higher than 10(-9) m/s when compacted at optimum water content of standard Proctor energy, but which can be reduced by means of appropriate compaction techniques or additives. Batch, column, and diffusion tests were carried out with monospecies synthetic solutions at pH 1, 3, and 5.5. Measured diffusion coefficients varied between 0.5 and 4 X 10(-10) m(2)/s. Retardation factors show that cadmium, a very mobile cation, is not adsorbed at pH I but is significantly retained at pH 3 and pH 5.5, whereas lead is retained at all tested pH values though slightly at pH 1. Estimated retardation factors from batch tests were 1.3-2.3 times those resulting from column tests and at its highest when obtained by diffusion tests; whereas batch tests allow a more complete exposure of the soil grains to the solution, time-dependent nonspecific adsorption may take longer to occur. The importance of contact time was observed and should be considered in further investigations. Its significant retention of metals suggests a promising utilization of this soil as a bottom liner for wastes landfills.

In the past few years, libraries have started to design public programs that educate patrons about different tools and techniques to protect personal privacy. But do end user solutions provide adequate safeguards against surveillance by corporate and government actors? What does a comprehensive plan for privacy entail in order that libraries live up to their privacy values? In this paper, the authors discuss the complexity of surveillance architecture that the library institution might confront when seeking to defend the privacy rights of patrons. This architecture consists of three main parts: physical or material aspects, logical characteristics, and social factors of information and communication flows in the library setting. For each category, the authors will present short case studies that are culled from practitioner experience, research, and public discourse. The case studies probe the challenges faced by the library—not only when making hardware and software choices, but also choices related to staffing and program design. The paper shows that privacy choices intersect not only with free speech and chilling effects, but also with questions that concern intellectual property, organizational development, civic engagement, technological innovation, public infrastructure, and more. The paper ends with discussion of what libraries will require in order to sustain and improve efforts to serve as stewards of privacy in the 21st century.

Esta dissertação apresenta um estudo que analisa o grau de satisfação de clientes, pacientes e acompanhantes, acerca da assistência nutricional hospitalar da ISCMPA, que utiliza o índice de satisfação de clientes como indicador de desempenho, demostrando a preocupação em elevar a qualidade do serviço a níveis de excelência. Foi aplicado um questionário, realizando entrevista pessoal com 215 clientes, resultando num estudo descritivo, de corte transversal, de perfil qualitativo e quantitativo. A análise de resultados foi apresentada em três blocos: características dos envolvidos na investigação, nível de satisfação e grau de importância por constructos. Aspectos como percepção dos clientes da imagem da Instituição, posição frente à comunidade, fatores que os levariam a trocar de Hospital e os sentimentos expressos pelos respondentes na ocasião da pesquisa, também foram abordados. O índice de 95 % de satisfação dos clientes, meta da Instituição, foi atingido em quatro dos seis contructos avaliados: Comportamento e Atuação de Funcionários, Comunicação com a Equipe por Telefone, Contato Inicial com a Equipe de Nutrição e Higiene de Material da Copa. Os constructos que não alcançaram a meta foram Refeições e Adequação de Material. O conhecimento do grau de satisfação dos clientes permitiu estabelecer um plano de intervenção, definindo ações corretivas e de melhorias para constructos que não alcançaram a meta e para pontos críticos identificados.

This work has as main objective the development of a key factors¿ model for the quality of Home Broker systems. An explanatory research was performed, based on a quantitative approach. To achieve this goal, some theoretical models of technology acceptance (TAM, TRA, TPB and IDT), reliability and quality of service were reviewed. It was proposed an extended key factors¿ model and developed a questionnaire, which was the research instrument used in this study. The questionnaire was applied over the Internet, from which was obtained a participation of 113 valid respondents, all of them users of Home Broker system. Once performed the data collection, statistical tests were used for the Factorial Analysis in order to achieve a definitive model. The key factors found were Perceived Usefulness, Perceived Ease of Use, Subjective Norms, Compatibility, Reliability and Relative Advantage. Some hypotheses from the model were also tested to investigate the relationship between the importance given to the factors and the resulting degree of satisfaction about quality of service. As a result of the study, a key factors¿ model for the quality of Home Broker systems was established, and identified that the factor Compatibility" has more explanatory power than the others."

O tema do presente estudo consiste na análise da demanda do mercado de taxistas de Curitiba frente à possibilidade de utilização de um combustível alternativo menos oneroso e poluente: o Gás Natural Veicular – GNV. O objetivo principal deste trabalho foi de analisar o mercado atual do GNV entre os taxistas de Curitiba, verificando a possibilidade de ampliar do uso do GNV por esta frota através de uma pesquisa de mercado. Os dados da pesquisa servirão de subsídios para campanhas de marketing orientadas à ampliação acelerada do uso desta tecnologia. Para tanto, realizou-se uma pesquisa de satisfação para levantar os fatores determinantes para a tomada de decisão dos taxistas de Curitiba de utilizarem ou não um combustível alternativo menos oneroso e poluente como o Gás Natural Veicular – GNV. O estudo visou obter a percepção dos taxistas de Curitiba. Com base nos resultados da pesquisa foi possível estabelecer subsídios para campanhas de marketing orientadas à ampliação acelerada do uso desta tecnologia. Para realização da pesquisa, utilizou-se uma pesquisa exploratória, com entrevistas individuais da cúpula das organizações que trabalham com o Gás Natural, prospectando novos projetos para o futuro e as perspectivas de evolução do mercado de GNV em Curitiba, a qual forneceu subsídios para a pesquisa quantitativa junto aos taxistas da cidade de Curitiba. Constatou-se que somente 36% do público estudado é usuário de gás natural veicular, mas entre os não usuários, 82% já pensou em instalar o kit gás. As principais razões pelas quais os taxistas pesquisados não utilizam o GNV são a perda de espaço no porta-malas e a perda de potência do veículo. Entre os taxistas que utilizam o GNV, no que tange aos atributos relacionados à rede de distribuição, a maior insatisfação refere-se à localização dos postos e a maior satisfação às eventuais filas para abastecimento. Nesses mesmos atributos o que obteve maior índice de importância foi justamente a localização dos postos. Entre os atributos relacionados ao produto, o maior índice de satisfação foi o nível de poluição ambiental e a maior insatisfação espaço para armazenamento, sendo o desempenho do veículo considerado o mais importante. Quanto aos atributos relacionados aos equipamentos, verificou-se a segurança com o maior índice de satisfação e de importância.

o estudo pretende contribuir para o entendimento do problema da evasao de funcionários do Banco Central do Brasil. Partindo da idéia de que a permanência de funcionário na organização alicerça-se no tripé: satisfação no trabalho, satisfação com o sistema de recompensas e influência do ambiente externo, procurou-se determinar os fatores desses eI lementos capazes de influenciar a decisâo de permanência ou evasão. Utilizaram-se dois tipos de análise:~ descritiva e de contingência. A primeira visou a apresentar ~s caracteristicas da amostra e os fatores do ambiente externo associados ã permanência ou evasão do Banco. A segunda visou a determinar os fatores que melhor explicam satisfação no trabalho e satisfaçao com osiétema de recompensas. Uma das conclusões do estudo: há um grande anseio por parte do funcionalismo na efetiva pa~ ticipação na vida organizacional, o que significa não so ouvir e ser ouvido na definição de metas e diretrizes organizacionais, como também poder planejar e implementar a carreira. Espera-se que as análises realizadas possam servir para repe~ sar os anseios, dGvidas, frustrações e realizações do funcionário no contexto da organização em que, no sentido amplo do termo, ele vive.

O objetivo deste trabalho é verificar se há ou não congruência entre a ideia de lealdade, manifestada no discurso dos bancos, e o seu conceito em marketing de relacionamento, identificando qual o sentido desse constructo nas mensagens das instituições bancárias, além de identificar, também, quais os fatores que levam os clientes a manterem relacionamento duradouro com bancos. O estudo é de caráter exploratório, e foi conduzido com entrevistas individuais com clientes pessoa física e gerentes de bancos públicos e privados de Brasília/DF, com o intuito de explorar o relacionamento entre cliente-banco, para uma melhor orientação na direção das análises dos dados obtidos. Os dados foram coletados junto a 11 entrevistados de ambos os sexos, residentes em Brasília/DF, durante os meses de março e abril de 2011. Para atingir os objetivos propostos, o método de pesquisa adotado foi qualitativo, com foco no valor informacional da mensagem propriamente dita, das palavras, argumentos e idéias nela expressos, utilizando-se uma forma interpretativa para a análise dos dados. Os resultados demonstraram o afastamento conceitual entre a ideia de lealdade dos bancos, definida como algo ligado ao entendimento de que o cliente, confiante em seu banco, está satisfeito e não o deixa, e o seu conceito em marketing de relacionamento, que a define como um profundo compromisso do cliente em recomprar um produto/serviço consistentemente no futuro, pois constatou-se que os clientes bancários, independentemente de fatores como o tempo gasto ou o esforço associados à troca de fornecedores, são sensíveis a aumento substancial da tarifação, não tem compromisso de recompra com banco e nem de compra junto a um único banco. Em decorrência, se pode concluir que não são fiéis/leais. Como fatores responsáveis por relacionamento duradouro com bancos, verificou-se, como principais, a qualidade do atendimento prestado pelo banco e a reciprocidade existente no relacionamento, ambos são constituintes dos sentimentos de satisfação e confiança nos clientes bancários. Conclui-se o trabalho, fazendo-se recomendações com a intenção de beneficiar e desenvolver os gestores deste segmento.
